Expression Of The Aspergillus Bimg Gene In Neurospora Crassa, B Csóka, T Kókai Zeke, J Doonan, H Fox, Z Fehér, V Dombrádi

In A. nidulans the bimG gene codes for the catalytic subunit of protein phosphatase 1. The wild type bimG gene was transformed into N. crassa and expressed under the direction of the alcA promoter. The heterologous bimG mRNA and protein were detected in the transformants by RT-PCR and Western blotting, respectively. However, the transformation did not result in detectable changes in phenotype. This work demonstrates that the alcA promoter, a conditional gene expression system widely used in both Aspergillus and higher plants, also functions in N. crassa.

This paper reports an action research study involving preservice elementary teachers enrolled in a college science methods course (ie. the course focussed on how to teach science at elementary school). There was an interactive science center nearby which many local elementary classes regularly visited, so I decided to set my students the task of visiting the center and reporting on it. However, I was unsure as to what outcomes the students would gain from the experience. Selected College Students' Knowledge And Perceptions Of Biotechnology Issues Reported In The Mass Media, Gary J. Wingenbach, Tracy A. Rutherford, Deborah W. Dunsford

The purpose of this study was to determine college students' awareness of and attitudes toward biotechnology issues reported in the mass media. Future agricultural communicators (N = 330) representing 11 land-grant universities in 10 states recorded their knowledge and perceptions of biotechnology issues as reported in the mass media. Respondents were mostly seniors (46%), female (55%), and considered themselves "8" average students (60%). Students achieved only 30% correct responses (M = 3.05) in a knowledge assessment of biotechnology practices, illustrating a lack of knowledge. This study explicates Critical Thinking (CT) in the respiratory care profession. An exploratory factor analysis was performed and seven measured CT behaviors were narrowed to two factors. A cluster analysis followed and three groups of practitioners were found. This empirical typology considerably expands the conception of CT in the professions.

Distance education has made great strides toward enfranchising nontraditional learners. Yet, while technology has continued to evolve and develop, student support services, especially those focused on technology training and assessment, are still a critical need. This study examined a representative sample of undergraduate, graduate, and postbaccalaureate agricultural students to determine the effect of demographics and prior distance education experience on their perceptions of the need for distance education assessment and training and their comfort level with distance learning technologies. Hermann Hesse's novels commonly represent characters' struggles through ideological opposition and conflict towards resolution. The majority of his critics attribute Hesse's interest in, and expression of, this struggle to his lifelong study of Eastern philosophy. However, Hesse's interest in things Eastern need not be taken as the exclusive determinant of the theme of individuation represented in his fiction. This essay argues that Hesse's predilection for elaborating the ideological crises and resolutions of his characters may also be interpreted as reflecting the Western, Hegelian concept of an Absolute Spirit that proceeds through exhaustive dialectical permutations before it becomes conscious of its …

Although Luisa Valenzuela and Borges were friends for many years, it is only recently that we find traces of that friendship in her narrative. "La calesita" and "El otro libro," published in 1998 and 1999 respectively, evoke many of the elements we find in Borges's narrative. The latter story, which is the main focus of this study, employs nearly all the narrative elements we have come to associate with the Argentine master. Nonetheless, a single sign is changed as Borges's male characters are replaced by females in Valenzuela's work. The end result is a tribute to Borges but one that …

This article reports findings from an evaluation study of the USDA Science and Education Impact Fact Sheet Program. The program was established in 1995 to communicate tangible effects of USDAExtension programming to help maintain investment in land-grant and USDA research and educational programs. The single-page Impact Fact Sheets, which address such topics as agriculture and the environment, parenting, and waste management, have been distributed to stakeholders through mail and personal visits and distributed to states for their own use. Field windbreaks have the potential of sequestering large amounts of carbon. Predicting how much carbon would be sequestered in a newly planted windbreak after ten or more years is of interest. The amount of carbon in a tree depends on its biomass. In a pilot study of Nebraska windbreaks, a Markov random field was used to predict the biomass of green ash in windbreaks as a function of soil and climate conditions. Previous research has shown that yield of cotton increases with plant density until density reaches 16 to 20 plants m-2. In 1997 and 1998 cotton was planted at various densities ranging from 2.5 to 22.5 plants m-2 in a RCB design. Ten row feet (3.048 m) of cotton were cut at ground level in each plot and placed into separate bags. Plant density for each plot was found by counting the number of plants in the bag representing that plot. Researchers studying acclimation of cattle to heat stress want to know if exposure to heat stress in controlled chambers will help cattle adjust to climatic conditions in the field. The four parameter nonlinear PET model is used to study the relationship between core body temperature and ambient temperature. This model works well when cattle are challenged by heat stress but the model is less useful for thermoneutral conditions. Studies of the relationship between animal body temperature and air temperature suggest body temperature is essentially unresponsive until a threshold is reached, then it responds dramatically to increasing air temperature. The goal is to estimate the threshold between the thermoneutral plateau and the beginning of the heat stress challenge. One approach is to fit a polynomial to estimate the knot position and use spline functions to perform linear least squares piecewise polynomial fitting. Yellow starthistle is a dominant weed of north-central Idaho canyon grasslands. The distribution of yellow starthistle can be affected by general landscape characteristics, such as land use, as well as specific terrain related features such as elevation, slope, and aspect. Slope and aspect can be considered as indicators of plant community composition and distribution. Hence, these variables may be incorporated into prediction models to estimate the likelihood of yellow starthistle occurrence. This paper addresses measurement issues involved in estimating the benefits from a university wheat breeding program. The conceptual foundation is to estimate the spatial distribution of farm-level yields for varieties in each year, based on Nebraska winter wheat variety test data from 1972 to 2001, with 9-20 locations per year. We can then estimate the state average yield difference that would occur between the portfolios of varieties that farmers actually grew, and the portfolio of varieties excluding those varieties from the university breeding program. We have assessed the impact of 13 different data transformation methods on the performance of four types of clustering methods (partitioning (K-mean), hierarchical distance (Average Linkage), multivariate normal mixture, and non-parametric kernel density) and four cluster number determination statistics (CNDS) (Pseudo F, Pseudo t2, Cubic Clustering Criterion (CCC), and Bayesian Information Criterion (BIC), using both simulated and real gene expression profile data. We found that Square Root, Cubic Root, and Spacing transformations have mostly positive impacts on the performance of the four types of clustering methods whereas Tukey's Bisquare and Interquantile Range have mostly negative impacts.
