Sugarcane (Saccharum spp.) is an important perennial grass crop for both sugar and biofuel industries. The Louisiana sugarcane breeding program is focused on improving sugar yield by incrementally increasing genetic gain. With the advancement in genotyping and (highthroughput) phenotyping techniques, genomic selection is a promising marker-assisted breeding tool. In this study, we assessed ridge regression best linear unbiased prediction (rrBLUP) and various Bayesian models to evaluate genomic prediction accuracy using a 10-fold cross validation on 95 commercial and elite parental clones from the Louisiana sugarcane breeding program. Datasets (individual and pooled in various combinations) were constructed based on soil type …

The potential of genomic selection (GS) to increase the efficiency of breeding programs has been clearly demonstrated; however, the implementation of GS in rice (Oryza sativa L.) breeding programs has been limited. In recent years, efforts have begun to work toward implementing GS into the Louisiana State University (LSU) Agricultural Center rice breeding program. One of the first steps for successful GS implementation is to establish a suitable marker set for the target germplasm and a reliable, cost-effective genotyping platform capable of providing informative marker data with an adequate turnaround time. The objective of this study was to develop a …

Pro-opiomelanocortin (POMC) neurons are important for the regulation of body weight and glucose balance. The inhibitory tone to POMC neurons is mediated primarily by the GABA receptors. However, the detailed mechanisms and functions of GABA receptors are not well understood. The α5 subunit of GABAA receptor, Gabra5, is reported to regulate feeding, and we found that Gabra5 is highly expressed in POMC neurons. To explore the function of Gabra5 in POMC neurons, we knocked down Gabra5 specifically from mature hypothalamic POMC neurons using the clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 strategy. This POMC-specific knock-down of Gabra5 did not affect …

Recent long-term studies in protected areas have revealed the loss of biodiversity, yet the ramifications for ecosystem health and resilience remain unknown. Here, we investigate how the loss of understory birds, in the lowest stratum of the forest, affects avian biomass and functional diversity in the Amazon rainforest. Across approximately 30 years in the Biological Dynamics of Forest Fragments Project, we used a historical baseline of avian communities to contrast the avian communities in today's primary forest with those in modern disturbed habitat. We found that in primary rainforest, the reduced abundance of insectivorous species led to reduced functional diversity, …

In this Article, we reported genome sequencing data from five Sumatran and five Bornean orang-utans. In re-analysing these data, Banes et al.1 found that eight of the ten samples were mislabelled, and we wish to correct the original publication according to their findings. In addition, Banes et al.1 find that one of the orang-utans that we had labelled as a male Sumatran is in fact a female Tapanuli orang-utan (an orang-utan species that was unknown at the time of publication of our original Article and that was first reported in 2017 (ref. 2)). While we here correct the original tables …

The estuarine environments surrounding coastal Louisiana create favorable conditions for microbially mediated mercury (Hg) methylation and subsequent bioaccumulation by biota. In 2010, the Deepwater Horizon (DWH) oil spill released large amounts of oil which, despite having low Hg concentrations, had the potential to influence methylmercury (MeHg) bioavailability in the coastal zone. To explore this possibility, we assessed Hg concentrations and trophodynamics in the coastal Louisiana food web prior to and immediately following the DWH oil spill and compared these metrics with an adjacent coastal ecosystem in the northern Gulf of Mexico. We found no differences in MeHg concentrations between oysters …

Role Of Nucleobindin-2/Nesfatin-1 In The Regulation Of Energetic Status And Maternal Mouthbrooding In The Cichlid Fish, Astatotilapia Burtoni, Saachi Chugh

LSU Master's Theses

Feeding and energetics are fundamental processes in all species that must also be balanced with reproductive investment. The peptide hormone nesfatin-1 is encoded by the nucleobindin-2(nucb2) gene and has recently emerged as a key player involved in both feeding and reproduction across vertebrates. How nesfatin-1 might help regulate trade-offs between feeding and reproduction, however, are relatively unexplored. The female cichlid fish Astatotilapia burtoniis an ideal system to examine the role of nesfatin-1 in regulating metabolic and reproductive functions because this species cycles between a gravid feeding state while preparing to mate, followed by a forced starvation …

Louisiana is one of the most hazard-prone states in the U.S., and many of its people are engaged directly or indirectly in agricultural activities that are impacted by an array of weather hazards. However, most hazard impact research on agriculture to date, for Louisiana and elsewhere, has focused on floods and hurricanes. This research develops a method of future crop loss risk assessment due to droughts, extreme low and high temperatures, hail, lightning, and tornadoes, using Louisiana as a case study. This approach improves future crop risk assessment by incorporating historical crop loss, historical and modeled future hazard intensity, cropland …

Assessing The Current State Of Louisiana's Crawfish Fishery: Trends And Challenges In Wild Capture And Aquaculture, Mahala G. Gambill

LSU Master's Theses

Crawfish are found throughout the southern U.S.; however, Louisiana is by far the largest producer. The state’s industry includes harvesting wild crawfish from natural habitats such as bayous, swamps, and marshes, and crawfish reared in outdoor ponds. Although commercial harvest records are available, characterization of Louisiana’s wild capture crawfish fishery in recent years is undocumented, and analysis of environmental variables that could be correlated with annual harvest totals have not yet been explored. Using trip ticket data from the Louisiana Department of Wildlife and Fisheries, I characterized monthly and annual trends within Louisiana basins to assess changes in crawfish harvest …

Lower ambient temperature (Ta) requires greater energy expenditure to sustain body temperature. However, effects of Ta on human energetics may be buffered by environmental modification and behavioral compensation. We used the IAEA DLW database for adults in the USA (n = 3213) to determine the effect of Ta (−10 to +30°C) on TEE, basal (BEE) and activity energy expenditure (AEE) and physical activity level (PAL). There were no significant relationships (p > 0.05) between maximum, minimum and average Ta and TEE, BEE, AEE and PAL. After adjustment for fat-free mass, fat mass and age, statistically significant (p < 0.01) relationships between TEE, BEE and Ta emerged in females but the effect sizes were not biologically meaningful. Temperatures inside buildings are regulated at 18–25°C independent of latitude. Hence, adults in the US modify their environments to keep TEE constant across a wide range of external ambient temperatures.

A generalized additive modeling (GAM) framework was used to characterize fish-habitat relationships and develop habitat suitability maps to predict spatiotemporal and ontogenetic shifts in Gulf sturgeon distribution within an impacted estuary in the northern Gulf of Mexico, Lake Pontchartrain. Gulf sturgeon (n = 103) were fitted with acoustic transmitters in the Pearl River during the early summer and fall from 2013 to 2018, and an array of acoustic receivers (n = 81) was used to monitor Gulf sturgeon habitat use and movement in the estuary from 2016 to 2019. Daily presence data from the telemetry array were paired with environmental …

Elucidating Factors Affecting Cercosporin Production In Cercospora Leaf Blight Of Soybean To Develop Better Tools For Disease Management, Maria Izabel Costa De Novaes Aug 2022

Elucidating Factors Affecting Cercosporin Production In Cercospora Leaf Blight Of Soybean To Develop Better Tools For Disease Management, Maria Izabel Costa De Novaes

LSU Doctoral Dissertations

Fungal toxins are pathogenicity/virulence factors for many plant diseases, including Cercospora leaf blight (CLB) of soybean (Glycine max (L.) Merr.). The pathogen, Cercospora cf. flagellaris, produces a perylenequinone toxin, cercosporin, which is activated by light to produce damaging reactive oxygen species. To identify novel solutions for CLB management, we studied factors that may impact the biology of cercoporin: fungal lipid droplets (LDs) and mineral and trace element composition (ionome) of the host. Furthermore, since artificial inoculation with the pathogen has been challenging, we also explored the usefulness of a previously developed cercosporin-based assay to study disease resistance. LDs …

Investigating Preferences And Behavior Within Environmental Economics, Sapana Bastola Aug 2022

Investigating Preferences And Behavior Within Environmental Economics, Sapana Bastola

LSU Doctoral Dissertations

To respond to the environmental problems posed by disposable plastic bags, several mechanisms exist, such as bag taxes and bans, but such policies are infeasible in several U.S. states and municipalities. This study uses a social quasi-experiment to examine the effect of a voluntary token-donation program, which reduces disposable plastic bag use by providing a small charitable donation incentive. We find that the token-donation program reduces the probability of disposable bag use by 11.4–12.9 percentage points, representing about a 30%–34% decrease in bag use. Our results demonstrate effective mechanisms to reduce plastic bags exist without government-mandated policy.

Despite the widespread …

The Effects Of Exercise On Innate Lymphoid Cell Metabolism And Function In Cancer, Eunhan Cho Aug 2022

The Effects Of Exercise On Innate Lymphoid Cell Metabolism And Function In Cancer, Eunhan Cho

LSU Doctoral Dissertations

Immunosenescence refers to the gradual decrease in immune competency over time, which is strongly associated with increased risk of cancer and a rise in morbidity and mortality among the elderly. Regular physical activity can be beneficial to the immune system by potentially decreasing the frequency of senescent immune cells and reducing mortality risk in cancer patients and survivors. However, less is known on the impact of acute exercise on “aggressive” cancers, such as hormone receptor negative breast cancers. Further, the mechanistic underpinnings of the exercise-associated improvements in the immune response to cancer remains unclear. In this context, past studies have …

In this paper I attempt to contribute to the developing field of “political philosophy of mind.” To render concrete the notion of “affective frame,” a social situation which pre-selects for salience and valence of environmental factors relative to a subject’s life, I conduct a case study of a deleterious socially instituted affective frame, which, during the early days of the COVID19 pandemic in the United States, produced individuated circumstances that came crashing down on “essential workers” who were forced into a double bind. We saw here an untenable and ultimately fatal situation that forced a choice between, on the one …

Application Of Organic Fertilizers For Lawn Establishment And Maintenance In Louisiana, Matthew Lambert

LSU Master's Theses

Landscape contractors are increasingly interested in organic products for Louisiana's residential and commercial turfgrass areas. However, the use, motivations, and barriers to adopting organic practices in the commercial turfgrass and landscape industry are undocumented. A survey with Louisiana Turfgrass Association (LTA) members to gauge their perceptions on current and future use of organic products in Louisiana was performed; and a field trial was conducted to evaluate organic fertilizers during turfgrass establishment. Substantial majorities of turfgrass professionals are currently applying organic fertilizers and believe organic product use will increase due to consumer demands, potential governmental regulation, and the belief that they …

The Association Of Cuzd1 And Stat5 In Adipocytes, Zachary Wigger Aug 2022

The Association Of Cuzd1 And Stat5 In Adipocytes, Zachary Wigger

LSU Master's Theses

Obesity is a metabolic disorder that has turned into a global epidemic over the past few decades. Adipocyte dysfunction brought on by obesity can lead to increased risk of metabolic diseases such as cardiovascular disease and type 2 diabetes mellitus (T2DM). Knowing how adipocytes function to regulate metabolic homeostasis is critical. CUZD1 (CUB and Zona Pellucida-like Domain-containing protein 1) has not been studied in the context of adipocyte biology but could play a role in several metabolic processes. CUZD1 has been suggested to be a biomarker for several inflammatory diseases and cancers but little functional data has been published. There …

Nuclear war would produce dire global consequences for humans and our environment. We simulated climate impacts of US-Russia and India-Pakistan nuclear wars in an Earth System Model, here, we report on the ocean impacts. Like volcanic eruptions and large forest fires, firestorms from nuclear war would transport light-blocking aerosols to the stratosphere, resulting in global cooling. The ocean responds over two timescales: a rapid cooling event and a long recovery, indicating a hysteresis response of the ocean to global cooling. Surface cooling drives sea ice expansion, enhanced meridional overturning, and intensified ocean vertical mixing that is expanded, deeper, and longer …

Atmospheric soot loadings from nuclear weapon detonation would cause disruptions to the Earth’s climate, limiting terrestrial and aquatic food production. Here, we use climate, crop and fishery models to estimate the impacts arising from six scenarios of stratospheric soot injection, predicting the total food calories available in each nation post-war after stored food is consumed. In quantifying impacts away from target areas, we demonstrate that soot injections larger than 5 Tg would lead to mass food shortages, and livestock and aquatic food production would be unable to compensate for reduced crop output, in almost all countries. Adaptation measures such as …

The Need For A Framework Addressing The Temporal Aspects Of Fish Sperm Motility Leading To Community-Level Standardization, Harvey Blackburn, Leticia Torres, Yue Liu, Terrence R. Tiersch Aug 2022

The Need For A Framework Addressing The Temporal Aspects Of Fish Sperm Motility Leading To Community-Level Standardization, Harvey Blackburn, Leticia Torres, Yue Liu, Terrence R. Tiersch

Faculty Publications

Motility is a widely available parameter that can be used to assess sperm quality of aquatic species. Sperm from fishes with external fertilization usually undergo a dynamic and short-lived period of motility after activation. The common practice of assigning a single value at an arbitrary peak of motility presents challenges for reproducibility, community-level standardization, and comparisons across studies. This study aimed to explore statistical approaches to standardize motility reporting, and to develop an initial framework for community-level standards. Sperm samples from 14 zebrafish (Danio rerio) with a total of 21,705 cells were analyzed by use of computer-assisted sperm analysis with …

Type 1 diabetes (T1D) is classified as an autoimmune disease where pancreatic b-cells are specifically targeted by cells of the immune system. The molecular mechanisms underlying this process are not completely understood. Herein, we identified that the Icam1 gene and ICAM-1 protein were selectively elevated in female NOD mice relative to male mice, fitting with the sexual dimorphism of diabetes onset in this key mouse model of T1D. In addition, ICAM-1 abundance was greater in hyperglycemic female NOD mice than in age-matched normoglycemic female NOD mice. Moreover, we discovered that the Icam1 gene was rapidly upregulated in response to IL-1β …

Obesity Management In The Primary Prevention Of Hepatocellular Carcinoma, Elizabeth R.M. Zunica, Elizabeth C. Heintz, Christopher L. Axelrod, John P. Kirwan

Division of Basic Science Faculty Publications

Hepatocellular carcinoma (HCC) is the most frequent primary hepatic malignancy and a leading cause of cancer-related death globally. HCC is associated with an indolent clinical presentation, resulting in frequent advanced stage diagnoses where surgical resection or transplant therapies are not an option and medical therapies are largely ineffective at improving survival. As such, there is a critical need to identify and enhance primary prevention strategies to mitigate HCC-related morbidity and mortality. Obesity is an independent risk factor for the onset and progression of HCC. Furthermore, obesity is a leading cause of nonalcoholic steatohepatitis (NASH), the fasting growing etiological factor of …

A broad diversity of biological organisms and systems interact with soil in ways that facilitate their growth and survival. These interactions are made possible by strategies that enable organisms to accomplish functions that can be analogous to those required in geotechnical engineering systems. Examples include anchorage in soft and weak ground, penetration into hard and stiff subsurface materials and movement in loose sand. Since the biological strategies have been 'vetted' by the process of natural selection, and the functions they accomplish are governed by the same physical laws in both the natural and engineered environments, they represent a unique source …

Midlife women experience changes in cardiometabolic, physical, and psychosocial health during menopause that negatively impacts their overall quality of life. Factors that contribute to these increases in cardiometabolic risk include weight gain as well as increases in fat mass (particularly abdominal adiposity), insulin resistance, and vascular dysfunction. Other deleterious changes in physical health (e. g. reduced sleep health, bone density, and balance) as well as changes in psychosocial health (e. g. mood, anxiety, and depression) often coincide and are linked to these increases in cardiometabolic risk. Physical activity and exercise are important lifestyle components that have been demonstrated to improve …

Qualifying Factors That Predict Weight Loss Success Within Diet Interventions, Jordan Losavio Jul 2022

Qualifying Factors That Predict Weight Loss Success Within Diet Interventions, Jordan Losavio

LSU Master's Theses

Evidence-based diet strategies lead to weight loss for some, while for others, the same diet approach could result in trivial weight loss or even in weight gain. This prompts the underlying research question: what factors predict that a given diet strategy will be efficacious for some, but not for others? This research was a secondary analysis of clinical weight loss studies to determine which factors influence an individual’s weight loss success for three diet intervention strategies: low-carbohydrate, low-fat, and low-calorie. A combination of physiological, psychological, behavioral, and dietary factors was evaluated for each diet study. Multiple linear regression and logistic …
