Anhydrobiotic organisms such as the brine shrimp Artemia franciscana have developed strategies to survive and thrive under conditions of limited water availability. One strategy is the accumulation of late embryo genesis abundant (LEA) proteins. Cells of Drosophila melanogaster (Kc167), which had previously been genetically modified to express a group 1 LEA protein (LEA1.3) found in A. franciscana, were used to test the impact of water stress due to freezing to -80°C. The purpose of this study was to examine whether a combination of a low volume, moderately-heavy (80% 1RM) back squat combined with whole body vibration would optimize the post activation potentiation (PAP) response compared to a set of low volume, moderately-heavy back squat without vibration in Division I collegiate female volleyball players. This thesis approaches the construction of race through the vantage of one agrarian magazine, the Prairie Farmer. It analyzes the rhetoric of the people who wrote for this magazine to distinguish changing attitudes toward whiteness and blackness in the rural and agricultural Midwest from the end of the Civil War to the Great Migration. While whiteness was equated with what the Prairie Farmer saw as the active, progressive farmer, blackness was associated with stupidity, laziness, and threat to property. From this, the thesis argues we can build a base of knowledge from which to analyze the roots of racism …

Flathead catfish, Pylodictis olivaris, are a popular game and commercial fish in the Midwestern United States. In the lower 322 km of the Wabash River, flathead catfish are commercially and recreationally harvested by Illinois and Indiana fishers. Current management regulations are different between the two states; Illinois and Indiana have minimum length limits of 381 and 254 mm, respectively. Indiana has proposed changing their minimum length limit to 381 mm and allowing only one fish> 889 mm to be harvested per day. The Caterpillar Game: A Classroom-Wide Behavior Intervention, Angela L. Boyle

The Caterpillar Game, which was developed specifically for this study, is a classroom-wide behavior management system that uses reinforcement principles. The purpose of the study was to examine the effectiveness of the Caterpillar Game on first grade students' disruptive behavior. A concurrent multiple-baseline across settings was used to evaluate this intervention. Changes in disruptive behavior and teacher praise were evaluated through live classroom observations. Results suggest that the Caterpillar Game decreased student disruptive classroom behavior and increased teacher praise. Teacher satisfaction for the Caterpillar Game was also positive.

New science standards, the Next Generation Science Standards (NGSS), recently have been released which require students to use modeling and other process skills and cross-cutting concepts to show understanding of core concepts. Understanding by design approach was used to develop an ecology curriculum in line with the NGSS framework, using field studies to involve students in real science. A schoolyard habitat area on Neoga School District property was used for the field studies. The United States Department of Health and Human Services (USDHH; 2011) has declared that the need for organ transplant in minorities is disproportionately high compared to Caucasians. Of the minority groups represented in the United States, African-Americans are the largest group in need of organ transplantation (USDHH, 2011) and, this group spends more time on the donor organ waiting list than any other minority group (Moore, 2007). George Gerbner (1998) dedicated a significant amount of this life attempting to understand the creation and impact of mass media messages on viewers. Out of this research sprang cultivation theory, which holds viewers' realities can be impacted over time by the media environment they inhabit and its overarching messages. While often applied to television, cultivation theory has largely ignored other mediums, specifically video games. Video games have evolved as a medium since the public arcades of the 1980's and now run on high-powered, internet-enabled consoles. Animals possessing tolerance to extreme water stress are termed anhydrobiotes. Many desiccation tolerant organisms respond to water stress by intracellular accumulation of selected sugars such as trehalose and larger macromolecules such as Late Embryogenesis Abundant (LEA) proteins and thereby maintain the cell viability. Evidence indicates that the presence of trehalose and Late Embryogenesis Abundant (LEA) proteins may work synergistically to confer cellular protection during drying in eukaryotic cells. We evaluated any increase in cellular desiccation tolerance by expressing different LEA proteins in a non-desiccation tolerant insect cell line Drosophila melanogaster (fruit fly) (Kc167 cells) in the presence of trehalose.

In sum, when considering the findings of all three chapters of this thesis cumulatively, several implications are worth noting in regards to the relationship between environmental policy and economic competitiveness. First, the notion that adopting stringent environmental standards will impede on a state's ability to remain economically competitive was shown to be the exception, rather than the rule in all of the cases surveyed in this thesis. Moreover, the cumulative research throughout this thesis has shown the depth of complexity that exists between environmental and economic policy networks. Late Embryogenesis Abundant Proteins Protect Human Hepatoma Cells During Acute Desiccation, Shumin Li, Nilay Chakraborty, Apurva Borcar, Michael A. Menze, Mehmet Toner, Steven C. Hand

Expression of late embryogenesis abundant (LEA) proteins is highly correlated with desiccation tolerance in anhydrobiotic animals, selected land plants, and bacteria. Genes encoding two LEA proteins, one localized to the cytoplasm/nucleus (AfrLEA2) and one targeted to mitochondria (AfrLEA3m), were stably transfected into human HepG2 cells. A trehalose transporterwas used for intracellular loading of this disaccharide. Cellswere rapidly and uniformly desiccated to low water content (<0.12 g H2O/g dry weight) with a recently developed spin-drying technique. Immediately on rehydration, control cells without LEA proteins or trehalose exhibited 0% membrane integrity, comparedwith 98% in cells loaded with trehalose and expressing AfrLEA2 or AfrLEA3m; surprisingly, AfrLEA3m without trehalose conferred 94% protection. Cell proliferation across 7 d showed an 18-fold increase for cells dried with AfrLEA3m and trehalose, compared with 27-fold for nondried controls. LEA proteins dramatically enhance desiccation tolerance in mammalian cells and offer the opportunity for engineering biostability in the dried state.

Mindfulness can be most comprehensively defined as a multifaceted, present-moment awareness intervention that capitalizes on self-perceptions (Gehart & McCollum, 2007; Nanda, 2009). Native American practices can be easily translated into counseling mindfulness techniques to aide in the healing journey of multicultural clients (Burks & Robbins, 2011; Durtschi,Rybak & Decker-Fitts, 2009; Garrett et al., 2011; Turner & Pope, 2009). A Medicine Wheel, or Sacred Hoop, is just one of the many ways to help individuals heal through mindfulness practices. Through the use of creating and discussing a healing wheel with clients, counselors can promote a context for understanding, respecting, and valuing …

Induction of HIF-1α by oxygen limitation promotes increased phosphorylation and catalytic depression of mitochondrial pyruvate dehydrogenase (PDH) and an enhanced glycolytic poise in cells. Cobalt chloride and desferrioxamine are widely used as mimics for hypoxia because they increase the levels of HIF-1α. We evaluated the ability of these agents to elicit selected physiological responses to hypoxia as a means to metabolically precondition mammalian cells, but without the detrimental effects of hypoxia. We show that, while CoCl2 does increase HIF-1α in a dose-dependent manner, it unexpectedly and strikingly decreases PDH phosphorylation at E1α sites 1, 2, and 3 (Ser293, Ser300, and …
