© 2016 International Association of Sedimentologists. The Marinoan panglaciation (cα 650 to 635 Ma) is represented in north-east Svalbard by the 130 to 175 m thick Wilsonbreen Formation which contains syn-glacial carbonates in its upper 100 m. These sediments are now known to have been deposited under a CO2-rich atmosphere, late in the glaciation, and global climate models facilitate testing of proposed analogues. Precipitated carbonates occur in four of the seven facies associations identified: Fluvial Channel (including stromatolitic and intraclastic limestones in ephemeral stream deposits); Dolomitic Floodplain (dolomite-cemented sand and siltstones, and microbial dolomites); Calcareous Lake Margin (intraclastic dolomite and …

To increase our understanding of the genetic basis of adiposity and its links to cardiometabolic disease risk, we conducted a genome-wide association meta-analysis of body fat percentage (BF%) in up to 100,716 individuals. Twelve loci reached genome-wide significance (Po5108), of which eight were previously associated with increased overall adiposity (BMI, BF%) and four (in or near COBLL1/GRB14, IGF2BP1, PLA2G6, CRTC1) were novel associations with BF%. Seven loci showed a larger effect on BF% than on BMI, suggestive of a primary association with adiposity, while five loci showed larger effects on BMI than on BF%, suggesting association with both fat and …

A Dose-Schedule Finding Design For Phase I-Ii Clinical Trials, Beibei Guo, Yisheng Li, Ying Yuan

Faculty Publications

Summary: Dose finding methods aiming at identifying an optimal dose of a treatment with a given schedule may be at a risk of misidentifying the best treatment for patients. We propose a phase I-II clinical trial design to find the optimal dose-schedule combination. We define schedule as the method and timing of administration of a given total dose in a treatment cycle. We propose a Bayesian dynamic model for the joint effects of dose and schedule. The model proposed allows us to borrow strength across dose-schedule combinations without making overly restrictive assumptions on the ordering pattern of the schedule effects. …

Quantifying Demographic Uncertainty: Bayesian Methods For Integral Projection Models, Bret D. Elderd, Tom E.X. Miller

© 2016 by the Ecological Society of America. Integral projection models ( IPM s) are a powerful and popular approach to modeling population dynamics. Generalized linear models form the statistical backbone of an IPM . These models are typically fi t using a frequentist approach. We suggest that hierarchical Bayesian statistical approaches offer important advantages over frequentist methods for building and interpreting IPM s, especially given the hierarchical nature of most demographic studies. Using a stochastic IPM for a desert cactus based on a 10- year study as a worked example, we highlight the application of a Bayesian approach for …

Change In Obesity Prevalence Across The United States Is Influenced By Recreational And Healthcare Contexts, Food Environments, And Hispanic Populations, Candice A. Myers, Tim Slack, Corby K. Martin, Stephanie T. Broyles, Steven B. Heymsfield

Faculty Publications

Objective To examine change in county-level adult obesity prevalence between 2004 and 2009 and identify associated community characteristics. Methods Change in county-level adult (≥20 years) obesity prevalence was calculated for a 5-year period (2004-2009). Community measures of economic, healthcare, recreational, food environment, population structure, and education contexts were also calculated. Regression analysis was used to assess community characteristics associated (p< 0.01) with change in adult obesity prevalence. Results Mean±SD change in obesity prevalence was 5.1±2.4%. Obesity prevalence decreased in 1.4% (n = 44) and increased in 98% (n = 3,060) of counties from 2004-2009. Results showed that both baseline levels and increases in physically inactive adults were associated with greater increases in obesity prevalence, while baseline levels of and increases in physician density and grocery store/supercenter density were related to smaller increases in obesity rates. Baseline levels of the Hispanic population share were negatively linked to changing obesity levels, while places with greater Hispanic population growth saw greater increases in obesity. Conclusions Most counties in the U.S. experienced increases in adult obesity prevalence from 2004 to 2009. Findings suggest that community-based interventions targeting adult obesity need to incorporate a range of community factors, such as levels of physical inactivity, access to physicians, availability of food outlets, and ethnic/racial population composition.

The response and adaption to salt remains poorly understood for beach morning glory [Ipomoea imperati (Vahl) Griseb], one of a few relatives of sweetpotato, known to thrive under salty and extreme drought conditions. In order to understand the genetic mechanisms underlying salt tolerance of a Convolvulaceae member, a genome-wide transcriptome study was carried out in beach morning glory by 454 pyrosequencing. A total of 286,584 filtered reads from both salt stressed and unstressed (control) root and shoot tissues were assembled into 95,790 unigenes with an average length of 667 base pairs (bp) and N50 of 706 bp. Predation on bacteria and accompanying mortality are important mechanisms in controlling bacterial populations and recycling of nutrients through the microbial loop. The agents most investigated and seen as responsible for bacterial mortality are viruses and protists. However, a body of evidence suggests that predatory bacteria such as the Halobacteriovorax (formerly Bacteriovorax), a Bdellovibrio-like organism, contribute substantially to bacterial death. Until now, conclusive evidence has been lacking. The goal of this study was to better understand the contributors to bacterial mortality by addressing the poorly understood role of Halobacteriovorax and how their role compares with that of viruses. The results revealed …

Efficient ethanol production from lignocellulosic biomass requires highly degradable feedstock; therefore, there is a similarity between forage crop production for ruminant animals and ethanol production from lignocellulosic biomass. Feed value analysis techniques may be used to estimate lignocellulosic biomass quality. Because lignin and its derivatives in cell walls are major compounds interrupting biomass degradation, fiber analysis and in vitro incubation tests were conducted with switchgrass (Panicum virgatum) and energycane (Saccharum spp.) biomass collected at 100 and 120g lignin (acid detergent lignin) kg-1 DM (dry matter). Mean NDF (neutral detergent fiber) in switchgrass was consistently greater than that of energy cane …

Reduced glomerular filtration rate defines chronic kidney disease and is associated with cardiovascular and all-cause mortality. We conducted a meta-analysis of genome-wide association studies for estimated glomerular filtration rate (eGFR), combining data across 133,413 individuals with replication in up to 42,166 individuals. We identify 24 new and confirm 29 previously identified loci. Of these 53 loci, 19 associate with eGFR among individuals with diabetes. Using bioinformatics, we show that identified genes at eGFR loci are enriched for expression in kidney tissues and in pathways relevant for kidney development and transmembrane transporter activity, kidney structure, and regulation of glucose metabolism. Chromatin …

© 2016 the American Physiological Society. Adiponectin is a hormone secreted from adipocytes that plays an important role in insulin sensitivity and protects against metabolic syndrome. Growth hormone (GH) and prolactin (PRL) are potent STAT5 activators that regulate the expression of several genes in adipocytes. Studies have shown that the secretion of adiponectin from adipose tissue is decreased by treatment with PRL and GH. In this study, we demonstrate that 3T3-L1 adipocytes treated with GH or PRL exhibit a reduction in adiponectin protein levels. Furthermore, we identified three putative STAT5 binding sites in the murine adiponectin promoter and show that …

Oleic-acid stabilized iron oxide nanocrystals were prepared by thermal decomposition of iron(III) acetylacetonate. Their crystallinity, morphology, and superparamagnetism were determined using X-ray diffraction, transmission electron microscopy, and a superconducting quantum interference device. Monodisperse core-shell particles were produced in the silica-coating of iron oxide via microemulsion synthesis. Surface modification of these particles was performed in two steps, which included the reaction of silica-coated iron oxide particles with 3-bromopropyltrichlorosilane, followed by azido-functionalization with sodium azide. Monoalkylated porphyrins …

Impact Of The 2010 Deepwater Horizon Oil Spill On Population Size And Genetic Structure Of Horse Flies In Louisiana Marshes, Claudia Husseneder, Jennifer R. Donaldson, Lane D. Foil

Faculty Publications

The greenhead horse fly, Tabanus nigrovittatus Macquart, is frequently found in coastal marshes of the Eastern United States. The greenhead horse fly larvae are top predators in the marsh and thus vulnerable to changes in the environment, and the adults potentially are attracted to polarized surfaces like oil. Therefore, horse fly populations could serve as bioindicators of marsh health and toxic effects of oil intrusion. In this study, we describe the impact of the April 2010 Deep Water Horizon oil spill in the Gulf of Mexico on tabanid population abundance and genetics as well as mating structure. Horse fly populations …

© 2016 Nature America, Inc. Rationale: Caveolin-1 (Cav-1) negatively regulates endothelial nitric oxide (NO) synthase-derived NO production, and this has been mapped to several residues on Cav-1, including F92. Herein, we reasoned that endothelial expression of an F92ACav-1 transgene would let us decipher the mechanisms and relationships between caveolae structure and intracellular signaling. Objective: This study was designed to separate caveolae formation from its downstream signaling effects. Methods and Results: An endothelial-specific doxycycline-regulated mouse model for the expression of Cav-1-F92A was developed. Blood pressure by telemetry and nitric oxide bioavailability by electron paramagnetic resonance and phosphorylation of vasodilator-stimulated phosphoprotein were …

Molar Tooth Carbonates And Benthic Methane Fluxes In Proterozoic Oceans, Bing Shen, Lin Dong, Shuhai Xiao, Xianguo Lang, Kangjun Huang, Yongbo Peng, Chuanming Zhou, Shan Ke, Pengju Liu

Faculty Publications

Molar tooth structures are ptygmatically folded and microspar-filled structures common in early- and mid-Proterozoic (â 1/42,500-750 million years ago, Ma) subtidal successions, but extremely rare in rocks Here, on the basis of Mg and S isotopes, we show that molar tooth structures may have formed within sediments where microbial sulphate reduction and methanogenesis converged. The convergence was driven by the abundant production of methyl sulphides (dimethyl sulphide and methanethiol) in euxinic or H 2 S-rich seawaters that were widespread in Proterozoic continental margins. In this convergence zone, methyl sulphides served as a non-competitive substrate supporting methane generation and methanethiol inhibited …

Effects Of The Deepwater Horizon Oil Spill On Coastal Marshes And Associated Organisms, N N. Rabalais, R. Eugene Turner

Faculty Publications

Oil gushed from the Macondo Mississippi Canyon 252 well into the Gulf of Mexico for 87 days after the Deepwater Horizon drilling rig exploded and sank. A concern, after widespread dispersant use offshore on surface waters and at the wellhead, was that the oil/dispersant mixture would reach valuable, and vulnerable, coastal ecosystems. Standardized oil spill response methodology identified 1,773 km of the 7,058 km of surveyed shoreline as oiled, with 1,075 km oiled in Louisiana. This paper synthesizes key results of published research on the oiling effects on coastal habitats and their inhabitants from microbes to vertebrates. There were immediate …

Qualitative inferences and sparse bay-wide measurements suggest that shoreline erosion increased after the 2010 BP Deepwater Horizon (DWH) disaster, but quantifying the impacts has been elusive at the landscape scale. We quantified the shoreline erosion of 46 islands for before and after the DWH oil spill to determine how much shoreline was lost, if the losses were temporary, and if recovery/restoration occurred. The erosion rates at the oiled islands increased to 275% in the first six months after the oiling, were 200% of that of the unoiled islands for the first 2.5 years after the oiling, and twelve times the …

Physiological information is rarely used in descriptions of maturity for managed, wild fish species; however, the use of physiological data holds great promise to provide important detail on the complexities of oocyte development and maturity. Investigating southern flounder (Paralichthys lethostigma)—an overfished commercial and recreational fishery resource—we examined pre-spawn physiological changes in females to provide further detail of the maturation process. Given that adults of this species complete maturation and spawn in unknown offshore locations, information on pre-spawn physiological changes is particularly informative for both size- and age-based patterns of maturity. We evaluated seasonal and ontogenetic changes in hormone concentrations in …

Therapeutic strategies have been reported that depend on synthetic network devices in which a urate-sensing transcriptional regulator detects pathological levels of urate and triggers production or release of urate oxidase. The transcription factor involved, HucR, is a member of the multiple antibiotic resistance (MarR) protein family. We show that protonation of stacked histidine residues at the pivot point of long helices that form the scaffold of the dimer interface leads to reversible formation of a molten globule state and significantly attenuated DNA binding at physiological temperatures. We also show that binding of urate …

One useful approach is to examine congeners varying in invasiveness in a comparative framework across native and invaded ranges. Acacia species have been widely introduced outside their native range of Australia, and a subset of these species have become invasive in multiple parts of the world. Within specific regions, the invasive status of these species varies. Our study examined whether a key mechanism in the life history of Acacia species, the legume-rhizobia symbiosis, influences acacia invasiveness …

Two homologous cyclindependent kinase inhibitor genes, SIAMESE (SIM) and SIM-RELATED 1 (SMR1), were recently shown to regulate Arabidopsis (Arabidopsis thaliana) defense based on phenotypes conferred by a sim smr1 double mutant. However, whether these two genes play differential roles in cell-cycle and defense control is unknown. In this report, we show that while acting synergistically to promote endoreplication, SIM and SMR1 play different roles in affecting the ploidy of trichome and leaf cells, respectively. In addition, we …

The cytotoxicity and uptake of four di(amino acid) chlorin e6 conjugates were investigated in human HEp2 cells, and compared with chlorin e6. The most cytotoxic and most taken up conjugates …
