Placental Adaptation To Maternal Malnutrition, M. Carey Satterfield, Ashley K. Edwards, Fuller W. Bazer, Kathrin A. Dunlap, Chelsie B. Steinhauser, Guoyao Wu

Faculty Publications

Maternal malnutrition gives rise to both short- and long-term consequences for the survival and health of the offspring. As the intermediary between mother and fetus, the placenta has the potential to interpret environmental signals, such as nutrient availability, and adapt to support fetal growth and development. While this potential is present, it is clear that at times placental adaptation fails to occur resulting in poor pregnancy outcomes. This review will focus on placental responses to maternal undernutrition related to changes in placental vascularization and hemodynamics and placental nutrient transport systems across species. While much of the available literature describes placental …

Genetic Knockouts Indicate That The Abcc2 Protein In The Bollworm Helicoverpa Zea Is Not A Major Receptor For The Cry1ac Insecticidal Protein, Omaththage P. Perera, Nathan S. Little, Heba Abdelgaffar, Juan Luis Jurat-Fuentes, Gadi V.P. Reddy

Faculty Publications

Members of the insect ATP binding cassette transporter subfamily C2 (ABCC2) in several moth species are known as receptors for the Cry1Ac insecticidal protein from Bacillus thuringiensis (Bt). Mutations that abolish the functional domains of ABCC2 are known to cause resistance to Cry1Ac, although the reported levels of resistance vary widely depending on insect species. In this study, the function of the ABCC2 gene as a putative Cry1Ac receptor in Helicoverpa zea, a major pest of over 300 crops, was evaluated using CRISPR/Cas9 to progressively eliminate different functional ABCC2 domains. Results from bioassays with edited insect lines support that mutations …

Stability Analysis Using Generalized Sup-Delay Inequalities, Frederic Mazenc, Michael Malisoff, Miroslav Krstic Oct 2021

Stability Analysis Using Generalized Sup-Delay Inequalities, Frederic Mazenc, Michael Malisoff, Miroslav Krstic

Faculty Publications

We provide a new generalization of a recent trajectory-based approach for proving global asymptotic stability properties, using new sup-delay inequalities. Our generalization eliminates the usual contractivity requirement from previous sup-delay inequality approaches. We show how the requirements of our generalization hold under less restrictive Halanay's conditions than those that were previously reported. While the usual Halanay's inequality requires the decay rate to exceed the gain, our less restrictive conditions allow the gain to exceed the decay rate. We apply our work to systems with switching delays and to a continuous-discrete system.

Leptin Receptor Signaling Is Required For Intact Hypoglycemic Counterregulation: A Study In Male Zucker Rats, Christopher D. Morrison, Marina A. Duvall, Cristal M. Hill, Redin A. Spann, David H. Mcdougal

Division of Basic Science Faculty Publications

Hypoglycemia is a major barrier to clinical management of persons with diabetes. Emerging evidence supports a role for leptin in gating hypoglycemic counterregulation. This work demonstrates that male leptin receptor null, Zucker (fa/fa), rats display severe impairments in hypoglycemic counterregulation. Thus, augmenting leptin levels may have clinical utility for preventing hypoglycemia.

Faculty Publications

In 2010, the Deepwater Horizon oil spill released an estimated 4.9 million barrels of oil into the Gulf of Mexico, damaging coastal ecosystems. Seaside Sparrows (Ammospiza maritima)—a year-round resident of Gulf Coast salt marshes—were exposed to oil, as shown by published isotopic and molecular analyses, but fitness consequences have not been clarified. We monitored nests around two bays in Plaquemines Parish, Louisiana, USA from 2012–2017 to assess possible impacts on the nesting biology of Seaside Sparrows. A majority of nests failed (76% of known-fate nests, N = 252 nests, 3521 exposure-days) during our study, and predation was the main cause …

Effect Of Aerobic Exercise-Induced Weight Loss On The Components Of Daily Energy Expenditure, Nicholas T. Broskey, Corby K. Martin, Jeffrey H. Burton, Timothy S. Church, Eric Ravussin, Leanne M. Redman

Clinical Research Faculty Publications

Introduction Exercise usually results in less weight loss than expected. This suggests increased energy intake and/or deceased expenditure counteract the energy deficit induced by exercise. The aim of this study was to evaluate changes in components of daily energy expenditure (doubly labeled water and room calorimetry) after 24 wk of exercise training with two doses of aerobic exercise. Methods This was an ancillary study in 42 (29 women, 13 men) sedentary, middle-age (47.8 ± 12.5 yr) individuals with obesity (35 ± 3.7 kg·m-2) enrolled in the Examination of Mechanisms of Exercise-induced Weight Compensation study. Subjects were randomized to three groups: …

Analysis Of A New Multispecies Tumor Growth Model Coupling 3d Phase-Fields With A 1d Vascular Network, Marvin Fritz, Prashant K. Jha, Tobias Köppl, J. Tinsley Oden, Barbara Wohlmuth

Faculty Publications

In this work, we present and analyze a mathematical model for tumor growth incorporating ECM erosion, interstitial flow, and the effect of vascular flow and nutrient transport. The model is of phase-field or diffused-interface type in which multiple phases of cell species and other constituents are separated by smooth evolving interfaces. The model involves a mesoscale version of Darcy's law to capture the flow mechanism in the tissue matrix. Modeling flow and transport processes in the vasculature supplying the healthy and cancerous tissue, one-dimensional (1D) equations are considered. Since the models governing the transport and flow processes are defined together …

Modeling And Simulation Of Vascular Tumors Embedded In Evolving Capillary Networks, Marvin Fritz, Prashant K. Jha, Tobias Köppl, J. Tinsley Oden, Andreas Wagner, Barbara Wohlmuth

Faculty Publications

In this work, we present a coupled 3D–1D model of solid tumor growth within a dynamically changing vascular network to facilitate realistic simulations of angiogenesis. Additionally, the model includes erosion of the extracellular matrix, interstitial flow, and coupled flow in blood vessels and tissue. We employ continuum mixture theory with stochastic Cahn–Hilliard type phase-field models of tumor growth. The interstitial flow is governed by a mesoscale version of Darcy's law. The flow in the blood vessels is controlled by Poiseuille flow, and Starling's law is applied to model the mass transfer in and out of blood vessels. The evolution of …

Mitochondria undergo continuous cycles of fission and fusion to promote inheritance, regulate quality control, and mitigate organelle stress. More recently, this process of mitochondrial dynamics has been demonstrated to be highly sensitive to nutrient supply, ultimately conferring bioenergetic plasticity to the organelle. However, whether regulators of mitochondrial dynamics play a causative role in nutrient regulation remains unclear. In this study, we generated a cellular loss-of-function model for dynamin-related protein 1 (DRP1), the primary regulator of outer membrane mitochondrial fission. Loss of DRP1 (shDRP1) resulted in extensive ultrastructural and functional remodeling of mitochondria, characterized by pleomorphic enlargement, increased electron density of …

Zeta Potential Measurements For Surface Modification Of Plastic Substrates For Nanofluidic Biosensors, Qinhan Liu Sep 2021

Zeta Potential Measurements For Surface Modification Of Plastic Substrates For Nanofluidic Biosensors, Qinhan Liu

LSU Master's Theses

Polymeric materials have revealed their great potentials in micro- and nano- fluidic devices for biosensing. Their advantages include low cost, various physicochemical properties, well-developed fabrication, and surface modification protocols comparing with traditional materials such as silicon or glass. Surface modification consists an essential step in fabrication of polymer-based biosensors. A proper characterization is required to justify the effectiveness of modifications on polymer surface. For nanofluidic devices, special phenomena such as overlapped electrical double layer, ion rectification and electroosmosis flow can dominate the behavior of single molcules. The concept of surface charge density plays a key role in nanofluidic devices due …

A Modular Open-Technology Device To Measure And Adjust Concentration Of Sperm Samples For Cryopreservation, Nikolas C. Zuchowicz

LSU Master's Theses

Repositories for aquatic germplasm can safeguard the genetic diversity of species of interest to aquaculture, research, and conservation. The development of such repositories is impeded by a lack of standardization both within laboratories and across the research community. Protocols for cryopreservation are often developed ad hoc and without close attention to variables, such as sperm concentration, that strongly affect the success and consistency of cryopreservation. The wide dissemination and use of specialized tools and devices can improve processing reliability, provide data logging, produce custom hardware to address unique problems, and save costs, time, and labor. The goal of the present …

Blue Carbon In South Florida's Mangroves: The Role Of Large Roots And Necromass, Zoë I. Shribman Sep 2021

Blue Carbon In South Florida's Mangroves: The Role Of Large Roots And Necromass, Zoë I. Shribman

LSU Master's Theses

Blue carbon sequestration and storage in mangroves largely result from belowground biomass allocation in response to flooded anaerobic soil conditions and nutrient availability. Biomass allocation to belowground roots is a major driver of mangrove soil formation and organic matter accumulation leading to blue carbon storage potential. Belowground biomass sampling in mangroves is labor intensive, limiting data availability on biomass stocks, particularly for large roots (>20 mm diameter) and necromass (dead roots). The mangrove nutrient model (NUMAN) uses mostly literature values to parameterize a soil cohort approach to simulate depth distribution of root mass and organic carbon concentration. We evaluated …

Corrosion inhibitors can be utilized to decrease the corrosion kinetics and therefore increase the durability of reinforced concrete structures. Recently, a green synthesized organic compound, 1-benzyl-4-phenyl-1H-1,2,3-triazole (BPT), was shown to be a successful green organic corrosion inhibitor for mild steel. Studies suggested that the BPT adsorbs chemically onto the steel and acts as a mixed inhibitor, suppressing both the anodic and cathodic corrosion kinetics of steel. In addition, microcapsules have shown to be an efficient way for a controlled inhibitor release in reinforced concrete structures. On the other hand, geopolymers (GPs) comprised of a long range of covalently bonded alumino-silicates, …

The rapid and widespread evolution of fungicide resistance remains a challenge for crop disease management. The demethylation inhibitor (DMI) class of fungicides is a widely used chemistry for managing disease, but there has been a gradual decline in efficacy in many crop pathosystems. Reliance on DMI fungicides has increased resistance in populations of the plant pathogenic fungus Cercospora beticola worldwide. To better understand the genetic and evolutionary basis for DMI resistance in C. beticola, a genome-wide association study (GWAS) and selective sweep analysis were conducted for the first time in this species. We performed whole-genome resequencing of 190 C. beticola …

Novel strategies for the management of soybean looper, Chrysodeixis includens (Walker) (Lepidoptera: Noctuidae), are needed due to its increasing importance, costs, and challenges of management in soybeans (Glycine max [L.] Merr.; Fabaceae) in the Americas. Pheromone-mediated mating disruption may be 1 such novel strategy, but an effective tool for estimating mating disruption success must be found. The technique of trapping male moths using pheromone baited traps is a common method for assessing mating disruption technology. This paper reports on the testing of 2 trap types and 3 commercially available lures containing (Z)-7-dodecenyl acetate as the major active component. Trapping was …
