Increasing populations and temperatures are expected to escalate food demands beyond production capacities, and the development of maize lines with better performance under heat stress is desirable. Here, we report that constitutive ectopic expression of a heterologous glutaredoxin S17 from Arabidopsis thaliana (AtGRXS17) can provide thermotolerance in maize through enhanced chaperone activity and modulation of heat stress-associated gene expression. The thermotolerant maize lines had increased protection against protein damage and yielded a sixfold increase in grain production in comparison to the non-transgenic counterparts under heat stress field conditions. The maize lines also displayed thermotolerance in the reproductive stages, resulting in …

The southern green stink bug (SGSB) Nezara viridula (L.) (Heteroptera: Pentatomidae), has become a significant pest of soybean and cotton in southern Central America and in the lower mid-southern U.S. A laboratory colony of SGSB was used to evaluate the effect of two isolates of Beauveria bassiana (Balsamo-Crivelli) Vuillemin (Hypocreales: Cordycipitaceae) including the commercial strain GHA and the Mississippi Delta native NI8 strain on the fecundity of this pest. Water control, Tween-80, and four concentrations of each strain (n × 104, 105, 106, and 107) were evaluated. Both native and commercial isolates with the highest concentrations were susceptible to the …

Resource selection in sexually dimorphic ungulates is at least partially explained by sex-specific resource requirements and risk aversion strategies. Females generally spend more time in areas with less risk and abundant, high-quality forage due to their smaller body size. However, demographically variable responses to risk are context dependent, and few have concurrently quantified male and female behavior within areas with the same resource base. We captured 111 (54 males, 57 females) adult white-tailed deer (Odocoileus virginianus) from 2009 to 2018 on a site in South Carolina, USA, where hunters were the primary source of adult mortality. We fit each deer …

Importance: It is unclear how effective intermittent fasting is for losing weight and body fat, and the effects may depend on the timing of the eating window. This randomized trial compared time-restricted eating (TRE) with eating over a period of 12 or more hours while matching weight-loss counseling across groups. Objective: To determine whether practicing TRE by eating early in the day (eTRE) is more effective for weight loss, fat loss, and cardiometabolic health than eating over a period of 12 or more hours. Design, Setting, and Participants: The study was a 14-week, parallel-arm, randomized clinical trial conducted between August …

The Fertilizer Recommendation Support Tool (FRST) will perform correlations between soil nutrient concentrations and crop response to fertilization from user-selected datasets in the FRST national database. Yield response for the nutrient of interest in a particular site-year is presented as relative yield (RY), a ratio of unfertilized yield to the maximum attainable yield (A). Several methods exist in the literature for estimating A and calculating RY but the effect of method choice on soil test correlation outcomes is undocumented. We used six published methods to calculate RY from site-year yield data for five published correlation datasets, and fit a generalized …

Objective: The 10-year intensive lifestyle intervention (ILI) of the Look AHEAD study left a legacy of relative deficits in cognitive function among participants who entered the clinical trial with obesity or a history of cardiovascular disease. We hypothesized that altered levels of two weight-sensitive proangiogenic cytokines, leptin and vascular endothelial growth factor (VEGF), accounted for this concerning finding. Methods: Serum leptin and VEGF concentrations were determined in 1,279 Look AHEAD participants at baseline, proximal to cessation of the interventions (Epoch 1), and an average of 4 years later (Epoch 2). Up to four standardized assessments of attention, executive function, and …

Pro-opiomelanocortin (POMC) neurons are important for the regulation of body weight and glucose balance. The inhibitory tone to POMC neurons is mediated primarily by the GABA receptors. However, the detailed mechanisms and functions of GABA receptors are not well understood. The α5 subunit of GABAA receptor, Gabra5, is reported to regulate feeding, and we found that Gabra5 is highly expressed in POMC neurons. To explore the function of Gabra5 in POMC neurons, we knocked down Gabra5 specifically from mature hypothalamic POMC neurons using the clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 strategy. This POMC-specific knock-down of Gabra5 did not affect …

Food Insecurity Between Two Southern Louisiana Universities: Prevalence And Qualitative Analysis Of Stakeholders' Attitudes, Elizabeth Moore
LSU Doctoral Dissertations
Emerging research has demonstrated high food insecurity rates among college students in the United States (US). The goal of this dissertation is to expand upon the literature evaluating food insecurity on college campuses to improve relief efforts. A literature review and mixed methods were used to measure and compare food insecurity prevalence rates and institutional perceptions of this issue among two Louisiana colleges, Louisiana State University (LSU) and McNeese State University (MSU).
Students who were more likely to be food insecure were from low income households, nonwhite, first generation college students, and/or had past reliance on federal assistance. Food insecure …

Lower ambient temperature (Ta) requires greater energy expenditure to sustain body temperature. However, effects of Ta on human energetics may be buffered by environmental modification and behavioral compensation. We used the IAEA DLW database for adults in the USA (n = 3213) to determine the effect of Ta (−10 to +30°C) on TEE, basal (BEE) and activity energy expenditure (AEE) and physical activity level (PAL). There were no significant relationships (p > 0.05) between maximum, minimum and average Ta and TEE, BEE, AEE and PAL. After adjustment for fat-free mass, fat mass and age, statistically significant (p < 0.01) relationships between TEE, BEE and Ta emerged in females but the effect sizes were not biologically meaningful. Temperatures inside buildings are regulated at 18–25°C independent of latitude. Hence, adults in the US modify their environments to keep TEE constant across a wide range of external ambient temperatures.

Elucidating Factors Affecting Cercosporin Production In Cercospora Leaf Blight Of Soybean To Develop Better Tools For Disease Management, Maria Izabel Costa De Novaes
LSU Doctoral Dissertations
Fungal toxins are pathogenicity/virulence factors for many plant diseases, including Cercospora leaf blight (CLB) of soybean (Glycine max (L.) Merr.). The pathogen, Cercospora cf. flagellaris, produces a perylenequinone toxin, cercosporin, which is activated by light to produce damaging reactive oxygen species. To identify novel solutions for CLB management, we studied factors that may impact the biology of cercoporin: fungal lipid droplets (LDs) and mineral and trace element composition (ionome) of the host. Furthermore, since artificial inoculation with the pathogen has been challenging, we also explored the usefulness of a previously developed cercosporin-based assay to study disease resistance. LDs …
The Association Of Cuzd1 And Stat5 In Adipocytes, Zachary Wigger
The Association Of Cuzd1 And Stat5 In Adipocytes, Zachary Wigger
LSU Master's Theses
Obesity is a metabolic disorder that has turned into a global epidemic over the past few decades. Adipocyte dysfunction brought on by obesity can lead to increased risk of metabolic diseases such as cardiovascular disease and type 2 diabetes mellitus (T2DM). Knowing how adipocytes function to regulate metabolic homeostasis is critical. CUZD1 (CUB and Zona Pellucida-like Domain-containing protein 1) has not been studied in the context of adipocyte biology but could play a role in several metabolic processes. CUZD1 has been suggested to be a biomarker for several inflammatory diseases and cancers but little functional data has been published. There …

Marketing influences consumers’ dietary purchases. However, little is known about marketing environments in Supplemental Nutrition Assistance Program (SNAP)-authorised stores. The present study explored SNAP-authorised store marketing environments in Louisiana by rurality, store ownership and store type (n 42). Sampling methods were designed to include randomly selected stores in each geographic area of the state. The GroPromo was used to measure placement, promotion, and child-focused aspects of marketing strategies used for healthier (fruits and vegetables) and less healthy products (chips, candy, sugar-sweetened beverages, child-focused cereal) in medium- and high-prominence marketing areas. In using multivariate analysis of variance (MANOVA) (P < 0⋅05) for data analysis, variations in GroPromo scores were found among SNAP-authorised stores by rurality (P < 0⋅05) and store ownership (P < 0⋅001); no differences were found by store type (P > 0⋅05). Type 1 diabetes (T1D) is classified as an autoimmune disease where pancreatic b-cells are specifically targeted by cells of the immune system. The molecular mechanisms underlying this process are not completely understood. Herein, we identified that the Icam1 gene and ICAM-1 protein were selectively elevated in female NOD mice relative to male mice, fitting with the sexual dimorphism of diabetes onset in this key mouse model of T1D. In addition, ICAM-1 abundance was greater in hyperglycemic female NOD mice than in age-matched normoglycemic female NOD mice. Moreover, we discovered that the Icam1 gene was rapidly upregulated in response to IL-1β …

Male military personnel conducting strenuous operations experience reduced testosterone concentrations, muscle mass, and physical performance. Pharmacological restoration of normal testosterone concentrations may attenuate performance decrements by mitigating muscle mass loss. Previously, administering testosterone enanthate (200 mg/wk) during 28 days of energy deficit prompted supraphysiological testosterone concentrations and lean mass gain without preventing isokinetic/isometric deterioration. Whether administering a practical dose of testosterone protects muscle and performance during strenuous operations is undetermined. The objective of this study was to test the effects of a single dose of testosterone undecanoate on body composition and military-relevant physical performance during a simulated operation. After a …

Background: Substantive previous work has shown that both gait speed and global cognition decline as people age. Rates of their decline, as opposed to cross-sectional measurements, could be more informative of future functional status and other clinical outcomes because they more accurately represent deteriorating systems. Additionally, understanding the sex and racial disparity in the speed of deterioration, if any, is also important as ethnic minorities are at an increased risk of mobility disability and dementia. Method: Data from 2 large longitudinal intervention studies were integrated. Rates of decline were derived from individual-level measures of gait speed of 400-m walk and …

Hepatocellular carcinoma (HCC) is the most frequent primary hepatic malignancy and a leading cause of cancer-related death globally. HCC is associated with an indolent clinical presentation, resulting in frequent advanced stage diagnoses where surgical resection or transplant therapies are not an option and medical therapies are largely ineffective at improving survival. As such, there is a critical need to identify and enhance primary prevention strategies to mitigate HCC-related morbidity and mortality. Obesity is an independent risk factor for the onset and progression of HCC. Furthermore, obesity is a leading cause of nonalcoholic steatohepatitis (NASH), the fasting growing etiological factor of …

The aim of this study was to compute intra-class correlations (ICCs) for weight-related and patient-reported outcomes in a cluster randomized clinical trial (cRCT) for weight loss. Baseline and follow-up data from the Promoting Successful Weight Loss in Primary Care in Louisiana (PROPEL) cRCT were used in this analysis. ICCs were computed for baseline and follow-up measures, and changes in body weight, cardiometabolic risk factors and health-related and weight-related quality of life at 6, 12, 18 and 24 months. Baseline ICCs ranged from 0 for PROMIS measures of anxiety and fatigue to 0.055 for total cholesterol (median = 0.019). The ICCs …
Qualifying Factors That Predict Weight Loss Success Within Diet Interventions, Jordan Losavio
Qualifying Factors That Predict Weight Loss Success Within Diet Interventions, Jordan Losavio
LSU Master's Theses
Evidence-based diet strategies lead to weight loss for some, while for others, the same diet approach could result in trivial weight loss or even in weight gain. This prompts the underlying research question: what factors predict that a given diet strategy will be efficacious for some, but not for others? This research was a secondary analysis of clinical weight loss studies to determine which factors influence an individual’s weight loss success for three diet intervention strategies: low-carbohydrate, low-fat, and low-calorie. A combination of physiological, psychological, behavioral, and dietary factors was evaluated for each diet study. Multiple linear regression and logistic …
Integration Of Remote Sensing Approaches For In-Season Nitrogen Management, Murilo De Santana Martins
Integration Of Remote Sensing Approaches For In-Season Nitrogen Management, Murilo De Santana Martins
LSU Doctoral Dissertations
Nitrogen (N) management is being conducted at flat rate in Louisiana due to practicality and convenience, but the price of N fertilizer and high breakeven costs are forcing producers to find ways to reduce costs and optimize N application. In this scenario, precision agriculture technologies, specifically the use of optical sensors on board of unmanned aerial systems (UAS) to variable rate N application on farm is showing a promising approach to save inputs and reduce environmental impacts. However, the general goal of this research was to develop and evaluate in-season N management approaches for N recommendation in corn (Zea …
Defining Toxicological Endpoints Of Aphis Gossypii Feeding, Flinn O'Hara
Defining Toxicological Endpoints Of Aphis Gossypii Feeding, Flinn O'Hara
LSU Master's Theses
The cotton aphid, Aphis gossypii Glover (Hemiptera: Aphididae), is a destructive agricultural pest, due to photosynthate removal and plant virus transmission. This project consisted of two main goals. First, we employed toxicity bioassays and electrical penetration graph (EPG) methodology to quantify changes to feeding behavior and toxicity of A. gossypii after exposure to commercialized aphicides. Commercialized aphicides containing flupyradifurone, sulfoxaflor, thiamethoxam, thiamethoxam + lambda cyhalothrin, and bifenthrin induced > 90% aphid mortality within 4 hours of exposure. This was supported by our EPG results that showed a significant reduction in the proportion of aphids that continued to probe on cotton 4 …

Fresh and fresh-cut fruits and vegetables have been associated in several foodborne illness outbreaks. Although investigations from those outbreaks reported that the contamination with pathogenic microorganisms may occur at any point in the farm to fork continuum, effective control strategies are still being widely investigated. In that direction, the concept of hurdle technology involving a sequence of different interventions have been widely explored. Among those interventions, ultraviolet (UV) light alone or in combination with other treatments such as use of organic acids or sanitizer solutions, has found to be a promising approach to maintain the microbiological safety and quality of …
Development Of A Non-Dairy Yogurt Alternative Using High Protein Brown And White Rice Flour, Anita Morris
Development Of A Non-Dairy Yogurt Alternative Using High Protein Brown And White Rice Flour, Anita Morris
LSU Doctoral Dissertations
There is a great demand for plant-based yogurt alternatives (YAs) due to societal prevalence of milk sensitivities and allergies as well as some consumers abstaining from animal-derived products. Producing YAs from rice flour has considerable potential because (1) rice is not a major food allergen, (2) using rice flour is more economical compared to plant milks, the main ingredient in commercially available YAs, and (3) there are no rice-based yogurt alternatives in today’s marketplace. The overall goals of this study were to develop probiotic YAs from brown and white rice flours, and to evaluate their storage stability, probiotic’s survival in …
Genetic Dissection And Qtl Pyramiding To Enhance Salinity Tolerance In Rice (Oryza Sativa L.), Sandeep Chapagain
Genetic Dissection And Qtl Pyramiding To Enhance Salinity Tolerance In Rice (Oryza Sativa L.), Sandeep Chapagain
LSU Doctoral Dissertations
Salinity is emerging as a major threat to rice production worldwide due to climate change and anthropogenic sources. We performed three different population development approaches using exotic salt tolerant donors in US rice backgrounds to identify the salt tolerant QTLs/genes and tolerant breeding lines. First approach involved development of a biparental introgression lines (ILs) population by utilizing a salinity tolerant donor ‘TCCP’ in an elite rice cultivar ‘Cheniere’ background and mapping of QTLs at germination, seedling, and reproductive stages A total of seven QTLs for five germination traits, 16 QTLs for seven seedling stage traits, and 30 QTLs for ten …
Aphid Herbivory On Industrial Hemp, Cannabis Sativa L., And The Impact Of Plant-Soil Feedbacks On Hemp Performance In Louisiana, Alex Leede
LSU Master's Theses
Industrial hemp, Cannabis sativa L., is defined as having less than 0.3% delta-9-tetrahydrocannabinol by dry weight. It can be grown for fiber, food, oil, and cannabinoids. As a newly legalized crop, little research exists regarding the cultivation or pest management practices in Louisiana. The versatility of industrial hemp may allow for substantial future production in Louisiana. This project set out to understand the impact of previous crops on hemp performance and soil microbial communities. Soybean, Glycine max L. (Merr), grain sorghum, Sorghum bicolor (L.) Moench, and sweetpotato, Ipomeae batatas (L.) Lam, were selected as soil conditioning crops because they are …
