An increasing number of laboratories are evaluating sample quality via motility analysis by means of computer-assisted sperm analysis (CASA) after sperm activation by manual dilution and mixing. Even with the use of CASA, due to user variation, there is a lack of control over the activation process, resulting in inconsistent motility analysis. Low sample volume (~1-2µL), and a short motility duration (burst motility of less than 15s) add to the complexity of these difficulties. To provide for a weed-free seedbed, Louisiana crop producers typically apply a burndown herbicide four to six wk prior to seeding summer annual crops; however, these treatments often provide inadequate henbit (Lamium amplexicaule L.) control. Research was conducted in Louisiana to evaluate henbit emergence from north to south, compare growth of henbit accessions based on emergence date, and control with fall-applied residual herbicides. For the emergence study during the weeks of Oct 17 to Dec 12 at all locations in all years, soil temperatures at 2.5 cm averaged between 10 and 18.5 C. Henbit at densities of at least 50 …

Obesity can have numerous detrimental consequences, namely metabolic syndrome, type 2 diabetes, cardiovascular disease, cancer, and Alzheimer’s disease. The pathogenesis and physiologic consequences of obesity are unknown, but they are often associated with increased inflammation and oxidative stress in both the body and in the brain. One factor that has been implicated in causing inflammation associated with a high fat diet is the enzyme NADPH oxidase, or NOX, specifically the subunit NOX2. The state of Louisiana has both temperate and subtropical climates and includes six ecoregions. Two ecoregions in south-central Louisiana were surveyed for members of the order Blattodea to better describe the diversity, life histories, and taxonomy of adult and nymphal cockroaches. Blattodea, excluding the newly included epifamily Termitoidae, is an understudied order of insects in spite of the importance species that are infamous peridomestic pest. The order Blattodea currently contains 9 families comprising of 460 genera. There are approximately 7570 described species 4641 of which are cockroaches and 2929 are termites. The northern Gulf of Mexico (GOM) contains approximately 2,500 oil and gas platforms, resulting in one of the largest de facto artificial reef systems in the world. As of 2013, 1,227 additional structures had ceased to produce oil and gas and have been decommissioned and removed. While active platforms are lit by high-pressure mercury vapor lights, inactive platforms are only minimally lit for navigation. The positively phototaxic behavior of many fish species causes lit oil platforms to act as fish attraction devices, especially at night. Louisiana leads all U.S. states in blue crab (Callinectes sapidus) landings, but high fuel and bait costs have hindered commercial fishing productivity of Louisiana in recent years. The primary baitfish, Atlantic menhaden (Brevoortia tyrannus), has steadily increased in price and decreased in availability, while crab prices remain low. To reduce costs for fishermen, an alternative bait was developed that incorporates shrimp waste into a semi-rigid alginate matrix. Lab testing and preliminary field tests show that shrimp-alginate bait may be a suitable alternative to menhaden for Louisiana crab fishermen. Patterns from hurricane damage give an indication that longleaf pine (Pinus palustris) is more windfirm than loblolly pine (Pinus taeda). Tree windfirmess has been attributed to many factors including species and material properties like wood strength and stiffness. Because longleaf pine wood is stronger and stiffer than loblolly pine wood, this study used static winching methodology to see if these properties account for differences in windfirmness by measuring bending force required to break stems (MMAX). Lignocellulosic biomass is a promising renewable resource for the production of biofuels and biochemicals. Energycane is considered a lignocellulosic biomass characterized by its high fiber content and cold tolerance. It can be planted on marginal land and does not need to compete with the food supply. Wet storage in combination with sodium hydroxide (NaOH) or white-rot fungi was applied to energycane bagasse to preserve the lignocellulosic polymeric sugars (cellulose and hemicellulose) during short-term storage (60 days) and to make them accessible for conversion into biofuels and biochemicals. Considerable attention has been given to the measureable differences that exist between different human populations in the size and shape of the pelvis, with Africans having a narrower pelvis than Europeans. By collecting data on sacral breadth from a South African skeletal population, this study (1) tests the hypothesis that African blacks possess a narrower sacrum, and by inference pelvis, than whites and (2) considers whether the size variation between blacks and whites is due to nutritional, historical and social differences, to a genetic basis related to climatic adaptation, or to both. Iron-sulfur ([Fe-S]) proteins are widely distributed in the biological systems. These proteins are involved in diverse fundamental life processes such as photosynthesis, respiration, ribosome biogenesis, DNA synthesis, and repair. Dysfunction of these proteins or their biogenesis has been implicated in causing cardiovascular diseases, neurodegenerative diseases, and various cancers. Iron-sulfur proteins are highly sensitive to nitric oxide (NO) and readily form dinitrosyl iron complexes (DNICs) upon exposure. Restoration of the iron-sulfur cluster in the NO-modified protein requires a two-step repair process. The first step is to remove DNICs from proteins. The objective of this research was to determine differences in carcass evaluation, yield, and meat quality from representative fresh domestic (FSD), frozen domestic (FZD), and frozen imported Australian (FIA) goat carcass sides (n=30 from each source). The U.S. carcasses were split into sides and the right side frozen and stored for 6 weeks. Left sides were fabricated into standardized primal cuts. After frozen storage, right sides and FIA carcasses were thawed for 3 days at 4°C. The weak Raman signals are enormously enhanced in the presence of a nano-patterned metallic surface next to the specimens. This dissertation describes a technique to fabricate a novel, low cost, high sensitive, disposable, and reproducible metallic nanostructure on a transparent substrate for Surface Enhanced Raman Scattering (SERS). Cysteine proteases (CPs) are enzymes with a nucleophilic thiol in their active sites. Inhibitors of cysteine proteases (ICPs) occur naturally in bacterial pathogens and some protozoa. In parasites, ICPs are often virulence factors, contributing to the formation and survival of amastigotes within host cells. These amastigotes have higher CP activity, therefore making both ICPs and CPs potential drug targets. Despite great genetic variability, ICPs contain highly conserved structural features, including a series of defined loops that play a significant role in binding CPs. Thiol-acrylate materials have been demonstrated to have therapeutic potential as biocompatible scaffolds for bone tissue regeneration due to their osteoconductivity, biodegradability, and well-suited mechanical properties. This study connects the mechanical properties and stability of thiol-acrylate polymer with cell adhesion and proliferation of human adipose derived stromal cells. The polymer presented in this study, trimethylolpropane ethoxylate triacrylate-co-trimethylolpropane tris (3-mercaptopropionate) (TMPeTA-co-TMPTMP), was synthesized by an amine-catalyzed Michael addition reaction. Physical, mechanical, and chemical characterizations were performed on the polymeric scaffold, followed by preliminary in vitro cytocompatibility tests. Live/dead staining assays showed significant differences in cell adhesion for TMPeTA (692 and 912 MW). …

In the first part of this dissertation, the adsorption isotherms, resonance coupling, and fluorescence of molecular dyes on the surface of plasmonic nanoparticles are investigates. SHG is also used to investigate the growth of a silver shell on the surface of gold nanoparticles in real time. Additionally, gold-silver-gold core-shell-shell nanoparticles possess extinction peak wavelengths that can be controlled over the visible and near-infrared regions and enhanced photothermal effect. Transient absorption measurements determine that the phonon-phonon scattering lifetime is considerably faster in the core-shell-shell nanoparticles than in the gold nanospheres and gold nanorods, which contributes to the higher photothermal efficiencies. The goal of my dissertation is to elucidate the origins and mechanisms underlying the recent successful population expansion of the invasive coral Oculina patagonica in the Mediterranean Sea. Research suggests that the cyclic AMP (cAMP) signaling pathway including CREB-CRE regulated expression of various genes is implicated in the predisposition to and development of alcoholism in humans. Regulatory elements are DNA sequences which have specialized activities that coordinate the functions of the genome. Promoters, enhancers, locus control regions, boundary elements (or insulator elements) are examples of DNA sequences that have regulatory properties. In transgenic assays insulator elements have been shown to block communication between regulatory regions, such as enhancers and promoters, when placed between these sequences and also protect genes from position effects when bracketing them, thereby affecting gene expression. Photosystem II (PSII) is a light-driven, water plastoquinone oxidoreductase present in all oxygenic photosynthetic organisms. The oxygen evolution process is catalyzed by the Mn4CaO5 cluster and an ensemble of intrinsic and extrinsic proteins which are associated with the photosystem. This metal cluster is stabilized and protected from exogenous reductants by the extrinsic proteins, PsbO, PsbP and PsbQ in higher plants, which are present on the lumenal face of PSII. Sensor arrays have evolved as powerful approaches for providing detection and discrimination of volatile organic compounds (VOCs) as required across numerous analytical applications. Such systems typically comprise a number of cross reactive sensor elements, which generate analyte specific response patterns upon exposure to VOCs, and are known as multisensor arrays. When evaluated using statistical methods, these response patterns facilitate classification of VOCs. As an alternative, a single dynamically operated sensor could also be used to generate analyte specific response patterns. Several of Alfred Hitchcock’s movies feature characters with disabilities. Often, these characters are protagonists, and Hitchcock systematically manipulates his audiences to identify with these characters through the editing process, sound effects, and music. This dissertation will analyze the ways music represents various disabilities in three Hitchcock films. Vertigo (1958) addresses obsession and phobia as its main themes, whereas Psycho (1960) investigates obsession and madness. Finally, The Man Who Knew Too Much (1956) explores muteness, hysteria, and identity in the context of two pieces of diegetic music.
