Treatment of municipal and rural wastewater using subsurface flow constructed wetlands (SFCW's) is growing in popularity. These systems often consist of an excavated cell containing 2-3 in. rock with the wastewater level maintained below the media surface. A SFCW is a natural form of wastewater treatment which removes organics and nutrients based on the attached growth of microorganisms on crushed rocks where biological reactions take place. The currently accepted design procedure is based on the first order plug flow model, although tracer studies have shown that flow conditions are not well represented by the plug flow condition. Oxygen And Ph Regulation Of Protein Synthesis In Mitochondria From Artemia Franciscana Embryos, Kurt E. Kwast, Steven C. Hand

To identify factors responsible for the down-regulation of mitochondrial biosynthetic processes during anoxia in encysted Artemia franciscana embryos, the effects of oxygen limitation and pH on protein synthesis were investigated in isolated mitochondria. At the optimal pH of 7.5, exposure of mitochondria to anoxia decreases the protein synthesis rate by 79%. Rates were suppressed by a further 10% at pH 6.8, the intracellular pH (pH,) measured under anoxia in vivo. Matrix pH, measured under identical conditions, was 8.43 ± 0.01 at an extramitochondrial pH of 7.9 (mean ± S.E.M. n = 3), 8.05 ± 0.01 at pH 7.5, and 7.10 …

Carbohydrate Mobilization During Germination Of Post-Diapausing Gemmules Of The Freshwater Sponge Eunapius Fragilis, Stephen H. Loomis, Louis F. Ungemach, Bruce R. Branchini, Steven C. Hand, Paul E. Fell

Post-diapausing gemmules of the freshwater sponge Eunapius fragilis were found to contain sorbitol and glycogen as their primary carbohydrates. The sorbitol probably acts to increase the tolerance of the gemmules to freezing and desiccation. During germination, average sorbitol levels-measured as micromoles of sorbitol per gram of fresh weight of gemmule tissue (μmol/gfw)-declined from a control value of 36 μmol/gfw to about 4 μmol/gfw. Concomitantly, average glycogen levels increased from a control value of 29 μmol/gfw to a steady-state level of 62 μmol/gfw. A discontinuous ice coverage and lake temperature record of 36-yr duration has been compiled for Colour Lake (79°25'N, 90°45'W) on Axel Heiberg Island, Northwest Territories. About once every 6 yr, the ice cover remains through the entire summer, creating a residual ice cover the following winter. Residual ice covers are more frequent in the last 10 yr (1986-1995) than in the 20 yr from 1959 to 1978, indicating a reduction in climate factors controlling ice decay. These factors are identified as the number of thawing degree-days, suggesting a tendency toward cooler summers over the last decade. We report year-round meteorology …

Seedlings of Taxodium distichum L., Quercus lyrata Walt. and Q. falcata var. pagodaefolia Ell. were grown for 22 days in a rhizotron system providing two soil redox potential regimes, +170 mV (low Eh) and +560 mV (high Eh). Leaf chlorophyll concentration and gas exchange, root alcohol dehydrogenase (ADH) activity, root and leaf ethylene production, and growth and biomass partitioning were measured. In response to the low Eh soil treatment, stomatal conductance was reduced in Q. falcata and Q. lyrata but not in T. distichum, whereas net photosynthesis was reduced significantly in all species; however, net photosynthesis in T. distichum began …

Automatic Correlation And Calibration Of Noisy Sensor Readings Using Elite Genetic Algorithms, R. R. Brooks, S. S. Iyengar, J. Chen

This paper explores an image processing application of optimization techniques which entails interpreting noisy sensor data. The application is a generalization of image correlation; we attempt to find the optimal gruence which matches two overlapping gray scale images corrupted with noise. Both tabu search and genetic algorithms are used to find the parameters which match the two images. A genetic algorithm approach using an elitist reproduction scheme is found to provide significantly superior results.

Studies were conducted in Louisiana to determine the spatial and temporal distribution of rice water weevil (RWW), Lissorhoptrus oryzophilus Kuschel, adults, eggs, larvae and pupae in drill-seeded rice. In addition, three sequential sampling plans, Kuno's fixed precision sequential sampling plan, the sequential probability ratio test (SPRT), and 2-SPRT, were developed and evaluated to establish an accurate and economically efficient sampling plan for RWW larvae in drill-seeded rice in Louisiana. Values obtained for the aggregation indices, b, from Taylor's power law, $\beta$, from Iwao's patchiness regression and, k, from the negative binomial distribution indicated that RWW larvae were nearly randomly distributed …

Three objectives that could lead to improved understanding and management of Pythium root rot of sugarcane were addressed. First, the potential of amending soil with different organic wastes to suppress root rot was evaluated, and the biological and chemical properties of wastes that may contribute to disease suppression were identified. Secondly, herbicide interactions with root rot affecting sugarcane growth were assessed. Finally, the role of weeds in disease epidemiology was investigated. Greenhouse and laboratory studies indicated soil amendment with some organic wastes offers a sustainable disease management option through biological control. Microplot experiments were established in 1992, 1993, and 1994 to investigate the relationships between the fungal pathogen Macrophomina phaseolina, and a nematode community comprised of Criconemella xenoplax and Tylenchorhynchus annulatus on grain sorghum in Louisiana. A factorial treatment arrangement was employed and consisted of two sorghum hybrids (De Kalb DK 50 and Pioneer hybrid 8333), three levels of M. phaseolina (0, 10, and 100 colony forming units (cfu)/g soil) and three nematode inoculum levels (0, X, and 2X). Application of composted municipal waste to agricultural lands has potential benefits to improve soil fertility and crop yield. It may lead to accumulation of heavy metals within soils and leaching of nutrients and heavy metals into the ground water. This project was performed in order to determine whether exercise-induced increases in plasma cortisol concentrations may be responsible for enhanced lymphokine activated killer (LAK) cell activity. Secondly, this project included preliminary experments which were designed to test the hypothesis that stimulatory effects of cortisol on LAK cell activity may be due to cross-talk between signalling systems important to glucocorticoid effects and stimuli which act through nuclear activator protein-1 (AP-1). Blood samples were collected from resting horses in order to determine whether the animals had normal plasma glucocorticoid concentrations. A PCR method with primers for t-RNA gene spacers was used for detection of Xanthomonas albilineans (Xa), the causal agent of leaf scald disease of sugarcane. A DNA segment of approximately 170 bp was amplified for Xa strains, but also for X. campestris pv. campestris, X. campestris pv. translucens and two sugarcane bacterial endophytes. Good agreement between PCR and isolation to detect Xa from sugarcane sap was observed. PCR was faster allowing pathogen detection in less than one day. Both methods resulted in low detection frequencies from asymptomatic plants, which may contain Xa below detection limits. The lowest limit for …

Pythium dimorphum Hendrix & Campbell and other Pythium species were determined to be pathogenic fungi that cause the death of longleaf pine seedlings during cold storage. Pathogenicity of these Pythium isolates as determined by infecting newly-germinated slash pine seedlings in vitro was confirmed in field evaluations. An in vitro dual culture screening system was effectively used for selecting Trichoderma isolates capable of killing P. dimorphum. Selected Trichoderma isolates and Gliocladium virens produced volatile and/or non-volatile antibiotics on solid media and in liquid cultures, but antibiotic production could be affected by medium type, pH, and culture age, depending on species or …

Eddy heat flux variability over the Louisiana-Texas shelf was investigated using satellite-derived surface velocity and temperature data between October 1993 and October 1994. Assuming the product of sea water density and specific heat is relatively constant, velocity-temperature covariance reflects eddy heat flux (the fluctuating part of the 1 x 1 degree, 90 day mean heat flux). Available velocity and temperature fields, however, are not synchronous. Temperature "observations" at velocity positions were optimally estimated using the Gauss Markoff Theorem. Extended sleep deprivation has been shown to produce impairments in sustained attention and vigilance, especially if the deprivation period is greater than 48 hours. However little is known about the effects of sleep deprivation on performance of cognitive tasks considered to be measures of higher cortical functioning such as cognitive flexibility and the capacity to shift response set. These two activities are associated with intact functioning of the frontal lobes of the cerebral cortex while attention and vigilance tasks are not considered to be part of this type of cognitive activity and are not associated with frontal lobe function. A series of experiments were designed to determine the cyclooxygenase (COX) isoform responsible for prostaglandin release during ovulation and luteolysis. Initially, a reliable, serum-free culture system was developed to produce large numbers of bovine granulosa cells originally harvested from the small antral follicles of abattoir ovaries. This method allowed adequate numbers of cells to be obtained for the subsequent study involving cyclooxygenase regulation. Lipid peroxidation is the process by which lipids in the cell membrane undergo free radical-initiated oxidation. The resulting cell damage has been implicated in the onset of many diseases, including as cancer, aging, and stroke. Lipid oxidation has been studied in a variety of membrane models, including homogeneous solution, micelles, liposomes, and microsomes. The kinetics for free radical reactions proven to occur during lipid oxidation in homogeneous solution have been thoroughly elucidated. Currently there is no cure for HIV/AIDS. The only way to deter progression of this disease is through regular prophylactic and curative treatment regimens. Thus, it is critical that patients are compliant with their outpatient medical appointments. Despite this, high rates of noncompliance have been reported among patients with HIV/AIDS. In addition, little attention has been directed toward establishing causal factors of this noncompliance. The present study represents one of the first efforts to elucidate psychosocial, illness, and demographic factors associated with compliance with outpatient, HIV-specific treatments. Stress has been related to increased plasma epinephrine (E), norepinephrine (NE), and glucocorticoids. These compounds produce immunomodulatory effects by interacting with hormone receptors on or in various immune cells. This project assessed the effects of stress on equine influenza virus (EIV)-specific and mitogen-induced lymphoproliferation of peripheral blood mononuclear cells (PBMCs) in relation to stress-induced changes in E, NE, and cortisol. In vitro effects of amines on proliferative responses and glucocorticoid receptor (GR) assays were done as a secondary measure of hormone and neurotransmitter interactions with the immune system. This study investigated the steroidogenic enzymes of the elasmobranch interrenal. First, biochemical characteristics of cytochrome P450c21 hydroxylase (P450c21) and 3$\beta$-hydroxysteroid dehydrogenase (3$\beta$-HSD) were determined with incubations of the microsomal fraction of Carcharhinus limbatus (blacktip shark) interrenal tissue. Second, molecular reagents were obtained by isolating and sequencing partial cDNA clones encoding the elasmobranch steroidogenic enzymes cytochrome P450 side chain cleavage (P450scc) and 3$\beta$-HSD from blacktip shark and Daysatis americana (southern stingray) interrenal. An Examination Of The Effects Of The Mother's Education And Household Exposure To Disease On Childhood Diarrheal Morbidity In Sudan., Dawood Hussein Sultan

This dissertation examines the effect of increases in the mother's level of education on the likelihood of diarrheal morbidity among Sudanese children aged 6 to 23 months. Maximum likelihood logistic regression models are used in the analysis. The logit estimates obtained from these models revealed that children of mothers who are illiterate run on average more than 10 times the risk of diarrhea as that run by children to mothers with a senior secondary or higher education. Slight increases in the mother's education, as indicated by an incomplete primary school education, reduce the likelihood of diarrhea by more than 50%. …

This dissertation explores the link between technology, capitalism, and the female body in 20th century cinema. It argues that commodified pleasure, or "engineered enjoyment," is always produced by establishing technological control over the female body. The scope of this dissertation is limited to films produced during the post-World War II era. In Chapter One I define "engineered enjoyment" and show how film is the prototypical example of such. In Chapter Two I examine how the cinematic apparatus works as a pleasure-producing system and explore what happens when that system breaks down, as in the 1947 film Lady in the Lake. …

Corbicula fluminea (Muller) maintains hydromineral balance in dilute media by active transepithelial transport of Na and Cl. Cl uptake depended on external (Cl) and displayed saturable kinetics. The transport capacity in pondwater(PW)-acclimated animals was 7.00 $\pm$ 0.51 $\mu$eq(g dry tissue $\cdot$ h)$\sp{-1}$ and the transport affinity 0.21 $\pm$ 0.08 mM. Prolonged salt depletion increased Cl transport capacity without changing the affinity. In PW-acclimated C. fluminea, Na and Cl transport were independent, and were stimulated by serotonin. In salt depleted (SD)animals, Na transport was partially Cl-dependent but Cl transport was Na-independent. Acetazolamide increased Na and Cl efflux. Both serotonin and acetazolamide …

This psychoanalytic study complicates prevailing notions about William Wordsworth's representations of mothers. Wordsworth does not invariably conflate mothers with Nature or consistently construct women as silent objects of male quest. Rather, he explores a variety of mothers' voices, often associating them with language acquisition and poetic composition. In early work he acknowledges mothers' significance directly and creates more vocal mothers, while in later work and revisions he often conceals mothers' significance and depicts more object-like mothers. In the 1805 version of book two of The Prelude, Wordsworth recalls himself as a "blessed babe" who recognizes his mother as a separate …

Cecropins are basic, amphipathic peptides with antibacterial activity against many Gram-positive and Gram-negative bacteria. By retaining the positively charged, amphipathic characteristic of cecropin B, a cecropin analog S1, in which 54% of amino acid residues were substituted, was synthesized and shown to possess a broad spectrum of antimicrobial activity. To determine expression of the cecropin analog S1 in Sf9 cells, two gene constructs were designed, and inserted into Autographa californica nuclear polyhedrosis virus under the control of the polyhedrin promoter. While brown rice is nutritionally superior to white rice, it is not shelf stable, becoming rancid in a matter of weeks. Preservation by refrigeration or freezing is expensive and not universally available. Therefore, other preservation methods, such as aqueous ethanol extraction of lipids, have been studied to overcome this problem, but these methods also have limitations. Young adults are two to three times more likely to be unemployed than older adults, and African-American and poorly educated youths are disproportionately unemployed (U.S. Bureau of Labor Statistics, 1995a). The goal of the present study was to identify precursors of youth unemployment that could be targets of interventions. This longitudinal study explored the relationship between individual, family, and neighborhood factors, measured during junior high and high school, and the employment status of 19-year-old males. This dissertation explores the figure of the vampire in the nineteenth century as a metaphor of disorder, especially as interpreted through the root metaphor of the second law of thermodynamics, also known as entropy. The theoretical approach used in the reading of several vampire texts is an entropy theory of literature, a mostly aesthetic approach of relevance to a comparatist study on vampirism and entropy. Chapter One considers the female vampire in one of her earliest nineteenth-century incarnations, the seductive Clarimonde of Theophile Gautier's La Morte amoureuse.
