Malate is a key intermediate in the citric acid cycle, an enzymatic cascade that is central to cellular energy metabolism and that has been applied to make biofuel cells. To enable real-time sensing of malate levels, we have engineered a genetically encoded, protein-based fluorescent biosensor called Malon specifically responsive to malate by performing structure-based mutagenesis of the Cache-binding domain of the Citron GFP-based biosensor. Malon demonstrates high specificity and fluorescence activation in response to malate and has been applied to monitor enzymatic reactions in vitro. This study addresses the complexity of assessing river water quality, a multifaceted process influenced by numerous water quality parameters (WQPs) characterized by inherent uncertainties and diverse judgment information from decision-makers. These uncertainties and diverse judgment information can be effectively represented and simulated using Neutrosophic sets. In this study, we propose an effective water pollution rating system, the Neutrosophic water quality index (NWQI), to derive a water pollution score (NWQI-score) for rating water pollution levels. Identifying similar DNA sequences is crucial in various biological research endeavors. This paper delves into the intricate workings of a specific algorithm designed for this purpose. We provide a systematic explanation, exploring how the algorithm handles user input, reads stored DNA sequences, utilizes the Word2Vec model for vector representation, and calculates sequence similarity using diverse metrics like Cosine Similarity and Neutrosophic Distance. There is extensive evidence that network structure (e.g., air transport, rivers, or roads) may significantly enhance the spread of epidemics into the surrounding geographical area. A new compartmental modeling framework is proposed which couples well-mixed (ODE in time) population centers at the vertices, 1D travel routes on the graph’s edges, and a 2D continuum containing the rest of the population to simulate how an infection spreads through a population. The edge equations are coupled to the vertex ODEs through junction conditions, while the domain equations are coupled to the edges through boundary conditions. Animals within social groups respond to costs and benefits of sociality by adjusting the proportion of time they spend in close proximity to other individuals in the group (cohesion). Variation in cohesion between individuals, in turn, shapes important group-level processes such as subgroup formation and fission–fusion dynamics. Although critical to animal sociality, a comprehensive understanding of the factors influencing cohesion remains a gap in our knowledge of cooperative behavior in animals. At Swarthmore College, we began developing an Introductory Physics for Life Sciences (IPLS) course nearly 20 years ago. The electricity, magnetism and optics semester was launched in 2008 and both semesters have been offered regularly since Fall 2015. Two primary goals of this curriculum are (1) to prepare students to effectively use physical models and quantitative reasoning in biological and biomedical contexts, and (2) for students to come to view physics as relevant to the life sciences. Background
Polymorphisms are common in nature, but they are rarely shared among closely related species. Polymorphisms could originate through convergence, ancestral polymorphism, or introgression. Although shared neutral genomic variation across species is commonplace, few examples of shared functional traits exist. The blue-orange petal color polymorphisms in two closely related species, Lysimachia monelli and L. arvensis were investigated with UV-vis reflectance spectra, flavonoid biochemistry, and transcriptome comparisons followed by climate niche analysis.
Results
Similar color morphs between species have nearly identical reflectance spectra, flavonoid biochemistry, and ABP gene expression patterns. Transcriptome comparisons reveal two orange-specific genes directly involved in both blue-orange …

Low-dimensional hybrid organic–inorganic perovskites (HOIPs) are promising electronically active materials for light absorption and emission. The design space of HOIPs is extremely large, as a variety of organic cations can be combined with different inorganic frameworks. This not only allows for tunable electronic and mechanical properties but also necessitates the development of new tools for in silico high throughput analysis of candidate materials. In this work, we present an accurate, efficient, and widely applicable machine learning interatomic potential (MLIP) trained on 86 diverse experimentally reported HOIP materials. An intelligent system must balance generalizing across similar experiences with maintaining the distinctiveness of each experience. This thesis explores how the hippocampus manages this balance through its neural representations to support adaptive behavior. In Chapter 1, I provide an overview of key hippocampal phenomena that contribute to this process, including remapping, splitter signal, and replay. In Chapter 2, I challenge the concept of random remapping by showing that it is possible to predict, better than chance, how a given experience will be encoded in the hippocampus across different subjects. Many parts of the world face severe and prolonged drought conditions, stressing the sustainability of water resources and agriculture. Transitioning to water-efficient crops is one strategy that can help adapt to water scarcity. An emerging drought-tolerant crop of interest is chia (Salvia hispanica). Yet, no study has compared its large-scale water use dynamics to those of widely established crops across the globe. Here, we use satellite data over multiple years to assess the water use efficiency of chia, alfalfa, corn, and soybean globally. Work and energy are fundamental topics in physics. However, enhancing students’ critical thinking skills in these areas remains a challenge. This research aimed to develop and assess the validity and readability of e-modules designed with PjBL-STEAM and virtual simulation media to address this issue. The development process followed a 4D model, and the resulting e-modules were evaluated as strongly valid category. The validation tests yielded percentage scores of 91.6% for media, 89.3% for content, and 88.0% for lesson plans. Additionally, the readability test achieved a score of 84.6%, indicating a excellent level of comprehensibility. These findings suggest that the e-modules …

The weakness in students’ scientific argumentation, particularly in providing evidence-based arguments, has become a significant concern in education. This study aims to assess students’ scientific argumentation skills specifically on the topic of static fluids. The survey was conducted with 78 twelfth-grade students from private high schools in East Java, Indonesia, who had previously studied this topic. A scientific argumentation test instrument, consisting of an essay format with five questions, was employed to collect data. This study investigates the factors influencing the achievement of competencies in the field of Fiber Optic Installation Engineering (TIFO) among vocational school graduates in Indonesia and their subsequent performance in the workforce. Utilizing a quantitative approach, the research examines the relationship between several key variables, including national examination scores, quality of infrastructure, teaching competence, and telecommunication systems subject scores. Path analysis structural equation modeling was employed to determine the direct and indirect effects of these variables on TIFO competency achievement and graduates’ performance. This study investigates the impact of implementing a Problem-Based Learning (PBL) model integrated with Blended Learning on enhancing students’ collaborative abilities. Conducted in class XI of SMA Negeri Plus Riau Province during the 2021/2022 academic year, the research utilized a control group design to compare the collaborative skills of students in an experimental class, which employed the PBL model, with those in a control class. The study assessed four key indicators of collaborative ability: working productively, valuing opinions, compromising, and sharing responsibility. This research aimed to investigate the effectiveness of virtual laboratories compared to real laboratories in improving students’ conceptual understanding of Archimedes’ principle. A quantitative approach was employed using a pretest-posttest design. The measurement instrument used in this study was a concept mastery test, consisting of 15 multiple-choice questions. Data were analyzed using an independent sample T-test. The research involved students from two XI IPA classes, each consisting of 33 students. The findings indicated that both virtual and real laboratories in the learning process could enhance students’ conceptual understanding. Mycetoma is currently considered as a neglected tropical disease. The incidence of mycetoma is unknown but most of the worldwide cases are present in the “mycetoma belt” including countries like Mexico, India, Senegal, and others. The treatment of mycetoma depends on the etiological agent responsible for the case. The Florida Everglades is a critically important, but highly threatened ecosystem that is becoming increasingly susceptible to the invasion of non-native species. This study investigated the ecological role of the invasive peacock eel (Macrognathus siamensis) within this ecosystem using 15 years of electrofishing data and stable isotope analysis. We investigated the population trends of peacock eels at the marsh-mangrove ecotone of the Shark River Estuary, the environmental factors contributing to their abundance, and the potential interactions they may have with native fish assemblages and coastal food webs. The synthetic peptide TAT-I24 (GRKKRRQRRRPPQCLAFYACFC) exerts antiviral activity against several double-stranded (ds) DNA viruses, including herpes simplex viruses, cytomegalovirus, some adenoviruses, vaccinia virus and SV40 polyomavirus. In the present study, in vitro profiling of this peptide was performed with the aim of characterizing and improving its properties for further development. As TAT-I24 contains three free cysteine residues, a potential disadvantageous feature, peptide variants with replacements or deletions of specific residues were generated and tested in various cell systems and by biochemical analyses. Before 2016, the Eskişehir city landfill was an irregular landfill. Since then, it has been transformed into a regulated landfill. This study aimed to investigate the presence of pollution in the landfill drainage area. For this purpose, water samples were collected from the landfill drainage area and the Kadirbey Farm Spring, upstream of the landfill area, during the rainy and dry seasons of 2021. The pharmacological activities and hepatotoxicity significantly influence the success or failure of new drugs during drug discovery and development. Traditional experimental methods, such as animal models, are costly and time-consuming for chemical testing. Background: Breast cancer outcomes are worse among Black women in the U.S. compared to White women. While extensive research has focused on risk factors contributing to breast cancer; the role of genomic elements in health disparities between these racial groups remains unclear. This study aims to identify genomic variants and socioeconomic status (SES) determinants influencing racial disparities in breast cancer survival through multiple mediation analyses. Methods: Our investigation is based on the NIH-supported All of Us (AoU) program and analyzes 7452 female participants with malignant tumors of breast, including 5073 with genomic data. Due to the unsettled weather conditions caused by typhoons, it is not possible to forecast the time when a typhoon will arrive or depart accurately. The uncertainty regarding the duration of typhoon disruptions makes it challenging for transportation carriers to efficiently adjust their schedules. In this study, to deal with the problem of airline fleet rerouting, flight rescheduling, and passenger reassignment for uncertain disruption periods caused by a typhoon, we utilize network flow techniques with the concept of probability to construct a stochastic model for minimizing the total operating costs. This dissertation focuses on efforts to understand the mechanisms behind this selective inhibition. A versatile synthetic approach to these natural product analogs and a potential method for evaluating their biological activity on the microelectrode array are presented. Chapter 1 provides an overview of Gq/11 proteins, detailing their roles in cellular signaling. Mutations lead to constitutively active oncogenic Gq/11 proteins, contributing to uveal melanoma. Pro-regenerative lipid mediator 1 (PreM1) is a specialized pro-resolving lipid mediator that promotes wound healing and regenerative functions of mesenchymal stem cells (MSCs), endothelial cells, and macrophages. The healing of third-degree (3°) burns and regenerative functions of MSCs are enhanced by ACgel1, an arginine-and-chitosan-based protein-mimic hybrid hydrogel. Adipose-tissue derived microvascular fragments (MVFs) are native vascularization units and a rich source of MSCs, endothelial cells, and perivascular cells for tissue regeneration. Here we describe an innovative PreM1-MVFs-ACgel1 construct that incorporated PreM1 and MVFs into ACgel1 via optimal design and fabrication. Novel near-infrared ratiometric molecules (probes A and B) produced by linking formyl-functionalized xanthene and methoxybenzene moieties, respectively, onto a xanthene-hemicyanine framework are detailed. Probe A exhibited a primary absorption peak at 780 nm and a shoulder peak at 730 nm and exhibited fluorescence at 740 nm↓ (signifies a downward shift in intensity upon acidification) in a pH 9.3 buffer and 780 nm↑ at pH 2.8 under excitation at 700 nm.
