Bio-degradable poly (l-lactic acid) (PLLA) scaffolds were prepared by using thermally induced phase separation (TIPS) method. A solution of PLLA-Dioxane was formed by dissolving PLLA in dehydrated 1,4-Dioxane at three wt/vol percentages, specifically 3, 7 and 10%. This PLLA-Dioxane solution was then frozen in borosilicate glass vials (5mL) at three cooling rates (1, 10 and 40 ˚C/min) in a commercially available controlled rate freezer (CRF). The frozen solution was freeze-dried to sublimate the Dioxane. The microstructural properties of the resulting PLLA scaffolds were determined utilizing Scanning Electron Microscopy (SEM) images and uni- axial compressive testing. Theonellamides A-F were isolated from Theonella swinhoei by Fusetani and co-workers. Despite considerable synthetic effort to produce theonellamide F by the Shioiri group in the early 1990’s, the total synthesis of a theonellamide has yet to be reported. We report herein our efforts toward some of the required amino acid residues and construction of the western ring of theonellamide C. We describe the synthesis of an uncoded amino acid, (2S,4R)-ɣ-hydroxy-α-amino adipic acid (Ahad), a building block for theonellamide C. The relationship between pollution and per capita income generally appears as an inverted U-shaped curve. This inverted U-shaped curve is known as the environmental Kuznets curve (EKC). The shape of the curve, however, is very sensitive to the data, location and pollutant considered in the analysis. Since the early 1990s, there has been an exponential growth in the number of empirical studies in this field, but many refute the inverted U-shaped nature of the curve for pollutants across different time periods and geographical regions. This study examines several different linkages between the nucleic acid binding thermodynamics and the functions of three different DNA polymerases. The focuses are correlation of the DNA binding thermodynamics and the functional behavior of Klenow and Klentaq polymerases (from Escherichia coli and Thermus aquaticus, respectively), identification of factors that influence the proofreading activity of Klenow, and examination of HIV reverse transcriptase (HIV-RT) binding to different primer/template nucleic acid constructs. Cancer research is centered on the discovery of new biomarkers that could unlock the obscurities behind the mechanisms that cause cancer or those associated with its spread (i.e., metastatic disease). Circulating tumor cells (CTCs) have emerged as attractive biomarkers for the management of many cancer-related diseases due primarily to the ease of securing them from a simple blood draw. However, their rarity (~1 CTC per mL of whole blood) makes enrichment analytically challenging. DNA protection during starvation (Dps) proteins are important for bacterial oxidative stress responses. This study aims to understand the roles and characteristics of the two Dps homologs in Deinococcus radiodurans: Dps-1 and Dps-2. Dps-2 contains a predicted signal peptide and in vivo localization of Dps-2 reveals that its location is non-cytoplasmic. β-galactosidase assays show that the Dps-2 promoter is upregulated in the presence of H2O2 and the results from the DNA protection assay show that Dps-2 is able to protect DNA efficiently against reactive oxygen species (ROS). Photosynthetic microalgae optimize the utilization of inorganic carbon by active uptake and concentration of inorganic carbon (Ci) around the carbon-fixing enzyme Rubisco. This process, mostly induced under limiting carbon conditions is called the Carbon Concentrating Mechanism or CCM. The photosynthetic green alga Chlamydomonas reinhardtii serves as an excellent model organism for the study of eukaryotic CCMs. However, unlike the prokaryotic cyanobacterial CCM, which has a well-characterized Ci uptake system, the Ci uptake and transport system of the C. reinhardtii CCM is not well understood. I apply our current understanding to further explore the relationships and evolution of the North American snake tribe Thamnophiini, as well as to address current topics in phylogenetic and taxonomic methodology. Chapter 1 of this Dissertation presents a brief introduction to the basic properties of porphyrins and their derivatives (hydroporphyrins), their abundance and functions in the nature, and their applications in photodynamic therapy. Chapter 2 reports the synthesis, characterization, conformational analysis and cellular studies of novel mono conjugated regioisomers of chlorin e6 derivatives that are promising photosensitizers for photodynamic therapy. All three regioisomers were synthesized from pheophytin a, which was extracted from the alga Spirulina pacifica. In vitro investigations using human carcinoma HEp2 cells show that the 152-lysyl regioisomers accumulate the most within cells, and the most phototoxic …

Late Embryogenesis Abundant (LEA) proteins are highly hydrophilic, intrinsically disordered proteins whose expression has been correlated with desiccation tolerance in anhydrobiotic organisms. Embryos of the brine shrimp, A. franciscana, contain high titers of group 3 LEA proteins during desiccation-tolerant stages such as diapause and pre-emergence development. Here I report the sequencing of three novel variants of AfrLEA3m mRNA (Afrlea3m_47, Afrlea3m_43 and Afrlea3m_29), whose deduced protein sequences are are predicted to localize to the mitochondrion. A review of recent publications in source identification revealed that there are very limited numbers of research in modeling methods for rivers. This study asks 1) What is the relationship between art, creativity, and social justice? 2) How do the theories of John Dewey, Maxine Greene, and Jane Piirto inform our understanding of this relationship? 3) What is the role of the arts in contemporary curriculum? To answer these questions, the study chronicled the various roles of art in Western society, from Classical Greece through the present day, before exploring the aesthetic theories of Dewey, Greene, and Piirto. Group of Uniform Materials Based on Organic Salts (GUMBOS) are a class of molten salts that have been focused to be specifically tailored towards applications. Primitively, from Davy to Walden molten salts were desirable for organic synthesis due to the properties. GUMBOS have been synthesized into nanoparticles (nanoGUMBOS) by various methods to be used for lanthanide luminescence and hyperthermal cancer therapeutics. Lanthanide photochemistry has been frequently studied for its high luminescence intensity, narrow emission band, and stable luminescent lifetime decay. Glycogen is the storage polysaccharide in a spectrum of organisms ranging from yeast to humans. Glycogen has a highly branched structure somewhat resembling a tree or shrub, with the branch points formed by glucose molecules being joined by α 1,6-glycosidic linkages. These linkages are formed by the action of branching enzyme. Branching of glycogen is critical to health, with a lack of branching causing glycogen storage disease type IV, which is fatal in humans by age 2. Although water coning is well understood, it is difficult to control in field operations resulting in low recovery and large volumes of waste produced water. A solution - proposed here - is a multi-functional well with the in-situ bottom water drainage and injection installations - Downhole Water Loop (DWL). Theoretically, DWL greatly improves well performance (for example, a two-fold increase of DWL well’s water drainage rate would increase the critical (water-free) oil rate by 80%). The investigation of polypeptide composite particles, PCPs, is described. The production of mesoporous and polycolloid architectures is also addressed. PCPs were prepared by both growing form and grafting to method. They consist of a core made of silica, silica-coated magnetite or silica-coated cobalt. Some of the particles were covalently labeled with fluorescein isothiocyanate. Several polypeptides were chosen for attachment on the core: poly(γ-stearyl-L-glutamate), PSLG, poly(Nε-carbobenzyloxy-L-lysine), PCBL and poly(o-benzyl-L-tyrosine), PBTY. Attachment of a well-characterized alkyne-end terminated PSLG and PCBL to an azide- functionalized particle yielded PCPs with a desired sparse coverage. It is becoming more important to detect ultra-low concentrations of analytes for biomedical, environmental, and national security applications. Equally important is that new methods should be easy to use, inexpensive, portable, and if possible allow detection using the naked eye. Detection of low concentrations of analytes generally cannot be achieved directly, but requires signal amplification by catalysts, macromolecules, metal surfaces or supramolecular aggregates. Porphyrins and metalloporphyrins have unique chemical and electronic properties and thus provide useful model structures for nanoscale studies of the role of chemical structure for electronic properties. Porphyrins have been proposed as viable materials for molecular-based information-storage devices, gas sensors, photovoltaic cells, organic light-emitting diodes and molecular wires. The function and efficiency of porphyrins in devices is largely attributable to molecular architecture and how the molecules are self-organized. Self-efficacy, gender, and motivation are subjective predictors of students' academic achievement. Self-assessment prompted by knowledge surveys involves metacognition, i.e., students' awareness of how they learn. Knowledge surveys can be used to measure changes in students' achievement level and to assist students in content review and inspire reflection on one's ability to learn. This dissertation addresses the little studied but socially salient processes through which Latino migrant laborers find work, travel, and obtain documentation using transnational social networks spanning between their places of origin and destinations in the United States. This project focuses on the creation and maintenance of these transnational linkages with a particular interest in their expansion into locations throughout American South, the region with the highest growth rates of Hispanic populations. Autism Spectrum Disorder (ASD) is currently defined using criteria from the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition, Text Revision (DSM-IV-TR). Alu elements are a primate-specific non-autonomous retrotransposon of ~300 base pairs (bp). A Short INterspersed Element (SINE), Alu elements are found in high copy number in genomes across the primate radiation. Alu elements have a history of effective use as a phylogenetic marker due to the presence-absence nature of their retrotransposition, and have been implicated in the cause of a number of genetic diseases. Previous research demonstrated environmentally persistent free radicals (EPFRs) will form on particulate surfaces under combustion conditions (temperature range of 150-400 °C) from reactions of organic precursors with redox-active transition metals. With an understanding of how these EPFRs form, it is necessary to determine how they behave in a natural environment after emission. To better understand this, the nature of EPFRs in ambient PM2.5 under simulated atmospheric conditions was investigated. Ambient PM2.5 samples were collected at a roadside ambient monitoring site near heavy interstate traffic and major industrial activity. Abstract Underwater excavations were conducted at an Early Classic (A.D. 300-600) and a Late Classic (A.D. 600-900) submerged ancient Maya salt works in Paynes Creek National Park, Belize. The archaeological investigations included underwater excavations, artifact analysis, marine sediment chemical testing, and source identification of obsidian—an important indicator of trade. The excavation and analysis of the salt works in Paynes Creek add a new perspective on ancient Maya craft production and the economies of other ancient civilizations. The first part of the thesis investigates the use of theoretical quantum calculations for the study of EPFRs as the initial and fundamental step in the formation of polychlorinated dibenzo-p-dioxin and dibenzofuran. The computational model comprised of density functionals (B3LYP, PBE1PBE, and M06) and two types of basis set namely: LANL2DZ for all atoms and GEN (LANL2DZ for metals and aug-cc-pVDZ for non-metals). Full mechanisms of EPFRs formation over (CuO)1-8, and aluminum oxide clusters were studied. A novel microfluidic device was developed for bacterial cell culturing using mass spectrometry as the detector. One of the challenges in proteomics is to achieve high sensitivity in the identification of proteins in complex samples with widely varying concentrations. The main limitations for proteomic studies are relatively slow and labor-intensive steps such as cell culturing and protein digestion of small sample quantities. Microfluidics is a promising approach to increase throughput and to reduce the time-consuming steps that are necessary for proteomics. ABSTRACT Research into young children’s leadership skills is sparse and focused on leadership in classroom contexts.
