Soft-sediment benthic environments are amongst the largest marine ecosystems in the world and play important roles in many ecosystem functions. In recent years, exploitation of resources and unintentional impacts on deep-sea benthic environments has increased. The Deepwater Horizon oil spill of 2010 in the northern Gulf of Mexico (GoM) represented a prime example of this. The oil spill not only highlighted deficiencies of data and information on baseline conditions, but also represented an opportunity to learn more and develop better methods for the future. Adaptive Divergence And Speciation Across Depth In A Caribbean Candelabrum Coral, Carlos Andrés Prada Montoya

Since Darwin, biologists wonder how organisms cope with environmental variation, why there are so many species, and how species form. In my dissertation, I explore how species of long-lived, clonal reef organisms originate across depth gradients. In Chapter 2, I evaluate the strength of depth to isolate populations by comparing the genes and morphologies of pairs of depth-segregated populations of the candelabrum coral Eunicea flexuosa across the Caribbean. Eunicea flexuosa is a long-lived clonal cnidarian that associates with an alga of the genus Symbiodinium. Genetic analysis revealed two depth-segregated lineages, each genetically well-mixed across the Caribbean. Survivorship data, combined with …

Scholarship has failed to explore adequately how Honoré de Balzac evokes the human condition’s universal elements through his most infamous criminal mastermind: Jacques Collin, alias Vautrin. Unlike analyses of Collin that I have encountered, this thesis takes all three novels and the obscure play in which Collin appears into account, challenges the transparency of his statements and the narration’s descriptions of him, explores the conservative position framing Balzac’s critique of early nineteenth-century Paris, and actively focuses on evidence of Collin’s typical subjectivity (i.e. his insatiable desire and fallibility). This aim of this research project is to probe the activation process of zebrafish spermatozoa. Zebrafish are a model species for biological engineering applications, and the cryopreservation of their reproductive cells allows for inexpensive cataloging and maintenance of valuable biological material. Evaluation of cryopreservation protocols for aquatic sperm cells is typically accomplished by motility analysis after subjecting cells to a cryopreservation treatment. In zebrafish sperm cells, motility is initiated when cells come into contact with a hypo-osmotic environment. Between 1865 and 1920, new gender expectations in the postbellum period, as well as the willingness to use the state to intervene in marriages led to social and legal reform that provided a mechanism to empower women and enforce their right to be free from violence. Women emerged from the Civil War more aware about the drawbacks of dependency. Evidence suggests that 15-lipoxygenase-2 (15-LOX-2) plays an important role in the progression of atherosclerosis. Due to the medical need to discover isoform specific inhibitors of 15-LOX-2, a novel human enzyme, our goal was to determine the 3-dimensional structure. Using bacterial expression, we were able to obtain a usable quantity of enzyme with a sufficient amount of the catalytic iron bound to the enzyme, however it was sensitive to cleavage by proteases. This led to a ratio of roughly 1:1 of cleaved and uncleaved protein after the final step of purification. Solving the crystal structure revealed the reason for the protease …

Understanding human long bone anatomy is an important concept to master for post-secondary students that major in medical fields since skeletal structures assist in locating a pulse, conducting clinical procedures, and identifying injection sites. Transition metal oxide nanoparticles contained in fly ash are known to catalyze the formation of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/Fs) during the waste incineration process. The potential catalytic activity of silica-supported NiO, CuO, and NiO-CuO nanoparticles for the formation of PCDD/Fs will be discussed in this dissertation. The successful synthesis of silica-supported NiO, CuO, and NiO-CuO nanoparticles as surrogates of combustion-generated nanoparticles was important to this study. Distracted driving continues to remain a cause of concern for a number of bodies, including government agencies, traffic safety advocacy groups and law enforcement agencies, because of its traffic safety risks. The driving simulator continues to be popular with researchers in collecting data on performance variables that provide scientific knowledge of the effects of distracted driving. Several of these performance variables can be used to quantify a single distracting effect, resulting in a multivariate dataset. Deinococcus radiodurans has a significantly more robust DNA repair response than Escherichia coli, which helps it survive high amounts of oxidative stress. D. radiodurans DNA Polymerase I (DrPolI) and RecA protein (DrRecA) play essential parts in this DNA repair capability. Structural stability studies of the DrPolI large fragment (Klendra) and examination of the DNA binding characteristics of Klendra and RecA are presented herein. In the absence of cofactors DrRecA binds to dsDNA more than 20 fold tighter than EcRecA, and binds ssDNA up to 9 fold tighter. Binding to dsDNA in the absence of cofactor presumably monitors DNA end binding. …

A water soluble, non-ionic, rodlike polymer, PEGL, was synthesized and characterized in efforts to synthesize a novel model polymer to study stiff polymers in solution. The rodlike system is likely stiff due to a high relative percent helicity in water up to 50 °C, measured by circular dichroism, the observation of liquid crystalline domains in water, and from the slope measured from conformation plots supplied by GPC/MALS; however, it also apparent the system is aggregating in both a 2 mM azide solution and a buffer (200 mM NaNO3 + 20 mM NaH2PO4 + 2 mM NaN3), in corroboration of negative …

Major Earth system perturbations in the deep past and today are recorded in sulfate sulfur and oxygen isotopes, as examined here in three cases. (1) Sedimentary sulfate record of the “Marinoan Oxygen-17 Depletion” (MOSD) event, implies ultra-high CO2 atmosphere at ~635 Ma after global glaciation. MOSD duration is constrained by correlating its most complete record to radiometric dates. Barium sulfate layers in South China sediments show the MOSD in lower layers but persistently absent up section. Carbon-13 correlation locates the MOSD within dated intervals from other sites, yielding a 0 - 0.99 Myr duration. Thus, sedimentary constraint on this non-steady-state …

Gulf killifish, Fundulus grandis, shows promise for commercial development as a marine baitfish species. Significant markets for F. grandis already exist throughout the Gulf Coast, with the vast majority of supply coming from wild-harvests, which can be hampered by seasonal availability and inconsistent fish health and size. Pers.) collected from 17 Louisiana sugarcane (Saccharum spp. hybrids) fields and two sites outside sugarcane-growing area was evaluated for genetic diversity, growth characteristics and response to glyphosate. Random Amplified Polymorphic DNA (RAPD) genetic analysis and Jacard’s similarity coefficient, a dedrogram, based on unweighted pair group mean average (UPGMA) identified two cluster groups based on presence of common alleles. The outer surface lipoprotein (osp) layer forms an interface between the internal and the external environment of the Lyme disease spirochete, Borrelia burgdorferi. The homeostatic maintenance of the osp layer effectuates adaptation of B. burgdorferi as it gets transmitted from the tick vector to a mammalian host and vice-versa. However, the regulation of the outer surface lipoproteins (osps) is still a conundrum for borrelia scientists. Part of this dissertation inquires about the homeostatic maintenance of the osp layer. Polonium-210 (t1/2=138 d) is the most common among the 33 known radioisotopes of Polonium found in the natural environment. It is produced by the radioactive decay of its long-lived grandparent Lead-210(t1/2=22.3 d) via Bismuth-210 (t1/2=5.012 d) and forms as part of the natural Uranium-238 decay series. The primary hazard associated with Polonium-210 is its radioactivity, as an alpha particle emitter. Marine organisms receive their maximum radioactive dose in the natural environment from Polonium-210. Severe bacterial sepsis leads to a pro-inflammatory condition that can manifest as septic shock, multiple organ failure, and death. Neutrophils are critical for the rapid elimination of bacteria, however, the role of neutrophil chemoattractant CXCL1, pattern recognition receptors (PRRs)- NLR protein 3 (NLRP3) and alcohol in bacterial clearance during sepsis remains elusive. We demonstrate that CXCL1 plays a pivotal role in mediating host defense to polymicrobial sepsis following cecal ligation and puncture (CLP) in gene-deficient mice. Scientific explanation and scientific argument are important aspects of science inquiry and science education. Though students of all ages find construction of written scientific explanations and arguments difficult, explicit instruction in scientific explanation and argument can improve student ability. This quantitative research study used a non-equivalent comparison group design to investigate the affect of explicit instruction of scientific explanation and argument in an undergraduate introductory biology laboratory course, using a college level appropriate modified CER Framework (McNeill et al., 2006). Previous researchers have demonstrated that Autism Spectrum Disorder (ASD) is related to increased prevalence of other psychological disorders. Comorbid disorders increase the difficulties in socialization and communication already experienced by children with ASD. These symptoms often require additional intervention techniques and should be identified as early as possible to beget the best prognosis. The Autism Spectrum Disorders – Child Version (ASD-C) is an informant based assessment battery specifically designed to measure symptoms of ASD, comorbid disorders, and challenging behavior in children with ASD. Burkholderia glumae is the major causal agent of bacterial panicle blight of rice, a disease that can cause significant yield losses. This bacterial pathogen produces a number of virulence factors that contribute to disease development. The phytotoxin, toxoflavin is required for full virulence and is the most important virulence factor in B. glumae. The known regulatory cascade of toxoflavin production involves the TofI-TofR quorum-sensing system, the transcriptional activators ToxJ and ToxR, and the toxoflavin biosynthesis and transport genes. Transposon mutagenesis of toxoflavin-producing B. glumae strains generated a number of mutants with increased toxoflavin production. Several of these mutants contained …
