Notes On Behaviors Of Free-Ranging Black-Footed Ferrets: Conata Basin, South Dakota, 2007–2009, David A. Eads

Western North American Naturalist

During 2007–2009 in the Conata Basin, South Dakota, field studies provided opportunities to accumulate incidental observations of wild-born black-footed ferrets (Mustela nigripes) and their interactions with other species. Here, I describe maternal behaviors by female ferrets; interactions between a female ferret and a rattlesnake (Crotalus viridis viridis); and attacks by ferrets on a horned lark (Eremophila alpestris), a deer mouse (Peromyscus sp.), and a cottontail rabbit (Sylvilagus sp.). Additionally, I present hypotheses for future testing and discuss conservation implications.

Breeding System And Interaccessional Hybridization Of Purshia Tridentata Plants Grown In A Common Garden, Rosemary L. Pendleton, E. Durant Mcarthur, Stewart C. Sanderson Aug 2012

Breeding System And Interaccessional Hybridization Of Purshia Tridentata Plants Grown In A Common Garden, Rosemary L. Pendleton, E. Durant Mcarthur, Stewart C. Sanderson

Western North American Naturalist

Purshia spp. (Rosaceae) comprise a widespread western North American species complex that is important as landscape dominants, wildlife habitat, browse for wild and domestic ungulates, and seed reserves for small mammals. This study examined aspects of the phenology, compatibility, pollination biology, and progeny fruit characteristics of multiple accessions of bitterbrush (Purshia tridentata), as well as a putative hybrid between bitterbrush and Purshia stansburiana. Except for open- and wind-pollination treatments, mass pollination was accomplished at anthesis within white paper sacks, which were used to isolate treatment branches. Bitterbrush appears to be largely self-incompatible and requires some agent, either wind …

Black-Footed Ferrets And Recreational Shooting Influence The Attributes Of Black-Tailed Prairie Dog Burrows, Dean E. Biggins, Shantini Ramakrishnan, Amanda R. Goldberg, David A. Eads Aug 2012

Black-Footed Ferrets And Recreational Shooting Influence The Attributes Of Black-Tailed Prairie Dog Burrows, Dean E. Biggins, Shantini Ramakrishnan, Amanda R. Goldberg, David A. Eads

Western North American Naturalist

Black-tailed prairie dogs (Cynomys ludovicianus) plug burrows occupied by black-footed ferrets (Mustela nigripes), and they also plug burrows to entomb dead prairie dogs. We further evaluated these phenomena by sampling connectivity and plugging of burrow openings on prairie dog colonies occupied by ferrets, colonies where recreational shooting was allowed, and colonies with neither shooting nor ferrets. We counted burrow openings on line surveys and within plots, classified surface plugging, and used an air blower to examine subsurface connectivity. Colonies with ferrets had lower densities of openings, fewer connected openings (suggesting increased subsurface plugging), and more surface …

Black-Footed Ferret Home Ranges In Conata Basin, South Dakota, Travis M. Livieri, Eric M. Anderson

Western North American Naturalist

We estimated annual home ranges and core areas of black-footed ferrets (Mustela nigripes) in Conata Basin, South Dakota, by collecting 834 locations of 28 ferrets (20 females, 8 males) through spotlighting from October 1997 to September 2000. Area-per-observation curves showed that a minimum of 23 locations were needed to estimate fixed-kernel home-range size. Mean 95% and 50% fixed-kernel annual home-range sizes of females (95%: 64.7 ha, SE = 11.6; 50%: 12.7 ha, SE = 3.0) were significantly smaller and less variable than those of males (95%: 131.8 ha, SE = 40.3; 50%: 35.6 ha, SE = 16.5). Minimum …

Evidence For An Historical Occurrence Of The Meadow Jumping Mouse (Zapus Hudsonius Luteus) In The Verde River Watershed, Arizona, Jennifer K. Frey Aug 2012

Evidence For An Historical Occurrence Of The Meadow Jumping Mouse (Zapus Hudsonius Luteus) In The Verde River Watershed, Arizona, Jennifer K. Frey

Western North American Naturalist

The meadow jumping mouse (Zapus hudsonius luteus) is a riparian obligate with a fragmented distribution in the American Southwest. Because of recent rapid declines in populations, it is a candidate for federal protection under the Endangered Species Act. In Arizona, its known distribution has been restricted to the White Mountains region in the east central part of the state. I present evidence for an historical occurrence of jumping mice in the Verde River watershed, Yavapai County, in west central Arizona. Evidence includes record of a specimen collected by Edgar Mearns in the 1880s, a 1944 report of parasites …

Notes On The Distribution Of Bailey's Eastern Woodrat (Neotoma Floridana Baileyi) In Nebraska, Zebadiah R. Graham, Keith Geluso, Russell A. Benedict

Western North American Naturalist

The eastern woodrat (Neotoma floridana) occurs in eastern and central parts of the United States, with 3 subspecies inhabiting Nebraska. Herein, we report on distributional records of N. f. baileyi—an isolated, disjunct subspecies in north-central Nebraska. We searched for diagnostic sign of woodrats in 4 counties in Nebraska and 1 county in South Dakota. We obtained distributional records for N. f. baileyi east, west, and north of known distribution limits in Nebraska, but none in South Dakota. Most distributional records represent range extensions because of lack of prior mammalian surveys in those areas. Observations of woodrats in …

Transgene Delivery Via Microelectromechanical Systems, Aubrey Marie Mueller Wilson Aug 2012

Transgene Delivery Via Microelectromechanical Systems, Aubrey Marie Mueller Wilson

Theses and Dissertations

The invention of pronuclear microinjection initiated the field of transgenic research. Over 30 years later microinjection remains the most straight-forward and most commonly used transgene delivery option. In this work we address the current progress of microelectromechanical systems (MEMS) used as transgenic delivery mechanisms. The nanoinjector is a specially designed MEMS device which uses electrostatic charge to manipulate transgene molecules. The process of nanoinjection was designed as an alternative to microinjection which causes less damage to developing embryos, improves embryo survival, birth rates, and overall efficiency of injections. In vivo testing of nanoinjection demonstrates it is both safe and effective. …

Mathematical Model And Experimental Exploration Of The Nanoinjector Lance Array, Nathan C. Toone Jul 2012

Mathematical Model And Experimental Exploration Of The Nanoinjector Lance Array, Nathan C. Toone

Theses and Dissertations

The Nanoinjector Lance Array has been developed to inject foreign material into thousands of cells at once using electrophoresis to attract and repel particles to and from the electrically-charged lances. A mathematical computer model simulating the motion of attracted or repelled proteins informs the design of the nanoinjection lance array system. The model is validated by accurately predicting protein velocity in electrophoresis experiments. A complete analysis of parameters is conducted via simulations and specific research questions regarding the counter electrode of the nanoinjector lance array system are explored using the model. A novel technique for fabricating lance arrays from collapsed …

Design And Testing Of A Biological Microelectromechanical System For The Injection Of Thousands Of Cells Simultaneously, Gregory Herlin Teichert Jul 2012

Design And Testing Of A Biological Microelectromechanical System For The Injection Of Thousands Of Cells Simultaneously, Gregory Herlin Teichert

Theses and Dissertations

The ability to inject DNA and other foreign particles into cells, both germ cells (e.g. to produce transgenic animals) and somatic cells (e.g. for gene therapy), is a powerful tool in genetic research. Nanoinjection is a method of DNA delivery that combines mechanical and electrical methods. It has proven to have higher cell viability than traditional microinjection, resulting in higher integration per injected embryo. The nanoinjection process can be performed on thousands of cells simultaneously using an array of microneedles that is inserted into a monolayer of cells. This thesis describes the needle array design requirements and the fabrication process …

A Longitudinal Study Of The Effect Of At Birth Adoptions On Anxiety, Stress Hormones And Adolescent Alcohol Intake: A Nonhuman Primate Model, Whitney Faith Maxwell

Theses and Dissertations

Adopted individuals have an increased risk for a variety of psychopathological disorders. Studies of the effects adoption in humans are difficult to perform because of the difficulty separating genetic risk and treatment effects. This is a developmental study investigating the effects of at birth adoption using a nonhuman primate model. Three experimental paradigms were used to assess maternal treatment, stress-related behavior, and physiology late in infancy and again later in life. Rhesus monkeys were reared for their first six months of life by either their biological mother or an unrelated, lactating adult female. Adoptions occurred immediately following birth. At six …

Evaluating The Performance Of Computational Approaches For Identifying Critical Sites In Protein-Coding Dna Sequences, Matthew Lewis Bendall Jul 2012

Evaluating The Performance Of Computational Approaches For Identifying Critical Sites In Protein-Coding Dna Sequences, Matthew Lewis Bendall

Theses and Dissertations

The ability to link a particular phenotype to its causative genotype is one of the most challenging objectives for biological research. Although the genetic code provides an explicit formula for determining the sequence of amino acid phenotypes produced by a given nucleotide sequence, identifying specific residues that are functionally important remains problematic. Many computational approaches have been developed that use patterns observed in DNA sequences to identify these critical sites. However, very few research studies have used empirical data to test whether these approaches are truly able to identify sites of interest. In most empirical studies, the actual protein function and …

Homogeneous Viologens For Use As Catalysts In Direct Carbohydrate Fuel Cells, Dane C. Hansen Jul 2012

Homogeneous Viologens For Use As Catalysts In Direct Carbohydrate Fuel Cells, Dane C. Hansen

Theses and Dissertations

Deriving electrical energy from glucose and other carbohydrates under mild conditions is an important research objective because these biomolecules are abundant, renewable, and can provide 12 to 24 electrons per molecule, yielding substantial electrical power. It was previously observed that disubstituted viologens, salts of N,N'-disubstituted 4,4'-bipyridine, are able to oxidize glucose under alkaline conditions. Building on that initial result, the objective of this work was to understand and quantify the effectiveness and utility of viologens as catalysts for use in direct carbohydrate fuel cells. The extent that viologens oxidize carbohydrates, the conditions under which that oxidation occurs, and the mechanism for …

Stoneflies Of Unusual Size: Population Genetics And Systematics Within Pteronarcyidae (Plecoptera), John S. Sproul

Theses and Dissertations

Chapter 1. The family Pteronarcyidae (Plecoptera) is a highly studied group of stoneflies and very important to a wide variety of aquatic studies. Several phylogenies have been proposed for this group recent decades, however there is little congruence between the various topologies. The present study revises the phylogeny of the group by combining molecular data from mitochondrial cytochrome oxidase subunit II, ribosomal subunit 12S, ribosomal subunit 16S, and nuclear loci ribosomal subinit 18S and Histone H3, with published morphological data in a parsimony-based total evidence analysis. The analysis produced a well-supported phylogeny with novel relationships within the genus Pteronarcys. …

Resolving Disulfide Bond Patterns In Snap25b Cysteine-Rich Region Using Lc Mass Spectrometry, Nozomi Ogawa Jul 2012

Resolving Disulfide Bond Patterns In Snap25b Cysteine-Rich Region Using Lc Mass Spectrometry, Nozomi Ogawa

Theses and Dissertations

A global analysis of the human proteome demonstrates that there are ~5500 tryptic fragments that contain four cysteines in close proximity. Elucidating whether they form disulfide bonds in vivo under different conditions is particularly important because cysteines are known to be a vital cellular redox sensor as well as a catalytic site for important biochemical reactions. However, currently there are no methods that can resolve disulfide patterns in closely-packed cysteine residues from a complex sample. In order to address this problem, we have developed a novel mass-spectrometry-based method to identify the different disulfide bonding patterns possible, using SNAP25B cysteine-rich region …

Evaluating The Effectiveness Of A Workshop/Support Group For Parents Raising Children With Type 1 Diabetes, Christina Marie Yazzie

Theses and Dissertations

The purpose of this study was to evaluate the effectiveness of a workshop/support group for parents raising children with type 1 diabetes mellitus (T1DM). Diagnosis of a chronic disease in a child generally arouses acute anxiety and stress in parents. Parental stress when raising a child with type 1 diabetes is often centered on worry about their child's health and parents' ability to maintain the care activities needed by these children. Parents are the primary caregivers of children with Type 1 diabetes, and the effect of stress on parents can impact the health of both children and parents. The present …

Culvert Roughness Elements For Native Utah Fish Passage: Phase Ii, Suzanne Kim Monk Jul 2012

Culvert Roughness Elements For Native Utah Fish Passage: Phase Ii, Suzanne Kim Monk

Theses and Dissertations

Native fishes have become an increasingly important concern when designing fish passable culverts. Many operational culverts constrict waterways which increase velocities and prevent upstream passage of small fish species. The current method to ensure fish passage is to match the average cross sectional velocity to the sustained swim speed of the fish. This study investigates the passage rates of leatherside chub (Lepidomeda aliciae) and speckled dace (Rhinichthys osculus) at three sites (an arch culvert with substrate bottom, box culvert with bare bottom, and a stream section with no culvert) located on Salina Creek near Salina, UT. It was found that …

Characterization And Preliminary Demonstration Of Microcantilever Array Integrated Sensors, Ryan R. Anderson Jul 2012

Characterization And Preliminary Demonstration Of Microcantilever Array Integrated Sensors, Ryan R. Anderson

Theses and Dissertations

I characterize the behavior of microcantilever arrays which utilize the in-plane photonic transduction that I've previously developed and evaluate the performance of the microcantilever arrays in simple sensing scenarios with integrated microfluidics. First the thermal responses of microcantilevers with a variety of patterns of deposited gold films are compared. Using a scanning electron microscope, I observe the deflection thermal sensitivities of 300 µm long microcantilevers to be -170.82 nm/K for a full gold coating and -1.93 nm/K for no gold coating. Using the photonic transduction method I measure a thermal sensitivity of -1.46 nm/K for a microcantilever array with no …

Plant Establishment And Soil Microenvironments In Utah Juniper Masticated Woodlands, Kert R. Young Jul 2012

Plant Establishment And Soil Microenvironments In Utah Juniper Masticated Woodlands, Kert R. Young

Theses and Dissertations

Juniper (Juniperus spp.) encroachment into sagebrush (Artemisia spp.) and bunchgrass communities has reduced understory plant cover and allowed juniper trees to dominate millions of hectares of semiarid rangelands. Trees are mechanically masticated or shredded to decrease wildfire potential and increase desirable understory plant cover. When trees are masticated after a major increase in tree population density and associated decrease in perennial understory cover, there is a risk that invasive annual grasses will dominate because they are highly responsive to the increased resource availability that commonly follows removal of the main resource user. To determine if tree mastication increases …

Liquid Dielectric Spectroscopy And Protein Simulation, Brett Lee Mellor Jul 2012

Liquid Dielectric Spectroscopy And Protein Simulation, Brett Lee Mellor

Theses and Dissertations

Protein electrical properties have been studied using dielectric relaxation measurements throughout the past century. These measurements have advanced both the theory and practice of liquid dielectric spectroscopy and have contributed to understanding of protein structure and function. In this dissertation, the relationship between permittivity measurements and underlying molecular mechanisms is explored. Also presented is a method to take molecular structures from the Protein Data Bank and subsequently estimate the charge distribution and dielectric relaxation properties of the proteins in solution. This process enables screening of target compounds for analysis by dielectric spectroscopy as well as better interpretation of protein relaxation …

Investigation Of Internal Wave Spectra Due To Observed Interactions, Benjamin Lee Hillyard Jul 2012

Investigation Of Internal Wave Spectra Due To Observed Interactions, Benjamin Lee Hillyard

Theses and Dissertations

Observational data are analyzed and decomposed to reveal internal ocean waves and their interactions with one another. Particularly, the interaction of small-scale internal waves with a large inertia wave packet is examined. Using the governing internal wave equations, an analysis is made of the energy propagation of a small scale internal wave with a large-scale inertia wave. With that, an assessment is made of the frequency of occurrence of various encounter types. Next, the possibility of energy transfer during an interaction is explored. The relative energy of the small wave before interacting with the large-scale inertia wave is calculated and …

Three Dimensional Characterization Of Vocal Fold Fluid Structure Interactions, Joseph R. Nielson Jul 2012

Three Dimensional Characterization Of Vocal Fold Fluid Structure Interactions, Joseph R. Nielson

Theses and Dissertations

Voice quality is strongly linked to quality of life; those who suffer from voice disorders are adversely affected in their social, family, and professional relationships. An effort has been made to more fully understand the physics behind how the voice is created, specifically the fluid structure interactions that occur during vocal fold vibration. Many techniques have been developed and implemented to study both the motion of the vocal folds and the airflow that creates the motion. Until recently these techniques have sought to understand a highly three-dimensional phenomenon with 1D or 2D perspectives. This research focuses on the development and implementation …

Tissue Specific Porcupine Deletion Reveals A Novel Role For Ectodermal Wnts In Musculotendon Development, Aaron P. Smith Jul 2012

Tissue Specific Porcupine Deletion Reveals A Novel Role For Ectodermal Wnts In Musculotendon Development, Aaron P. Smith

Theses and Dissertations

The Wnt family of secreted proteins consists of 19 family members (in the mouse) and is known to signal through multiple pathways that regulate crucial processes in the development of almost all tissues. Dissecting the roles of individual Wnts has been hampered due to functional redundancy that exists between family members. We made use of a conditional allele of the acyltransferase, Porcupine (Porcn), that is required for the secretion of all Wnt ligands, and the Msx2Cre deleter to eliminate the secretion of all Wnt ligands from the ventral limb ectoderm, ventral abdominal ectoderm, and urogenital ectoderm. Phenotypically the …
