Purdue University Libraries Open Access Publishing Fund

Circulating albumin is expected to play a significant role in the trafficking of plasma free fatty acids (FFA) between tissues, such as FFA transfer from adipose tissue to the liver. However, it was not yet known how disrupting FFA binding to albumin in circulation would alter lipid metabolism and any resulting impacts upon control of glycemia. To improve understanding of metabolic control, we aimed to determine whether lack of serum albumin would decrease plasma FFA, hepatic lipid storage, whole body substrate oxidation, and glucose metabolism. Male and female homozygous albumin knockout mice and C57BL/6J wild type controls, each on a …

The Crucial Role Of Sestrins In Oxidative Stress Response And Disease: An Analysis Of Phosphorylation Mutation S110a In Hsesn2, Julianne Fay

Honors Program: Senior Projects (Public)

Sestrins are highly conserved proteins which are induced under environmentally stressful conditions to adjust the cell’s response to such disturbances. This protein possesses two domains critical for its dual function in reducing reactive oxygen species (ROS) and regulating the mTORC1 complex. Sestrins may use diverse transcription factors, such as Nrf2, peroxiredoxin reductase activity, and a catalytic cysteine to regenerate oxidized species; however this was confirmed to be through a mechanism likely different than AhpD, for which there was distance sequence similarity. Furthermore, Sestrins are important regulators of TORC1 and AMPK, which are crucial energy sensors able to control cellular metabolism …

Gap Junction-Dependent And -Independent Functions Of Connexin43 In Biology, Yi Zhu

Children’s Nutrition Research Center Staff Publications

Simple Summary

Connexin43 is one of the 21 members of a large protein family that forms intercellular gap junction complexes. It plays a critical role in development by allowing two adjacent cells to exchange cellular content. Mouse deletion studies have demonstrated its critical roles in many organs, including cardiac muscle, blood vessels, bone, adipose tissue and more. However, as the most expressed and most studied connexin in the family, Connexin43 surprisingly shows many gap-junction-independent roles, spanning from the forming of hemichannels, to regulating membrane trafficking, to regulating mitochondrial function. Connexin43 is also unique in the family by forming many smaller …
