Ejaculates from sires were examined by polymerase chain reaction to determine percentage of sperm bearing the Y chromosome. Results were verified by examining the percentage of male calves per ejaculate used in artificial insemination (AI) and the percentage of male piglets per litter from a controlled mating program. Spermatozoal DNA was amplified by polymerase chain reaction with specific primers for the Y chromosome. Image analysis measured the fluorescent intensity of the 194-bp band. Ejaculates were compared with a pooled standard of spermatozoal DNA equated to a 50% Y-bearing sperm ejaculate. Calving data were obtained from information collected for the National …

Changes in substrate oxidation with isoenergetic high-carbohydrate (HC) and high-fat (HF) diets in male nonathletic subjects, aerobically trained athletes, and weight-trained athletes were examined in a crossover study. A whole-room respiration chamber was used to measure 24-h energy expenditure (EE) and substrate oxidation with control, He, or HF diets for 7 d. The nonathletic group had higher 24-h EE (P < 0.05), exercise EE (P < 0.03), and resting metabolic rate (P < 0.04) than did the aerobically trained athletes when these measurements were corrected for lean body mass. Fat oxidation was significantly correlated with lean body mass and diet. However, athletic status had no effect on substrate oxidation. Carbohydrate oxidation across groups increased acutely by 23% after 24 h of the HC diet (P < 0.0001). Carbohydrate balance increased significantly over time with the HC diet (P < 0.002) and decreased acutely after return to the control diet (P < 0.0001). With the HF diet, carbohydrate balance increased and was significantly different from balance with the control diet by day 7 (P < 0.03). Fat balance decreased significantly with both the HF (P < 0.04) and HC (P = 0.0075) diets by day 7. Carbohydrate oxidation correlated with carbohydrate intake with both the control (r = 0.61, P < 0.01) and HC diets (r = 0.59, P < 0.02), but not the HF diet. Fat oxidation was not correlated with fat intake. In conclusion, substrate oxidation in a respiration chamber is significantly affected by diet, but not by prior athletic training.

A sensitive methodology for the determination of RNA and DNA concentrations was developed which allowed for the quantification of nanogram amounts of both DNA and RNA. The method uses a combination of thiazole orange, a non-specific nucleic acid stain, and the RNA degrading enzyme RNase. Thiazole orange had high fluorescence yield. The fluorescence was additive for combinations of RNA and DNA. The addition of proteinase-K increased the fluorescence of the tissue sample by 100%. Recovery of nucleic acid standards added to tissue extracts yielded $\sim$100% recovery. The method is compatible with assays for protein concentration and electron transport system (ETS) …

There is special concern about the potential health effects of by-products released into the environment after combustion or conversion of fossil fuels. Polycyclic aromatic hydrocarbons (PAH), while initially released largely into the atmosphere, are subsequently deposited in soil and water and on foods. Although the significance of PAH contamination with regard to environmentally caused diseases and cancer in humans is difficult to estimate, it is suggested that the potential hazards may be underestimated. Three experiments were conducted to determine the effects of dietary nitrogen on the energy status of ruminants and the feasibility of using L-carnitine to prevent the negative effects associated with overfeeding nitrogen. The objectives of the first experiment were to monitor the nitrogen and energy status of cows grazing ryegrass pastures through the measurement of key blood metabolites and insulin and to determine the protein supplementation scheme which would maximize milk yield and maintain plasma urea nitrogen levels below dangerous levels. Disposal of crawfish processing wastes poses a challenging problem to the rapidly expanding crawfish industry. Co-composting is examined as a waste management alternative to landfill disposal. Wood chips, rice hulls, bagasse, and bark were evaluated for use as bulking agents in composting crawfish processing residuals. Use of bagasse as a bulking agent led to the largest reduction in volatile solids, organic-C, particle size, and compost volume. Finished compost using bagasse contained the greatest concentration of N. Self-heating patterns and decomposition of crawfish residuals were satisfactory using all four bulking agents. The present study used a longitudinal design in an attempt to answer the following questions: (a) what is the prevalence of sedentary lifestyle in a low-income primary care female population? (b) does minor stress predict depressive symptoms after controlling for major stress? and (c) does leisure-time physical activity moderate the association between stress and depressive symptoms? The sample included 150 randomly selected adult female patients recruited from primary care clinics at a public hospital in the state of Louisiana. In recent years, there have been a few reports of the relationship between eating disorders in mothers and the psychological adjustment of their children. Most of these reports have been based upon anecdotal case studies, however a wide range of prenatal, perinatal, and postnatal complications, including behavioral and feeding difficulties in children of these mothers has been described. This study advanced research in this area by incorporating both normal and depression comparison groups. Despite increasing institutional focus in aquaculture, sustainability remains an amorphous and much debated concept. Little consensus has been identified on the issue beyond the general recognition that the concept should contain environmental, economic, and sociological considerations. This study addressed the specific, relevant question of whether politically diverse aquacuiture interests can collectively develop and agree on production-level goals and indicators of aquaculture sustainability. The research partitioned sustainability into its three subcategories: environmental, economic and social. Dysfunction in homeostatic mechanisms of cell death and proliferation are considered to be important in carcinogenesis. The p53 gene has been implicated in the regulation of cell death and proliferation. The host response was measured through sequential immunohistochemical (IHC) detection of apoptosis, PCNA, and p53 in livers of Fundulus grandis fish exposed to MNNG or to 2-AF, two known carcinogens. Studies performed determined the stability of MNNG in saltwater, the suitability of different fixatives on IHC detection of apoptosis, PCNA-PC10, and p53 (various clones), and frequencies of expression of apoptosis, PCNA, and p53. Freshly milled raw rice bran was stabilized with microwave heat for 3 min. Half of raw and treated bran were packed in Ziplock(c) bags and the other half vacuum packed. Half of the samples were stored at 4--5°C and the other half at 25°C for 16 weeks. The free fatty acid (FFA) content increased rapidly in raw rice bran samples vacuum packed and samples packed in Ziplock(c) bags store at 25°C from an initial amount of 2.5% to 54.3% and 48.1%, respectively. This study associated climatic and environmental factors with the incidence of visceral leishmaniasis (calazar) in Northeast Brazil. Remote sensing (RS) techniques permitted evaluation of spatial and temporal landscape features to stratify the region and define the target population for this vector-borne disease. The Municipality of Caninde, Ceara, Brazil was divided into 873-- 2 x 2 km2 squares centered on coordinates from a Universal Transverse Mercator projection (scale 1:100,000, 1994) and geo-referenced with 2 Landsat T.M. A high purity $\gamma$-oryzanol was obtained from crude rice bran oil using a normal phase preparative scale HPLC. It was used in implementing a reverse phase HPLC method for separating each individual component of $\gamma$-oryzanol present in rice bran oil. Ten fractions were isolated in the reverse phase HPLC method. The ten components of $\gamma$-oryzanol were identified. Three of these, cycloartenyl ferulate, 24-methylene cycloartanyl ferulate, and campesteryl ferulate, were major components of $\gamma$-oryzanol. Edwardsiella ictaluri, causative agent of enteric septicemia of catfish (ESC), is the primary bacterial pathogen of commercially produced channel catfish (Ictalurus punctatus). The purpose of this study was to make progress towards development of a successful ESC vaccine by generating and characterizing a pool of E. ictaluri antigens and determining if they were protective against ESC in catfish. Antigenic protein expression was evaluated in E. ictaluri cells under different conditions of growth using two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and western blotting with pooled convalescent catfish serum (CCS). It is the goal of this dissertation research to reveal some aspects of the physical nature of spinach carbonic anhydrase as a representative $\beta$CA using the techniques of sequence comparison, molecular biology, and biophysics. Though both $\alpha$ and $\beta$ carbonic anhydrases are zinc dependent metalloenzymes, it is clear that the two isoforms do not adopt the same mechanism for coordinating the active site metal. While $\alpha$CA binds zinc through three histidine ligands, $\beta$CA cannot due to a lack of evolutionarily conserved histidines. Instead, the $\beta$ family has adopted a ligand scheme incorporating a single histidine and two cysteines. This has …

In Exp. 1, 42 calves were fed milk replacer and then starter either with or without 1 ppm supplemental chromium (Cr) as Cr-picolinate. Neither Cr nor sex affected growth performance. With the exception of plasma nonesterified fatty acid concentrations (NEFA), weekly plasma metabolites were not affected by Cr or sex. Pre- and postfeeding plasma NEFA concentrations were lower in Cr-fed calves, but were not affected by sex. AU calves seemed to become less insulin sensitive with age, as plasma glucose became lower and insulin higher as calves became older. During i.v. glucose tolerance tests (IVGTT), calves cleared glucose faster at …

This farm-raised catfish quality study measured desirable flavor attributes, Chickeny, Nutty, Fat Complex, Corn, Sweet, with the objective to determine effects of feed ingredients, time-on-diet and genetic strain. Overall flavor impact differences were tested by triangle tests. The diets were Casein-base reference feed by itself or partially substituted with: 10% catfish meal, 10% meat and bone meal, 10% rice bran, or 3% menhaden oil and commercial feed formulation. Fish were grown indoors to minimize environmental flavors for 70, 160, 210 and 318 days. The genetic strains evaluated were channel, albino channel, blue, hybrid channel x blue, and black bullhead catfish. …

"Airbirds" are defined as those birds that are adapted to almost constant life on the wing and do not perch, except for breeding and roosting. They use the air as the only medium in which to find and gather food. One taxon meets that definition: the swifts (Apodidae). Foraging zones of airbirds can be segregated in terms of clutch size, commuting foraging distance, nestling period, and nestling feeding rate. There is evidence in the literature that suggests a possible worthwhile role of antioxidants during athletic endeavors and exercise performance. Several studies have evaluated the antioxidant ability of beta-carotene and demonstrated a beneficial influence. However, these studies have investigated beta-carotene's effects within an antioxidant mixture which included other antioxidant vitamins. Consequently, this study attempted to isolate the antioxidant role of beta-carotene during exercise. Three parameters of beta-carotene influence were measured using eleven well-trained runners: serum malondialdehyde (MDA) concentration (a marker of free radical generation), 5000 meter race performance, and maximum oxygen consumption (VO2max). The plant pathogen responsible for halo blight in legumes, Pseudomonas syringae pathovar phaseolicola, produces an alginate exopolysaccharide which prevents desiccation and enhances the "water soaking" effect associated with pathogenesis. These bacteria demonstrated elevated extracellular alginate lyase activity during log growth and stationary phases in batch cultures. An analysis of the substrate specificity indicated enzymatic activity on both acetylated and non-acetylated alginate. The release of the enzyme(s) in log growth phase and the subsequent decrease in production in stationary phase have not been previously reported. In the present project the use of ammoniation and fermentation as an integrated process was studied in order to give aflatoxin-contaminated corn an alternative use. Aflatoxin-contaminated corn can be fermented to obtain fuel alcohol and decontaminated distiller's dried solid grains (DDSG) can be used for animal feed. The efficacy of a fermentation process, with the inclusion of ammonium persulfate, for the decontamination of aflatoxin B$\sb1$-contaminated yellow corn was studied, as well as the effect of peroxides on the ethanol production. Addition of 2.0% ammonium persulfate decreased the aflatoxin B$\sb1$ levels by 87%. Peroxides did not have an effect on ethanol …

The current study evaluated interactions between the mycotoxins aflatoxin B$\sb1$ (AFB$\sb1$) and fumonisin B$\sb1$ (FB$\sb1$) in pure systems as well as in corn. During storage at 25$\sp\circ$C with high moisture (ca. 50%), corn contaminated with Aspergillus flavus (L13) and/or Fusarium moniliforme (A00102) presented different aflatoxin and fumonisin levels. Toxin production/degradation varied over storage time (0-45 days). Evaluation of the mutagenicity of unifungal or bifungal corn extracts with the Salmonella/microsomal assay (tester strains TA-100 and TA-98) with metabolic activation (S9) showed positive mutagenic responses. Different ratios of pure AFB$\sb1$/FB$\sb1$ produced divergent mutagenic responses (tester strains TA-100 and TA-98) with S9. The …

Characterization And Utilization Of Flavors From Undersized Crawfish., Feng Chen

Undersized processed crawfish are proposed to be a resource for effective recovery of crawfish flavors. Tail puree from undersized crawfish was hydrolyzed using protease APL-440 through response surface methodology to enrich flavor production. Optimal hydrolytic conditions were determined to be pH $8{\sim}9,\ 65\sp\circ$C, 0.3% (V/W) enzyme/substrate ratio with the reaction maintained at 33.3% puree concentration over 60 min period. Effects of storage time and extraction method on the crawfish flavor profile were evaluated. Shorter storage time gave superior flavor stocks with less oxidized flavors. The pest status of the fall armyworm, Spodoptera frugiperda (J. E. Smith), including development and survival on conventional and transgenic Bacillus thuringiensis (Bt) cotton and host-strain influences, was examined in laboratory and field studies. Neonates (1st instars) were fed leaves from conventional (cv. DP 5415 and cv. DP 5690) and transgenic Bt (cv. NuCOTN 33$\rm\sp{B}$ and cv. NuCOTN 35$\rm\sp{B})$ cotton. There were no significant differences $(P>0.05)$ between cultivars in larval mortality and survival to the pupal and adult stages. However, times to pupation and adult eclosion were significantly longer $(P<0.05)$ for larvae fed transgenic Bt cotton compared to conventional cotton. In addition, larvae weighed significantly less $(P<0.05)$ when fed transgenic Bt cotton compared to conventional cotton. A study was conducted to determine when cotton bolls become tolerant to fall armyworm injury. Fifth instars were caged individually on bolls of various ages from conventional (cv. DP 5415) and transgenic Bt (cv. NuCOTN 33$\rm\sp{B})$ cotton to define the period of boll susceptibility to larval injury. Raw milk samples (n = 246) and pasteurized milk samples (n = 104) were analyzed to determine adenosine triphosphate (ATP) and compared with the standard plate count (SPC). An ATP filtration method was used to filter milk samples prior to ATP determination, which was measured in relative light units (RLU). The ATP assay took approximately 7 minutes to complete and could allow rapid prediction of SPC which is a measure of raw milk quality. Preweaning data collected on 1,180 calves in generation five of a crossbreeding project were analysed. The systems involved were first-cross (F$\sb1$, three-breed terminal, two-, three-, and four-breed rotational and rotational-terminal. Angus (A), Brahman (B), Charolais (C), and Hereford (H) breeds were used in the F$\sb1$ and rotational phases, while Gelbvieh (G) and Simmental (S) were the terminal sire breeds. The traits studied were birth weight (BWT), preweaning gain (ADG), 205 d weight (WWT), and weaning height (WHT). Saccoglossus bromophenolosus King et al., 1994 and Protoglossus graveolens Giray and King, 1996 contain high concentrations of 2,4-dibromophenol (DBP), the function of which is uncertain. Mature enteropneusts that were collected from the field and maintained in vitro without bromide retained DBP, which is inconsistent with active DBP secretion into the burrow environment. DBP was also conserved during field manipulations that decreased food availability in situ. Further, DBP did not deter predation in feeding experiments with the anomuran crab Pagurus longicarpus and the polychaetes Glycera dibranchiata, Nereis virens and Nepthys incisa. The excess of hypertension among blacks has been recognized since early in this century and explains a substantial portion of the black health disadvantage. In a cohort study begun in the 1970s, hypertension accounted for 20% of all-cause mortality among blacks, compared to 10% among whites. National data on trends in hypertension (140/90 mm Hg or treatment) prevalence from 1960 to 1990 suggest a decline from 44% to 32%, although differences in survey technique likely account for this pattern. During this period the prevalence ratio of black:white remained constant at 1.5, suggesting that secular trends in causal factors, if any, …
