Yeast Crf1p: An Activator In Need Is An Activator Indeed, Sanjay Kumar
Yeast Crf1p: An Activator In Need Is An Activator Indeed, Sanjay Kumar
LSU Doctoral Dissertations
Ribosome biogenesis is an energetically costly process, and tight regulation is required for stoichiometric balance between components. This requires coordination of RNA polymerases I, II, and III. Lack of nutrients or the presence of stress leads to downregulation of ribosome biogenesis, a process for which mechanistic target of rapamycin complex I (mTORC1) is key. mTORC1 activity is communicated by means of specific transcription factors, and in yeast, which is a primary model system in which transcriptional coordination has been delineated, transcription factors involved in regulation of ribosomal protein genes include Fhl1p and its cofactors, Ifh1p and Crf1p. Ifh1p is an …
Multichannel Low-Cost Sensor For Iaq Monitoring: Implementation And Guidelines, Isaac Ntiamoah
Multichannel Low-Cost Sensor For Iaq Monitoring: Implementation And Guidelines, Isaac Ntiamoah
LSU Master's Theses
Indoor air quality (IAQ) is adversely affected by both indoor and outdoor environments, and rental homes occupied by low-income families suffer the most. Poor indoor air can be detrimental to the health of occupants, especially for susceptible populations like those with respiratory diseases such as asthma. Although IAQ monitoring is necessary, it is a challenging, costly, and time-intensive task. Moreover, the protocol for assessing IAQ in residential settings varies, and additional research is needed to establish a standard operating procedure that incorporates new monitoring technologies, such as low-cost air sensors. However, there is a dearth of comprehensive, evidence-based protocols for …
Integrated Pest Management And Behavior Of Stored Grain Beetles, Ethan Doherty
Integrated Pest Management And Behavior Of Stored Grain Beetles, Ethan Doherty
LSU Doctoral Dissertations
The lesser grain borer, Rhyzopertha dominica, and the rice weevil, Sitophilus oryzae, are two of the most destructive pests of stored rice. Their management has relied heavily upon fumigant insecticides. However, amid growing concerns that fumigant-resistant pests are becoming widespread, there is an urgent need for diversified management techniques and integrated pest management. Here, we explored management techniques and the behavior of these beetles. Host plant resistance toward both beetles was evaluated for 13 varieties of rice. We found large differences among varieties for both beetles in all resistance measures. We found that while nutritional components of a …

Becoming Avian: Amazonian Featherworks From The John P. O’Neill Collection, Madeline R. Blanchard
LSU Master's Theses
In 1998 ornithologist John P. O’Neill donated an ethnographic collection of 434 objects he was gifted from researcher Charles Fugler or purchased from persons in Pucallpa, Peru, during his time there studying Amazonian birds. I evaluate 18 feathered objects. According to O’Neill, the cultures responsible for the items are the Cashinahua, Aguaruna, Achual, and Arawak. Eighteen of these items are beautifully crafted arrangements of feathered clothing and objects. The collection includes five headdresses, five bouquets, a hat, a necklace, three tassels, a backrack, a scarf, and a hair tie.
The objects and the seventeen species of bird used are active …
Computational Investigations Of Polymer Electrolytes In Aqueous Solutions, Hishara Keshani Gallage Dona
Computational Investigations Of Polymer Electrolytes In Aqueous Solutions, Hishara Keshani Gallage Dona
LSU Doctoral Dissertations
Polymer electrolyte membranes, an intriguing category of soft materials, are characterized by polymer backbones tethered with negatively or positively charged functional groups, finding applications from biological macromolecules to electrochemical separations in fuel cells. This dissertation focuses on computational investigations of polymer electrolytes in aqueous solutions using polypeptoids and ion exchange membranes. Polypeptoids, highly tailorable peptidomimetic polymers lacking backbone hydrogen bonding or chirality, were used as model systems. Micelles formed from these polypeptoid polymers exhibited electrostatic repulsion, which competes with charge-sodium interactions, adding complexity to predicting micelle shapes. This research refined the design strategy, enhancing micelle shape and structure through precise …

Structural And Functional Characterizations Of The Two Mini-Ferritins In Pseudomonas Aeruginosa Revealed The Coexistence Of A Classic Dps (Pa0962) And A Dps-Like (Pa4880) Protein, Rajapaksha Pathirannahalage Nimesha Hansani Rajapaksha
LSU Doctoral Dissertations
In the P. aeruginosa genome, gene PA0962 codes for a probable DNA-binding protein from starved cells (Pa Dps). Pa Dps is assembled from 12 monomers, each consisting of a four-helix bundle fold typical of Dps proteins. A subunit dimer of Pa Dps harbors two ferroxidase centers. Assembly of a dodecameric structure of Pa Dps is favored by the presence of divalent metal ions and at pH 6.0 in the absence or presence of divalent cations. Pa Dps is unable to form the large DNA-protein complexes typical of many Dps proteins. Instead, Pa Dps acts on plasmid DNA to form relaxed …

LSU Master's Theses
To solve the Climate Crisis, a global Energy Transition is required. The primary contemporary tool for tackling these two massive problems is modeling, both biophysical climate models and IAMS (Integrated Assessment Models) which combine socioeconomic and technological progress with Climate Change. However, the proliferation and complexity of modeling systems make it difficult for most people to contribute substantively to solutions. In particular, stakeholders, policymakers, nonspecialists and activists require sufficient understanding of modeling to impact outcomes.
In this thesis, after summarizing the Climate Crisis and potential Energy Transition solutions, I review current modeling approaches. Models can be considered in terms of …

Rangewide Migratory Connectivity And Migration Ecology In Swainson's Warblers (Limnothlypis Swainsonii) Derived From Multisensor Geolocators, Garrett Rhyne
LSU Master's Theses
Successful conservation of Neotropical migratory passerines requires a detailed understanding of their annual cycle ecology, including wintering and breeding distributions, migratory pathways and stopovers, and migratory phenology. It is also important to evaluate the extent of geographic isolation between separate breeding populations during the nonbreeding season, known as migratory connectivity. Swainson’s Warbler (Limnothlypis swainsonii) is a species of conservation concern that, due to their secretive nature and selection of dense forest habitats, is poorly studied and lacks migratory information. Our objective was to track Swainson’s Warblers across their breeding populations (Mississippi Valley, Coastal Plains, and Appalachian) to detail …

Continuous spin excitations are widely recognized as one of the hallmarks of novel spin states in quantum magnets, such as quantum spin liquids (QSLs). Here, we report the observation of such kind of excitations in K2Ni2(SO4)3, which consists of two sets of intersected spin-1 (Ni2+) trillium lattices. Our inelastic neutron scattering measurement on single crystals clearly shows a dominant excitation continuum, which exhibits a distinct temperature-dependent behavior from that of spin waves, and is rooted in strong quantum spin fluctuations. Further using the self-consistent-Gaussian-approximation method, we determine that the fourth- and fifth-nearest-neighbor exchange interactions are dominant. These two bonds together …

Adipose tissue is a major modulator of metabolic function by regulating energy storage and by acting as an endocrine organ through the secretion of adipokines. With the advantage of next-generation sequencing-based single-cell technologies, adipose tissue has been studied at single-cell resolution, thus providing unbiased insight into its molecular composition. Recent single-cell RNA sequencing studies in human and mouse models have dissected the transcriptional cellular heterogeneity of subcutaneous (SAT), visceral (VAT), and intramuscular (IMAT) white adipose tissue depots and revealed unique populations of adipose tissue progenitor cells, mature adipocytes, immune cell, vascular cells, and mesothelial cells that play direct roles on …

Background: Intramuscular fat (IMF) and intramuscular connective tissue (IMC) are often seen in human myopathies and are central to beef quality. The mechanisms regulating their accumulation remain poorly understood. Here, we explored the possibility of using beef cattle as a novel model for mechanistic studies of intramuscular adipogenesis and fibrogenesis. Methods: Skeletal muscle single-cell RNAseq was performed on three cattle breeds, including Wagyu (high IMF), Brahman (abundant IMC but scarce IMF), and Wagyu/Brahman cross. Sophisticated bioinformatics analyses, including clustering analysis, gene set enrichment analyses, gene regulatory network construction, RNA velocity, pseudotime analysis, and cell–cell communication analysis, were performed to elucidate …

Hemp has been cultivated for thousands of years. It was grown widely in the United States until the mid-1900s, when laws limited the growth of hemp and later banned it entirely. Since the 2018 Farm Bill removed legal barriers to growing the crop, Louisiana created a structure to regulate hemp farming in 2019.
Because little information existed about hemp in Louisiana, the LSU AgCenter created the Industrial Hemp Working Group to study the crop, create production practices and help growers interested in this ancient plant.
This issue of Louisiana Agriculture focuses on the work of these researchers and the growers …

The tarnished plant bug, Lygus lineolaris (Hemiptera: Miridae), has a wide host range of over 700 plant species, including 130 crops of economic importance. During early spring, managing the field edges with weeds and other wild hosts is important in preventing early-season infestations of L. lineolaris in cotton to prevent damage to the squares and other fruiting structures. Scouting fields for L. lineolaris is time- and labor-intensive, and end-user variability associated with field sampling can lead to inaccuracies. Insect traps that combine visual cues and pheromones are more accurate, sustainable, and economically feasible in contrast to traditional insect detection methods. …

Nonobese diabetic (NOD) mice are the most commonly used rodent model to study mechanisms relevant to the autoimmunity and immunology of type 1 diabetes. Although many different strains of mice have been used as controls for studies comparing nondiabetic lines to the NOD strain, we hypothesized that the parental strain that gave rise to the NOD line might be one of the best options. Therefore, we compared female ICR and NOD mice, which are matched at key major histocompatibility complex (MHC) loci, to understand their metabolic and immunologic similarities and differences. Several novel observations emerged: 1) NOD mice have greater …

Particulate matter (PM) containing environmentally persistent free radicals (EPFRs) results from the incomplete combustion of organic wastes which chemisorb to transition metals. This process generates a particle-pollutant complex that continuously redox cycles to produce reactive oxygen species. EPFRs are well characterized, but their cardiopulmonary effects remain unknown. This publication provides a detailed approach to evaluating these effects and demonstrates the impact that EPFRs have on the lungs and vasculature. Combustion-derived EPFRs were generated (EPFR lo: 2.1e−16 radical/g, EPFR hi: 5.5e−17 radical/g), characterized, and verified as representative of those found in urban areas. Dry particle aerosolization and whole-body inhalation were established …

Understanding the potential impacts of internal waves on phytoplankton communities in oligotrophic oceans remains an important research challenge. In this study, we elucidated the impact of internal waves on phytoplankton communities through a comprehensive 154-hr time-series of observations in the South China Sea (SCS). We identified distinctive variations in phytoplankton pigment biomass and composition across the upper, middle, and lower layers of the euphotic zone, which we attributed to the perturbations triggered by internal waves. Phytoplankton other than Prochlorococcus in the lower, nutrient-replete layer likely benefitted from allochthonous nutrients introduced by internal waves, but their growth rates were constrained by …

Maternal care is crucial for the survival and development of offspring. Oxytocin modulates maternal behavior by binding to oxytocin receptors (OXTRs) in various parts of the brain. Previously, we showed that OXTRs are expressed in the anteroventral periventricular nucleus (AVPV) of female, but not male mice. Because the AVPV is involved in the regulation of maternal behavior and oxytocin enhances its induction, this finding leads to the hypothesis that the female specific population of OXTR neurons in the AVPV regulates maternal behavior. To address this hypothesis, OXTR-Venus reporter mice were used to assess if expression levels of OXTR in the …

There is considerably greater variation in metabolic rates between men than between women, in terms of basal, activity and total (daily) energy expenditure (EE). One possible explanation is that EE is associated with male sexual characteristics (which are known to vary more than other traits) such as musculature and athletic capacity. Such traits might be predicted to be most prominent during periods of adolescence and young adulthood, when sexual behaviour develops and peaks. We tested this hypothesis on a large dataset by comparing the amount of male variation and female variation in total EE, activity EE and basal EE, at …

[HCo(CO)x(bisphosphine)](BF4), x = 1-3, is a highly active hydroformylation catalyst system, especially for internal branched alkenes. In situ infrared spectroscopy (IR), electron paramagnetic resonance (EPR), and nuclear magnetic resonance studies support the proposed catalyst formulation. IR studies reveal the formation of a dicationic Co(I) paramagnetic CO-bridged dimer, [Co2(μ-CO)2(CO)(bisphosphine)2]2+, at lower temperatures formed from the reaction of two catalyst complexes via the elimination of H2. DFT studies indicate a dimer structure with square-pyramidal and tetrahedral cobalt centers. This monomer-dimer equilibrium is analogous to that seen for HCo(CO)4, reacting to eliminate H2 and form Co2(CO)8. EPR studies on the catalyst …

Conductive hydrogels have gained a great deal of interest in the flexible electronics industry because of their remarkable inherent properties. However, a significant challenge remains for balancing hydrogel’s conductivity, self-healing, and strength properties. Herein, double network ionic hydrogels were fabricated by concurrently introducing borax into dicarboxylic cellulose nanofiber (DCNFs) and polyacrylamide (PAM) hydrogels. The incorporation of borax provided a superabsorbent feature to the PAM/DCNF hydrogels (without borax) with the equilibrium water absorption rate increased from 552 to 1800% after 42 h. The compressive strength of the prepared hydrogel was 935 kPa compared to 132 kPa for the PAM hydrogel, with …

Bioremediation of oil spills occurs when indigenous microbes metabolize petroleum hydrocarbons. Rapid proliferation of hydrocarbon-consuming bacteria near the oil-water interface is vital to optimizing the oil biodegradation rate. Adding nanoparticles (NPs) to microbial systems can enhance the bacterial growth rate. However, the role of NP source material and surface charge on their ability to impact bacterial growth and oil-water interface characteristics is poorly understood. Here, we investigate the role of NP composition and surface chemistry on the growth rate of hydrocarbon-consuming bacteria and its attachment to the oil-water interface. We use Alcanivorax borkumensis as a model organism because of its …
