Hurricane Katrina devastated areas of New Orleans and caused the evacuation of most of the city’s residents. Many people were exposed to dangerous storms and flooding and lost many of their possessions. One of the most common psychological disorders following a disaster is Posttraumatic Stress Disorder. This study describes the PTSD symptom endorsement of a sample of women who experienced Hurricane Katrina. In addition, many of these women had previous trauma histories which are also described. The incidence of foodborne outbreaks linked to fresh produce has increased in the United States. Particularly noteworthy was the 2006 Escherichia coli O157:H7 outbreak associated with pre-packaged baby spinach. Factors affecting the contamination of spinach leaves with E. coli O157:H7 are not yet well understood. This study aimed to determine whether E. coli O157:H7 would be present in the aerial leaf tissue of a growing spinach plant when introduced at various plant maturities and different inoculum levels in the growth media in a greenhouse setting. In nature, self-assembly processes of biologically active organic molecules often occur, resulting in formation of dimers and higher oligomers of various and sometimes complex structures. This natural occurrence of self-organization has been the subject of research, with the aim of understanding and possibly modulating the aggregation behavior of biological molecules. The meso-tetrakis(4-phosphonatophenyl) porphyrin, H2TPPP was synthesized, purified, and characterized. Its self-assembly was studied in aqueous solutions as a function of pH and time. This project assessed short-term temperature effects on total ammonia nitrogen (TAN) utilization rates in a batch laboratory-scale recirculating system. The tank system was designed for experiments on short term steady state and diurnal temperatures. A set of numerical models was developed to simulate observed results. The performance of the biofilters was determined with three tank replicates at fixed temperatures of 13, 20 and 300C; and at diurnal transient (sinusoidal) temperature regimes of (20 ± 30C; 30 ± 30C). Sediments from the Gulf of Papua were examined to estimate particle flux and sediment mass accumulation rates on multiple timescales. Patterns of sediment mass accumulation and inventory-derived 210Pb fluxes display regional variations, decreasing seaward, and along isobaths seaward from the northeastern shelf edge. The amount of terrigenous sediment load being discharged annually from the shelf and accumulating in Pandora Trough is approximately 7-14×106 tonnes. The existence of possible turbidity-current transport and deposition have been documented in deeper parts of the Gulf of Papua. High excess 210Pb fluxes estimated from seabed inventories at the shelf edge and upper slope are consistent …

Understanding the thermodynamics of substrate selection by DNA Polymerase I is important for characterizing the balance between replication and repair for this enzyme in vivo. Due to their sequence and structural similarities, Klenow and Klentaq, the “large fragments” of the Pol I DNA polymerases from Escherichia coli and Thermus aquaticus, are considered functional homologues. Klentaq, however, does not have a functional proofreading site. Examination of the DNA binding thermodynamics of Klenow and Klentaq to different DNA structures: single-stranded DNA (ss-DNA), primer-template DNA (pt-DNA), and blunt-end double-stranded DNA (ds-DNA) show that the binding selectivity pattern is similar when examined …

Natural self-assembly processes provide nanofabrication capabilities for designing surfaces with nanoscale control of surface chemistry and relative orientation of the nanomaterials on the surfaces. Particle lithography was used to produce periodic arrays of protein nanostructures. Monodisperse mesoparticles can be applied to rapidly prepare millions of uniform protein nanostructures on flat surfaces using the conventional benchtop chemistry steps of mixing, centrifuging, evaporation and drying. The purpose of developing this microsystem is to identify acute infectious disease and prevent contagious outbreak in a matter of time. Rickettsia felis is a gram-negative bacterium predominantly described in the cat flea, Ctenocephalides felis. Since first described in 1990 in a commercial cat flea colony in the United States, R. felis has been detected in numerous arthropod species in 28 countries around the world. Additionally, as the etiologic agent of flea-borne rickettsiosis, R. felis can cause disease in humans, with patients presenting with clinical symptoms typical of rickettsial diseases including: fever, headache, and myalgia. Phytoplankton and benthic microalgae (BMA) may both contribute to the sediment organic matter pool available to benthic consumers. Recent studies have highlighted the presence and ecological roles of benthic microalgae (BMA) on the continental shelf but studies from the north-central Gulf of Mexico are limited. Here we use photosynthetic pigment analysis and microscopic examination of sediment microalgae to investigate how the biomass, composition, and degradation state of sediment-associated microalgae varies along the Louisiana inner shelf across a continuum of water column and sediment conditions. Platinum has been used as an inorganic medicinal agent with various applications such as anti cancer agents. Several new targeted Pt(II) conjugates were synthesized having the NGR and LHRH targeting motif. The platinum conjugates bearing the NGR motif revealed selective delivery and destruction of cancer cells that had CD13 receptors compared to untargeted carboplatin drug. The upper Breton Sound estuary was hydrologically reconnected to the Mississippi River via the Caernarvon freshwater diversion structure in 1991. The Caernarvon structure can provide controlled freshwater pulses to the upper Breton Sound estuarine ecosystem, replicating historic freshwater pulsed events, although the original authorization was to control salinity isohalines at specific locations in the estuary. However, unlike historic freshwater pulsed events prior to the construction of levees, the current freshwater pulse contains an unprecedented amount of inorganic nitrogen, predominately as nitrate (annual average 71.4 µM NO3-). Transient periods of unsteady loading pose a challenge in design and operation of bioreactors for air pollution control. As a potential means of overcoming some of these challenges, a systematic study of granular activated carbon (GAC) sorption was conducted to further evaluate its potential as a load equalization mechanism for unsteady loading of volatile organic carbons (VOCs). The interactions of fine and ultrafine particles with chemicals play a dominant role in determining the mobility and availability of pollutants in the environment. Fine particles in sediments can sequester chemicals from the water column, and release volatile and semi-volatile organic compounds to the gas phase upon exposure to air. Iron-sulfur ([Fe-S]) clusters represent one of natures most diverse and ubiquitous protein prosthetic groups. [Fe-S] proteins are integral for diverse biological processes. Reconstitution of apo-proteins can occur by exogenously adding excess iron and sulfide. However, due to the toxicity of iron and sulfide, it is most likely that proteins coordinate [Fe-S] cluster assembly in cells. Important processes governing the fate and transport of light petroleum hydrocarbons in the Lower Mississippi River and other river delta systems are not adequately represented in existing oil spill models. Laminitis is a crippling and often life-threatening disease of the equine foot. Soft tissue damage characteristic of this disease has been associated with increased MMP activity. Therefore, it seems likely that MMPIs could be potential therapeutic agents for laminitis. Further characterization of equine MMPs and evaluation of the effectiveness of MMPIs in the horse are needed. Equine MMP-9 was harvested from neutrophils, purified by affinity chromatography, and evaluated using western blotting and gelatin zymography. For over a century, nucleation for all systems was thought simplistically to be a process that advances through the formation of critical clusters with a well-defined composition. Our results show intriguing nucleation mechanisms that challenge the aforementioned notion. We employed the simple TraPPE-UA (transferable potential for phase equilibria – united atom) force field and the AVUS-HR approach (a combination of aggregation-volume-bias Monte Carlo, umbrella sampling, and histogram reweighting), to investigate the homogeneous vapor-to-liquid nucleation of various nucleating systems. Ozone (O3), one of the most powerful oxidants known, is phytotoxic at high levels in the troposphere, or ground-level. Effects of acute ozone exposure for two consecutive days was examined on Bermudagrass (Cynodon dactylon), centipedegrass (Eremochloa ophiuroides), zoysiagrass (Zoysia japonica), St. Augustinegrass (Stenotaphrum secundatum), Liriope muscari ‘Big Blue’, Liriope muscari ‘Aztec’, and Ophiopogon japonicus. Zoysiagrass, St. Augustinegrass, Liriope muscari ‘Big Blue’ were used in the second study based on the differential responses found in the study. Genomic imprinting is a process that leads to the silencing of one allele of a gene in a parent of origin specific manner. Genes that are involved in this process are often regulated in clusters, one of which is the Peg3 (Paternally expressed gene 3) imprinted domain. We investigated this region for both CpG islands and long antisense transcripts, two common features of imprinted gene clusters. First, we performed a systematic survey of DNA methylation status of the CpG islands in this region of the mouse, cow, and human genomes. Supercritical fluid extraction (SFE) of oxeye daisy (Chrysanthemum leucanthemum L.) seed, heartwood of Port-Orford cedar (POC) (Chamaecyparis lawsoniana), Alaska yellow cedar (AYC) (Chamaecyparis nootkatensis), and Eastern red cedar (ERC) (Juniperus virginiana L), and Chinese tallow (Sapium sebiferum L. Roxb. ) seed was investigated in this study and compared with other extraction methods. For the oxeye daisy seed extraction, Soxhlet extraction (SE) with hexane, microwave assisted extraction (MAE), and supercritical fluid extraction (SFE) were conducted. A Bayesian-updating approach is developed to the estimation of the total uncertainty-based Margin of Safety (MOS) for Total Maximum Daily Load calculations using the watershed modeling tool HSPF. In order to determine the prior, likelihood, and posterior distributions of uncertainties involved in Bayesian approach, various probability density functions are derived. The temperature measurement induced uncertainty in dissolved oxygen simulation is found to be normally distributed. The temporal scale uncertainty in weather data decreases with decreasing temporal resolution. The temporal-scale uncertainties in the rainfall and temperature data follow a Weibull and general extreme value distributions, respectively. The spatial-resolution uncertainty in simulated …

Processing biomass in near- and supercritical water has garnered increasing attention because of its ability to accept a variety of wet feedstocks, energy efficiency, and ability to regulate the solubility and separation of components. Very few studies of biomass hydrothermal conversion provide a comprehensive evaluation of multiple process parameters and various additives and their effects on a single product phase. This research examined the influence of temperature, residence time, biomass concentration, and particle size on volatiles production from the hydrothermal conversion of sugarcane bagasse.
