Race, aging, and organ impairment are population intrinsic factors that affect drug pharmacokinetic (PK) possibly through changing in function and/or expression of drug metabolizing enzymes and transporters. The impacts of aging and chronic kidney disease (CKD) on CYP3A, but not drug transporters, were demonstrated in Thai population receiving a microdose cocktail containing 5 probe substrates (midazolam, dabigatran etexilate, pitavastatin, rosuvastatin, and atorvastatin). This study therefore aimed to further investigate the impacts of race, aging, and CKD on the drug transporters in Thai subjects, using the physiologically-based pharmacokinetic (PBPK) approach. The population PBPK models were newly developed specifically to 3 groups …
Notch2 Activation Of Full-Length And Truncated Recombinant Jagged1 Fused With Human Igg1 Fc Produced In Mammalian Expression System, Y Tran Nhu
Chulalongkorn University Theses and Dissertations (Chula ETD)
Osteoporosis associated with old ages has emerged in 21st century as an increasing issue. To stimulate the mineral deposition in bone cells, Jagged1 (Jag1) protein has been demonstrated to trigger mineralization via Notch signaling pathway by binding to Notch2 receptor. Therefore, the Jag1 protein is one of promising candidates for osteoporosis treatment. However, Jag1 consisting of 1218 amino acids is infeasible to apply in clinical uses. Here we engineered Jag1 into truncated proteins containing key domains, Delta–Serrate–Lag-2 domain, N-terminal C2 domain and several epidermal growth factor–like domains which could be responsible for the binding of Notch receptor. To ensure the …

Application Of Crispr-Cas12a-Based Detection Of G6pd Mutations In The Study Of Impact Of G6pd Deficiency And G6pd Mutations On The Diagnosis Of Diabetes Using Hemoglobin A1c In Thailand Cohort, Punchalee Mungkalasut
Chulalongkorn University Theses and Dissertations (Chula ETD)
Glucose 6-phosphate dehydrogenase (G6PD) deficiency, a common inherited enzymopathy, is characterized by decreased enzyme activity, affecting hemolytic anemia and shortening the lifespan of erythrocytes. This study investigated the association between G6PD deficiency and its mutations (G6PD ViangchanG871A and G6PD MahidolG487A) with HbA1c levels in a Thai population. We found that fasting plasma glucose (FPG) and fructosamine (FA) in G6PD deficiency (FPG 94.89±31.49 mg/dl, p=0.321 FA 248.65±45.63 µmol/dl p=0.099) including G6PD ViangchanG871A (FBG 87.22±7.11 mg/dl, p=0.536; FA 246.73±20.44 µmol/dl, p=0.076) and G6PD MahidolG487A (FBG 88.87±13.59 mg/dl, p=0.890; FA 243.73±45.21 µmol/dl, p=0.0732) were not significantly different compared with G6PD normal (FBG 88.85±13.25 …

Development Of Polymer Blends Of Poly(Methyl Methacrylate) And Poly(Lactic Acid) As Denture Base Material, Taksid Charasseangpaisarn
Chulalongkorn University Theses and Dissertations (Chula ETD)
Nowadays, poly(lactic acid) (PLA) usage in dentistry is increasing due to its mechanical and physical properties close to those of poly(methyl methacrylate) (PMMA). However, the thermal properties of PLA is poor due to its low glass transition temperature, which leads to distortion when used in high temperatures. The blending of polymers is an easy and cost-effective method to improve the desired properties of polymers. The objective of this study was to develop the PMMA/PLA blend in various compositions for use as denture base materials. The three PMMA/PLA blend compositions (M75, M50, and M25) were conducted by melt blending with a …

Nail Cortisol For Welfare Assessment In Shelter And Owned Cats, Minh Pham Quoc Anh
Chulalongkorn University Theses and Dissertations (Chula ETD)
Shelter cats are vulnerable to stress due to the challenging circumstances they face, including confinement, unfamiliar environments, people, and animals. This vulnerability often leads to welfare issues, emphasizing the importance of assessing and improving their well-being. Measuring cortisol levels, a stress hormone, from nails has emerged as a non-invasive method to evaluate the stress response in humans and other animals. This study aimed to apply nail cortisol for welfare assessment in cats by measuring and comparing nail cortisol concentrations (NCCs) of owned cats and shelter cats living in different conditions. Nails of 31 owned cats (OC), 29 shelter cats living …
