High-temperature geothermal springs are ecosystems that resemble early Earth, offering insights into the origins and early evolution of life. In these unique ecosystems, thermophilic microorganisms thrive. However, our knowledge of thermophiles remains limited. A major challenge is that many thermophiles remain uncultivated. The primary goal of this dissertation was to explore the diversity, metabolism, and ecological functions of thermophiles using cultivation-dependent and cultivation-independent approaches. Chapter 1 provides a broad overview of geothermal spring ecosystems and thermophiles, along with a roadmap of the dissertation. Understanding how charge carrier move through conjugated polymers is crucial for optimizing the functionality and efficiency of various applications, from organic solar cells and light-emitting diodes (LEDs) to field-effect transistors. Previous studies have demonstrated that tracking the fluorescence centroid displacement provides a method to determine the position of single charge carriers in a conjugated polymer nanoparticle. This method allows the observation of charge carrier trajectories over time and allow researchers to obtain insights into the dynamic processes. Both the federal government and photojournalism have played a substantial role in the “othering” of Appalachia while simultaneously attempting to save it. Examining both institutions demonstrates the exploitative processes that alter the perception of rural communities and their surrounding environment. Stacey Kranitz’s archival exhibit, As It Was Give(n) To Me, internalizes this precarious role of the camera in a region heavily journalized. Emotions are biological responses to stimuli that allow individuals to derive meaning, appraise experiences, and prepare to respond. However, individuals perceive emotions differently based on emotion socialization which not only dictates the way emotions are viewed and managed but also has been directly linked with mental health outcomes. Furthermore, research shows emotion socialization is informed by demographic variables such as gender such that the expectations of emotionality differ; where women are taught to express emotions, men are taught to conceal. This dissertation consists of two projects. The first one involves nonparametric methods on Continuous Time Markov Chains (CTMCs). The second one is centered around Bayesian shrinkage models for detecting prognostic and predictive biomarkers in high-dimensional clinical data. Both these projects build on methods from across the frequentist and Bayesian paradigm to offer novel solutions. Drought, a prevalent and consequential natural disaster, poses widespread, indirect challenges across environmental and societal dimensions. Despite considerable focus on monitoring meteorological and hydrological drought and studying their characteristics, there is a gap in assessing its multifaceted impacts, especially on societal sectors. The dissertation comprises three research essays utilizing artificial intelligence to quantitatively study multi-dimensional drought impacts. The first essay leveraged deep learning and natural language processing to predict multi-dimensional drought impacts from textual datasets, including social media, news media, and citizen scientist reports. Eukaryotic cells are highly heterogeneous. These cells are arranged into different compartments, carrying out separate functions and facilitating biological processes. Proteins are the effector biomolecules targeted to subcellular locations that help fulfill specific tasks in living organisms. Spatial proteomics can help unravel molecularly how protein abundance and localization are altered in cells, which is not feasible in traditional bulk-scale proteomics. Recent advancements in artificial intelligence (AI) have accelerated the prediction of unknown protein structures. However, accurately predicting the three-dimensional (3D) structures of fusion proteins remains a difficult task because the current AI-based protein structure predictions are focused on the WT proteins rather than on the newly fused proteins in nature. Following the central dogma of biology, fusion proteins are translated from fusion transcripts, which are made by transcribing the fusion genes between two different loci through the chromosomal rearrangements in cancer. In this study, we investigate the influence of cold-plasma-induced enhanced performance and efficiency of SAPO-34 membranes in the separation of CO2 and CH4 mixtures. Placing the herein presented research in a broader context, we aim to address the question of whether cold plasma can significantly impact the membrane performance. We subjected SAPO-34 membranes to plasma mild disturbances and analyzed their performance in separating CO2 and CH4. Our findings reveal a notable enhancement in membrane efficiency and sustained performance when exposed to cold plasma. The capability of curcumin and its analogs to inhibit various processes involved in cancer development and progression has been extensively studied. Curcumin is also a photoactive compound, which can be activated with blue light to initiate photochemistry. Motivated by this and its inherent anti-cancer properties, we examined the practicality of using a commercially available nano-cumin formulation as a photosensitizer for the photodynamic therapy (PDT) treatment of oral cancers. This article critically analyzes the ethical and legal implications of adopting predictive analytics by the French administrative justice system. It raises a key question: Is it wise to integrate artificial intelligence into the administrative justice system, considering its potential benefits, despite the associated risks, ethical dilemmas, and legal challenges? Hydrothermal systems have been proposed as environments for prebiotic chemistry on early Earth. Ancient Mars was an ocean world and could also have had hydrothermal vents supporting biological or prebiotic processes. The Strytan Hydrothermal Field (SHF) in Iceland is a basalt-hosted alkaline vent that forms massive hydrothermal Mg-saponite chimneys and is a potential analog to basalt-hosted alkaline vents that may have existed at the Eridania basin on Mars, where saponite deposits are thought to have formed from ancient hydrothermal activity. Glycogen synthase kinase 3β (GSK3β) is a multifunctional serine/threonine kinase involved in several key signaling pathways, including glycogen metabolism, WNT/β-catenin, and Hedgehog signaling. Hyperactivity of GSK3β has been linked to Alzheimer’s disease, bipolar disorder, type II diabetes, and some cancers. Therefore, GSK3β is of interest as a target for therapeutics. Lithium ion (Li+) is a classical inhibitor of GSK3β. Structurally similar beryllium ion (Be2+) is ~1000-fold more potent. Climate change is considered amongst the most severe threats to terrestrial and aquatic ecosystems globally. Ecosystems in the southwestern United States have specifically been impacted by intense drought conditions since 2000. Higher temperatures combined with altered precipitation stresses many ecosystems; however, ecosystem specific responses to such stressors may vary. Here, the effects of climate change on semi-arid ecosystems are analyzed for some of the most vulnerable ecosystems in the southwestern United States: lacustrine, riparian, and dryland ecosystems.

Cold-adapted organisms use antifreeze proteins (AFPs) and antifreeze glycoproteins (AFGPs) to enable survival in freezing habitats. AF(G)Ps have been proposed to prevent leakage of intracellular metabolites by stabilizing cellular membranes, a property that could be of great physiological and biomedical relevance. The generality of this effect is not understood, and for the known examples of membrane leakage prevention by AF(G)Ps, the mechanisms remain unclear. Here, we report a comprehensive evaluation of the effects of different AFPs on the leakage of ions and dyes from model membrane systems. Presenting whole slide images (WSIs) as graph will enable a more efficient and accurate learning framework for cancer diagnosis. Due to the fact that a single WSI consists of billions of pixels and there is a lack of vast annotated datasets required for computational pathology, the problem of learning from WSIs using typical deep learning approaches such as convolutional neural network (CNN) is challenging. Silicon photonic integrated circuits enable the capability of producing on-chip processing of photonic signals, including both emission and detection. Emission materials, however, have been limited for monolithic integration due to high processing temperatures, indirect bandgap materials, and material integration complications. The (Si)GeSn material system has recently attracted attention due to the bandgap transition from the Si and Ge, naturally indirect bandgap, to direct bandgap with sufficient Sn incorporation. Rapid population growth and development in Western states are exerting strain on the region’s limited water resources. Urbanization exacerbates this issue by increasing impervious surfaces, limiting infiltration of precipitation during storm events and snowmelt, which results in changes to hydrologic conditions with higher runoff volumes and higher peak flows. Stormwater transports pollutants as it flows across impervious surfaces, discharging high volumes of runoff and elevated loads of urban contaminants into receiving waters. Prior research has established a role for both social isolation and exposure to high fat Western diets in altering a range of behaviors from reduced memory performance to increased depression-like behaviors. The present study scrutinizes the interplay among these variables during the peri-adolescent developmental phase, utilizing Long-Evans rats as the experimental model. Our overarching hypothesis is that rats exposed to either social isolation, a high-fat diet, or both will result in heightened pain sensitivity, diminished cognitive flexibility, and increased neuroinflammatory responses within brain regions implicated in sociability, cognition, memory, and pain processing. Four-membered cyclic amides, commonly known as β-lactams, have played a crucial role in drug discovery research since its discovery by Alexander Fleming in 1928 from Penicillium notatum. The β-lactam antibiotics are broadly effective against several bacterial infections through acylating the transpeptidase involved in cross-linking peptides to form peptidoglycan or periplasmic murein, which is a fundamental constituent of the bacterial cell wall for both the gram-positive (ten or more layers) and gram-negative (one or two layers) bacteria. Tillandsia usneoides in epiphytic bromeliads takes up water through absorptive trichomes on the shoot surface under extreme environmental conditions. Although previous studies revealed the way by which T. usneoides absorbs water and prevents water loss, its water transport remains unclear. Uses of T. usneoides L. have been documented going back to pre-Columbian and late archaic societies. Between the 1800s and 1900s. T. usneoides was utilized in fruit packing and polish, among other industrial goods. This study addresses the critical environmental issue of elevated nitrate levels in water bodies, primarily due to excessive use of nitrogenous fertilizers and improper waste disposal. It focuses on reducing nitrate concentrations in polluted water to permissible levels through the effectiveness of hybrid materials in nitrate adsorption. We synthesized nine amino-functionalized adsorbents using grafting and sol-gel techniques. Batch adsorption tests confirmed the high nitrate adsorption capacities of these adsorbents, with sol-gel materials showing the highest efficiency due to their abundant amino group contents. Compared to unmodified carbons and even some metal materials, nitrogen-doped carbons have been found to exhibit better performance for reducing oxygen-oxygen bonds, a key step in electroreduction of both O2 (an important reaction in energy applications) and H2O2 (an important reaction in sensing and biosensing). Previous studies from our lab revealed that thermal decomposition of urea in the presence of carbon fiber (CF) results in N-doped that exhibited good electrocatalytic properties for H2O2 reduction. Due to their semi-aquatic lifestyle, pinnipeds are an ideal sentinel group used to study anthropogenic threats to the marine environment. Microplastics, primarily transported to the ocean through river discharge or weathering of larger plastics, are a threat to both pinnipeds and humans. Bioaccumulation of microplastics within the marine food web has been observed, with pinnipeds indirectly ingesting microplastics through their prey. As generalists, gray seals (Halichoerus grypus) are a pinniped species that can provide information on microplastic exposure to many lower trophic level organisms. Recent declines in bee populations and ranges have been cause for concern due to the valuable pollination service that they provide. Several factors have been proposed to contribute to these declines, including habitat loss, pathogen spread, and pesticide usage, so many pollinator conservation schemes have involved the addition of pollinator-friendly habitat through wildflower plantings and artificial nesting sites. Because of this, many efforts have been made to enhance bee populations across different landscape types, including natural, agricultural, urban, and industrial areas. A multi-color flow cytometry method coupled with fluorescent staining was developed to detect and quantify E. faecalis concentrations in coastal waters as an alternative to the traditional membrane filtration method. The developed method was used to characterize E. faecalis during batch growth and yielded consistent results across the various physiological states of the cells. Additionally, the application of propidium iodide (i.e., a viability stain), afforded the method the ability to differentiate between cells with intact and compromised cell membranes, which raises the potential of quantifying cells that are in viable-but-non-culturable (VBNC) state.
