We longitudinally explored the relationship of body size and adiponectin levels in 393 community-dwelling Afro-Jamaicans. Adiponectin levels were greater in women, increased with age and declined with abdominal adiposity. Multivariate regression analyses suggest that subcutaneous fat in women may contribute significantly to the variance in their adiponectin levels. © 2010 Elsevier Ireland Ltd.

The search for longevity-determining genes in human has largely neglected the operation of genetic interactions. We have identified a novel combination of common variants of three genes that has a marked association with human lifespan and healthy aging. Subjects were recruited and stratified according to their genetically inferred ethnic affiliation to account for population structure. Haplotype analysis was performed in three candidate genes, and the haplotype combinations were tested for association with exceptional longevity. An HRAS1 haplotype enhanced the effect of an APOE haplotype on exceptional survival, and a LASS1 haplotype further augmented its magnitude. These results were replicated in …

Distribution Of Isoflavones And Antioxidant Activities Of Soybean Cotyledon, Coat And Germ, Xiaohua Yue, Ahmed Moursy Abdallah, Zhimin Xu
Faculty Publications
Soy isoflavones in the cotyledon, coat and germ of four soybean varieties from the southern U.S.A. were determined and compared. Isoflavones with aglycone, daidzein, genistein, glycitein and aglycone, withβ-glucoside, genistin, daidzin and glycitin, were found in the four varieties. The range of total soy isoflavones in cotyledon, coat and germ extracts from the varieties was 2.73-9.71, 5.56-16.94 and 27.76-81.43 mg/g, respectively. Soy germ was the richest source of isoflavones among the three soybean parts. Distribution of the isoflavones in soy cotyledon was different from that in soy coat or germ. Daidzin and glycitin were the major isoflavones in the germ …
Beneath The Salt Marsh Canopy: Loss Of Soil Strength With Increasing Nutrient Loads, R. Eugene Turner
Beneath The Salt Marsh Canopy: Loss Of Soil Strength With Increasing Nutrient Loads, R. Eugene Turner
Faculty Publications
Although the broadly observed increase in nutrient loading rates to coastal waters in the last 100 years may increase aboveground biomass, it also tends to increase soil metabolism and lower root and rhizome biomass—responses that can compromise soil strength. Fourteen different multiyear field combinations of nutrient amendments to salt marshes were made to determine the relationship between soil strength and various nitrogen, phosphorus, and nitrogen+phosphorus loadings. There was a proportional decline in soil strength that reached 35% in the 60- to 100-cm soil layer at the highest loadings and did not level off. These loading rates are equivalent to those …

Background: Chronic disease accounts for nearly three-quarters of US deaths, yet prevalence rates are not consistently reported at the state level and are not available at the sub-state level. This makes it difficult to assess trends in prevalence and impossible to measure sub-state differences. Such county-level differences could inform and direct the delivery of health services to those with the greatest need. Methods: We used a database of prescription drugs filled in the US as a proxy for nationwide, county-level prevalence of three top causes of death: heart disease, stroke, and diabetes. We tested whether prescription data are statistically valid proxy …

Records from a colony of captive Indian rhesus macaques (Macaca mulatta) were used to estimate heritability for a number of reproductive traits. Records were based on a total of 7,816 births by 1,901 females from 1979 to 2007. Heritability was estimated with a linear animal model using a multiple trait derivative free REML set of programs. Because no male parents were identified, the numerator relationship matrix contained female kinships established over six generations. Reproductive traits included female age at the birth of the first, second and last infant, age at death, inter-birth intervals, number of infants born per female and …

Emerging evidence suggests that increases in activated T cell populations in adipose tissue may contribute toward obesity-associated metabolic syndrome. The present study investigates three unanswered questions: 1) Do adipose-resident T cells (ARTs) from lean and obese mice have altered cytokine production in response to TCR ligation?; 2) Do the extralymphoid ARTs possess a unique TCR repertoire compared with lymphoid-resident T cells and whether obesity alters the TCR diversity in specific adipose depots?; and 3) Does short-term elimination of T cells in epididymal fat pad without disturbing the systemic T cell homeostasis regulate inflammation and insulin-action during obesity? We found that …

Endoreplication, also known as endoreduplication, is a phyogenetically widespread modified version of the cell cycle in which DNA replication is not followed by cell division. The SIAMESE (SIM) gene of Arabidopsis thaliana encodes the founding member of a novel class of plant-specific cyclin-dependent kinase (CDK) inhibitors and is a key regulator of endoreplication during the development of trichomes (shoot epidermal hairs). Here, we have identified mutations in the CCS52A1 gene as genetic modifiers of the multicellular trichome phenotype of sim mutants. Loss-of-function ccs52A1 mutations dramatically enhance the multicellularity of sim mutants trichomes in double mutants, whereas overexpression of CCS52A1 completely …

The objective of this work was to determine the genetic effects that control the inheritance of characters associated with the phosphorus utilization efficiency (PUE) in maize, and to determine the relationship among them. A diallel between six maize cultivars was performed and 15 hybrid combinations were obtained from it. These cultivars were evaluated in high and low phosphorus availability in the greenhouse and field. Plant response to nutrient availability (RU) and traits associated to utilization and translocation efficiency were measured. There were significant interactions between hybrid combinations and phosphorus availability for plant dry matter mass (MSP), P concentration in plants …

Symbiosis In Subterranean Termites: A Review Of Insights From Molecular Studies, Claudia Husseneder
Faculty Publications
The symbiotic relationship of termites and their eukaryotic and prokaryotic gut microbiota is a focal point of research because of the important roles symbionts play in termite nutrition. The use of molecular methods has recently provided valuable insights into the species diversity and the roles of microorganisms in the guts of termites. This paper provides a review of the current knowledge of symbiont species inventories, genome analysis, and gene expression in the guts of subterranean termites. Particular emphasis is given to the termite genera Reticulitermes and Coptotermes (Isoptera: Rhinotermitidae), because they contain pest species of global impact in their native …

A mutation of AtSOS1 (Salt Overly Sensitive 1), a plasma membrane Na +/H+-antiporter in Arabidopsis thaliana, leads to a salt-sensitive phenotype accompanied by the death of root cells under salt stress. Intracellular events and changes in gene expression were compared during a non-lethal salt stress between the wild type and a representative SOS1 mutant, atsos1-1, by confocal microscopy using ion-specific fluorophores and by quantitative RT-PCR. In addition to the higher accumulation of sodium ions, atsos1-1 showed inhibition of endocytosis, abnormalities in vacuolar shape and function, and changes in intracellular pH compared to the wild type in root tip cells under …

Application of low doses of alum to treatment wetlands to reduce elevated outflow winter phosphorus concentrations were tested in mesocosms vegetated with either Typha domingensis, Schoenoplectus californicus, or submerged aquatic vegetation (SAV) (Najas guadalupensis-dominated). Alum was pumped to experimental units at a rate of 0.91 g Al m-2 d-1 and water quality monitored for 3 months. The alum application significantly improved the outflow water quality and overall the growth of the plants was unaffected by the alum application. Biomass and growth varied between species and through time, but no significant effects of alum application were detected. The concentrations …

Phosphate (Pi) availability is a major constraint to plant growth. Consequently, plants have evolved complex adaptations to tolerate low Pi conditions. Numerous genes implicated in these adaptations have been identified, but their chromatin-level regulation has not been investigated. The nuclear actin-related protein ARP6 is conserved among all eukaryotes and is an essential component of the SWR1 chromatin remodeling complex, which regulates transcription via deposition of the H2A.Z histone variant into chromatin. Here, we demonstrate that ARP6 is required for proper H2A.Z deposition at a number of Pi starvation response (PSR) genes in Arabidopsis (Arabidopsis thaliana). The loss of H2A.Z at …

The brain controls reproduction in response to relevant external and internal cues. Central to this process in vertebrates is gonadotropin-releasing hormone (GnRH1) produced in neurons of the hypothalamic-preoptic area (POA). GnRH1 released from the POA stimulates pituitary release of gonadotropins, which in males causes sperm production and concomitant steroid hormone release from the testes. Kisspeptin, a neuropeptide acting via the kisspeptin receptor (Kiss1r), increases GnRH1 release and is linked to development of the reproductive system in mammals and other vertebrates. In both fish and mammals, kiss1r mRNA levels increase in the brain around the time of puberty but the environmental …

Development Of A Prototype Bayesian Network Model Representing The Relationship Between Fresh Market Yield And Some Agroclimatic Variables Known To Influence Storage Root Initiation In Sweetpotato, Arthur Villordon, Julio Solis, Don Labonte, Christopher Clark
Faculty Publications
A prototype phenology-driven Bayesian belief network (BBN) model, named BxNET, was developed to represent the relationship between fresh market yield (U.S. #1 grade) and agroclimatic variables known to influence the critical storage root initiation stages in 'Beauregard' sweetpotato. This data-driven model was developed from experimental data collected over 3 years of field trials in which management variables were kept as uniform as possible. The BBN was developed assuming that soil moisture measured at the 15-cm depth was not a limiting variable during the first 20 days after transplanting, during which the onset of storage root initiation determined the majority of …

The relevance of loci associated with blood lipids recently identified in European populations in individuals of African ancestry is unknown. We tested association between lipid traits and 36 previously described single-nucleotide polymorphisms (SNPs) in 1,466 individuals of African ancestry from Spanish Town, Jamaica. For the same allele and effect direction as observed in individuals of European ancestry, SNPs at three loci (1p13, 2p21, and 19p13) showed statistically significant association (p < 0.05) with LDL, two loci (11q12 and 20q13) showed association with HDL cholesterol, and two loci (11q12 and 2p24) showed association with triglycerides. The most significant association was between a SNP at 1p13 and LDL cholesterol (p = 4.6 × 10-8). This SNP is in a linkage disequilibrium region containing four genes (CELSR2, PSRC1, MYBPHL, and SORT1) and was recently shown to relate to risk for myocardial infarction. Overall, the results of this study suggest that much of the genetic variation which influences blood lipids is shared across ethnic groups. © 2010 Springer-Verlag.
