The Purification And Characterization Of Methylmalonyl Coa Mutase From Bovine Brain, Damon Martin

Methylmalonyl CoA mutase has been purified over 2300-fold from bovine brain using fractional precipitation, ion exchange resins, and gel filtration procedures. The crude extract had an equal mixture of mutase in the halo- and apoenzyme form. After the final purification step the ratio had changed to 86% holo-enzyme and 14% apoenzyme. The mutase enzyme had a pH optimum of 7. 0 in Tris-HCI buffer. The Km values for L-methylmalonyl CoA and succinyl CoA were 7.7 x 10^-4 M and 1.8 x 10^-4 M respectively. Cobalt-substituted carboxypeptidase and carbonic anhydrase were chosen as candidates for resonance Raman spectroscopy because they possess visible absorption due to the Co(II)-protein complex. However, no evidence for resonance-enhanced peaks due to the cobalt-containing chromophore was found with laser excitation near the visible absorption bands (514.5 nm) or closer to the ultraviolet absorption bands (457.9nm).

Arsanilazocarboxypeptidase and model azotyrosine compounds were selected for a Raman spectroscopic investigation because they have intense absorption bands in the visible region. All of these substances yielded similar resonance Raman spectra. Although there was no evidence for specific zinc-ligand vibrations, shifts in vibrational frequencies of the …

The Effects Of Pentachlorophenol On The Electrical Conductivity Of Lipid Bilayer Membranes, William Harvey Perman

The effects of pentachlorophenol (PCP), a widely used pesticide, on the electrical characteristics of lipid bilayer membranes has been studied. When a small amount of PCP (even at a concentration of a few micromoles per liter) is present in the electrolytic solution surrounding the membrane, the electrical conductivity of the membrane significantly increases. The present work was concerned with detailed measurements of the changes in the conductivity caused by PCP under chemically controlled conditions. The results of a survey, concerning environmental education taken of the educators of the Nelson County School System, indicated a need for environmental education workshops and classes.

The status of environmental education in the nation and in Kentucky were studied and compared. A research was done of the status of environmental education in Nelson County, and implications were drawn for implementing environmental education in the Nelson County School System.

A tentative program for implementing environmental education in the Nelson Country schools was given, and a bibliography of resources and materials to be used in said program was included.

Two Texas barrier islands, Boca Chica and Brazos, have unique coastal vegetational communities that may be Influenced by several environmental factors. These environmental factors include temperature, precipitation, wind speed, sunshine exposure, day length, and dune height. The purpose of this investigation was to determine the total number of coastal plant species per station, season, and year and to consider environmental factors that may significantly affect the number and pattern of coastal species. Coverage comparison between aerial and land sampling and aerial vegetational zonation was also considered.

Results indicated the presence of 21 angiosperm families and a total of 54 species. …

This bulletin describes nutrient and energy composition of beef cattle waste fractions as a function of the ration roughage level and type of feedlot. Results can be used to plan engineering system approaches and research to develop handling and processing equipment necessary for recovery of the most valuable portion of waste.

Feeding And Food Preferences By Three Sympatric Species Of Cyprinodontid Fishes, Lawrence J. Baer

The feeding and food utilization by three sympatric species of cyprinodontid fishes was examined using a modified Ivlev Electivity Co-efficient. Results indicated that although Fundulus heteroalitus and Fundulus majalis are sympatric in the area examined,their usage of the available food source varied noticeably. The third investigated species, Cyprinodon variegatus, fed mainly upon a vegetal detritus food source varied by the consumption of invertebrate and vertebrate food items. Slight intra-specific variances of diet between crespuscular periods was also discussed.

Considerable conflict has existed for many years between professional fishing, recreational usage and development in the west and south coast estuaries of Western Australia. One of the areas where intense conflict exists is Wilson Inlet.

The Government's task of advising the various interests concerned in these conflicts has been difficult, due to the lack of knowledge of estuarine fauna and environment in relation to usage and development.

The fresh-water mussel industry of the Tennessee River is nearing an end. Overharvesting, habitat alterations, and pollution are the major contributors to the depletion of the mussel resource, upon which the shell industry is based. A history of unconcern by shell harvesters and weak conservation enforcement by governmental agencies, has left the major waterways of the United States nearly void of commercial clams. The lower Tennessee River presently supplies the mussel industry with nearly all the important species of mollusks. In April and June 1968, the Pacific Ocean Biological Survey Program (POBSP) of the Smithsonian Institution conducted surveys on breeding marine birds and pinnipeds on various Mexican islands. Between 18 to 26 April and 21 to 29 June, pinniped populations were surveyed at Islas de Guadalupe, San Benito, Cedros, and Natividad off Baja California. The consumption of alcohol is a problem not only in this country (U.S.), but also in many other places in the world. Thus we find the Indians in Peru, Bolivia and other neighboring countries (Ecuador and northern part of Argentina) consuming alcohol to the point that they become alcoholics, in addition to this they also chew coca leaves, plant from which cocaine is extracted.

In an effort to establish the cause for this behavior, we put Sprague-Dawley albino rats on either a poor or good basal diet. Once every other day the rats were injected with a solution of cocaine. …

The western phosphate field produced about 15 percent, in 1970, of the nation's domestically consumed phosphate rock and contains an estimated 58 percent of the nation's indicated phosphate reserves. The central portion of the western phosphate field, which contains the Dry Valley area of southeastern Idaho, encompasses the greatest amount of mineable phosphate reserves in the smallest geographic area within the western field.

Exploration of properties in the area should follow an orderly sequence of literature search, area reconnaissance, detailed target appraisal, and detailed three-dimensional sampling to effectively evaluate each property. Sheep with esophageal fistulas were used to determine the daily intake, nutritive content and digestibility of forage at three periods and two stocking intensities during the spring of 1972 on a typical foothill range in northern Utah.

Heavy grazing under a season-long regime did not influence the concentrations of dietary chemical components when compared to moderate grazing; however, it did depress the digestibility of cellulose and organic matter. There was a significant decline in the dietary chemical components due to forage maturation. Digestibility of organic matter and cellulose were significantly higher in the early spring as compared to late spring. …

In-stream concrete structures were studied through model tests and river tests. The model studies indicated that four designs provided good habitat in the model stream. These structures were the inverted weir, the "V" structure, the slab with legs and the cylinder. Through the river studies it was determined that these structures did not influence enough of the total river area to be effective in providing good fish habitat. Also an appreciable amount of yearly maintenance would be required to free the structures from silting in, debris, and vandalism. The slab with legs was the only promising structure.

The Environmental Information System Office (EISO) at Oak Ridge National Laboratory (ORNL) provides information support for researchers and administrators involved with energy and environmental policy and progress. Multiple EISO activities for various governmental agencies have resulted in establishment of compatible data bases concerned with energy and environmental information, methods for effectively developing these, development and computer display of numerical data summaries, and reports evaluating published information. Direction is provided by continuing dialogue between users and information system staff.

Growth and reproduction may be stimulated by increased temperature in the cooling system and thermal plume during seasons when ambient water temperature is less than optimum, but growth, reproduction and survival are reduced when the elevated temperatures become excessive. Some fish species congregate in the warm thermal plumes during cold seasons but are excluded from this living space by temperatures above their preference in the summer. However, the warm refuge provided by a thermal plume in cold seasons can be a death trap if a power plant shuts down suddenly and exposes the fish to cold shock exceeding their lower …

Most of the waste products of human society end up in the oceans and the number and quantities of pollutants entering the oceans have been increasing at exponential rates. Recognition of these trends has resulted in expansion of international environmental attention to include sources of marine pollution other than vessels. Source, in this sense, refers to the pathway through which pollutants reach the sea. Land-based sources accounts for the vast bulk of the pollutants reaching the oceans. Effective protection of the marine environment, therefore, must include regulation of land-based sources of marine pollution.

It is now clear that the capacity of the oceans to accept some of the by-products of civilization is limited. The oceans can no longer be the world's sink, it we are to maintain the ecological viability of the biosphere. The dangers of the present situation are compounded by the difficulties in defining "marine pollution." Today's concern with pollution and its effects upon our way of life requires that the impact of ships on the marine environment be considered in order to minimize the risk that waterborne cargoes, particularly petroleum products, present to the ecology.
