The Effects Of Gibberellic Acid And Kinetin On Overcoming The Effects Of Juglone Stress On Seed Germination And Seedling Growth, İrfan Terzi̇, İsmai̇l Kocaçalişkan
Turkish Journal of Botany
Juglone (5-hydroxy-1,4-naphthoquinone) is an allelochemical responsible for walnut allelopathy. The effects of gibberellic acid (GA_{3}) and kinetin (KIN) on overcoming the effects of juglone stress on seed germination and seedling growth were investigated in barley, wheat, cucumber, alfalfa, and tomato. Seeds pre-treated with plant growth regulators were used to test their effects on the alleviation of juglone stress. It was observed that seed germination in tomato and wheat was inhibited by juglone and that the plant growth regulators alleviated it significantly. Elongation and dry weight of the seedlings of all the species used in the study were reduced significantly by …

Using Vegetation Units As Salinity Predictors In The Lower Cheliff Algeria, Adda Ababou, Mohammed Chouieb, Mohammed Khader, Khalladi Mederbal, Djamel Saidi
Turkish Journal of Botany
The Lower Cheliff plain is among the largest salted soils in north-western Algeria. In order to establish the relationships between the main soil factors and plant species in this stressed ecosystem, 133 relevés related to soil and vegetation were realized. Soil variables measured included electrical conductivity and calcium carbonate. The 133 relevés were arranged in clusters using k-means classification. The similarity analysis used to examine the variation in vegetation assemblage structure showed significant differences in taxonomical composition among groups of relevés. The phi coefficient of fidelity used then to extract the different vegetation units and to measure species concentration in …

Towards Cloning A Tyrosinase Gene From Pitayas, Kurt J. Chamberlain
Towards Cloning A Tyrosinase Gene From Pitayas, Kurt J. Chamberlain
Honors Program Theses
Betalains, such as those produced by pitayas, are water-soluble nitrogen-containing pigment molecules (Gandia-Herrero et al., 2005). In most families of the Caryophyllales, betalains replace anthocyanins, which are the pigment molecules found in all other families of flowering plants (Strack et al., 2001). Betalains accumulate in the flowers, fruits, and occasionally vegetative tissues of these plants (Strack et al., 2001). Depending on the structure of the molecule, betalains are classified as either betacyanins or betaxanthins. Betacyanins contribute to red-violet coloration, while betaxanthins contribute to the yellow-orange coloration found only in a limited number of plant species (Gandia-Herrero et al., 2005). The …

A Comparison Of Soil And Corn Kernel Aspergillus Flavus Populations: Evidence For Niche Specialization, Rebecca Ruth Sweany
LSU Master's Theses
Aspergillus flavus is an opportunistic fungal pathogen that infects peanuts, cotton, corn and tree nuts. Aspergillus flavus is a major problem globally due to the production of acutely toxic and carcinogenic aflatoxins. Louisiana climatic conditions lead to annual threats of corn aflatoxin contamination. The purpose of this study was to determine the specific ability of different strains of A. flavus to infect corn. Five soil samples and 10 corn ears were collected from each of seven corn fields throughout Louisiana. In addition, Francis Deville of Monsanto Company collected 7, 6, 2, and 4 soil samples and corn ears from four …

Agronomy, Horticulture and Plant Science Faculty Publications
A potential pest of switchgrass, Panicum virgatum L., was first detected in South Dakota in 2004, where death of partially emerged leaves was noted in a small proportion of tillers. Similar “dead heart” symptoms were observed in switchgrass in Illinois during 2008 and adults of a stem-boring caterpillar were collected and identified as Blastobasis repartella (Dietz). In 2009, a survey of the central United States was used to estimate the distribution and abundance of this insect. In eight northern states, B. repartella was consistently found in both cultivated plots and natural stands of switchgrass. In four southern states, B. repartella …

Increased Weed Diversity, Density And Above-Ground Biomass In Long-Term Organic Crop Rotations, Samuel E. Wortman, John L. Lindquist, Milton J. Haar, Charles A. Francis
Department of Agronomy and Horticulture: Faculty Publications
While weed management is consistently a top priority among farmers, there is also growing concern for the conservation of biodiversity. Maintaining diverse weed communities below bioeconomic thresholds may provide ecosystem services for the crop and the surrounding ecosystem. This study was conducted to determine if weed diversity, density and biomass differ within and among organic and conventional crop rotations. In 2007 and 2008, we sampled weed communities in four long-term crop rotations near Mead, Nebraska, United States using seedbank analyses (elutriation and greenhouse emergence) and above-ground biomass sampling. Two conventional crop rotations consisted of a corn (Zea mays) …
Molecular Studies Of 5-Azacytidine-Induced Early-Flowering Lines Of Flax, Megan A. House
Molecular Studies Of 5-Azacytidine-Induced Early-Flowering Lines Of Flax, Megan A. House
Theses and Dissertations (Comprehensive)
Several early-flowering flax (Linum usitatissimum L.) lines were derived from treatment of germinating seeds with 5-azacytidine in 1990. These lines are also shorter, have fewer leaves, and their DNA is hypomethylated, relative to their corresponding controls. The work presented in this thesis used early-flowering and control lines of the Royal (R) flax genotype, and the Large (L) flax genotroph. Firstly, levels of cytosine methylation were measured over a 24-hour period in the early-flowering line RE2 and its control (RC), using an HPLC method. Secondly, to determine the response of the flax lines to short-day conditions, control and early-flowering …

Chromium, Copper, And Arsenic Concentration And Speciation In Soil Adjacent To Chromated Copper Arsenate (Cca) Treated Lumber Along A Topohydrosequence, Donald Roy Schwer Iii
University of Kentucky Master's Theses
Arsenic (As), Chromium (Cr), and Copper (Cu) are ubiquitous in soils as a result of anthropogenic and geogenic processes. The fate of As, Cr, and Cu in the environment is largely governed by their speciation, which is influenced by soil physiochemical properties. This study investigated the influence of soil physiochemical properties and landscape position on As, Cr, and Cu concentration and speciation in soils adjacent to Chromated Copper Arsenate (CCA) treated lumber fence posts. Concentration gradients showed elevated total As and Cu adjacent to the three fence posts, which decreased with increasing distance from the posts. In addition, As and …
Low Co-Cultivation Temperature At 20oc Improved Agrobacterium Tumefaciens-Mediated Transformation Of Tobacco Leaf Disks, Guiying Su
LSU Master's Theses
I investigated the effect of co-cultivation temperature and binary Ti vectors on the fresh weight yield of leaf disks of tobacco (Nicotiana tabacum L. cv. Xanthi (nn, Smith) after Agrobacterium tumefaciens-mediated transformation. I concluded that the temperature at 20ºC during co-cultivation of leaf disks with A. tumefaciens was the most critical factor on the reproducible enhancement of fresh weight yield. The highest fresh weight increase and presumably transformation efficiency was achieved under the following experimental conditions: A. tumefaciens strain LBA4404 containing pLSU binary Ti vector was grown overnight at 28oC in liquid Agrobacterium medium and the concentration was adjusted to …

Insights Into The World Of Pea Nodulation Using The Low Nodulator R50, Scott Clemow
Theses and Dissertations (Comprehensive)
Cytokinin oxidase (CKX) is the enzyme responsible for the degradation of cytokinin, a class of adenine-based plant hormones that stimulate cell division, among other physiological processes. The pea mutant R50 is characterized by having a pale leaf phenotype, dwarf stature, few lateral roots, low nodule formation, and elevated levels of endogenous cytokinins in its shoots, roots and nodules. When compared to that of the wild-type Sparkle, total CKX activity is low but the transcript levels of PsCKX1 are significantly higher. In this study, I investigated the expression of PsCKX1 throughout the development of a nodule and the localization of PsCKX1 …

The Pleiotropic Mutant R50 (Sym16): A Link Between Cytokinin And Starch Metabolism May Explain Its Seed Phenotype, Chengli Long
Theses and Dissertations (Comprehensive)
Cytokinins are a class of adenine-based plant hormones that stimulate cell divisions and are therefore essential for organ growth and development. The role that cytokinin plays in nodulation and that of R50 (syml6) in particular is very complex. In this study, a cytokinin antagonist and a cytokinin oxidase (CKX) inhibitor were used as tools to study the effects of cytokinin perception or cytokinin degradation on nodulation in pea, the cytokinin transduction pathway, especially that induced by CRE1/AHK4, and CKX play a significant role in pea nodulation. Comparison of the full length sequences PsCKX2 cDNAs from Sparkle and R50 …
Don't Judge A Book By Its Cover: The Curious Case Of Wild Ginger Pollination, W. John Hayden
Don't Judge A Book By Its Cover: The Curious Case Of Wild Ginger Pollination, W. John Hayden
Biology Faculty Publications
What pollinates wild ginger? This seems like an easy question. The inconspicuous little flowers are held close to the forest floor, often completely hidden by a dense canopy of ginger leaves above. Flower color is rather drab, dominated by brown and maroon hues. Wind pollination seems completely unlikely and flowers pollinated by bees, butterflies, moths, or hummingbirds are always much more showy and accessible to these flying creatures. Flies, however, given their natural inclination to seek carrion as a food source for their babies (i.e. maggots), are often attracted to brown and maroon flowers. And because their actual quarry, animal …

Physiological And Biochemical Attributes Of Red-Fleshed Dragon Fruit, Hylocereus Polyrhizus., Punitha R. Vasanthan
Student Works (2010-2019)
An understanding of the postharvest parameters is essential for the successful market introduction of red-fleshed dragon fruit, Hylocereus polyrhizus, which has been recently developed as fruit crop. In this study, fruit at three different harvest maturities were evaluated in terms of physiological and biochemical aspects. Fruit which were harvested at complete red stage maintained their quality for up to one week. On the other hand, fruit harvested at early maturity attained the physical properties of fruit ripened on the plant (firmness reduced, peel colour changes, pH increased) but they contained noticeably lower concentrations of soluble sugars suggesting that fruit can …

Marine Algae As A Source Of Photosensitisers For Photodynamic Therapy Of Cancer., Tang Yee Voon
Student Works (2010-2019)
Photodynamic therapy (PDT) involves the intravenous administration of a photosensitiser that preferentially localises in cancerous tissues more than the surrounding normal tissues. Subsequent illumination of photosensitiser generates reactive oxygen species that cause irreversible photo-damage to the cancer cells. To date, most photosensitisers that have been approved for clinical use or are currently in clinical trials are cyclic tetrapyrroles which are derived or inspired by naturally occurring chlorophyll and its degradation products. Some non-tetrapyrrolic photosensitisers have also been found in nature suggesting that there are potential photosensitisers with interesting structures in nature. To this end, this thesis outlines the collection of …

Biolistic Transformation Of Oil Palm Using The Phosphomannose Isomerase (Pmi) Gene As The Positive Selectable Marker., Bahariah Bohari
Student Works (2010-2019)
The selectable marker system based on the Escherichia coli phosphomannose isomerase (pmi) gene was adapted for genetic transformation of oil palm. This system makes use of the pmi gene that encodes phosphomannose isomerase, which converts mannose-6-phosphate to fructose-6-phosphate and uses mannose as the selection agent. This is to anticipate the future requirement of using non-antibiotic resistance genes for the commercialization of transgenic oil palm. The use of antibiotic or herbicide-based selection systems has caused much public concern due to inadequate knowledge of the agents’ impact on the environment and on human health. In this study, four transformation vectors, namely pMI3, …
