Gene Expression Analysis Reveals Dysregulation In Traumatic Brain Injury Leading To Tauopathy, Kristina M. Tosi

USF Tampa Graduate Theses and Dissertations

Traumatic Brain Injury (TBI) causes a cascade of detrimental events within neurons, contributing to a host of neurological diseases relating to dementia. In this study we have performed a meta-analysis comparing differentially expressed genes (DEG) in specific cell types such as neurons and microglia revealing inflammation as a common driver of neurological deficiencies post TBI. Inflammation and hypoxia are well characterized features of TBI; however, the molecular mechanisms causing tau hyperphosphorylation by tauopathy associated genes (TAG) post-TBI are currently poorly understood. Furthermore, there are limited in-vitro neuroinflammatory models that mimic primary and secondary TBI. We hypothesize that inducing hypoxia and …

Seed-based revegetation is increasingly pursued in wetland restoration, but seed and seedling mortality is high and can be further limited by environmental stressors, like hydrology and salinity. The interactive effects of hydrology and salinity on native plants, especially in the presence of invasive plants, are less clear. In greenhouse and mesocosm experiments, we tested different hydrologic and salinity conditions on the growth of native seed mixes and Phragmites australis, a widespread wetland invader in North America. The greenhouse experiment tested two mixes (forb mix, graminoid mix), while mixes in the mesocosm experiment combined forbs and graminoids chosen for broad …

Open Access archive

Introduction The limited access to temporal fine structure (TFS) cues is a reason for reduced speech-in-noise recognition in cochlear implant (CI) users. The CI signal processing schemes like electroacoustic stimulation (EAS) and fine structure processing (FSP) encode TFS in the low frequency whereas theoretical strategies such as frequency amplitude modulation encoder (FAME) encode TFS in all the bands. Objective The present study compared the effect of simulated CI signal processing schemes that either encode no TFS, TFS information in all bands, or TFS only in low-frequency bands on concurrent vowel identification (CVI) and Zebra speech perception (ZSP). Methods Temporal fine …

Label-Free Visualization Of Unfolding And Crosslinking Mediated Protein Aggregation In Nonenzymatically Glycated Proteins, Darshan Chikkanayakanahalli Mukunda, Shaik Basha, Meagan Gail D'Souza, Subhash Chandra

Open Access archive

Nonenzymatic glycation (NEG) unfolds and crosslinks proteins, resulting in aggregation. Label-free evaluation of such structural changes, without disturbing molecular integrity, would be beneficial for understanding the fundamental mechanisms of protein aggregation. The current study demonstrates the assessment of NEG-induced protein aggregation by combining autofluorescence (AF) spectroscopy and imaging. The methylglyoxal (MG) induced protein unfolding and the formation of cross-linking advanced glycation end-products (AGEs) leading to aggregation were evaluated using deep-UV-induced-autofluorescence (dUV-AF) spectroscopy in proteins with distinct structural characteristics. Since the AGEs formed on proteins are fluorescent, the study demonstrated the possibility of autofluorescence imaging of NEG-induced protein aggregates. Autofluorescence spectroscopy …
