The DNA-bending protein TF1 is the Bacillus subtilis bacteriophage SPO1- encoded homolog of the bacterial HU proteins and the Escherichia coli integration host factor. We recently proposed that TF1, which binds with high affinity (K(d) was ~3 nM) to preferred sites within the hydroxymethyluracil (hmU)-containing phage genome, identifies its binding sites based on sequence-dependent DNA flexibility. Here, we show that two hmU-A base pair steps coinciding with two previously proposed sites of DNA distortion are critical for complex formation. An insertion/deletion (I/D) polymorphism of the angiotensin I converting enzyme gene influences the level of serum angiotensin converting enzyme activity and has been associated with risk of several cardiovascular conditions. The relationship to blood pressure remains uncertain, however. We conducted a population-based survey in Kingston, Jamaica, to examine the association between angiotensin converting enzyme genotype, angiotensin converting enzyme serum activity and blood pressure. Serum angiotensin converting enzyme activity was measured and genotyping performed for the I/D polymorphism in 500 community residents. The overall prevalence of the D allele was 59.3%. The cytochrome (cyt) b subunit of ubihydroquinone: cytochrome c oxidoreductase(bc1 complex) contains four invariant glycine (G) residues proposed to be essential for proper packing of the high and low potential (b(H) and b(L)) hemes of the bc1 complex. One of these residues, G146 located in the transmembrane helix C of cyt b of Rhodobacter capsulatus, was substituted with A and V using site-directed mutagenesis, and the effects of these substitutions on the properties of the ubiquinone oxidation (Q(o)) site and heme b(L) of the bc1 complex were analyzed. For many years, the concept that the heparin-binding sequence is sequestered within vitronectin and exposed upon denaturation of the protein has guided experimental design and interpretation of related structure- function studies on the protein. To evaluate binding of heparin to both native and denatured/renatured vitronectin, methods for monitoring binding in solution have been developed. A fluorescence method based on changes in an extrinsic probe attached to heparin has been used to evaluate heparin binding to native and denatured/renatured vitronectin. This approach indicates that there are not major differences in intrinsic heparin-binding affinities between native and renatured protein and invalidate the …

Wetland Loss In The Northern Gulf Of Mexico: Multiple Working Hypotheses, R. Eugene Turner

I examined four hypotheses about causes for the dramatically high coastal wetland losses (0.86% yr−1) in the northern Gulf of Mexico: an extensive dredged canal and spoil bank network, a decline in sediments in the Mississippi River during the 1950s, Mississippi River navigation and flood protection levees, and salinity changes. Natural factors contributing to these habitat changes include eustatic sea-level rise and geological compaction, which appear to have remained relatively constant this century, although variation does occur. These four hypotheses were tested using data on land-to-water changes in 15-min quadrangle maps inventoried for four intervals between the 1930s and 1990. …

Plasminogen activator inhibitor-1 (PAI-1), the primary inhibitor of tissue-type plasminogen activator and urokinase, is known to convert readily to a latent form by insertion of the reactive center loop into a central β- sheet. Interaction with vitronectin stabilizes PAI-1 and decreases the rate of conversion to the latent form, but conformational effects of vitronectin on the reactive center loop of PAI-1 have not been documented. Mutant forms of PAI-1 were designed with a cysteine substitution at either position P1' or P9 of the reactive center loop. A number of studies have demonstrated that tumor necrosis factor-α (TNF-α) is associated with profound insulin resistance in adipocytes and may also play a critical role in the insulin resistance of obesity and non- insulin-dependent diabetes mellitus. Reports on the mechanism of TNF-α action have been somewhat contradictory. GLUT4 down-regulation has been implicated as a possible cause of insulin resistance as has been the reduced kinase function of the insulin receptor. General circulation models predict that freshwater discharge from the Mississippi River (USA) to the coastal ocean would increase 20 % if atmospheric CO2 concentration doubles. Here we use a coupled physical-biological 2-box model to investigate the potential impacts of increased freshwater and nutrient inputs on the production and decay of organic matter in the coastal waters of the northern Gulf of Mexico. Model results for a doubled CO2 climate indicate that the annual net productivity of the upper water column (NP, 0 to 10 m) is likely to increase by 65 g C m(-2) yr(-1), relative to a 1985-1992 average …

Many species of the ubiquitous pennate diatom genus Pseudo-nitzschia have recently been discovered to produce domoic acid, a potent neurotoxin which causes Amnesic Shellfish Poisoning (ASP). Pseudo-nitzschia spp. were extremely abundant (up to 10(8) cells l(-1); present in 67% of 2195 samples) from 1990 to 1994 on the Louisiana and Texas, USA, continental shelves and moderately abundant (up to 10(5) cells l(-1); present in 18% of 192 samples) over oyster beds in the Terrebonne Bay estuary in Louisiana in 1993 and 1994. Studies on sexual behavior in Africa have shown that the proportion of women with premarital sexual experience is high, but the factors associated with premarital sexual behavior have not been clearly identified. This is partly due to the separation between theory and research. This study attempts to fill this gap by constructing a new conceptual framework in which hypotheses of rational adaptation, social disorganization, and patrilineal bias theories can be empirically tested. Based on a life history approach, the present framework emphasizes the role of three dimensions of family background in shaping individual behavior: financial capital, human capital, and social …

Field studies were conducted over three years to evaluate preplant postemergence herbicides for red rice control in rice under no-till water-seeded culture. Application of paraquat or sulfosate plus thiobencarb and paraquat alone 3 days before seeding (DBS) resulted in the greatest red rice control 11 weeks after seeding (WAS). Rice yield was increased and percent red rice in rice grain decreased when paraquat or sulfosate was applied in combination with thiobencarb compared with the nontreated control. Rice yield was increased when herbicides were applied 3 and 7 DBS as compared with 14 and 28 DBS. Aqueous extracts of cursed buttercup, …

In the literature, self-injurious behavior and aggression are often lumped together collectively as maladaptive behaviors. This study examined the social skills characteristics between persons with self-injurious behavior, aggression and a control group using the Matson Evaluation of Social Skills in Persons with Severe and Profound Retardation (MESSIER). Persons engaging in self-injurious behavior were significantly different from persons engaging in aggression as well as from the control group on five of the six subscales. The self-injury group had lower scores on subscales measuring positive behaviors and higher scores on subscales measuring negative behaviors. As expected for persons engaging in aggression, scores …

In this interdisciplinary study, I apply the materials of family systems theory to the study of five twentieth-century literary texts, each written by a Southern white woman. Arranged in the order they will appear in this study, the five texts are The Keepers of the House (1964) by Shirley Ann Grau; Net of Jewels (1992) by Ellen Gilchrist; The Golden Apples (1949) by Eudora Welty; The Voice at the Back Door (1956) by Elizabeth Spencer; and Can't Quit You, Baby (1988) by Ellen Douglas. Beef tenderness and color stability were evaluated in conventional and modified atmosphere packaging (MAP) systems. Equal portions of inside rounds removed immediately (PM0) or at 48 h postmortem (PM48) were vacuum packaged (VP) whole or cut into steaks and packaged in 80%N$\sb2$:20%CO$\sb2$ MAP. On day 12, MAP environments were exchanged for 80%O$\sb2$:20%CO$\sb2$. Calpastatin activity was lower at 2 d postmortem for PM48 steaks than PM0 steaks. Electrophoresis indicated more degradation of 50 and $>$250 kDA compounds at 2 d postmortem for PM48 than PM0 steaks. Steaks from PM48 required less shear force than PM0 steaks. Progeny testing is practiced to select the best crosses (families) prior to individual plant selection and to guide decisions relative to hybridizing and seedling planting in many crops including sugarcane (Saccharum sp. hyb.). Research to optimize the progeny testing methodology examined its effectiveness relative to family, within family, environment, replication, and intrarow plant spacing. Variance-component analysis indicated that family by environment (locations, years) interaction was a minor inhibitor of selection effectiveness. Within-family variance was the largest source of variation for the five traits considered. For stalk weight, stalk length, and stalk diameter, a small majority of this variance was due …

Chlamydomonas reinhardtii, a unicellular green alga, grows photoautotrophically at very low concentrations of inorganic carbon due to the presence of an inducible CO2-concentrating mechanism. During the induction of the CO2-concentrating mechanism at low-CO2 growth conditions, at least five polypeptides that are either absent or present in low amounts in cells grown on high-CO2 concentrations are induced. One of these induced polypeptides with a molecular mass of 36 kD, LIP-36, has been localized to the chloroplast envelope. Peg3 (paternally expressed gene 3) is the first imprinted gene detected in the proximal region of mouse chromosome 7. Because imprinting is a trait that is generally conserved among mammals, and imprinted domains generally encompass several adjacent genes, expression patterns and chromosomal environment of the human counterpart of Peg3 are of special interest. In this study we have localized human PEG3 ~ 2 Mb proximal of the telomere of chromosome 19q, within a region known to carry large numbers of tandemly clustered Kruppel-type zinc finger-containing (ZNF) genes. Boundary elements are thought to define the peripheries of chromatin domains and to restrict enhancer-promoter interactions to their target genes within their domains. We previously characterized a cDNA encoding the BEAF- 32A protein (32A), which hinds with high affinity to the scs' boundary element from the Drosophila melanogaster 87A7 hsp70 locus. Here, we report a second protein, BEAF-32B, that differs from 32A only in its amino terminus. Unlike 32A, it has the same DNA binding specificity as the complete BEAF activity affinity purified from Drosophila. We characterize three domains in these proteins. Regional, Holocene records hold particular relevance for understanding the reciprocal nature of global environmental change and one of its major human dimensions: "sustainable agriculture", i.e., food production strategies which entail fewer causes of and are less susceptible to environmental change. In an epoch of accelerating anthropogenic transformation, those records reveal the protracted regional causes and consequences of change (often agricultural) in the global system as well as informing models of prehistoric, intensive agriculture which, because of long tenures and high productivities, suggest strategies for sustainable agricultural in the present. We characterized the glycoprotein K (gK)-null herpes simplex virus type 1 [HSV-1] (KOS) ΔgK and compared it to the gK-null virus HSV-1 F-gKβ (L. Hutchinson et al., J. Virol. 69:5401-5413, 1995). Δgk and F-gKβ mutant viruses produced small plaques on Vero cel monolayers at 48 h postinfection. F-gKβ caused extensive fusion of 143TK cells that was sensitive to melittin, a specific inhibitor of gK-induced cell fusion, while ΔgK virus did not fuse 143TK cells. A recombinant plasmid containing the truncated gK gene specified by F-gKβ failed to rescue the ICP27-null virus KOS (d27-1), while a plasmid with the ΔgK deletion …

Development Of A Protein-Free Chemically Defined Culture Medium For The Propagation Of The Oyster Pathogen Perkinsus Marinus, J. F. La Peyre, M. Faisal

In the present study we describe a protein-free, chemically defined culture medium, designated JL-ODRP-3, which supports the propagation of Perkinsus marinus, a parasite of the eastern oyster, Crassostrea virginica. P. marinus adapted rapidly to the defined medium and the growth rate of the protozoan increased significantly following a few subcultures. Two isolates of P. marinus, one from the Chesapeake Bay (Virginia) and the other from the Gulf of Mexico (Texas) were cultured for at least ten passes. The doubling times for the isolates from Virginia and Texas, in log phase, were 18 ± 1.2 and 28.6 ± 3.2 hours respectively, …
