Production Of Transgenic Goats By Somatic Cell Nuclear Transfer, Brett C. Reggio

A series of experiments were conducted to produce transgenic goats by somatic cell nuclear transfer (NT). In all experiments, donor cells were electrically fused to enucleated metaphase II oocytes, then chemically activated. In a preliminary study to evaluate embryonic development following NT with proliferating or quiescent fibroblast or cumulus cells, significantly more embryos reconstructed with quiescent cumulus cells fused (77%) compared with proliferating cumulus cells (41%), proliferating fibroblasts (36%) or quiescent fibroblasts (37%). Improved development to the eight- to sixteen-cell stage was observed when fibroblast cells were serum starved (serum starved 39% vs. serum fed 15%). However, there was no …

Since the 1950s, car following phenomena have been studied and analyzed, resulting in various models and algorithms. In general, the car following process has been defined as a stimulus-response relationship in which the driver of the following vehicle reacts to the actions of the lead vehicle after a time lag. The visual determination of saccharides has been of interest for well over a century. Sugars are a relatively challenging class of compounds to analyze. They exhibit great structural similarity as well as transparency in the visible region. The sensing of specific saccharides could aid the monitoring of disease or industrial fermentation products. Restless Legs Syndrome (RLS) is a disorder characterized by disagreeable leg sensations, usually prior to sleep onset, which cause an almost irresistible urge to move the legs. A characteristic feature of this disorder is that the movements are partially or completely relieved with leg motions. Attempts to find the underlying pathology have been unsuccessful. Thus, there are no objective physiological tests to diagnose this condition. This research develops a comprehensive framework for interactive walkthrough involving one billion particles in an immersive virtual environment to enable interrogative visualization of large atomistic simulation data. As a mixture of scientific and engineering approaches, the framework is based on four key techniques: adaptive data compression based on space-filling curves, octree-based visibility and occlusion culling, predictive caching based on machine learning, and scalable data reduction based on parallel and distributed processing. Short peptides containing Cα,α-dipropylglycine (Dpg) at alternating sequence positions were synthesized and examined for conformational behavior. Peptide assembly was performed using Fmoc-solid-phase chemistry where the coupling with PyAOP could be significantly enhanced at elevated temperature. Circular dichroism (CD) and NMR conformational studies revealed that incorporation of Dpg residues induced folded structures into peptides. It was observed that Dpg residues adopted helical conformation in a helix-promoting sequence. The resulting helical structure was comprised of consecutive β-turns whose structure was stabilized by salt bridge in aqueous solution. Using a theoretical language grounded in post-structuralism and cultural studies, this examination hopes to demonstrate the primacy of performance and theatre in the formation of culture, Irish-American specifically, or otherwise. I examined patterns and timing of ontogeny and relative growth in five species of blenny (Teleostei: Blenniidae) from the northern Gulf of Mexico by assigning a suite of discrete character state scores to ontogenetic events (10 external traits; 218 total specimens). This is the first study to evaluate developmental patterns in reef-associated fishes relative to the timing of metamorphosis and settlement by applying scaling techniques and statistical methods to quantify, differentiate, and select criteria for defining intervals of development across taxa. The goal of this dissertation research is to investigate the defense systems of strawberry plant by characterizing the genes and their products involved in plant disease-resistance. Pathogenesis-related proteins, including hydrolytic enzymes chitinases and â-1,3- glucanases, have been known to be induced in plants upon infection with various pathogens. Highest total chitinase activity was found in strawberry crown, whereas root, petiole, fruit, leaf, and runner showed successively lower activities. Chitinase isoform analysis showed that up to six acidic and two basic isoforms were present in various organs. The total chitinase activity was stable at 50oC. A fluorescent binding assay was developed to investigate the effects of site-directed mutagenesis on the binding affinity and binding specificity of the chitin-binding domain of chitinase A1 from Bacillus circulans WL-12. The chitin-binding domain (ChBD) was genetically fused to the N-terminus of the green fluorescent protein, GFP. The polyhistidine-tagged hybrid protein was expressed in Escherichia coli under the dose-dependent regulation of the araBAD promoter and purified using metal affinity-, chitin- or ion-exchange chromatography. The purpose of this study was to investigate the influence of perception of social support, privacy preference and level of depression on marital adjustment for couples presenting for infertility treatment. Data was obtained from three infertility clinics in Louisiana from January 1, 2001 through October 1, 2001. Forty-seven couples presenting for infertility treatment completed four questionnaires measuring marital adjustment, perception of social support, privacy preference and level of depression. Demographic and descriptive information was gathered from an information sheet completed by the subjects. Wetland groundwater hydrology was investigated at different spatial scales to assess the usefulness of the information to coastal managers. Specific objectives were to: (1) review studies related to coastal groundwater discharge, evaluating techniques and identifying controls; (2) understand regional trends in groundwater flow along the U.S. East and Gulf coasts; (3) evaluate the applicability of naturally occurring radioisotopes as indicators of groundwater in a shallow deltaic system; and (4) evaluate groundwater-surface water exchange within Barataria Basin, Louisiana. Ehrlichia canis is the etiologic agent of “tropical canine pancytopenia”, or canine ehrlichiosis. The impetus for this research was to overcome the lack of any reliable means of elucidating the genetic profiles of these illusive and historically difficult to manipulate organisms. The use of a broad-host range plasmid greatly facilitated the determination of an electro-transformation protocol. The transforming plasmid possesses a chloramphenicol antibiotic resistance gene marker (chloramphenicol acetyltransferase [CAT] gene), and a visual reporter gene marker, the Green Fluorescent Protein (GFP) gene. Adsorption-desorption and transport behavior of pesticides are important processes that influence the amount retained by the soil and that which is susceptible to runoff or movement within the soil profile. Kinetic batch experiments were used to study the adsorption-desorption behavior of metolachlor (a non-ionic herbicide) and deltamethrin (a pyrethroid insectcide) in soils, and atrazine (a triazine) in sugarcane mulch residue. A series of spatially based models of Hawaiian streams and stream fish habitats were developed to aid in the conservation of native fishes. The spatial models focused on the quantification of habitat for native fishes at three levels within a spatial hierarchy. First, at the reach level, the models classified the major geomorphological features of a stream and its watershed to determine the distribution of fish habitat within the different sections of the stream. Predator-prey interactions are an important factor in energy transfer through food webs. To estimate effects of altered energy flow caused by costs and benefits of antipredator defenses and to study their implications for community stability, I used the planktivorous fish - Chaoborus - Daphnia system. In the laboratory, I found that clonal variance of Daphnia antipredator defenses was high, and that behavioral, morphological, and life-history defenses were uncoupled. Structural-time series models have not gained much ground in commodity market modeling despite the overwhelming popularity of time series approaches in forecasting and dynamic analyses. This dissertation contributes by applying developments in seasonal cointegration and structural-time series analysis (e.g., Zellner and Palm (1974); Hsiao (1997); Lee (1992); Franses and Kunst (1999); Ghysels and Osborn, 2001) to the study of agricultural commodity markets. This study examined several aspects of canopy structure and their influence on growth of loblolly pine (Pinus taeda L.) plantations. Foliage distribution, crown hydraulic architecture, and the effect of chronological changes in canopy structure on stand development were explored as possible components of a future process-based model intended for management purposes. A model based on the Johnson’s SB distribution was developed to predict leaf area distribution of loblolly pine trees. Unresolved phylogenetic problems in pyrenomycetes and population structure of an asexual plant pathogen (Discula destructiva) were investigated. Analyses based on DNA sequences of nuclear encoded small and large subunit RNA genes (SSU and LSU nrDNA) and deduced amino acid sequences of a subunit of RNA polymerase II gene (RPB2) indicated that five genera previously considered in Ceratostomataceae of Sordariales were related to Hypocreales. Melanospora (including the type) and a genus of Ceratostomataceae formed a basal clade, but monophyly of the five genera and of the genus Melanospora was rejected. There is an emerging market for functional oligosaccharides for use in foods. Currently, technology for the production of oligosaccharides is limited to extraction from plant sources, acid or enzymatic hydrolysis of polysaccharides or synthesis by transglycosylation reactions. Oligosaccharides also can be produced using a Leuconostoc fermentation and restricting the polymer size by the addition of maltose. Maltose limits the dextransucrase reaction, yielding high concentrations of a-glucooligosaccharides. Branched oligomers produced by this process were found to be readily catabolized by bifidobacteria and lactobacillus but were not readily utilized by either Salmonella sp., or E. coli, pointing toward their use in intestinal …

Unlike most prairie nesting ducks (Anas spp.), the North American population of northern pintails (A. acuta, hereafter pintails) has failed to respond as expected to improved wetland conditions on the U.S. and Canadian prairies during the mid to late 1990s. My primary objectives were to test the “ecological trap hypothesis” on a landscape level by examining pintail nest site selection and nest success in a highly agricultural environment in southern Saskatchewan. This dissertation reconceives curriculum through an historical approach that employs Ludwig Wittgenstein’s later philosophy. Curriculum is more than the knowledge taught in school. Curriculum, as I a theorist conceives it, is concerned with the broader intellectual and ideological ways a society thinks about education. Hence, the current school curriculum’s focus on specific learning outcomes offers a limited view of the knowledge fashioned by a society, thereby offering an intellectual and social history that is highly selective. This dissertation addresses the problem of the denigration of the voice in poststructural theory and contemporary performance criticism. The problem has antecedents in twentieth-century language philosophy. Saussure defines language as a compendium of arbitrary words recognized according to the degrees of phonetic difference between them. Since for Saussure the arbitrary words of language also designate arbitrary concepts, he concludes that the sounds of words cannot be thought constituent of their sense. After Saussure, structuralism dislodges the voice from its privileged position in the phonologic discourses of Western thought. Olfactory signal transduction is initiated by binding of odorous ligands to G protein-coupled odorant receptors, resulting in the activation of G protein and second messengers that culminates in the generation of action potentials to relay this signal to the brain for odor recognition. This signaling cascade comprises the primary olfactory response. Odorant receptors comprise the largest multigene family identified to date, enabling the olfactory system to respond to an enormous pool of potential odorant molecules. Evolutionary perspectives on human mating have provided testable hypotheses regarding what qualities people desire in their mates and why they want them. One study was conducted to replicate previous findings in mate preference using a more sophisticated paired comparison methodology to develop scales. Paired comparison scaling generally replicated gender differences in mate preferences consistent with evolutionary predictions. Further, decision-making reaction time (ms) suggested the presence of an underlying psychological continuum of selection criteria. Thiobencarb (TB), widely used for the control of broadleaf weeds, grasses, and sedges in rice fields, is considered safe for rice plants when used at recommended rates. TB's reductive dechlorination product, dechlorinated thiobencarb (DTB), is highly toxic to rice. TB is naturally transformed into DTB in field soils in certain areas in Japan and the United States. Knotgrass (Paspalum distichum L.) and torpedograss (Panicum repens L.) are perennial grasses that are extremely difficult to remove in established turfgrass. Studies were conducted to determine the potential for controlling these troublesome perennial weeds. Pot studies were conducted for two years to evaluate several herbicides with multiple modes of action for knotgrass control. In graminicides studies, 8 weeks after initial application (WAI) single or split applications of fluazifop or quizalofop provided excellent control (88 to 95%). Nucleolin is a nucleolar protein important for ribosome biogenesis. Nucleolin contains a conserved glycine arginine rich (GAR) domain near its carboxy terminus.
