Antibiotic resistance is one of the most pressing threats to public health. Intrinsic resistance to many antimicrobials in Gram-negative bacteria is accomplished by naturally occurring efflux pumps. Efflux pumps are transmembrane proteins that can extrude antimicrobials from bacterial cytoplasm. Efflux pumps recognize a wide range of compounds including antibiotics, antiseptics, pesticides, and hormones. The upregulation of efflux pump genes in response to increasing concentrations of substrates has shown to increase resistance to toxic substrates. We hypothesize that efflux pump substrates other than antimicrobials, such as steroid hormones, will also induce the expression of efflux pump genes and consequently increase resistance …

In the face of a global climate emergency, community parks & recreation and land management agencies are beginning to adopt a critical role in the fight against climate change. This study gathered information on efforts around the world and created a case study guide to examine the climate change mitigation and adaptation strategies of East Bay Regional Park District in the San Francisco Bay Area of California in the United States. The study was conducted in hopes of shedding light on how small agencies can be proactive and contribute to larger efforts, creating greater regional impacts.

In this paper, we investigate the release of heavy metals from sludge produced from an electrical industry using both organic and inorganic acids. Single and sequential extractions were conducted to assess heavy metals in different phases of the sludge. Metal release from sludge was investigated in the presence of three inorganic acids (nitric, sulfuric, and phosphoric) and three organic acids (acetic, malic, and citric) at concentrations ranging from 0.1 to 2.0 mol L−1. Sequential extraction indicated the presence of Cu primarily in the carbonate fraction, Pb in the residual fraction, and Ni in the FeMn oxide fraction. The cumulative release …

For my senior project, I perform data analysis using statistical methods to determine body metrics that correlate with blood glucose levels. Working with Dr. Tina Smilkstein, I take repeat measurements from 6 different volunteers to establish trends in bodily metric data. The data taken includes weight, body fat, pulse rate, VO2, blood glucose, blood pressure, hours slept, and quality of sleep. Using these values, I use the program MiniTab to view results.
A few examples of correlations with blood glucose found in this project are:

Alpine regions are among the hardest hit by global climate change, experiencing rising temperatures at a faster rate than lowland areas. Such fast rates of warming are predicted to cause rapid changes in alpine plant communities, including shifts in plant functional traits along elevation gradients. To investigate these effects within species, we examined the relationship between elevation, aspect, and functional traits of three common alpine plants in California; the shrub Holodiscus discolor var. microphyllus, the forb Calyptridium umbellatum, and the grass Trisetum spicatum. Montane meadows provide essential habitat for a variety of unique species and important ecosystem services in the western United States. Although important, meadows have experienced increased rates of conifer encroachment due to climate change, fire suppression and grazing. To combat meadow degradation from conifer encroachment, land managers have employed various restoration strategies one of which is conifer removal. California red scale (CRS), Aonidella aurantii, is an increasingly injurious insect pest for the California citrus industry due to insecticide resistance, changing weather patterns, and shifting trade regulations. The presence of the insect on fruit, damages the rind of the fruit and high populations can cause dieback of branches as well as lower yields. Pheromone mating disruption of CRS has the potential to alleviate population control concerns and reduce insecticide use. Impedance spectroscopy (IS) is an important tool for cell detection and characterization in medical and food safety applications. In this thesis, the Cal Poly Biofluidics Lab’s impedance spectroscopy system was re-evaluated and optimized for single-cell impedance spectroscopy. To evaluate the IS system, an impedance spectroscopy bioMEMS chip was fabricated in the Cal Poly Microfabcrication lab, software was developed to run IS experiments, and studies were run to validate the system. Losing Our Minds To Madness: Paradigm Changes In Western European Perceptions Of Mental Illness, James Michael Cecil
History
Academia and scholarship of the 20th-century bred a renewed interest in mental illness throughout history. Despite an increase in the literature within the discourse surrounding "madness," scholars have generally failed to understand how and why Western European societies have viewed mental illness in various ways throughout recorded history. This paper argues that there remains an inherent, human desire to reject anything different from humanity, particularly mental illness, which is nearly impossible to fully comprehend. This is especially true in the case of how societies have institutionalized, punished, and subjugated the "mad" individual.

Cell-free protein synthesis (CFPS) using crude lysates has developed into a robust platform technology over the last 60 years to express numerous types of recombinant proteins. The open-nature, elimination of reliance on cell viability, and focus of all energy towards production of the protein of interest represent substantial advantages of CFPS over in vivo protein expression methods. CFPS has provided new opportunities across a series of research fields that include metabolic engineering, therapeutic and vaccine development, education, biosensors, and many more. The intertidal zone is characterized by persistent, tidally-driven fluctuations in both abiotic (e.g., temperature, [O2], salinity) and biotic (e.g., food availability, predation) conditions, which makes this a very physiologically challenging habitat for resident organisms. The magnitude and degree of variability of these environmental stressors differs between intertidal zones, with the most extreme physiological stress often being experienced by organisms in the high intertidal. Given that many of the fluctuating conditions in this environment are primary drivers of metabolic rate (e.g., temperature, [O2], food availability), we hypothesized that sessile conspecifics residing in different tidal zones would exhibit …

The analysis of the spatiotemporal variability of wind power remains limited during the planning stage of an offshore wind farm. This study provides a framework to investigate how offshore wind power varies along the Central California Coast over diurnal and seasonal time scales, which is critical for reliability and functionality of the grid system. We find that offshore wind power in this region peaks during evening hours across all seasons and maximizes in spring and summer. The timing of peak offshore wind power production better aligns with that of peak demand across California than solar and land-based wind power production, …

Celebrating the 200th anniversary of Mary Shelly’s Frankenstein, the Cal Poly English Department and Kennedy Library organized a series of interdisciplinary events including FrankenReads, an all-day public reading of the novel. Spanning twelve hours, members of the Cal Poly community from all colleges participated in the celebration by volunteering to read portions of Frankenstein; or, The Modern Prometheus.

Nitrogen pollution is a growing problem that is detrimental to the environment and the economy. Traditional treatment of nitrogen is a multi-stage process, expensive, operationally intensive, and requires large land areas. This research studied the effects of BiOWiSH® Aqua (Aqua), a biological enhancement product, on the simultaneous nitrification and denitrification process in a membrane aerated bioreactor (MABR) to determine if it is a feasible application for wastewater treatment. The MABR used during experimentation was a small-scale batch reactor with a continuous flow of air through a silicone membrane.

The Linac Coherent Light Source (LCLS) is a free electron laser that is located at SLAC National Accelerator Laboratory. It fires 120 pulses per second, creating x-ray snapshots of materials at the atomic and molecular level as it undergoes any changes or processes. This resulted in many scientific discoveries in chemistry, biology, energy science, and technology. Now, there is a new vision at SLAC regarding LCLS: to develop Simulacrum. Simulacrum is a system that simulates LCLS and its control system. Santa Rosa Island, Channel Islands National Park, was grazed by cattle, sheep, pigs, elk, and deer for 154 years, degrading the island’s vegetation and causing erosion. In 1998, the livestock were removed, and in 2011 the remaining big game animals were removed, allowing ecosystem recovery to begin. This study evaluated growth of two native species planted in 2016, at the cloud forest restoration site on the Soledad Ridge. Baccharis pilularis (coyote brush) was planted as nursery stock from 2-inch pots, while Quercus pacifica (island scrub oak) was planted as sprouted acorns. The state-of-art model-free reinforcement learning algorithms can generate admissible controls for complicated systems with no prior knowledge of the system dynamics, so long as sufficient (oftentimes millions) of samples are available from the environ- ment. On the other hand, model-based reinforcement learning approaches seek to leverage known optimal or robust control to reinforcement learning tasks by mod- elling the system dynamics and applying well established control algorithms to the system model. Environmental elements such as changing rapidly changing temperature, prolonged periods of low to no water exposure, desiccation, predation and increased wave action can influence the diversity of microhabitats that inhabit the littoral zones. When making observations of various shorelines, specifically the physical conditions of the surrounding marine vegetation (i.e. Fucus distichus), an inquiry is made as to the role of Fucus in the amelioration of stressors on the marine habitat during low tide (MLLW), however, due to time considerations, temperature and humidity are the focus of this study. The Astrobiogeochemistry Laboratory (abcLab) is interested in exploring molecular and mineralogical biosignatures in ancient and recent Earth samples to provide evidence of early signs of life. Currently, in the abcLab, scientists are utilizing a lipid biomarker extraction and analysis method to remove organic material from geologic samples taken from several sites around the world including Australia, the United States and West Wales. Interactions between bacteria and phytoplankton are crucial for the cycling of organic matter in marine environments. Around 50% of organic carbon taken up by marine bacteria is converted into inorganic carbon. The uptake of organic carbon by marine bacteria exuded from phytoplankton is a key factor in regulating the marine carbon cycle. One such molecule that is exuded by phytoplankton and then uptaken by marine bacteria is called glycolate - the anion of glycolic acid, a two caron molecule. Planetary Protection involves the development and implementation of policies and requirements to protect solar system bodies from forward and backward contamination during human or robotic space missions. As stipulated by the United Nations Outer Space Treaty (Article IX) and interpreted by the Committee on Space Research, over 100 nations abide by the treaty and its objectives. Invasive species can lead to serious ecological changes. The San Francisco Bay area is one of the most invaded areas in the world due to the commercial shipping industry and recreational water vessels. While the intertidal is not widely invaded, the harbors and docks are. One exception to this is the invasive, colonial filter-feeding bryozoan Watersipora, which has been found at various rocky outer coast sites.
