Sperm cryopreservation is a highly efficient method for preserving genetic resources. It extends the reproductive period of males and significantly reduces costs normally associated with maintenance of live animal colonies. However, previous zebrafish (Danio rerio) cryopreservation methods have produced variable outcomes and low post-thaw fertilization rates. To improve post-thaw fertilization rates after cryopreservation, we developed a new extender and cryoprotective medium (CPM), introduced quality assessment (QA), determined the optimal cooling rate, and improved the post-thaw in vitro fertilization process. We found that the hypertonic extender E400 preserved motility of sperm held on ice for at least 6 h. We implemented …

The sterile insect technique (SIT) has been used for the control or eradication of target insect pests. To successfully apply SIT, it is very important to clarify the effect of irradiation on male reproduction in the target pest, because their mating and spermiogenesis abilities affect the success of eradication program. The sweetpotato weevil, Cylas formicarius (Fabricius) (Coleoptera: Brentidae) is a notorious and worldwide pest of sweet potato. Diarrhea remains the second leading cause of death among children under five globally, about one in five children’s deaths are attributable to it. In recent years,’ probiotics have been used as an alternative therapy measure to manage the situation. A promising delivery method that entraps probiotics in infant formula may help prevent or reduce diarrhea. © Yu et al. The adipokine leptin acts on the brain to regulate energy balance but specific functions in many brain areas remain poorly understood. Among these, the preoptic area (POA) is well known to regulate core body temperature by controlling brown fat thermogenesis, and we have previously shown that glutamatergic, long-form leptin receptor (Lepr)-expressing neurons in the POA are stimulated by warm ambient temperature and suppress energy expenditure and food intake. The state of Louisiana has become the lead producer of alligator skins and meat in the United States. More than 300,000 alligators are harvested annually from farm and wild sources. Simultaneously, the industry generates thousands of pounds of waste during the processing of alligators. This waste contains valuable fibrous proteins such as keratins. This study aimed to identify keratin in keratin-rich extracts prepared from alligator backstraps. Alligator backstrap samples were collected from alligator processing facilities located in Springfield, Louisiana. Changes to the embryonic environment that occur within the first few days of development can have significant effects on subsequent fetal development. The following studies investigate the effects of progesterone-deprivation during the early preimplantation period in the mare. The first study described embryo morphology (diameter, stage, and quality grade) collected from mares with induced aluteal cycles. The second study evaluated the effect of induced aluteal cycles on the future fertility of embryo collections as a potential method for cycle manipulation of embryo donor mares. Starch retrogradation is an important factor affecting cake firmness. Effects of hydroxy-propyl-methylcellulose (HPMC) at 0.5% (w/w) or extra virgin coconut oil (EVCO) at 20% (w/w), either alone or together, on physicochemical properties and starch crystallinity of sponge cake during storage was investigated. Addition of EVCO increased specific volume and expansion ratio, while addition of HPMC increased water activity of the freshly baked sponge cake. Changes in firmness, moisture, and spin-lattice relaxation time from nuclear magnetic resonance (T1) of sponge cakes followed a first-order kinetic reaction. Ulcerative colitis (UC) is a disabling inflammatory bowel disease. UC is characterized by chronic, relapsing inflammation of the colon and rectum. Current treatments such as monoclonal antibodies against TNF-α, IL-6, IL-12/p40, adhesion molecules, Janus kinases (JAK) inhibitors have side effects or lose their effects over time. Alternative approaches with fewer side effects for patients are needed. Montmorency tart cherries (Prunus cerasus) are a good source of anti-inflammatory flavonoids. Minimizing nitrogen (N) loss is critical for improving N use efficiency (NUE) in crop production and reducing its effects on the environment. Lake Pontchartrain provides wintering habitat for a continentally-significant proportion of lesser scaup (Aythya affinis). Mid-winter aerial surveys indicate dramatic variation in annual scaup abundance (0–1,194,907), though the mechanisms driving fluctuations have not been previously investigated. In response to natural and anthropogenic disturbances, species composition, abundance, and size structure of benthic invertebrate populations in Lake Pontchartrain exhibit drastic variability among years. Given that wintering scaup feed primarily on mollusks, environmental disturbances have the potential to influence scaup dynamics on the estuary by altering the benthic community. Bagged media products are readily available at garden retails. However, most need to be amended or have supplemental topsoil combined for use in raised beds. Three media mixes were evaluated for the growth and yields of lettuce, broccoli and cauliflower. Two tested media were commercially available purchased products and the third was developed at Louisiana State University Agriculture Center (LSU AgCenter) specially designed for raised bed vegetable production. The LSU AgCenter medium produced greater yield and plant growth in all three crops (p ≤ 0.05) as compared to the commercially available media. Since its first outbreak in sorghum in 2013, severe infestations have spread throughout the southern U.S. causing high economic losses. While insecticidal control has mitigated some of the impacts of this pest, a sustainable ecology-based management program is needed to reduce reliance on chemical control. Rivers carry terrestrial nutrients and other materials to estuaries, affecting biogeochemical cycles and ecosystems in coastal margins. This dissertation research incorporated five studies to investigate the dynamics of carbon, nutrients, and trace metals in four major coastal rivers in southwest Louisiana – the Sabine, Calcasieu, Mermentau, and Vermilion Rivers. The overarching goal of this research was to broaden the knowledge and understanding of biogeochemical processes in freshwater – saltwater mixing zones. Dense infestations of the free-floating aquatic fern giant salvinia (Salvinia molesta D.S. Mitchell) have been expanding across the southeastern United States for over two decades. Although herbicide programs have provided relief to giant salvinia infested areas, morphology and growth characteristics make management difficult. A series of mesocosm trials were conducted to explore alternative management strategies for giant salvinia. Evaluation of non-registered aquatic herbicides showed that rates of sulfometuron and metsulfuron controlled giant salvinia and provided a 98 to 100% biomass reduction. An outdoor mesocosm trial evaluated potential glyphosate resistance in giant salvinia in select water bodies throughout Louisiana. Differences …

OBJECTIVE Improved blood pressure control and use of renin-angiotensin-aldosterone system blockers have altered the clinical presentation or phenotype of chronic kidney disease (CKD) in U.S. adults with diabetes. These changes may influence mortality. RESEARCH DESIGN AND METHODS Data from theNationalHealth and Nutrition Examination Surveys (NHANES) 1988-2006 were used to examinemortality trends in adults with diabetes, defined as physician diagnosis, fasting glucose ≥126 mg/dL, HbA1c >6.5% (48 mmol/mol), or use of glucose-lowering medications. Background: To test the hypothesis that p62 is an optimal target for autophagy inhibition and Verteporfin, a clinically available drug approved by FDA to treat macular degeneration that inhibits autophagy by targeting p62 protein, can be developed clinically to improve therapy for advanced prostate cancer. Methods: Forced expression of p62 in PC-3 cells and normal prostate epithelial cells, RWPE-1 and PZ-HPV7, were carried out by transfection of these cells with pcDNA3.1/p62 or p62 shRNA plasmid. The primary purpose of this study was to examine Louisiana high school students’ perceptions towards consuming whole grain food products. The findings of this study are presented by objectives in the following sections. There is an obesity epidemic in America. Despite the wealth of information, this issue continues to have a negative impact on many adolescents particularly those of low socio-economic status in America. A possible cause for this problem is that adolescents may not have been taught to select the proper foods for consumption.
