© 2019 Picardo et al. Inspiratory breathing movements depend on pre-Bötzinger complex (preBötC) interneurons that express calcium (Ca 2+ )-activated nonselective cationic current (I CAN ) to generate robust neural bursts. Hypothesized to be rhythmogenic, reducing I CAN is predicted to slow down or stop breathing; its contributions to motor pattern would be reflected in the magnitude of movements (output). We tested the role(s) of I CAN using reverse genetic techniques to diminish its putative ion channels Trpm4 or Trpc3 in preBötC neurons in vivo. Adult mice transduced with Trpm4-targeted short hairpin RNA (shRNA) progressively decreased the tidal volume of …

Investigations focusing on host-ectoparasite interactions in animals have revealed asymptomatic to severe health and fitness consequences suggesting that species mobilize different interspecific response mechanisms. Fewer studies, however, have examined intraspecific responses to ectoparasitic burdens. In this study, we analyzed host health and fitness responses to increasing ectoparasite burdens along with the presence/absence of hemoparasites of free-ranging insular rock iguanas (Cyclura cychlura) in The Bahamas. Using hematology, plasma biochemistry, as well as body condition and growth rate comparisons, we failed to find significant associations of tick burdens with annual growth rate, corticosterone, packed cell volume, total white blood cell, and heterophil, …

The genetic resources of aquatic biomedical model organisms are the products of millions of years of evolution, decades of scientific development, and hundreds of millions of dollars of research funding investment. Genetic resources (e.g., specific alleles, transgenes, or combinations) of each model organism can be considered a form of scientific wealth that can be accumulated and exchanged, typically in the form of live animals or germplasm. Large-scale maintenance of live aquatic organisms that carry these genetic resources is inefficient, costly, and risky. In situ maintenance may be substantially enhanced and backed up by combining cryopreserved germplasm repositories and genetic information …

Eastern wild turkeys (Meleagris gallopavo silvestris) use a polygynous-promiscuous mating system, wherein males compete for mating opportunities and communicate with females via courtship behaviors. One courtship behavior is vocalization (gobbling), which attracts females and signals dominance to other males. However, temporal variation in gobbling activity may be influenced by external stimuli, environmental variation, and hunter activity. Gobbling activity is a key determinant of hunter satisfaction, and gobbling chronology is often used by state agencies to inform regulatory processes. To identify factors influencing gobbling activity, we evaluated daily gobbling chronology on 3 sites in South Carolina, USA (Webb Wildlife Management Area …

One Environmental Health has emerged as an important area of research that considers the interconnectedness of human, animal and ecosystem health with a focus on toxicology. The great whales in the Gulf of Maine are important species for ecosystem health, for the economies of the Eastern seaboard of the United States, and as sentinels for human health. The Gulf of Maine is an area with heavy coastal development, industry, and marine traffic, all of which contribute chronic exposures to environmental chemicals that can bioaccumulate in tissues and may gradually diminish an individual whale's or a population's fitness. We biopsied whales …

Long-duration spaceflights reportedly induce immune dysregulation, which is considered a risk to astronaut safety and mission success. Recent studies have examined the impact of spaceflight on markers of adaptive and innate immunity, but no study, to date, has comprehensively evaluated humoral immunity and serological markers of B cell function. The aim of this study was to characterize changes in B cell numbers and phenotypes, along with plasma Igs and polyclonal free light chains (FLCs)—near-“real-time” biomarkers of Ig synthesis—in response to an ~6-mo mission to the International Space Station (ISS). Whole-blood samples were collected before flight, during flight (“Early flight,” “Mid-flight,” …

Copyright © 2019 The Authors, Recent studies suggest that closely related species can accumulate substantial genetic and phenotypic differences despite ongoing gene flow, thus challenging traditional ideas regarding the genetics of speciation. Baboons (genus Papio) are Old World monkeys consisting of six readily distinguishable species. Baboon species hybridize in the wild, and prior data imply a complex history of differentiation and introgression. We produced a reference genome assembly for the olive baboon (Papio anubis) and whole-genome sequence data for all six extant species. We document multiple episodes of admixture and introgression during the radiation of Papio baboons, thus demonstrating their …

Habitat Usage Patterns Of Bottlenose Dolphins (Tursiops Truncatus) In Terrebonne And Timbalier Bays, Louisiana, Mary Allison Manning
LSU Master's Theses
I coupled fine-scale environmental data with observed behavior and group composition data to examine overall distribution within the bay system and to characterize the habitat associated with foraging and the presence of calves. Semi-isolated populations of bottlenose dolphins (Tursiops truncatus) along Louisiana’s coast are undergoing increased risks from boat traffic, oil spills, land subsidence, and planned water diversions (CPRA 2017). Characterizing the habitat use of bottlenose dolphins in Terrebonne and Timbalier bays, Louisiana, is important given the likely high site fidelity, small home ranges, and low exchange of individuals with nearby coastal populations (Lane et al. 2015, McDonald …

The ability to produce genetically superior livestock has established somatic cell nuclear transfer (SCNT) as an invaluable tool in commercial livestock production. Successful reprogramming of somatic cells towards pluripotency requires the epigenetic marks characteristic of the differentiated cell type first be erased in order to inactivate the somatic cell program and activate the embryonic program. Several small molecules have been shown to improve both the kinetics and efficiency of reprogramming. These chemical modifiers aid in overcoming the “roadblocks” encountered during the reprogramming process by inducing the necessary epigenetic modifications needed to silence the somatic cell genome and completely reactivate the …
Composition And Ecology Of Avian Communities Along Elevational Gradients In Borneo, Ryan Christian Burner
Composition And Ecology Of Avian Communities Along Elevational Gradients In Borneo, Ryan Christian Burner
LSU Doctoral Dissertations
A primary goal of ecology is to explain the diversity and distribution of organisms. Species distributions can be a function of abiotic factors, species interactions, dispersal limitations, and history, but the relative importance of these factors is widely debated. Elevational gradients are useful systems for studying these effects because many of these factors vary predictably with elevation, and because elevational gradients are replicated many times across the earth. However, few quantitative surveys of Southeast Asian bird communities have been conducted along elevational gradients. In this study, I surveyed birds using point counts and measured habitat and temperature across primary forest …

© 2019 The Author(s). Background: The efficiency of drug development defined as a number of successfully launched new pharmaceuticals normalized by financial investments has significantly declined. Nonetheless, recent advances in high-throughput experimental techniques and computational modeling promise reductions in the costs and development times required to bring new drugs to market. The prediction of toxicity of drug candidates is one of the important components of modern drug discovery. Results: In this work, we describe eToxPred, a new approach to reliably estimate the toxicity and synthetic accessibility of small organic compounds. eToxPred employs machine learning algorithms trained on molecular fingerprints to …

Investigations Into Aspergillus Flavus Infection Of Corn And Regulation Of Aflatoxin Production By Volatiles And Biocontrol Strains, Rebecca R. Sweany
LSU Doctoral Dissertations
Aspergillus flavus is an ascomycete fungus impacting agriculture, public and environmental health due to production of acutely-toxic, carcinogenic aflatoxins in oil seed crops, especially corn in Louisiana, and lung infections of immunocompromised patients resulting in a leading cause of AIDS patients’ deaths globally. The most effective aflatoxin mitigation strategy, biocontrol by atoxigenic A. flavus presents several concerns including: dissemination of a human pathogen, possible sexual reproduction and reliance on precise environmental conditions. Several aspects of A. flavus biology were investigated to potentially improve treatments. Studies investigated how genotypically diverse members within the A. flavus population interact with other population members …

There is a global need for access to oral health care. Proper oral health care is necessary for physiological and psychological health, as well as social and economic issues. Major oral diseases that must be screened include caries, cracked teeth, and impacted molars. These diseases are screened and diagnosed using a variety of medical imaging methods, computed tomography (CT) X-ray being the most popular and efficient method. The challenges in screening with X-ray technology include financial and time cost as well as health risk, including radiation exposure induced cancer (REIC). This thesis summarizes research to explore the possibility of overcoming …
Effects Of Shoal Margin Collapses On The Morphodynamics Of A Sandy Estuary, M. R. Hiatt
Effects Of Shoal Margin Collapses On The Morphodynamics Of A Sandy Estuary, M. R. Hiatt
Faculty Publications
Shoal margin collapses of several million cubic meters have occurred in the Western Scheldt estuary, the Netherlands, on average five times a year over the last decades. While these collapses involve significant volumes of material, their effect on the channel-shoal morphology is unknown. We hypothesize that collapses dynamicize the channel-shoal interactions, which could impact the ecological functioning, flood safety, and navigation in the estuary. The objective is to investigate how locations, probability, type, and volume of shoal margin collapse affect the channel-shoal dynamics. We implemented an empirically validated parameterization for shoal margin collapses and tested its effect on simulated estuary …

Coastal wetland restoration can be complex and expensive, so knowing long-term consequences makes it important to inform decisions about if, when, and where to conduct restoration. We determined temporal changes in land gain and loss in receiving basins and adjacent reference areas for two diversions of the Mississippi River in south Louisiana (Davis Pond and Caernarvon initiated in 1991 and 2002, respectively). Water from both diversions went into receiving basins with vegetated areas as did the adjoining reference areas. The results from two different types of satellite imagery analyses demonstrate a net land loss after diversions began. The results were …

Coastal marshes are important carbon sinks facing serious threats from climatic stressors. Current research reveals that the growth of individual marsh plants is susceptible to a changing climate, but the responses of different marsh systems at a landscape scale are less clear. Here, we document the multi-decadal changes in the phenology and the area of the extensive coastal marshes in Louisiana, USA, a representative of coastal ecosystems around the world that currently experiencing sea-level rise, temperature warming, and atmospheric CO2 increase. The phenological records are constructed using the longest continuous satellite-based record of the Earth's ecosystems, the Landsat data, and …

The diamondback terrapin is the only temperate turtle species that exclusively inhabits estuarine environments. Morphological, behavioral and physiological features contribute to the terrapin’s ability to regulate body fluid osmotic pressure in a euryhaline environment. Low integument permeability combined with aquatic–terrestrial shuttling behavior limits passive exchange of water and salts with the environment, and terrapins regulate active uptake of salts via alterations in drinking and feeding behavior. The lachrymal salt gland facilitates excretion of excess sodium (Na+) and chloride (Cl−) ions through active transport mechanisms. We investigated body fluid dynamics, oxygen consumption (V O2) and osmotic status …
