Ifn-Γ Increases The Expression Of Sars-Cov-2 Receptors On Vero E6 Cells, Bindu Madhavi Madabattula

Browse all Theses and Dissertations

Respiratory epithelial cells are the initial target for SARS-CoV-2 infection. IFN-γ is known to increase the expression of ACE-2, an initial receptor for SARS-CoV-2, on epithelial cells. This study focuses on examining the effect of IFN-γ for ACE-2, TMPRSS2, and neuropilin-1 on Vero E6 cells using two immunofluorescence methods, namely, direct (membrane) fluorescence method and Cytation5 method. Direct fluorescence was determined using an Accu-Scope and ImageJ analysis. Using this method, significance (p<0.023) was observed only for ACE-2 when Vero E6 cells were treated with IFN-γ. Cytation5 fluorescence was determined using a Bio-tek Cytation5 plate reader. The results showed that IFN-γ significantly increased (p<0.001) the expression of ACE-2, neuropilin-1, and TMPRSS2. These results indicate Cytation5 is a more sensitive method for determining the expression of receptors on Vero E6 cells. The elevated levels of SARS-CoV-2 receptors expression resulting from IFN-γ treatment makes the epithelial cells more susceptible targets of SARS-CoV-2 infection. IFN-γ is most likely provided by innate immune cells in the initial COVID-19 infection, consequently contributing to the severity of disease.

Nitrogen (N) losses from cropping systems negatively affect soil quality/fertility, crop yield, and the environment through its contribution to climate change and water pollution. İt possesses oil-water-atmospheric system challenges to both the too much and the too little N application countries. Soil and climatic factors are the critical drivers of N loss from agricultural systems as they influence major processes like nitrification and denitrification. Other N loss pathways include leaching, runoff, and volatilization. Nitrous oxide (N2O) and ammonia (NH3) gases are the two pollutants resulting from N fertilizers in crop production and represent N loss’ significant pathway. Nitrous oxide is …

Waste accumulation and microbial pollution are key environmental challenges in most developing countries. Land filling, the commonest waste disposal method, is not sustainable due to land scarcity and point source pollution. Thus, a large fraction of organic wastes is not properly disposed, yet the conventional methods of waste recycling are less effective. The degradation of waste using insect life stages (entomocomposting) has emerged as a novel tool that is fast, affordable, and sustainable in recycling organic wastes with maximum pathogen suppression. This bioconversion process also generates an organic fertilizer with potential to enhance soil remediation and provide additional benefits on …

Previous research on Cyrba algerina (Araneae, Salticidae) has shown this jumping spider expresses predatory specialisation with respect to spiders as prey as well as inter-population variation in responsiveness to prey-spider odour. However, this earlier research pertained to a single prey species (Oecobius machadoi) and only field-collected C. algerina individuals were tested. Here we extend the previous research by using laboratory-reared, as well as field-collected, individuals of C. algerina and also by using another prey-spider species, Zelotes thorelli, as well as O. machadoi. Two localities in Portugal are considered, Sintra where C. algerina and both prey species are abundant and Tavira …

We investigated the effect of bee species identity and harvesting methods on the chemical composition and antiradical activity of 53 honey samples, produced by six stingless bee species in western Kenya (Kakamega forest). Our results illustrate that none of the assayed parameters significantly varied between the honey samples harvested by “punching holes” (n = 25) and “squeezing” (n = 28) methods. By contrast, species identity drove significant differences in the assayed parameters. Positive correlations between the antiradical activity and the phytochemicals (phenols and flavonoids) were observed, and honeys from Liotrigona sp. exhibited the highest amounts of phenols (214 mg GAE/100 …

Production of entomopathogenic agents as biopesticides, mainly fungal and some viral agents, has been established in many countries of Asia, South America and Africa using low technology approach, often by small to medium scale producers. While these reduced technology systems can generate effective products, these systems require the application of appropriate production methodology and rigorous monitoring if the appropriate quality and safety standards are to be reached. The role of governments in creating and maintaining a policy and regulatory environment that promotes local entomopathogen (EP) production, marketing and use remains important for success. The wider adoption of EP products in …

Sex determination typically happens via the ploidy of individ-uals: haploids become males and diploids become females. Arrhenotoky is believed to be the ancestral reproduction mode in Hymenopterans, with haploid males produced parthenogenetically, and diploid females produced sexually. However, a number of transitions towards thelytoky (diploid females produced parthenogenetically) have appeared in Hymenopterans, and in most cases populations or species are either totally arrhenotok-ous or totally thelytokous. Here we present the case of Cotesia typhae (Fernandez-Triana), a Braconidae that produces parthenogenetic females at a low frequency. The …

Vector-borne diseases affecting livestock have serious impacts in Africa. Trypanosomosis is caused by parasites transmitted by tsetse flies and other blood-sucking Diptera. The animal form of the disease is a scourge for African livestock keepers, is already present in Latin America and Asia, and has the potential to spread further. A human form of the disease also exists, known as human African trypanosomosis or sleeping sickness. Controlling and progressively minimizing the burden of animal trypanosomosis (COMBAT) is a four-year research and innovation project funded by the European Commission, whose ultimate goal is to reduce the burden of animal trypanosomosis (AT) …
