Interactive Effects Of Co2, Temperature, And Nitrate Limitation On The Growth And Physiology Of Marine Cyanobacterium Synechococcus Sp. Ccmp 1334, Alyssa K. Sharbaugh

LSU Master's Theses

The marine cyanobacterium Synechococcus sp. CCMP 1334 was grown in a continuous culture system on a 12:12 h light:dark cycle at all combinations of low and high pCO2 (400 and 1000 ppmv, respectively), nitrate availability (nitrate-limited and nutrient-replete conditions), and temperatures of 21°C, 24°C, 28°C, 32°C, and 35°C. The maximum median nutrient-replete growth rate was ~1.15 d−1 at 32 –35°C. Median growth rates at 1000 ppmv pCO2 were higher than those at 400 ppmv at all temperatures, but most of the differences were statistically insignificant. Carbon:nitrogen ratios were independent of pCO2 at a fixed relative growth rate but decreased with …

Role Of Electronegativity In Environmentally Persistent Free Radicals (Epfrs) Formation On Zno, Syed Monjur Ahmed, Reuben A. Oumnov, Orhan Kizilkaya, Randall W. Hall, Phillip T. Sprunger, Robert L. Cook Mar 2024

Role Of Electronegativity In Environmentally Persistent Free Radicals (Epfrs) Formation On Zno, Syed Monjur Ahmed, Reuben A. Oumnov, Orhan Kizilkaya, Randall W. Hall, Phillip T. Sprunger, Robert L. Cook

Faculty Publications

Environmentally persistent free radicals (EPFRs), a group of emerging pollutants, have significantly longer lifetimes than typical free radicals. EPFRs form by the adsorption of organic precursors on a transition metal oxide (TMO) surface involving electron charge transfer between the organic and TMO. In this paper, dihalogenated benzenes were incorporated to study the role of electronegativity in the electron transfer process to obtain a fundamental knowledge of EPFR formation mechanism on ZnO. Upon chemisorption on ZnO nanoparticles at 250 °C, electron paramagnetic resonance (EPR) confirms the formation of oxygen adjacent carbon-centered organic free radicals with concentrations between 1016 and 10 …

Problem-Solving In The Preparation Of Teachers For Agricultural Education: A Mixed Methods Study Investigating The Effect Of Metacognitive Awareness And Cognitive Ability On Students Enrolled In An Introductory Agricultural Mechanics Course, Whitney Figland Mar 2024

Problem-Solving In The Preparation Of Teachers For Agricultural Education: A Mixed Methods Study Investigating The Effect Of Metacognitive Awareness And Cognitive Ability On Students Enrolled In An Introductory Agricultural Mechanics Course, Whitney Figland

LSU Doctoral Dissertations

Cognitive and metacognitive ability have been identified as a critical factor that influences individuals to successfully problem-solve. However, recent research has indicated that students do not have the necessary 21st-century skills to be successful in the workplace. This issue has been confounded by the lack of school-based agricultural education (SBAE) teachers’ limited knowledge regarding effectively teaching decision-making skills and the use of teaching methods that promote problem-solving. Therefore, this study sought to understand the effect that cognitive and metacognitive ability had on the problem-solving of students who are enrolled in an Introduction to Agricultural Mechanics course at Louisiana State University. …

The Impacts Of Nature-Based Sediment Restoration Practices On Coastal Wetland Soil Biogeochemistry, Jacob Z. Cheng Mar 2024

The Impacts Of Nature-Based Sediment Restoration Practices On Coastal Wetland Soil Biogeochemistry, Jacob Z. Cheng

LSU Master's Theses

Coastal marshes provide many valuable ecosystem services. However, these wetlands are threatened worldwide, being subject to sea level rise, sediment starvation, subsidence, coastal edge erosion, etc. Many efforts are being made to counteract this wetland loss and preserve the ecosystem services. Two such methods are sediment diversion and the thin layer placement (TLP) of dredged sediment.

Sediment diversion involves reconnecting the main channel of a river body to disconnected wetlands, supplying much needed soil-building materials. However, this method affects the salinity of the waters, introducing large amounts of freshwater into the system. Salinity changes can affect marsh biogeochemistry. Most ecological …

Physical Activity Barriers And Facilitators In African American Women: A Social Ecological Models Perspective, Leah Ann Carter Mar 2024

Physical Activity Barriers And Facilitators In African American Women: A Social Ecological Models Perspective, Leah Ann Carter

LSU Doctoral Dissertations

African American (AA) women experience poor health outcomes that can be addressed by increasing PA engagement. Understanding how PA barriers and facilitators occur at multiple levels of influence may help tailor PA interventions for AA women. Although prior work has established barriers to PA that are unique to African American women, few studies identify the sociocultural norms associated with these impediments. Little is known about the cultural implications of barriers such as haircare maintenance, and public policy.

The primary aim of this dissertation was to capture AA women’s voice about PA barriers and facilitators using the social ecological model framework. …

Atlantic tarpon (Megalops atlanticus) are capable of long-distance migrations (hundreds of kilometers) but also exhibit resident behaviors in estuarine and coastal habitats. The aim of this study was to characterize the spatial distribution of juvenile tarpon and identify migration pathways of adult tarpon in the northern Gulf of Mexico. Spatial distribution of juvenile tarpon was investigated using gillnet data collected by Texas Parks and Wildlife Department (TPWD) over the past four decades. Generalized additive models (GAMs) indicated that salinity and water temperature played a significant role in tarpon presence, with tarpon occurrences peaking in the fall and increasing over the …

During 25 surveys of global Phytophthora diversity, conducted between 1998 and 2020, 43 new species were detected in natural ecosystems and, occasionally, in nurseries and outplantings in Europe, Southeast and East Asia and the Americas. Based on a multigene phylogeny of nine nuclear and four mitochondrial gene regions they were assigned to five of the six known subclades, 2a-c, e and f, of Phytophthora major Clade 2 and the new subclade 2g. The evolutionary history of the Clade appears to have involved the pre-Gondwanan divergence of three extant subclades, 2c, 2e and 2f, all having disjunct natural distributions on separate …

Sweetpotato (Ipomoea batatas L.) is an important staple crop cultivated in over 100 countries, and the storage roots and vines provide food for humans and livestock. Sweetpotato consumption and demand for its value-added products have increased significantly in the last two decades and have led to new cultivar development, expansion in acreage, and increased demand in the United States and its export markets. Despite the known nutritional components and other health benefits, further research is needed to characterize the genetic diversity and chemical composition related to their storage root qualities, essential in developing consumer-preferred cultivars that offer host plant resistance …

Because of the importance of potassium efflux in inflammasome activation, we investigated the role of the two-pore potassium (K2P) channel TREK-1 in macrophage inflammasome activity. Using primary alveolar macrophages (AMs) and bone marrow-derived macrophages (BMDMs) from wild-type (wt) and TREK-1–/– mice, we measured responses to inflammasome priming [using lipopolysaccharide (LPS)] and activation (LPS þ ATP). We measured IL-1b, caspase-1, and NLRP3 via ELISA and Western blot. A membrane-permeable potassium indicator was used to measure potassium efflux during ATP exposure, and a fluorescence-based assay was used to assess changes in membrane potential. Inflammasome activation induced by LPS þ ATP increased IL-1b …

Humans have profoundly impacted the distribution of plant and animal species over thousands of years. The most direct example of these effects is human-mediated movement of individuals, either through translocation of individuals within their range or through the introduction of species to new habitats. While human involvement may be suspected in species with obvious range disjunctions, it can be difficult to detect natural versus human-mediated dispersal events for populations at the edge of a species' range, and this uncertainty muddles how we understand the evolutionary history of populations and broad biogeographical patterns. Studies combining genetic data with archaeological, linguistic and …

Poor fit between models of sequence or trait evolution and empirical data is known to cause biases and lead to spurious conclusions about evolutionary patterns and processes. Bayesian posterior prediction is a flexible and intuitive approach for detecting such cases of poor fit. However, the expected behavior of posterior predictive tests has never been characterized for evolutionary models, which is critical for their proper interpretation. Here, we show that the expected distribution of posterior predictive P-values is generally not uniform, in contrast to frequentist P-values used for hypothesis testing, and extreme posterior predictive P-values often provide more evidence of poor …

Identification Of A Chondrocyte-Specific Enhancer In The Hoxc8 Gene, Stephania A. Cormier, Claudia Kappen Mar 2024

Identification Of A Chondrocyte-Specific Enhancer In The Hoxc8 Gene, Stephania A. Cormier, Claudia Kappen

Division of Basic Science Faculty Publications

Hox genes encode transcription factors whose roles in patterning animal body plans during embryonic development are well-documented. Multiple studies demonstrate that Hox genes continue to act in adult cells, in normal differentiation, in regenerative processes, and, with abnormal expression, in diverse types of cancers. However, surprisingly little is known about the regulatory mechanisms that govern Hox gene expression in specific cell types, as they differentiate during late embryonic development, and in the adult organism. The murine Hoxc8 gene determines the identity of multiple skeletal elements in the lower thoracic and lumbar region and continues to play a role in the …

Advancing ecological restoration assessments requires a more detailed consideration of species interactions and ecosystem processes. Most restoration projects rely on a few metrics not always directly linked with ecological theory. Here, we used Odum's theory of ecosystem development to assess and compare the ecosystem structure and services of created marshes (4–6 years old) with preexisting, reference marshes in a brackish water region of the Mississippi River Delta. We built ecosystem models for created and reference marshes that integrated large datasets of stomach contents, stable isotopes, and taxa abundances. Despite strong resemblance in community structure, created marshes were at an earlier …

Genomic selection (GS) is revolutionizing plant breeding. However, its practical implementation is still challenging, since there are many factors that affect its accuracy. For this reason, this research explores data augmentation with the goal of improving its accuracy. Deep neural networks with data augmentation (DA) generate synthetic data from the original training set to increase the training set and to improve the prediction performance of any statistical or machine learning algorithm. There is much empirical evidence of their success in many computer vision applications. Due to this, DA was explored in the context of GS using 14 real datasets. We …

Assessing The Effectiveness Of Regional Storm Surge Reduction Strategies And Tank Level Structural Mitigation Measures For Aboveground Storage Tanks, Santosh Ghimire, Sabarethinam Kameshwar

Faculty Publications

The effectiveness of regional storm surge reduction strategies and tank-level structural mitigation measures in reducing the failure probability of aboveground storage tanks (ASTs) were studied. Given past failures during flood and hurricane events, several studies have developed fragility models for ASTs. However, the suitability of these fragility models for different hurricane hazard scenarios is unknown. Furthermore, to combat climate change and sea level rise, several regional storm surge reduction strategies are being proposed. However, the effectiveness of these strategies in improving the safety of ASTs is also unknown. So, herein, a framework was proposed that facilitates assessing the suitability of …

Producing Digital Reflections Of Reality Through Intercultural Filmmaking And Multisensory Engagement, Trai Thomas

LSU Doctoral Dissertations

This thesis analyzes the author’s production of three intercultural, multisensory films situated in New Orleans, encompassing elements both narrative and documentary genres. Intercultural cinema exists in a setting where people of different cultural backgrounds live together in power-inflicted spaces of diaspora, colonialism, and cultural apartheid and the development of the story is centered around exoticism, fetishization, and fascination with the other (Marks 2007, 1). Multisensory media primarily engages through visual perception and auditory input, which create cultural knowledge stored as long-term memory files in the brain (Marks 2007).

Calorie restriction (CR) with adequate nutrient intake is a potential geroprotective intervention. To advance this concept in humans, we tested the hypothesis that moderate CR in healthy young-to-middle-aged individuals would reduce circulating biomarkers of cellular senescence, a fundamental mechanism of aging and aging-related conditions. Using plasma specimens from the Comprehensive Assessment of Long-term Effects of Reducing Intake of Energy (CALERIE™) phase 2 study, we found that CR significantly reduced the concentrations of several senescence biomarkers at 12 and 24 months compared to an ad libitum diet. Using machine learning, changes in biomarker concentrations emerged as important predictors of the change …

It is well recognized that patients with severe obesity exhibit remarkable heterogeneity in response to different types of weight-loss interventions. Those who undergo Roux-en-Y gastric bypass (RYGB) usually exhibit more favorable glycemic outcomes than those who receive adjustable gastric banding (BAND) or intensive medical intervention (IMI). The molecular mechanisms behind these observations, however, remain largely unknown. To identify the plasma metabolites associated with differential glycemic outcomes induced by weight-loss intervention, we studied 75 patients with severe obesity (25 each in RYGB, BAND, or IMI). Using untargeted metabolomics, we repeatedly measured 364 metabolites in plasma samples at baseline and 1-year after …

Rice blast, the most destructive disease of cultivated rice world-wide, is caused by the filamentous fungus Magnaporthe oryzae. To cause disease in plants, M. oryzae secretes a diverse range of effector proteins to suppress plant defense responses, modulate cellular processes, and support pathogen growth. Some effectors can be secreted by appressoria even before host penetration, while others accumulate in the apoplast, or enter living plant cells where they target specific plant subcellular compartments. During plant infection, the blast fungus induces the formation of a specialized plant structure known as the biotrophic interfacial complex (BIC), which appears to be crucial for …

Temperature and precipitation have been identified as factors that potentially influence eastern wild turkey (Meleagris gallopavo silvestris) reproduction, but robust analyses testing the relationship between weather parameters and turkey nest success are lacking. Therefore, we assessed how weather influenced turkey daily nest survival using 8 years of data collected from 715 nests across the southeastern United States. We also conducted exploratory analyses investigating if weather conditions during or prior to nesting best predicted nest success. We then assessed the possible implications of climate change through 2041–2060 for future eastern wild turkey daily nest survival and nest success for variables determined …

Negative bias in mark-recapture abundance estimators due to heterogeneity in detection (capture) probability is a well-known problem, but we believe most biologists do not understand why heterogeneity causes bias and how bias can be reduced. We demonstrate how heterogeneity creates dependence and bias in mark-recapture approaches to abundance estimation. In comparison, heterogeneity, and hence estimator bias, is not as problematic for distance sampling and mark-resight methods because both techniques estimate detection probabilities based on a known quantity. We show how the introduction of a known number of individuals planted into a study population prior to a mark-recapture survey can reduce …
