Studies with the transmission electron microscope were used to detect and attempt to identify viruses infecting sweetpotato (Ipomoea batatas ) and other Ipomoea species. Flexuous-rods, short curved-rods, and spherical virus-like particles were observed in cells of symptomatic plants. Also, various cytopathic changes such as crystals, vesicles, fibril structures, and cylindrical inclusions were observed. Some of these cytopathic changes were associated with specific viral groups such as genus Crinivirus and Potyvirus and therefore helpful in diagnosis. Some molecular and biological properties of an isolate of Sweetpotato chlorotic stunt virus from the sweetpotato cultivar White Bunch (SPCSV-WB) were determined. Two species of …

Soybean [Glycine max (L.) Merr] is one of the most important oil crops. Genetic transformation techniques can provide new tools for soybean improvement. The production of transgenic soybean has been limited. This research integrated a modified soybean regeneration system into a transformation protocol to optimize the production of transgenic soybean. Soybean hypocotyls and cotyledonary nodes were cultured on Gamborg B5 medium containing thidiazuron (TDZ). TDZ at 0.16 muM was found superior to other concentrations for inducing multiple shoot formation. Explants isolated from seedlings germinated on medium containing 6-benzyl-aminopurine (BAP) greater than 7 muM produced more multiple shoots than when on …

Bioremediation and phytoremediation are in their infancy and this research addresses concerns for application of these technologies. This laboratory and mathematical modeling effort predicts the pollutant flux from soils and sediments to the overlying waters. Little work has been done on transport in surface waters such as the slow moving and stagnant areas of the southern U.S. Mass transfer is a proposed rate limiting factor for biological transformation when using phytoremediation or natural attenuation. Two types of wetland plants, a woody species, black willow ( Salix nigra), and a non-woody, the macrophyte three-square bulrush (Scirpus olneyi), were used in laboratory studies to evaluate relationships between sorption, plant uptake, translocation and transpiration of phenanthrene, a model polycyclic aromatic hydrocarbon and chlorobenzene, a model chlorinated benzene from the reversible and desorption-resistant compartment of a sediment for Phenanthrene (PHEN) and a wetland soil for chlorobenzene (CB). The activities of non-ribosomal nucleolar proteins are now understood to be important for the normal functions of both nucleoli and C&barbelow;ajal B&barbelow;odies (CBs). Although these proteins have been studied extensively in other eukaryotes, knowledge of non-ribosomal nucleolar proteins in Drosophila melanogaster lags far behind. The n&barbelow;ucleo&barbelow;lar p&barbelow;hosphop&barbelow;rotein of 140 kDa (Nopp140) may function to shuttle box C/D and box H/ACA s&barbelow;mall n&barbelow;ucleo&barbelow;lar (sno)RNAs from the nucleus to the nucleolus, where they function in the 2 '-O-methylation and pseudouridylation of rRNA, respectively. Genetic evidence indicating that Toll-like receptor 4 (TLR4) is the lipopolysaccharide (LPS) receptor in mice was reported. However, biochemical evidence that murine TLR4 confers LPS responsiveness has not been convincingly demonstrated. Inducible cyclooxygenase (COX-2) is selectively expressed in LPS-stimulated macrophages in part mediated through activation of NFkappaB. Thus, we determined whether murine TLR4 confers LPS responsiveness as evaluated by activation of NFkappaB and COX-2 expression. Expression of the constitutively active form (DeltaTlr4) of TLR4 in murine macrophage cell line (RAW 264.7) is sufficient to activate NFkappaB and COX-2 expression and to stimulate prostaglandin E2 synthesis. Urban storm water runoff mobilizes and transports significant loads of heavy metals. Promulgation of NPDES Phase II regulations has spurred development of Best Management Practices (BMPs) for in-situ control of heavy metals. In this study, Amphoteric materials such as manganese oxides and iron oxides on the various substrates including silica sands, polymeric beads and cementitious particles were investigated. Surface characterization results indicated that oxide coatings ranged from 20 to 200-mum in depth, and surface areas were significantly increased by greater than a factor of one thousand compared to uncoated media. Contaminant bioavailability depends on physicochemical processes such as adsorption/desorption, diffusion, and dissolution. The rates of sorption and desorption of trichloroethylene (TCE) and 1,3-Dichlororbenzene (1,3-DCB) were studied. Desorption kinetics of 3-month and 5-month old contaminated soils showed that progressively less amount of contaminant was available for facile desorption compared to freshly contaminated soil in an empirical non-linear model. Microcosm batch studies were conducted to study the biodegradation of 1,3-DCB in the aqueous and soil phase containing a freshly contaminated soil and a soil containing only the desorption resistant fraction. One of the cultural changes noted in American society in the last fifty years has been a noticeable increase in the public use of sexual profanity, particularly by women. Many commentators attribute this change to the sexual revolution of the nineteen-sixties and seventies, as well as the increasing emancipation of women from traditional gender roles. This dissertation examines the ideological foundations that have shaped both western sexual attitudes and the nature of modern use of sexual profanity, to question whether these changes are indicative of greater gender equity. This dissertation explores Nietzsche's political theory through an analysis of his major work, Thus Spoke Zarathustra. It is demonstrated that the narrative structure of Zarathustra follows from a politically-grounded critique of collective human existence. Nietzsche critiques the state and mass society as inherently repressive. He critiques the concept of the community and friendship as producing ressentiment and bad conscience. One of the central metaphors in Zarathustra proves to be that of nausea, arising from contempt for the mediocre masses of humanity and the realization of the chaotic and contingent nature of existence. Using the Pathfinder computer algorithm comparisons were made of participants' knowledge organization of sexual, sexually aggressive, and aggressive concepts. Three hundred and fifty-two heterosexual males (81 admitting a past history and future likelihood of sexual aggressive/coercive behavior; 111 admitting a past history of engaging in sexual aggression; 88 admitting no past history of or future likelihood of engaging in sexual aggression) rated all possible pairwise combinations of 18 words relevant to sex, aggression, sexual aggression, and emotion. The incidence of iguana-associated salmonellosis reported in humans during the 1990's is a public health concern. This research is the first attempt to describe the epidemiology of Salmonella in the green iguana. Seasonal prevalence of Salmonella in green iguanas and their environment were estimated at a commercial iguana farm in El Salvador. The prevalence of Salmonella was lower in adult iguanas (44%) than in yearlings (50%) (Odds ratio (OR): 0.77, 95% Confidence Intervals (CI): 0.6--0.96) and higher in male iguanas (50%) than females (41%) (OR: 1.5, 95% CI: 1.2--1.8). This is a series of 3 experiments in the area of vasoreactivity. The first study investigated the stability and reproducibility of brachial artery flow-mediated dilation (BAFMD). Twenty-six healthy volunteers underwent 3 scans on 2 days, performed by 2 ultrasonographers, and analyzed by 2 readers. All subjects were tested between 7 and 11am after refraining from food and exercise. Average baseline diameter was 3.48±0.53mm, which increased to 3.71±0.57mm (6.58±4.15%) at peak dilation. ICCC's for days, testers and readers were 0.9188, 0.9394, and 0.8982 respectively. To detect a difference in vasoreactivity of 60% (two-tailed), e.g. 5% vasodilation versus 8% vasodilation, at 90% …

An increasingly urgent task in the field of conservation biology is to identify changes in abiotic and biotic interactions that result when large areas of forest are converted to small fragments surrounded by anthropogenic landscapes. My research, conducted in lowland dipterocarp rain forest at the 2,500-ha Pasoh Forest Reserve in Peninsular Malaysia, investigated a novel but strongly negative edge effect - namely, a tremendous increase in the density of wild pigs (Sus scrofa) and the resulting deleterious impacts on the understory plant community. Polycyclic aromatic amines (arylamines) are a class of chemical carcinogens that are prevalent in environmental and industrial settings. In order to study the mechanism of their toxicity a quantitative and qualitative detection method was developed to measure the C8-adenine adducts of benzidine and 2-aminofluorene in DNA samples. To do this a novel synthetic method using a palladium catalyst was developed to prepare authentic and deuterated arylamine adducts to serve as standards. These standards were then used to develop a high performance liquid chromatography, electrospray, tandem mass spectrometry, isotope dilution, detection method. Meiofaunal colonization of artificial substrates was investigated in a shallow estuarine embayment. Meiofauna colonized various artificial substrates rapidly, at a rate of 1000 individuals collector-1 day -1 regardless of season, suggesting that high abundances of meiofauna commonly exist in the water column. These water-column meiofauna may be important in fish diets and may serve as a dispersing pool that aids recolonization after disturbance events. Colonization of artificial substrates by transient meiofauna of unknown origin(s) occurred through the water column. In particular, I examine how the clothing company United Colors of Benetton attempted to position itself within the AIDS crisis. Field studies were conducted to characterize flow fields and their effect on larval transport and retention within Barataria Bay, Louisiana. Drifter tracking and passive larval sampling was conducted monthly during the spring and summer months from 1994 through 1996. Fifteen drifter tracking sessions were conducted over full tidal cycles (diurnal). Drifter positions were determined approximately every thirty minutes using a GPS receiver. A spline interpolation was used to estimate drifter positions at synchronous five-minute intervals. Ultra-thin films of Au are formed on glass by thermal evaporation of Au onto a silane-modified glass surface. Monolayers of 3-aminopropyltriethoxysilane on glass (APS/glass) are outstanding surfaces for the formation of adherent, electrically conductive, smooth, and highly oriented Au films. Scanning tunneling microscopy images indicate that ultra-thin Au films are highly conductive and have a significant {111} texture, and X-ray diffraction studies confirm the latter. In addition, Au films on APS/glass surface are not removed by tape tests, pointing to a strong chemical interaction between the Au and amine surface. The general purpose of this study is to relate the fields of geography and religion at both the academic and the practical levels. Primarily, it investigates the New Testament description of the land and its human occupancy. Secondarily, it identifies New Testament attitudes toward geography and suggests implications for its study and practice. It describes the geography, both physical and human, for each of the five classes of the document's book in a separate chapter. This study examines the contrasting themes of land degradation and environmental stewardship of farming enterprises in the regional context of the Upper Pearl River Basin of Mississippi. Both the previous imagery and analysis of Southern Cotton Belt agriculture emphasize the profligate nature of farming practices. The contribution this study makes is to amend both the stock images and macro-analyses of Southern cotton culture. Analysis at the sub-regional level demonstrates that many farmers actually practiced sound land management techniques. The super-fast dynamics of solutes in polar solvents and at solid-liquid interface is investigated by femtosecond time-resolved Raman scattering (RRS) and computer simulation of molecular dynamics (MD). Femtosecond RRS is used to investigate four bipyridinium radicals in aqueous solution: methylviologen monocation MV+, benzylviologen monocation, 4,4'-bipyridinium-N,N'-di(propylsulfonate) monoanion and N,N'-ethylene-2,2' -bipyridinium monocation. Time-resolution of the dynamics of the four radicals is carried out by a pump and probe technique using the time-dependent transient intensity of Stokes and anti-Stokes RRS of C-C stretching mode. A field study focused on the effectiveness of ammoniated bagasse (ABG) to enhance the bioremediation of the contaminated wetland. This work demonstrates the use of pyrrole-terminated dendrimers (PTD)s for the oxidation-state-dependant encapsulation and release of various guest molecules. In order to achieve the goals of this research, a two-step approach was taken. The first portion of this doctoral work involved synthesis of low-generation (small number of peripheral groups) PTDs having a simple structure (DAB-Py x, X = 4, 8, 16) and studying the intramolecular oligomerization of the terminal pyrrole groups by well-known electrochemical methods. Sheath blight caused by fungal pathogen Rhizoctonia solani can cause significant yield loss in rice, especially in warm, high rainfall environments. Integrated pest management (IPM) is the key to sheath blight control. This system utilizes disease resistance, pesticides, cultural practices and biological control to control multiple pests. Silicon fertilization is an additional cultural practice that can reduce sheath blight severity and increase grain yield. Terminos Lagoon in the southern Gulf of Mexico has 5 ecological subsystems: (1) fluvial lagoon systems (FLS) with permanent river discharge, low salinity, high turbidity and nutrients. (2) Central basin (CB) between marine and river conditions, intermediate salinity and variable transparency. (3) Inner littoral of Carmen Island (ILCI) with strong marine influence, high transparency and extensive seagrass meadows. The knee joint is the most complex joint in the human body. A complete understanding of the physical behavior of the joint is essential for the prevention of injury and efficient treatment of infirmities of the knee. A kinematic model of the human knee including bone surfaces and four major ligaments was studied using techniques pioneered in robotic workspace analysis.
