The c-JUN N-terminal kinase (JNK) signaling pathway plays an important role in regulating the innate immune response. Immune signaling is governed by the coordinated activity of protein kinases counter-balanced by protein phosphatases; however, the importance of the latter family of enzymes is less well understood. c-JUN N-terminal kinase (JNK)-stimulatory phosphatase 1 (JSP1, also known as DUSP22) has been implicated as a positive regulator of JNK signaling, yet its role in innate immunity is not clear. Using a mouse model of the local Shwartzman reaction, we show that JSP1 is essential for LPS-TNF-alpha-induced vascular injury. JSP1-knockout mice exhibited reduced vascular hemorrhage. …

Why General Ai Inherited The Body: A Structural Account Of Embodied Modulation Under Monolithic Imitation, Griselda Poe

General AI has pursued the replication of human-level intelligence without first decomposing human cognition into structurally distinct components. Human cognition, however, is shaped by embodied constraints such as mortality, survival pressures, finite lifespan, and physiological states. This paper argues that when cognition is treated as a single undifferentiated whole, embodied modulation is not accidentally introduced into AI systems but structurally entailed. Any attempt to imitate “human intelligence” under a monolithic model necessarily incorporates variability shaped by mortal embodiment. The tensions observed in contemporary AI systems are better understood as consequences of copying an undecomposed target rather than isolated implementation errors. …

Carbon dioxide (CO₂) dynamics in caves are critical for karst processes, speleothem growth and dissolution, microclimate regulation, and caver safety. In cave atmospheres, CO₂ commonly derives from soil respiration, epikarst degassing, microbial activity, and, in tectonically active regions, deep-seated fault-related emissions. Fault-related CO₂ degassing may also track tectonic activity. However, continuous monitoring remains limited due to the cost and inflexibility of commercial instruments. Here we present an open-source, low-cost Arduino-based CO₂ datalogger designed for cave applications. Campaigns for ecological sustainability have gained a great currency in the contemporary academic discourse. Nonetheless, from time immemorial, Africans have pushed the ecological sustainability agenda through cultural taboos and regulated resource harvesting. Many African societies have embedded ecological consciousness in daily life expressions, and have intricately expressed it through literary and ritual texts. This study focuses on ecological awareness in the ritual texts of the Pangwa community. Ecosystem-based approach that integrates floods and environmental risk management is believed to reduce flood disaster risk while providing socio-economic and environmental benefits to floodplain occupants. This study examined the socio-economic and environmental benefits of the ecosystem-based approach in managing flood disaster risk in the Little Ruaha River Basin in the southern highlands of Tanzania. The study involved 157 participants and employed the mixed research design to collect quantitative and qualitative data. Genomic data sharing has become an essential component of biomedical research, enabling large-scale collaborations and accelerating discoveries in human genetics. To balance the need for accessibility with privacy concerns, several controlled-access mechanisms have been proposed, including genomic beacons. Genomic beacons answer simple presence/absence queries about specific genetic variants. However, prior work has demonstrated that beacons remain vulnerable to genome reconstruction attacks, where an adversary can recover large portions of participants’ genomes using summary statistics. Power distribution systems play a crucial role in transmitting electrical power from generation sources to end users. During transmission, significant power losses occur in the form of heat as the current flowing along the lines/cables has resistance. To minimize power losses, distribution network reconfiguration (DNR) has been widely adopted. This paper proposes optimal DNR based on metaheuristic techniques with discrete mutation feature targeting active power loss reduction, which subsequently lowers carbon emissions and operational costs. This study was performed to investigate the therapeutic potential of a polyherbal formulation containing Curcuma longa, Phyllanthus niruri, Ziziphus mauritiana, Nigella sativa, and Annona muricata as multi-target inhibitors against breast cancer protein targets using molecular docking and molecular dynamics in silico approach. Functionally graded materials (FGMs) are a highly advanced class of biomaterials with graded structure and properties, enabling the fabrication of physiologically and mechanically compatible materials for use in various medical devices. This study aims to produce a functional-grade material based on a cobalt-chromium-molybdenum alloy (F75) reinforced with 4% titanium (Ti) and hydroxyapatite (HA). This will enhance the material's mechanical properties, corrosion resistance, and bioactivity, making it suitable for use as a bone substitute. Background: An emerging feature in modern biomedical research is collecting and analyzing numerous variables. In the presence of many potential covariates, inference becomes challenging requiring both distinguishing a set of covariates truly associated with an outcome and estimating their corresponding regression coefficients consistently. Traditional statistical inference typically focuses on estimating coefficients assuming a pre-specified set of covariates. Further, advanced machine/statistical learning methods performing both selection and estimation predominantly focus on outcome prediction rather than association inference.

For over thirty years, patients have been visiting the Arkansas Epilepsy Program for diagnosis and treatment for seizures and seizure-like episodes. As part of their clinical evaluation, patients often undergo ambulatory EEG (electroencephalogram) monitoring. This routine process produces valuable data for treating the patient. In this study, over 2000 ambulatory EEG reports from 1998 to 2016 were reviewed. A large database of seizures was created from the reports, with information on 407 patients, 1611 EEG-confirmed seizures, and 1726 patient-reported seizures (IRB Protocol #17-093). Monoclonal gammopathy of undetermined significance (MGUS) is a necessary precursor condition to multiple myeloma (MM). Given the role of autophagy in modulating MM risk, we investigated whether genetic variation in autophagy-related genes influences susceptibility to MGUS. We analyzed the association of 34,042 common autophagy-related single nucleotide polymorphisms (SNPs) with MGUS across six independent cohorts, five from Europe and one from North America, comprising 2317 MGUS cases and 282,358 controls. We also assessed their impact on immune parameters, including absolute counts of 91 blood-derived immune cell subsets and 103 circulating immunological proteins. In this paper, we will establish the existence of positive periodic solutions for a class of first order iterative functional differential equations by using a powerful and effective technique based on the Krasnoselskii fixed point theorem together with some useful properties of a Green's function. The obtained results that complement some previous works, are illustrated with an example.

In this paper, we define multiplicative dual numbers, multiplicative dual vectors, scalars, and vector products associated with the multiplicative dual numbers. We give the base elements of multiplicative dual elliptic quaternions and their operational properties. We explain the rotation and translation motions in multiplicative dual elliptical quaternions. We provide the matrix representations of the screw motion using multiplicative dual elliptical quaternions. Thanks to these multiplicative dual elliptical quaternions, we explain the screw motion along with rotation and translation with theorems and proofs. We visualize this screw motion with mathematical programs by applying screw-ruled surfaces.

Measuring What Matters: Specifications Grading And Latin* Students’ Mathematics Identity, Luis Miguel Fernández, Mayra Ortiz Galarza, Cristina Villalobos, Martha Asare

This study examined Specifications Grading, an alternative grading system emphasizing clearly defined learning outcomes and revision, and mathematics identity among 846 Latin* Calculus I students at a Hispanic-Serving Institution. Mathematics identity, comprising competence/performance, recognition, and interest, was measured at the beginning and end of the semester. Repeated-measures analyses indicated stable competence/performance and recognition alongside declines in interest. Specifications Grading was associated with increased mathematics identity, and multilingual students experienced smaller declines than their peers overall.

Substrate channeling is a strategy for enhancing flux and yield in enzymatic cascades and is increasingly relevant for applications in biocatalysis, biotechnology, and bioelectrochemical systems. However, efforts to engineer channeling are limited by the lack of high-throughput methods to evaluate and optimize channeling efficiency. Here, we present a fluorescence-based screening assay to rapidly assess substrate channeling in a model system involving fumarase and malate dehydrogenase, two sequential enzymes from the Krebs cycle. Proton exchange membrane (PEM) fuel cells are promising for clean and efficient energy conversion across diverse applications. High-temperature PEM fuel cells (HT-PEMFCs) using phosphoric acid (PA)-doped polybenzimidazole (PBI) can operate up to 200°C, but at lower temperatures, water-induced PA loss often leads to performance degradation and reduced durability. In this work, we present a new class of main-chain ion-pair PBI membranes by integrating the imidazolium-biphosphate ion-pair units directly into the PBI backbone. Several ancient languages use linguistic structures to differentiate between singular, dual, and plural forms that are used when one, two, or many objects are referenced. (Some languages go further, adding a trial form to differentiate two from three.) In many modern languages, numerals are used to convey precise number. After reviewing a range of ways languages express number (including the interesting case of Pirahã which does not express precise number), the paper concludes with a proposed developmental path for the expression of quantity in languages.

Fluorescence anisotropy and single-molecule orientation-localization microscopy (SMOLM) are powerful techniques that quantify the rotational diffusion of dipole-like emitters, which is important for sensing molecular interactions and chemical environments at the nanoscale. Numerous theoretical and experimental studies have thoroughly characterized single-molecule rotations even when those rotations are much faster than the detector integration time. Here, we extend the theory of measuring rotational diffusion to situations where a single dipole rotates uniformly everywhere outside of an isotropic cone of a certain size, termed a negative cone. South Carolina has many water resources that provide tremendous opportunities for people to enjoy water-based recreational activities, such as swimming, boating, and fishing. Participation in these activities produces significant economic benefits and supports many jobs throughout the state. However, at times, water quality may be unsafe for immersion or skin contact due to high levels of pollutants, such as bacteria or cyanotoxins. It is important for people to be aware of potential concerns and understand methods for determining if a waterbody’s current conditions are likely to be safe for primary or secondary recreation.

Objective.Accurate lung function assessment is essential for diagnosing and managing diseases like chronic obstructive pulmonary disorder, pulmonary emboli, and lung cancer. Single-photon emission computed tomography (SPECT) provides valuable 3D functional imaging of ventilation and perfusion, but is limited by low spatial resolution, availability, additional radiation, and cost. Alternative methods, including CT-based perfusion (CT-P) and deep learning models, require large datasets to validate results that are often scarce. Pulmonary function tests (PFTs) offer rapid and noninvasive global lung function measures and are clinically widely used. Through characterization of the electronic excited-state topology, linear-SiCP is predicted to be photostable in the UV region, whereas linear-AlCP is predicted to undergo photodissociation to form Al+CP products after absorption of light in the 200–250 nm range, representing a catalytic process for freeing aluminum molecules from a solid dust grain. Both SiCP and AlCP are predicted to have electronic transitions with large absorption cross sections (∼10–17 cm2) and to undergo fluorescence from bound excited states. The electrospinning of the polymer of intrinsic microporosity-1 (PIM-1) poses challenges due to its limited solubility and tendency to form bead-like structures at high concentrations. This work details the synthesis and analysis of electrospun composite fibers made from polyacrylonitrile (PAN) and PIM-1 for electrochemical applications. The data analysis confirmed the successful incorporation of PIM-1 into the PAN matrix. The fiber characteristics were significantly influenced by PIM-1 loading (1–10%), resulting in fiber diameters ranging from 0.8 to 1.7 μm. Plasmodium falciparum, the deadliest causative agent of malaria, continues to evade eradication efforts through widespread drug resistance. The recent development of ADC-028, a 4-aminoquinoline antimalarial with excellent activity and pharmacokinetic properties, prompted the investigation of bisquinoline analogs featuring similar structural motifs. Here, we describe a structure-activity relationship study that guided the optimization of compounds with key features, including the 4-anilinoquinoline core and diverse bridging linkers. Several analogs exhibited potent in vitro activity (IC50 < 20 nM) against both drug-sensitive and multidrug-resistant P. falciparum strains, while maintaining favorable cytotoxicity profiles. Among them, 25 demonstrated improved intrinsic metabolic stability (t1/2 = 121 min) and potent in vivo efficacy (ED50 = 0.32 mg/kg/day), achieving complete curative protection at a reduced dose compared to ADC-028. While 25 showed moderately reduced oral bioavailability (F = 43%) and a shorter half-life (T1/2 = 27.2 h) relative to ADC-028, its enhanced in vivo efficacy underscores its therapeutic potential. This work highlights a promising path forward in developing antimalarials that retain the efficacy of legacy compounds while overcoming modern resistance mechanisms.
