This dissertation investigates the development and optimization of pathology-responsive polymersomes for simultaneous enhanced magnetic resonance imaging (MRI) and enzyme replacement therapy (ERT) in the treatment of GM1 gangliosidosis (GM1). While therapeutic strategies exist for similar, non-neuropathic lysosomal storage disorders, implementation towards GM1 faces significant challenges due to the blood-brain barrier.
Herein, we developed and characterized a range of low molecular weight hyaluronic acid-b-polylactic acid (HA-PLA) polymersomes (PSs) capable of encapsulating therapeutic enzymes in their hydrophilic core. Due to the incomplete development of their immune system, newly hatched chicks are vulnerable to a wide range of pathogenic microorganisms, including enteric bacterial pathogens during the early stage of life. To boost their immune system during this critical period, numerous studies suggest early feeding, either in ovo or immediately post-hatch, of nutrients and immunomodulators such as prebiotics, probiotics, phytobiotics, Toll-like receptors (TLRs) ligands, vitamins and minerals. Peanut ring nematode, Mesocriconema ornatum is a nematode pest of peanut which can cause necrotic lesions on pods, pegs, and roots. It has been reported to facilitate secondary infections in peanut and other crops resulting in more significant losses in yield and quality. Aspergillus flavus is a ubiquitous facultative saprophyte that produces aflatoxin, the most problematic mycotoxin for global health and economy. Foster care is a critical intervention for vulnerable children, with approximately 400,000 children entering the system annually in the United States. Parental substance use, second only to neglect, is now a leading cause of foster care entry. While previous research has explored the relationship between socioeconomic factors and foster care, few studies have incorporated spatial approaches at the neighborhood level to understand how factors such as economic inequality, healthcare disparities, and substance use influence foster care entries. Advancing our understanding of genetic, metabolic, and environmental factors in substance use disorder (SUD) could lead to more effective therapies. In 2022, 16.5% of the U.S. population was diagnosed with SUD, yet current treatments lack efficacy for all patients, making a precision medicine approach crucial. This dissertation explores SUD's relationship with tryptophan (TRP) metabolism through the kynurenine pathway (KP) as a potential therapeutic target. Chapter I reviews current knowledge on SUD and KP. This study examines the effectiveness of an Early Warning System (EWS) originally designed for high-poverty schools and its potential impact on improving outcomes for students living in poverty but attending a mid to low-poverty school. The focus is on reducing the graduation rate disparity between economically disadvantaged students and their more affluent peers through intentional parental involvement. The research explores how structured communication between parents, teachers, and students, combined with tailored interventions in the EWS, can enhance student engagement and academic performance. This study examined the financial impacts of the early COVID-19 pandemic among households with children and compared outcomes based on disability status and urban influence. Survey data were collected in fall 2020 by the Urban Institute from mostly rural households participating in the Meals-to-You program, which delivered food to eligible households with children in the U.S. due to school closures. Relative risk contingency analysis and multivariate logistic regression were used to assess financial hardship from the pandemic. Sampled households reporting at least one member with a disability had greater odds of food insecurity, delaying purchases, depleting savings, and difficulty paying …

Nematodes are a growing threat to the multi-billion-dollar strawberry industry in the US due to the lack of safe and effective management methods. Limited information on the distribution of nematodes in fumigated plasticulture strawberry fields further complicates their management. Anthropogenic stressors and climate change are contributing to the rapid deterioration of marine ecosystems and the important ecological services they provide to coastal communities; concomitantly, this allows infectious agents that cause disease to proliferate in marine environments. At the forefront of marine disease ecology are the impacts of pathogens and parasites on commercially and recreationally exploited decapod crustaceans, including the Caribbean spiny lobster Panulirus argus. This thesis aimed to evaluate the effects of interrupting the daily scotophase period on bone health and production in laying hens. Modern commercial laying hens are prone to high rates of bone demineralization due to the constant mobilization of calcium to withstand daily eggshell calcification mechanisms. The industry light standard for laying hens is 16 hours of continuous light (photophase) and 8 hours of continuous darkness (scotophase) daily. During scotophase, physical activity is reduced due to instinctive perching behaviors, and thus, feeding behaviors are halted. However, physiological processes, including digestion, excretion, and reproduction, remain active during scotophase.

Cryptococcus neoformans is a globally distributed fungal pathogen responsible for causing cryptococcal meningitis in immunocompromised individuals. This pathogenic yeast must adapt to changes in temperature upon entering the human host. Septin proteins are conserved filament-forming GTPases that assemble as higher-order complexes at the cell cortex to support cytokinesis and morphogenesis in fungal and animal cells. In C. neoformans, four septin homologs (Cdc3, Cdc10, Cdc11, and Cdc12) assemble at the mother-bud neck, contributing to cytokinesis through poorly understood mechanisms. C. neoformans strains lacking the septins Cdc3 or Cdc12 are viable at 25°C, but fail to proliferate at 37°C, and are …

Commercial propagators have reported relatively poor propagation success, e.g., <75% rooting, for many hemp (Cannabis sativa) cultivars. These rooting percentages have a significant economic impact due to the wasted labor, space and supplies. Also, the market demands 100% of the propagation cells in a tray to be fully rooted, thus repairing and replacing poorly rooted or unrooted cuttings requires significant labor cost. The goal of this thesis was to evaluate several factors that influence adventitious root formation of hemp stem cuttings.

Many newer peanut cultivars offer yield benefits but have larger canopies and are taking 10 or more days longer to reach desired maturity levels. Prohexadione calcium (PC) and twin row planting pattern have been reported to increase the amount of orange, brown or black pods compared to their respective alternatives. Four cultivars were selected and studied based on frequency of use in South Carolina then paired into an earlier maturing group that reached optimal maturity at 135 to 140 days after planting, and a later maturing group that reached optimal maturity at 150 or more days after planting.

High Pressure processing is an innovative technique for preserving a wide variety of foods, including meat, and this method is increasingly used to satisfy consumer demand for quality food products. High-pressure treatment can impact, negatively or positively, various aspects of meat quality, including color, texture, protein denaturation, and microbial stability, particularly when food products are processed at ambient or low temperatures. By minimizing thermal exposure, appropriate high-pressure ranges help to prevent quality and nutrition degradation, reduce microbial contamination, and enhance tenderness.

This two-chapter thesis evaluates the motivations and determinants of the central figures in farmers’ markets and organic markets. The first chapter explores the rise of farmers’ markets popularity through an in-person survey that was designed and distributed to 274 farmers' market consumers. Logit and ordered logit regressions were used to identify the motivations and characteristics that affect the likelihood of being a frequent farmers’ market customer (visiting 5 times or more per season) and spending over $50. Almost all students are expected to return nearly a full grade-level behind and would likely have to learn to navigate a completely new school environment. This study explored the mental health of healthcare practitioners in Kosovo during the COVID-19 pandemic, focusing on the prevalence of stress, depression, and anxiety, and the associated individual, family, and work-related factors. The research employed a cross-sectional design with a sample of 449 healthcare workers from two hospitals in Kosovo. The Depression, Anxiety, and Stress Scale (DASS-21) was used to assess mental health symptoms, while socio-demographic and work-related information was collected through a questionnaire.

Given the rising cost of poultry diets, alternative ingredients like sorghum are crucial for maximizing returns without compromising product quality. Sorghum's drought tolerance and adaptability, along with its nutritional profile, make it a viable corn substitute. This dissertation examines the nutrient composition, amino acid digestibility, and performance of poultry fed with American tannin-free grain sorghum. Chapter II evaluates the impact of sorghum-based diets, with or without phytase superdosing, on broiler performance, including body weight, weight gain, feed intake, and feed conversion ratio.
