Race 1 isolates collected from tomato roots were used to develop an orange mutant of Fusarium oxysporum f. sp. lycopersici (FOL) for use in competitive infection tests against nonpathogenic F. oxysporum isolates. Eighty one color mutants were induced by exposure to ultraviolet light. The mutant ORA-22 retained pathogenicity and growth habit similar to the wild-type. Preliminary competitive infection tests indicated differences within the nonpathogenic population in the ability to suppress or enhance infection by FOL orange mutant ORA-22. Four vegetative compatibility groups (VCGs) were tested to determine if competitive infection ability was associated with VCG designation. Results from these tests …

The analgesic and anti-inflammatory effects of the nonsteroidal anti-inflammatory drugs (NSAIDs) ketoprofen (2.2 and 3.63 mg/kg) and phenylbutazone (4.4 mg/kg) were compared in equine models of acute synovitis and chronic hoof pain. The eicosanoids, prostaglandin E$\sb2$ (PGE$\sb2$) and leukotriene B$\sb4$ (LTB$\sb4$), increased dramatically in synovial fluid after carrageenan-induced synovitis of the intercarpal joint. PGE$\sb2$ concentrations in untreated horses peaked at 9 hours while LTB$\sb4$ concentrations peaked in all horses at 3 hours. Synovial fluid concentrations of both eicosanoids returned to near baseline by 48 hours. Lameness, joint temperature, and synovial fluid volume, protein and nucleated cells increased at 3 to …

It was demonstrated, using a homologous transgenic rice system, $5\sp\prime$ distal and proximal cis-acting transcriptional regulatory elements were required for developmental control of a rice seed storage protein glutelin (Gt1) gene. The functional importance of proximal elements in the context of the 1.8 kb promoter was demonstrated by single substitution mutations in the TATA box ($-$28/$-$23), AACA motif ($-$73/$-$61), and protein-binding Boxes I ($-$103/$-$86), II ($-$124/$-$110), III ($-$175/$-$158), and IV ($-$200/$-$217). A simultaneous mutation of five protein-binding sites ($-$410/$-$86) essentially eliminated the activity of the 1.8 kb promoter. Bombesin is a tetradecapeptide first isolated from frog skin. This peptide has potent effects on core body temperature of rats when administered centrally. Bombesin-induced hypothermia at normal ambient temperatures appears to be dependent upon some pre-existent condition which may be related to nutritional status. Four experiments were conducted to examine if the pre-existing condition for bombesin-induced hypothermia at normal ambient temperatures is primarily related to modulation of sympathetic nervous system activation. Experiment I demonstrated that central bombesin resulted in hypothermia in rats tested at normal ambient temperatures under conditions of ad-libitum access to food only when injected peripherally with the …

In many dicotyledons, the nutrient reserves are stored in the cotyledons. Phaseolin, 7S globulin, is the major seed protein from common bean Phaseolus vulgaris. In vitro transcription assays and gene transfer studies indicate that phaseolin gene expression is regulated primarily at the transcriptional level. In this dissertation, I studied cis- and trans-acting factors that may play a role in the transcriptional regulation of the $\beta$-phaseolin gene. Gel mobility shift and exonuclease III protection assays identified four distinct DNA binding proteins, CAN, AG-1, CA-1, and TATA-box binding protein. This study was conducted to: (1) determine the effects of stocking density and dietary protein:energy ratio on survival, growth, feed consumption, feed conversion, lipo-somatic index, dress-out percentage, and productive protein value of common snapping turtles; and (2) determine the fatty acid composition of muscle, liver, and fat bodies of wild common snapping turtles and common snapping turtles fed a diet of known fatty acid composition. Hatchling common snapping turtles, Chelydra serpentina, were stocked at five or 10 animals per container (29 and 58 turtles/m$\sp2$, respectively), and fed one of seven prepared diets. In anticipation of the regional demand for tomatillo (Physalis ixocarpa Brot.) for the fresh market and sauce industry, four years of research trials (1990-1993) showed a significant adaptation of tomatillo to Louisiana planting conditions where, like tomato, it performed best in the cooler temperatures of spring and early fall. Field surveys indicated that virus diseases were major constraints on production. A foliar mosaic and yellow mottle found commonly affecting plants was caused by Physalis mosaic virus (PhyMV), identified by host reaction, electron microscopy, serology, and dsRNA analysis. Cucumber mosaic virus (CMV) and tomato spotted wilt virus (TSWV) were also found. …

The purpose of this study was to describe and compare funded and unfunded research proposals submitted to the Southern Region Sustainable Agriculture Research and Education Council between 1992 and 1994. Coding reliability and validity were assessed through a pilot study after which the actual coding was carried out. A census of 32 funded and a simple random sample of 54 unfunded proposals were used for this study. Descriptive statistics (frequencies) were used to describe research themes, sustainable agricultural practices, research categories, funds requested for research, origin of proposals, length of research period, research cooperators, and the occupation of principal investigators. …

This dissertation presents a model of strategic issue management that focuses on the mediational role that interpretation has between monitoring and scanning the environment and organizational response. Hypotheses concerning the relationships between several dimensions of issue interpretation and several dimensions of organizational issue response are posited. The strategic issue of consumer nutrition awareness within the restaurant industry is targeted, and a research methodology constructed for testing the hypotheses. Urgency, Understanding, and Capability all had significant relationships with the response variables of Magnitude, Locus, and Activeness, with Understanding's relationship tending to be curvilinear. In most of the United States, overwintering redclaw crayfish (Cherax quadricarinatus) is necessary because they cannot withstand winter temperatures. Consequently, hatcheries are required to produce juveniles for pond stocking. The objectives of this study were: (1) identify biological factors affecting production of juvenile redclaw, (2) determine production capabilities of a redclaw crayfish hatchery, and (3) determine the economic potential of redclaw hatcheries. Experiments were designed to assess fecundity and factors influencing spawning such as stocking density, male/female weight, diet, mortality, and cannibalism. The effect of caffeine ingestion upon endurance performance while walking at 45% VO$\sb{2\rm max}$ in a warm environment (27$\sp\circ$C, 48% rh) was examined. Eight moderately trained subjects (aged 21-25 years) walked on a treadmill on 4 separate occasions until exhaustion under 4 different treatment regimens. Two of the treatments consisted of caffeine-containing beverages, while the other 2 included non-caffeine-containing beverages. One hour prior to the start of exercise, the subjects consumed a GatorPro drink with either 5 mg/kg of caffeine, if caffeine was the randomized drink, or placebo (flour). Several human and animal studies of chromium (Cr) supplementation in the form of Cr picolinate (CrPic) have reported increases in lean body mass, often with concurrent body fat decreases. This 12-week study examined the effects of six dietary levels of CrPic (0, 75, 150, 300, 600, 1500 ppb Cr) upon the growth and body composition of the rat. Ninety male Harlan Sprague-Dawley outbred weanling rats arrived in three groups of 30 animals (referred to as Blocks 1, 2, and 3). The objective of the present study was to evaluate the effect of post-block mouse and bovine embryos on the development of pre-block embryos around the in vitro developmental block stage. The effect of the post-block embryos on the development of the pre-block embryos was evaluated in culture, in chimeras and in cytoplasmic hybrid embryos. The results indicate that post-block embryos were able to stimulate the development of the pre-block embryos in culture. Two researcher-produced, standard-length videos on photosynthesis (and accompanying ancillary materials) were developed with the input of educators and content experts. The control treatment video script contained the standard number of prototypical examples and graphics based high school textbooks. The experimental treatment video script was based on the theory of conceptual change. Two student focus groups met daily for one week and reviewed portions of the original experimental treatment script for clarity, brevity, and choice and placement of examples and nonexamples. The generation time of L. monocytogenes was calculated to be 72.2, 28.5, 17.01, and 6.9 h on crawfish tail meat held at 0, 4, 6, and 12$\sp\circ$C, respectively. Potassium sorbate (PS) (0.3%) spray treatments on crawfish held at 4$\sp\circ$C, extended the lag phase of the bacterium. However, once exponential growth began, a generation time not significantly different (P $>$ 0.05) than that of untreated crawfish was observed. Samples sprayed with 0.3% citric acid (CA) and held at 4$\sp\circ$C supported growth of the bacterium equally well as that of untreated samples. Agricultural terraces form a distinctive element in the cultural landscape of Fiji. These skillfully fashioned earthworks were developed for the irrigated cultivation of one specific cultigen--taro (Colocasia esculenta), a long domesticated edible aroid. Constructing pondfields on slopes represents an intensification of production, with taro yields responding favorably to higher levels of labor input. In Fiji, irrigated terraces on the leeward side of larger islands also served to overcome seasonal conditions of drought or soil moisture deficits. Nearly all of these intensive agrosystems have been abandoned. But the carefully sculpted hillsides endure, and serve as poignant reminders of past travail. Taro …

Intrinsically curved DNA upstream of the RNA polymerase binding site, the promoter, influences transcription in Bacillus subtilis. In this study, curved DNA upstream of the $-$35 region was examined for the ability to influence transcription in Escherichia coli and B. subtilis through the use of hybrid promoters. These promoters were constructed by substituting the curved DNA from two B. subtilis promoters, Bal129 and Alu156, for the DNA upstream of the $-$35 region in the lambda phage promoters $p\sb{\rm R}$ and $p\sb{\rm L}$. Three experiments were designed to provide a comprehensive reference for the responses of plasma metabolites and hormones to control and restricted intakes of protein and(or) energy and to feed deprivation. Field strains of soybean looper, Pseudoplusia includens (Walker), from Louisiana, Texas, and Puerto Rico were tested for permethrin synergism by piperonyl butoxide (PB) and S,S,S tributylphosphorotrithioate (DEF). Resistance was completely suppressed by 10 $\mu$g PB/larva in strains from Louisiana with 2.7 to 14.1-fold resistance. The Texas strain was 14.8-fold resistant and permethrin toxicity was increased 8.6-fold by PB. The Puerto Rico strain was highly resistant to permethrin (426.6-fold) and PB increased permethrin toxicity only 4.6 and 7.5-fold, respectively, at 10 and 20 $\mu$g/larva. The fall armyworm (Spodoptera frugiperda J. E. Smith) Lepidoptera: Noctuidae) consists of two genetically differentiated strains. The corn strain feeds on corn and the rice strain feeds primarily on rice and forage grasses. My research focused on host plant adaptations within each strain as assessed by larval performance and enzymatic responses to feeding upon different hosts. Three sets of experiments were conducted. The first experiment examined consumption and utilization in both strains reared on corn. The agriculturally important soils of the lower Mississippi alluvial plain typically display surprisingly high levels of nitrogen (N) fertility given the low amounts of organic matter and readily available forms of N characteristically present in these soils. The principal goal of this research was to investigate N dynamics in these soils with special attention devoted to the possible role of clay-fixed NH$\sb4\sp+$ in N fertility. White flesh sweetpotatoes (Ipomoea batatas (L.) Lam) are a major food crop in developing countries. This study was to evaluate the protein and carotenoid content and quality of various white fleshed African sweetpotatoes and Apios americana tubers for use as a weaning food. The sweetpotato and Apios are not normally considered sources of carotenoids and proteins, but the wide use of sweetpotatoes as a source of carbohydrate and the renewed interest in Apios warrant the study of carotenoids and proteins in these crops. Among food-borne pathogens, Listeria monocytogenes is one of the more resistant to food processing methods, including irradiation. USFDA approval for irradiation of processed meat and seafood products likely to contain Listeria spp. is $<$3 kGy gamma radiation. The incidence of L. monocytogenes in food products varies between 5-30% and at levels $<10\sp4$ CFU/g. With the capacity of this bacterium to multiply at refrigeration temperatures and the zero tolerance level mandated by the USFDA it becomes imperative to maximize the efficiency of the irradiation process. The first purpose of this study was to determine if initial cell concentration and/or temperature at time of irradiation influences the radiosensitivity of L. monocytogenes. Secondly, to determine whether split dose application would effect the radiation response of this bacterium. Concentrations of 10$\sp3,$ 10$\sp6$ and 10$\sp9$ cfu/ml of L. monocytogenes Scott A, were suspended in tryptic soy broth and exposed to 0-5 kGy gamma (1.25MeV) radiation at 20, 4, and -80$\sp\circ{\rm C}.$ Split doses were applied in equal portions to initial cell concentrations of 10$\sp6$ and 10$\sp9$ at the same temperatures. Surviving cells under the various conditions were enumerated and irradiation D-values were calculated from linear regression curves. At all temperatures, cell concentrations of 10$\sp3{\rm CFU/ml}$ were reduced to non recoverable levels with 2 kGy gamma radiation. The irradiation D-value of 0.43 kGy for frozen (-80$\sp\circ{\rm C})$ cultures of 10$\sp6$ CFU/ml was significantly lower (p $<$ 0.05) than the D-values (0.58 and 0.62 kGy) for the same cell concentration at the other two temperatures (20$\sp\circ$ and 4$\sp\circ{\rm C},$ respectively). For higher cell concentration of 10$\sp9$ cfu/ml, a D-value of 0.42 kGy was obtained for both 4$\sp\circ{\rm C}$ and -80$\sp\circ{\rm C}$ these were significantly lower (p $<$ 0.05) than 0.50 kGy for 20$\sp\circ{\rm C}$ suspensions. With both cell concentrations, the lowest D-values were obtained when cells were irradiated in the frozen state indicating a that primary cell damage was due to direct effects of irradiation. Split doses of 1 and 2 hours time between fractions approached the generation times for this bacterium and were significantly reduced at 20$\sp\circ{\rm C}.$ However, there was no consistent trend in reductions at 4$\sp\circ$ or -80$\sp\circ{\rm C}.$ Split dose application of irradiation to food pathogens should be further explored.

Impedance technology is currently used as a measurement of microbial activity. The purpose of the current study was to determine the feasibility of utilizing impedance technology to evaluate the quality of white shrimp (marine species) and channel catfish (freshwater species). Impedance analysis measurements (I-values) of raw shrimp increased for samples stored at 5, 25, and 35$\sp\circ$C; however, control samples stored at $-$20$\sp\circ$C displayed no significant changes in I-values. I-values from thermally processed shrimp had no direct relationship with odor scores, increased storage time, or increased storage temperature. Alginate has many industrial uses because of its unique colloidal behavior, and its ability to thicken, stabilize, emulsify, suspend, form films, and produce gels. Currently, all commercial alginate comes from brown algae, where it exists naturally as a structural material. Treatment with androgens and anabolic steroids in conditions of high bone turnover seen in osteoporotic postmenopausal females has resulted in bone density improvement. Research has demonstrated a link between muscle mass and bone due to the mechanical demands placed on bone by muscular contractile force associated with physical activity. Acetylene reduction and nitrogen difference assays were used to evaluate effects of defoliation by soybean looper, Pseudoplusia includens (Walker), during reproductive growth stages on symbiotic nitrogen fixation by soybean. Defoliation (50-55%) of group VI cultivar Lee from full bloom into pod development stages caused up to 85% reduction of nitrogenase activity, whereas lower defoliation levels (32%) during seed development did not affect nitrogenase activity. Defoliation (74-94%) of group IV cultivar Clark caused 80-100% reduction of nitrogenase activity, regardless of whether defoliation occurred during bloom and pod development or during seed development. The purpose of this study was to determine the effectiveness of using nontraditional referral and screening procedures to identify culturally different (minority) gifted children. The extreme disparity of representative frequencies of these groups within gifted programs nationally and in Louisiana prompted research in four parishes (counties) to compare nontraditional with traditional identification procedures. The nontraditional referral procedures included child search methods and parent and teacher training about the characteristics of giftedness among culturally different populations. Background: Androgen-dependent proteins in the lumen of the epididymis are required for sperm maturation. One of these is a retinoic acid binding protein, E-RABP, which binds both all-trans and 9-cis retinoic acid. The other retinoid-binding proteins whose structures are known do not bind 9-cis retinoids. Results: We describe the X-ray structure determination of E-RABP with and without bound ligand. The ligand binds deep in the β-barrel of the protein, the β-ionone ring innermost. The binding site, like the ligand, is amphipathic and the deepest part of the cavity is formed by a ring of aromatic amino acids. The isoprene tail …
