A Novel Process For The Production Of Clinical Dextran., Doman Kim

A new process for the production of clinical (or controlled size) dextran was developed. This process is simpler, cheaper and more economical than traditional methods. It involves the use of a mixed culture fermentation with a dextranase producing strain (Lipomyces starkeyi ATCC 74054) and a dextransucrase producing strain (Leuconostoc mesenteroides ATCC 10830). The new process produced 84% (w/w) of theoretical yield clinical dextran with an average polydispersity between 1.2 and 1.5. The mechanism of clinical dextran formation is the competitive production of acceptors from newly formed dextran molecules by dextranase. This purpose of this study is to assess the efficacy of performance as a means of social change by focusing on performances that have arisen out of the AIDS crisis. Working from a cultural studies perspective, I attempt to delineate both the possibilities and limitations for this perspective on social change. By comparing the various forms these performances have taken, I offer an explanation as to how these forms inhibit or enhance the ability of a performance to challenge the dominant discourse on AIDS, and create and mobilize communities that can act in the fight against AIDS. The text of this dissertation explores interconnections between cosmology and curriculum. I believe the demands of an Ecozoic Age will initiate a reconceptualizing of curriculum. Further, I believe the particular cosmological perspective I advance can help us with this reconceptualization. Chapter 1 utilizes the multi-faceted insights of scientific, historical, and speculative thought. The texts used in this chapter incorporate an engagement with past cultures (Ong, Eliade), with contemporary theoretical perspectives (Munitz, Haught, and Stafford), and with a scholarship that envisions alternative futures (Bohm). Predator-prey relationships between post-larval and juvenile teleosts are greatly influenced by the manner in which feeding behavior of fish interacts with the behavior of meiofaunal prey. Juvenile starry flounder (Platichthys stellatus Pallas) in a southeastern Alaskan bay fed disproportionately on the harpacticoid copepod Microarthridion littorale (Poppe), particularly on males. This apparent selectivity was not due to differing vertical distribution of harpacticoid species or to feeding in other locations. Comparison of ingested prey to harpacticoid assemblages collected by various techniques (including near-bottom collections) suggested that the prey selection shown by starry flounder may be the result of emergent behavior of its …

This study presents comprehensive organic geochemical evidence of organic-rich marine shales in the middle Eocene lower part of the Claiborne Group and the lower Eocene-Paleocene Wilcox Group of south Louisiana. The shales are the only post-Cretaceous age sediments in the northwest Gulf of Mexico basin that fit the conventional criteria for liquid hydrocarbon source rocks. Visual and geochemical evidence suggests that the Paleogene shales contain terrestrially derived, amorphous kerogen with high hydrogen indices whose oil potential has been upgraded by bacterial degradation. The Paleogene section is at immature to late mature levels with respect to petroleum generation, at present-day burial …

Studies dealing with the availability and speciation of arsenic (As) as affected by soil redox potential and pH were initiated because of the lack of information on As chemistry in flooded soils. The chemistry of native and applied As was studied in a Crowley silt loam soil (Typic Albaqualf). Arsenic uptake and its toxic effect on two rice cultivars as affected by As chemical form and concentration were also studied. Soil redox potential and pH were shown to affect speciation and solubility of both native and applied As. In a biochemical systematic study of the tribe Heliantheae, the isolation and structural elucidation of secondary metabolites from four species is reported. The aerial parts of Melampodium leucanthum afforded, besides the known melampolides melampodin A acetate, leucanthin B and leucanthinin, the new lactone leucanthinin-4-epoxide. Peracid oxidation of leucanthinin provided its monoepoxide derivative, the physical and spectroscopical data of which were identical with the new natural lactone. The isolation and structural elucidation of 7$\alpha$-hydroxydehydrocostus lactone, 13-hydroxy-11,13-dihydro-7,11-dehydro-3-desoxy-zaluzanin C and reynosin from Podachaenium eminens is reported. The isolation of the furanoheliangolides atripliciolide 2-methylbutyrate, atripliciolide tiglate and 17,18-dihydrobudlein A from Pappobolus nigrescens was accomplished. …

This work focuses on the production and characterization of crosslinked products obtained from PS II preparations, with the aim of better understanding the structural relationships between the manganese stabilizing protein (MSP) and the interior antenna protein CPa-1. Two oxygen evolving reaction center (OERC) preparations, OERC complex and OERC core, produced various crosslinked products with the hydrophobic crosslinker dithio-bis-(succinimidyl propionate) (DTSP). Crosslinking experiments with OERC complex demonstrated that a close association of MSP to CPa-1 occurs, as both proteins were found to be present simultaneously in various crosslinked products revealed by one- and two-dimensional electrophoretic and immunoblotting techniques. DRAINMOD-CREAMS model was used to simulate surface runoff and sediment loss from subsurface drained plot and non-subsurface drained plot in southern Louisiana. Seven years (1981-1987) of recorded data were used in this study. In general, the performance of the model in simulating surface runoff and sediment loss from subsurface drained plot is satisfactory. However, the DRAINMOD-CREAMS model did not simulate surface runoff and sediment loss accurately from non-subsurface drained plot. The CREAMS nutrient submodel was also tested on both plots. The CREAMS model seriously overestimates the nitrogen loss from non-subsurface drained plot. The sensitivity of the DRAINMOD-CREAMS model on erodibility …

Micellar electrokinetic capillary chromatography (MECC) is a rapidly growing technique allowing separation of electrically neutral compounds with the high separation efficiencies typically observed in capillary zone electrophoresis. This technique is still in its infancy and subsequently suffers from several inherent problems which must be addressed such as the limited range of solute elution and clear protocol for optimization of separations. Field-based studies and quantitative models provide new insights into the stratigraphic architecture of a developing continental extensional orogen. The early Miocene age Mojave Extensional Belt of southern California (MEB) exhibits a tripartite stratigraphy, consisting of (1) pre- to early synextension volcanic deposits, unconformably overlain by (2) syntectonic basement-derived megabreccia and breccia with local finer-grained units, overlain by (3) a posttectonic fining-upward sequence of gravel, sand, shale, and limestone. On a more local scale, stratigraphy and sediment dispersal within the MEB reflect the evolution of the major tectonic elements of the belt: (1) Breakaway zone. Laguna de Terminos is Mexico's largest coastal lagoon. There is a high habitat diversity including open water, mangroves, seagrasses and marshes, which supports Mexico's most economically important fisheries. Understanding ecosystem processes is essential for effective management of the lagoon's resources. This research developed landscape-level models for aquatic primary production (LAPPTER model), and fish migration (ROE model) in Laguna de Terminos. The ROE model was developed first as a theoretical approach, and later integrated into the LAPPTER model as a submodule which utilizes simulated production as a food source. Laboratory incubation studies were conducted to measure adsorption, movement, and transformations of urea and hydrolyzed ammoniacal N in flooded soil columns. Urea was added to the floodwater (750 mg N L$\sp{-1}$, equivalent to 150 kg N ha$\sp{-1}$) of Crowley silt loam soil columns and urea diffusion and urea hydrolysis were measured. Subsequent NH$\sb4\sp+$-N and NO$\sb3\sp-$-N distribution after urea application and hydrolysis were measured over a 30 d period under aerobic and anaerobic conditions. A Molecular Blueprint For The Pore-Forming Structure Of Voltage-Gated Calcium Channels, Anne Grove, John M. Tomich, Mauricio Montal

A protein that imitates the sequence of a highly conserved segment predicted to line the pore of dihydropyridine-sensitive L-type calcium channels was designed and synthesized. Single-channel conductance properties were studied in planar lipid bilayers. The synthetic protein emulates the ionic conductance, ionic selectivity, and pharmacological properties of the authentic calcium channel, including the stereospecific action of agonist and antagonist enantiomers of the dihydropyridine BayK 8644. The identified sequence is identical in L-type calcium channels from skeletal muscle and isoforms of cardiac muscle, brain, and aorta. The ontogenetic sequence of ossification of the cranium, including descriptions of the development of individual elements, from initial appearance of bone through adulthood, is described for two species of amphibian: the Spotted Salamander, Ambystoma maculatum, and the Tailed Frog, Ascaphus truei. This study is based on cleared-and-stained developmental series of 269 specimens of Ambystoma and 325 of Ascaphus. Nine stages of cranial development are defined for Ambystoma maculatum. By metamorphosis, all bones of the skull of Ambystoma except the articular have begun to ossify. The ossification sequence of Ambystoma shows little interindividual variation; only 6 specimens do not follow the …

We have investigated the distribution of several recently Inserted Alu family members within representatives of diverse human groups. Human population studies using 65 unrelated human DNA samples, as well as a familial study to test inheritance, showed that Individual Alu family members could be divided into three groups. The first group consisted of relatively older Alu family members which were monomorphic (homozygous) throughout the population tested (HS C3N1 and C4N6). The second group (HS C4N2, C4N5 and C4N8), apparently Inserted into other repetitive regions of the genome, resulting in inconclusive results in the PCR test used. However, it is clear …

Long-term water level changes in the northern Gulf of Mexico were examined using tide gauge records for this century. Strong coherence exists between the annual mean water changes at Galveston, Texas, and (1) the relatively geologically-stable west coast of Florida, (2) global mean sea level, and 93) the subsiding Louisiana coast. Water levels at the Galveston gauge, one of the longest records (81 yr), have risen steadily, but not accelerated over the long-term. The apparent acceleration of water rise in the recent two decades is within the historical pattern, and is probably driven by regional or global, but not local …

The national, local, and in-field distribution of Calonectria crotalariae (Loos) Bell and Sobers, as reflected in the distribution of Cylindrocladium black rot (CBR) of peanut, red crown rot (RCR) of soybean and microsclerotia densities in soil, respectively, are described. The current distribution of RCR in Louisiana is probably due to the introduction of the pathogen on indigo (Indigofera tinctoria (L.)) and to subsequent movement by man. RCR has been reported in 17 Louisiana parishes. In field studies, delayed soybean planting was efficacious in reducing RCR incidence. The objective of this experiment was to compare in vitro development of bovine embryos obtained from superovulation and in vitro fertilization (IVF) in two unique monolayer co-culture systems, bovine fetal spleen cell monolayers (BFS) and chick embryo fibroblast monolayers (CEF). To test the possible harmful effects of these co-culture systems, 225 murine morula-stage embryos were cultured for 72 hours with these monolayers. The percentage of hatched embryos was 62.7% in culture medium alone, 76% in BFS and 57.3% in CEF. BFS and CEF did not induce any detectable harmful effect on these developing murine embryos. Investigators have sought to clarify the Borderline Personality Disorder (BPD) category by comparing it with other personality disorders (e.g., Clarkin, Widiger, Frances, Hurt, & Gilmore, 1983; Zanarini, Gunderson, Grankenburg, & Chauncey, 1990), schizophrenia (see review by Siever & Gunderson, 1978), and major depression (see review by Gunderson & Phillips, 1991). The current study investigated the discriminative validity of the DSM-III-R (American Psychiatric Association, 1987) borderline criteria and qualitative differences in depression between borderlines (BPD) and patients diagnosed with major depressive episode (MDE). The 10 ligation species of human cyanomethemoglobin were previously found to distribute into three discrete cooperative free energy levels according to a combinatorial code (i.e., dependent on both the number and configuration of ligated subunits). Analysis of this distribution showed that the hemoglobin tetramer occupies a third allosteric state in addition to those of the unligated (T) and fully ligated (R) species. Acetate concentrations in marine and freshwater matrices were measured by an enzymatic technique which coupled the synthesis of acetyl coenzyme A to AMP production. The resulting AMP was assayed by a sensitive and relatively rapid high-pressure liquid chromatography method, using an aqueous, isocratic mobile phase for elution. The method was insensitive to the presence of seawater salts and required no sample prepurification or distillation. Propionate caused a minor, but statistically insignificant, interference when equimolar with acetate; butyrate caused no interference, even at relatively high concentrations. Detection limits for acetate were approximately 100 nM with a precision of about 5%. Pore …

Specific and sensitive amplification of major outer membrane protein (MOMP) gene DNA sequences of Chlamydia psittaci was achieved in a two-step polymerase chain reaction. First, oligonucleotide primers specific for 5' and 3' nontranslated regulatory regions of the MOMP gene were used in a polymerase chain reaction to amplify a DNA fragment of approximately 1,400 bp. A portion of this DNA fragment was amplified in a second reaction using a degenerate oligonucleotide primer specific for a DNA sequence contained within the 1,400-bp DNA fragment and one of the first-step primers. The entire nucleotide sequences of the spike glycoprotein (S) genes of the highly virulent bovine coronavirus (BCV) strain BCV-LY138, the avirulent BCV-L9 and related Norden Vaccine (BCV-Vaccine) strains were determined using the polymerase chain reaction (PCR) to amplify cDNAs obtained by reverse transcription of viral RNA, and to produce single strand cDNAs for DNA sequencing. The S gene sequences of these viral strains were compared with those of recently published strains BCV-Mebus, BCV-Quebec, and BCV-F15. Monoclonal antibodies (MAbs) against two major structural proteins of the cell-adapted Mebus strain of bovine coronavirus (BCV-L9) were produced and characterized. Seven MAbs reacted with the peplomeric glycoprotein, gp100/S, while three MAbs reacted with the nucleoprotein p53/N in Western blot analysis of BCV polypeptides. MAbs to gp100/S reacted with discontinuous epitopes of gp100/S in Westerns under mild but not under standard denaturing conditions. In contrast, MAbs to p53/N reacted in both types of Westerns, and those epitopes were thus continuous. Simplified Procedure For The Isolation Of Intact Chloroplasts From Chlamydomonas Reinhardtii, Catherine B. Mason, Sharon Matthews, Terry M. Bricker, James V. Moroney

A simple procedure that yields highly purified intact chloroplasts from Chlamydomonas reinhardtii is described. This procedure involves breakage of cell wall-deficient cells by passing them through a narrow bore syringe needle. The intact chloroplasts are then purified from the crude homogenate by differential centrifugation and Percoll gradient centrifugation. This procedure generates relatively high yields of chloroplasts capable of CO2 fixation. These chloroplasts were characterized by electron microscopy, marker enzyme analysis, and ferricyanide exclusion. Transmission electron microscopy indicates that these chloroplasts retain their pyrenoids and eyespots.
