Objectives: The human gastrointestinal tract (GI)may play a critical role in the prevention of obesity and metabolic diseases. Considering the importance of a healthy diet for optimal health, this study examined associations of diet quality assessed by three a priori-defined dietary patterns (Healthy Eating Index 2010 [HEI-2010],Mediterranean Eating Pattern for Americans [MEPA], and Dietary Approaches to Stop Hypertension [DASH] with gut microbiota, short-chain fatty acids (SCFA) and intestinal permeabilitymarkers in adult obese participants.

Milk exosomes (MEs) and their microRNA cargos constitute novel bioactive food compounds, and bovine MEs (BMEs) are being considered for use in drug delivery. The internalization of MEs by macrophages and degradation in lysosomes (in all cells) limit biological activities. Here, we determined whether BMEs internalized by murine bone marrow-derived macrophages (BMDMs) retrofuse with the intralumenal membrane in the multivesicular body (MVB) for subsequent release into the extracellular space or whether BMEs are destined for degradation in lysosomes.
Conclusions: The retrofusion of BME-derived intralumenal MVBs and secretion of BME into the extracellular space is quantitatively minor in BMDMs. The majority …

Objectives: To develop BMEs that evade elimination by BMDMs.
Conclusions: The elimination of BMEs UNL1 and UNL2 is significantly reduced compared unmodified BMEs in BMDM cultures. This is of great importance when using BMEs for delivering therapeutics.

Neurodegenerative diseases (NDs) are the age-related diseases which represent a major cause of mortality in aging population nowadays. Previous studies have shown that glutamate, the most abundant excitatory neurotransmitter, significantly increases in extracellular fluid of the patient’s brain and is the cause of neurodegeneration in NDs. The increase of extracellular glutamate elevates reactive oxygen species (ROS) accumulation and calcium (Ca2+) which exacerbate mitochondrial function, followed by inducing MAPK pathway and endoplasmic reticulum (ER) stress, respectively. Intuitive Eating (IE) is a food mindset focusing on honoring hunger and fullness, while rejecting the diet mentality. Research demonstrates a clear link between IE and health outcomes across many populations, including college students. Despite a growing body of literature, we still know very little about what complexities food insecurity may present for individuals trying to adopt IE behaviors. Aim: The primary aim of this thesis was to investigate the relationship between nutrition and frailty in Irish older adults. The nutritional status, dietary quality and perceived barriers/motivators to nutrition were also examined, and a frailty intervention protocol was designed. Methods: Cross-sectional study in Irish community-dwelling adults aged ≥65 years. Nutritional status, dietary intake, functional mobility and food choice motivators/healthy eating barriers were assessed using the Mini Nutritional Assessment – Short Form (MNA-SF), a food frequency questionnaire (FFQ), the Timed Up and Go (TUG) test and a custom designed 2-item questionnaire, respectively. I studied the age at first menstruation as a predictor of Western diet consumption among adults at The University of Akron and in Cleveland. I used stable carbon and nitrogen isotope compositions of human hair, as well as surveyed consumption of foods important to a Western diet (red meat, sugar, and dairy), to understand whether participants aligned with a Western dietary pattern. Age at survey and adult economic status were investigated as potential confounding variables. Objectives: Dietary γ -glutamyl peptides (γ -GPs) found in dry edible beans exhibit biological activities (antioxidant, anti-inflammatory) with potential benefits against chronic metabolic disorders. γ -GPs are responsible for the desirable Kokumi flavor through allosteric activation of the Calcium Sensing Receptor (CaSR) present in multiple mammalian tissues. In this study, we investigated the metabolic effects of γ -glutamyl valine (γ -EV) in diabetic obese mice.
Conclusions: γ -EV improved the diabetic condition of db/db mice via modulation of glucose and lipid metabolism.

Background: The COVID-19 pandemic brought new challenges affecting the wellbeing of individuals in communities with low income. Understanding where people live and how those environments can facilitate or hinder living a healthy lifestyle is essential for developing interventions that target behavior change and health promotion. Objective: This study compares Extension Nutrition Educators’ (NEs) perceptions of the barriers and facilitators impacting the healthfulness of the environment of communities with low income in eleven states before and during the COVID-19 pandemic.

INTRODUCTION: Appetite fluctuates across the menstrual cycle, suggesting ovarian hormones are involved in the regulation of appetite hormones and energy intake. Exercise is another regulator of appetite with higher intensity exercise generating a greater and more consistent suppression. มะเร็งท่อน้ำดีเป็นมะเร็งที่เกิดขึ้นบริเวณท่อทางเดินน้ำดี โดยมีความหลากหลายของเซลล์มะเร็งบริเวณเนื้อเยื่อมะเร็งและมีความรุนแรง มะเร็งท่อน้ำดีจัดเป็นสาเหตุหลักในการเสียชีวิตของผู้ป่วยในประเทศไทย เนื่องจากผู้ป่วยได้รับการวินิจฉัยเมื่อมะเร็งเจริญพัฒนาไปยังระยะท้ายและยังขาดวิธีการรักษาที่มีประสิทธิภาพ การรักษามะเร็งท่อน้ำดีด้วยวิธีมาตรฐานในปัจจุบันในผู้ป่วยที่ไม่สามารถผ่าตัดได้ หรือในผู้ป่วยหลังการผ่าตัด คือ การรักษาด้วยยาเคมีบำบัด ชนิดเจมไซทาบีนหรือยาเจมไซทาบีนร่วมกับยาซิสพลาติน แต่อย่างไรก็ตามผู้ป่วยส่วนใหญ่พบปัญหาการดื้อต่อยาเคมีบำบัดนำไปสู่การพยากรณ์โรคที่ไม่ดี ดังนั้นการพัฒนาการรักษาที่มีประสิทธิภาพมากยิ่งขึ้นถือว่ามีความจำเป็นเร่งด่วน โดยได้มีการศึกษาการตายของเซลล์มะเร็งแบบใหม่ที่ทำให้เซลล์มะเร็งตาย และกระตุ้นการทำงานของระบบภูมิคุ้มกันแบบจำเพาะ เรียกว่า การตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการตายของเซลล์แบบเนคโครปโทซิส จัดว่าเป็นการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยได้มีการศึกษาอย่างแพร่หลายในมะเร็งหลายชนิด โดยพบว่าการกระตุ้นให้เซลล์มะเร็งตายแบบเนคโครปโทซิสสามารถกระตุ้นระบบภูมิคุ้มแบบจำเพาะและยับยั้งการเจริญพัฒนาของก้อนมะเร็งในหนูทดลอง การศึกษาภายในห้องปฏิบัติการก่อนหน้าของคณะผู้วิจัยพบว่าการเหนี่ยวนำร่วมกันระหว่างยาเคมีบำบัดเจมไซทาบีนและสารสะแมคไมเมติกสามารถเหนี่ยวนำให้เซลล์มะเร็งท่อน้ำดีตายแบบเนคโครปโทซิสในสภาวะที่มีการยับยั้งการทำงานของเอนไซม์แคสเปส ทั้งนี้จากข้อจำกัดที่ยาเคมีบำบัดมีความเป็นพิษที่สูง การศึกษานี้จึงมีจุดมุ่งหมายเพื่อค้นหาสารที่สามารถนำมาทำการเหนี่ยวนำร่วมกับสารสะแมคไมเมติก โดยมีสมมติฐานของงานวิจัย คือ สารพฤกษเคมีที่มีฤทธิ์ทำให้เกิดการทำลายของดีเอ็นเอและหยุดวัฏจักรของเซลล์จะสามารถเสริมฤทธิ์กับสารสะแมคไมเมติก โดยกระตุ้นการตายของเซลล์แบบในคโครปโทซิสในเซลล์มะเร็งท่อน้ำดี โดยงานวิจัยนี้ได้ทำการศึกษาฤทธิ์ยับยั้งการแบ่งตัวและการตายเซลล์มะเร็งท่อน้ำดีของสารพฤกษเคมี ได้แก่ ชิโคนิน พลัมบาจิน เควอซิติน โดยพบว่าสารเควอซิตินมีค่าความเข้มข้นของสารที่ยับยั้งการรอดชีวิตของเซลล์ได้ 50% (IC50) ที่ต่ำที่สุดเมื่อเปรียบเทียบกับสารพฤกษเคมีชนิดอื่นๆ ในการวิเคราะห์ด้วยวิธีการทางชีวสารสนเทศน์พบว่าสารเควอซิตินมีเป้าหมายไปยังยีนที่มีบทบาทในการควบคุมวัฏจักรเซลล์ระยะ G2/M และยีนเป้าหมายของสารเควอซิตินมีการแสดงออกที่เพิ่มมากขึ้นอย่างมีนัยสำคัญในเนื้อเยื่อมะเร็งท่อน้ำดีเมื่อเปรียบเทียบกับเซลล์ปกติ การทดลองในเซลล์มะเร็งท่อน้ำดี (in vitro) พบว่าสารเควอซิตินสามารถหยุดวัฏจักรเซลล์ที่ระยะ G2/M นอกจากนี้สารเควอซิตินช่วงความเข้มข้นค่าต่ำที่มีความเป็นพิษน้อยในรูปแบบสารเดี่ยว เมื่อนำไปเหนี่ยวนำร่วมกับสารสะแมคไมเมติกชนิด LCL-161 แล้วพบว่าสามารถเสริมฤทธิ์กันกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดี โดยเมื่อทำการวิเคราะห์ชนิดการตายของเซลล์เพิ่มเติมพบว่ามีการแสดงออกที่เพิ่มขึ้นของโปรตีนเอ็มแอลเคแอลที่ถูกเติมหมู่ฟอสเฟต (phosphorylated MLKL; pMLKL) ซึ่งเป็นตัวบ่งชี้ที่จำเพาะต่อการตายแบบเนคโครปโทซิส นอกจากนั้นยังพบว่าการตายของเซลล์ถูกยับยั้งเมื่อมีการเติมตัวยับยั้งที่จำเพาะกับโปรตีนที่สำคัญต่อการเกิดเนคโครปโทซิส แสดงให้เห็นว่าการเหนี่ยวนำเซลล์ด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นให้เซลล์มะเร็งท่อน้ำดีตายผ่านเนคโครปโทซิส การศึกษาเพิ่มเติมเพื่อศึกษาการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการวิเคราะห์การเคลื่อนย้ายของโปรตีนเอชเอ็มจีบีวัน (HMGB-1) จากภายในนิวเคลียสมายังบริเวณไซโตพลาสซึมด้วยวิธีการย้อมแบบอิมมูโนฟลูออเรสเซนส์ พบว่าการเหนี่ยวนำเซลล์มะเร็งท่อน้ำดีด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นการตายของเซลล์ที่กระตุ้นภูมิคุ้ม กล่าวโดยสรุป การเหนี่ยวนำร่วมกันระหว่างสารเควอซิตินและสารสะแมคไมเมติกในสภาวะที่มีการยับยั้งเอนไซม์แคสเปสสามารถเสริมฤทธิ์กันโดยกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดีแบบเนคโครปโทซิสและการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน ดังนั้นงานวิจัยนี้อาจนำไปสู่การพัฒนากลยุทธ์การรักษามะเร็งท่อน้ำดีแบบใหม่ที่เพิ่มประสิทธิภาพในการรรักษาในอนาคต อย่างไรก็ตาม งานวิจัยนี้ยังต้องมีการศึกษาเพิ่มเติม เช่น การศึกษากลไกการเสริมฤทธิ์และการกระตุ้นการตายของเซลล์ การศึกษาฤทธิ์ในเซลล์มะเร็งท่อน้ำดีชนิดอื่นๆ และการเมินความเป็นพิษในเซลล์ปกติ เป็นต้น

BACKGROUND: Kwashiorkor is a childhood syndrome of edematous malnutrition. Its precise nutritional precipitants remain uncertain despite nine decades of study. Remarkably, kwashiorkor's disturbances resemble the effects of experimental diets that are deficient in one-carbon nutrients. This similarity suggests that kwashiorkor may represent a nutritionally mediated syndrome of acute one-carbon metabolism dysfunction. Here we report findings from a cross-sectional exploration of serum one-carbon metabolites in Malawian children.

Little is known about longitudinal patterns of welfare program participation among single mothers after they transition from employment to unemployment. To better understand how utilization patterns of these welfare programs may change during the 12 months after a job loss, we used the 2008 Survey of Income and Program Participation to examine the patterns of participation in Medicaid, the Supplemental Nutrition Assistance Program, Temporary Assistance for Needy Families, and unemployment insurance among 342 single mothers who transitioned from employment to unemployment during the Great Recession. Background: Clinical reports show a positive correlation between phytosterol concentrations and severity of cholestatic liver disease markers in infants during long-term administration of parenteral lipid emulsions. Establishing a causal link between phytosterols and cholestasis has been complicated by confounding factors of lipid emulsion load, fatty acid composition, and vitamin E in many of these studies. The goal of this study is to determine whether altering the phytosterol concentration within a common soybean oil-based emulsion will alter the onset and severity of cholestasis in parenterally fed preterm piglets.

Background: Stroke is one of the leading causes of mortality across the world. However, there is a paucity of information regarding mortality rates and associated risk factors in patients with acute ischemic stroke (AIS) undergoing endovascular thrombectomy (EVT). In this study, we aimed to clarify these issues and analyzed previous publications related to mortality in patients treated with EVT.

Background: High-grade glioma (HGG) is a malignant brain tumor that is common and aggressive in children and adults. In the current medical paradigm, surgery and radiotherapy are the standard treatments for HGG patients. Despite this, the overall prognosis is still very bleak. Studies have shown that platelet-derived growth factor receptor α (PDGFRA) is an essential target to treat tumors and inhibiting the activity of PDGFRA can improve the prognosis of HGG. Feeding styles of parents have been associated with dietary quality/intake and weight outcomes; however, much of the research to date has been cross sectional and the direction of influence unclear. This prospective longitudinal study evaluated the direction of effects between feeding styles and child appetitive traits over time in a sample of 129 Hispanic parent/child dyads that participated in a larger study. Data analyzed for the current study were collected when the children were 4-5 years old and again at ages 7-9 years. Objective: To validate the Family Health Behavior Scale (FHBS) for Brazilian families.
Methods: The sample included 272 children aged 5 to 12 years old. Caregivers and their healthy answered the FHBS and questions about physical activity. In addition, anthropometric measurements of the children's weight and height were performed, as well as the bioimpedance exam. Pro-opiomelanocortin (POMC) neurons are important for the regulation of body weight and glucose balance. The inhibitory tone to POMC neurons is mediated primarily by the GABA receptors. However, the detailed mechanisms and functions of GABA receptors are not well understood. The α5 subunit of GABAA receptor, Gabra5, is reported to regulate feeding, and we found that Gabra5 is highly expressed in POMC neurons. To explore the function of Gabra5 in POMC neurons, we knocked down Gabra5 specifically from mature hypothalamic POMC neurons using the clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 strategy. Phthalates are ubiquitous endocrine-disrupting chemicals, and research indicates that prenatal exposure to some phthalates may affect neurodevelopment. In a prospective birth cohort study, five first-morning urine samples collected across pregnancy were pooled and the following phthalate biomarkers assessed: sum of di-(2-ethylhexyl) phthalate metabolites (ΣDEHP), sum of diisononyl phthalate metabolites (ΣDINP), sum of dibutyl phthalate metabolites (ΣDBP), sum of anti-androgenic metabolites (ΣAA), monoethyl phthalate (MEP), and sum of all phthalate metabolites (ΣAll). The tissue-specific molecular mechanisms involved in perinatal liver and intestinal farnesoid X receptor (FXR)-fibroblast growth factor 19 (FGF19) signaling are poorly defined. Our aim was to establish how gestational age and feeding status affect bile acid synthesis pathway, bile acid pool size, ileal response to bile acid stimulation, genes involved in bile acid-FXR-FGF19 signaling and plasma FGF19 in neonatal pigs. The purpose of this study was to determine the knowledge, attitude and behavior influences of Registered Dietitians (RDNs) regarding Vitamin D (VD). An electronic questionnaire served as the data collection tool developed using previously validated questions and supplemented with behavior construct questions. Content and face validity was assessed by an expert panel; internal consistency was assessed using Cronbach’s Alpha.
