Development Of An On-Line Computer System For Enzyme Kinetics Studies, Jacqueline R. Thweatt

The PDP-11/10 minicomputer was interfaced to the Beckman 25 ultraviolet/visible spectrophotometer and this on-line computer system was applied to enzyme kinetics studies. This interface involved hardware equipment to locate, amplify, and transmit the analog signal to an analog-to-digital converter, and software to acquire and store the digital data. Data processing programs were used to calculate kinetic parameters and statistical information from the acquired data.
The Varian Cary 219 ultraviolet/visible spectrophotometer can be readily interfaced to the Apple II Plus microcomputer by hardware equipment which can be purchased from the manufacturer. Various software may also be purchased for use in enzyme …

Human serum high density lipoprotein subclasses, HDL2 and HDL3 , were isolated by preparative salt density gradient ultracentrifugation and further analyzed by electrophoresis on a 4 to 15% concentration gradient polyacrylamide gel.
The separation of the major classes of serum lipoproteins was achieved after a single ultracentrifugation for 272,000 g at 15°c in a swinging bucket rotor. High resolution concentration gradient gel electrophoresis was found to be particularly suitable for the separation of lipoproteins. The isolation of HDL by precipitation methods, heparin-manganese and dextran-magnesium have also been evaluated in this study, and were found not to completely separate …

The cloning of DNA fragments derived from the (beta)-like globin gene cluster of normal and (beta)('+)-thalassemic individuals is described. These and other cloned human DNA fragments are employed to define the strucure and transcription of a family of interspersed, repetitive RNA polymerase III transcription units in human DNA. These data allow unequivocal identification of these transcription units with the Alu family of interspersed, repetitive DNA sequences. Comparison of these sequences to those of other primate and rodent interspersed repeats reveals that the primate sequence resembles in many respects a dimer of the rodent sequence. Spontaneous mammary carcinoma in C3H mice is preceded by premalignant hyperplastic alveolar nodules. When a primary tumor is evident, examination of the other mammae will reveal a number of microscopic precancerous lesions in various stages of transformation to overt malignant tumors. In an effort to inhibit the development of additional malignant tumors after surgical removal of the first apparent tumor, we treated female C3H/HeJ mice with subcutaneous injections of levamisole (0.6 mg per kg of body weight) on a twice weekly regimen. Acid phosphatase was the first tumor marker to be measured in blood. Although it is used as a marker of prostatic carcinoma, serum levels are also elevated in many diseases of prostate, bone, and hematologic tissue. While serum acid phosphatase concentration is elevated in patients with prostatic carcinoma or infarction, it may also be increased merely with benign hypertrophy and after prostate massage. Recent studies demonstrate that levels are not significantly increased after routine rectal examination. Although of limited use as a screening test, the assay may be useful in staging, prognostication during hormonal therapy, and in making clinical decisions. …

Mechanical expansion of the maxilla, at the midline suture, is a widely accepted orthodontic procedure used to gain arch width in certain types of maxillary deficient arches.
In animal research on the mandible, techniques using surgical separation at the symphysis with immediate expansion up to 12 mm, fixation with a metal splint, and autogenous bone transplanted into the expanded area, have been reported to be successful.
To date, no experiment utilizing slow mechanical expansion of the mandible at the symphysis has been reported.
In this pilot study, the mandible of a female baboon was surgically separated A-P at the symphysis. …

The in vitro mammalian virus inductest is based on the measurement of induced viral gene expression from Simian virus 40 (SV40) transformed weanling Syrian hamster kidney cells (clone E). Upon challenge by many DNA-damaging chemical agents, infectious virions were produced which were quantitated by assay on a cell line permissive to SV40, such as the highly contact inhibited African green monkey kidney cells, CV-1P. Some known carcinogens and non-carcinogens were tested in this research including mitomycin C, aflatoxins Bl and Gl, sterigmatocystin and several polycyclic hydrocarbons.

The effects of a physiologically balanced mixture of amino acids on the synthesis of proteins has been investigated. The roles of leucine and tryptophan, both implicated in the regulation of protein synthesis, were also studied. The balance of amino acids is important under specific rate-limiting conditions; the physiological balance "protects" the protein synthesizing system from the stressed condition of leucine limitation. Leucine is an important regulator of protein synthesis and the tryptophan effect on translation is dependent on the concentration of leucine. Thus tryptophan is a secondary regulator. The purpose of this research was to determine the significance of attendance of Lamaze classes and(or presence in the delivery room on paternal attachment behaviors. Seventeen white, first-time fathers, ages 20 to 35, were observed in the mother's hospital room within 72 hours of birth using a modified paternal attachment tool. The behaviors observed included verbalization, inspection, holding, en face, smiling, and tactile response. Observations were scored on 30-second intervals for a total of 15 minutes. Fathers were then placed into one of the following groups: Group I (n 10) — Fathers from Lamaze classes and present in the delivery …

To determine if excessive systemic exposure to aluminum would be reflected in increased aluminum concentration in hair, rabbits were given a series of aluminum lactate injections. Hair was collected before the aluminum lactate administration from the site of injections and twice after the injections from this site as well as from an area adjoining the injection site. Aluminum was determined by flameless atomic absorption analysis of acid-digested samples. The concentration of aluminum in the hair increased after the injections in samples taken at both times from both sites. This paper reviews the literature cm rational-emotive therapy (RET). The focus was on literature published from 1975 to the present, with emphasis on literature reviews and theoretical studies. Background material by Albert Ellis was included. A historical perspective is given of RET's origin and development. Its philosophy,, theory, and techniques are surveyed. Articular attention was given to RET's application to emotional education and undisturbed populations, especially those who exhibit creativity, high productivity, and above-average capabilities.
Most writers confirm the validity of RET’s theoretical base and conclude that it is effective as a preventive and curative method of psychotherapy. Others criticize …

The literature concerning the effects of d, l-parachlorophenylalanine (PCPA) upon shock-induced aggression (SIA) is reviewed and found to be inconsistent. PCPA, a known serotonin depletor, has behavioral effects in a variety of other procedures which collectively suggest that PCPA should produce SIA enhancement. The present study was designed to analyze PCPA (300 mg/kg) effects upon SIA in rats restrained spatially close to an inanimate target and panel operandum. The results showed marked increases in both aggressive biting and panel pressing for several days following each PCPA treatment for each subject tested. These data were interpreted to indicate that serotonin depletion …

The effects of several levels of exercise on inappropriate behaviors of four trainable mentally impaired subjects were observed in a school setting. Treatment conditions, which occurred as the first daily activity, included (a) Phase I, daily alternating conditions of warm-up exercises and jogging at a moderate rate for a short distance; (b) Phase II, daily alternating conditions of no exercise and jogging at a vigorous rate for a moderate distance; and (c) Phase III, jogging at a vigorous rate for a moderate distance on consecutive days. This study attempted to investigate a sample of both parents' and college freshmens' attitudes toward sex education, and the content and timing of sex education in the Tidewater schools. Two Likert-type scales each containing ten statements were administered in order to survey attitudes. A statistically significant difference was found in scores on the attitude toward content and timing of sex education scale between parents and college freshmen. Parents demonstrated a more favorable attitude toward sex education as an integral component of the overall educational curriculum. The majority of parents indicated support for including important aspects such as reproduction and indicated …

The successful inpatient treatment of emotionally troubled adolescents must facilitate their achievement of developmentally appropriate independence while minimizing the maladaptive dependency that can be fostered by the hospitalization experience. Levels of interpersonal dependency were measured in seven adolescent psychiatric inpatients over a ninety day period. The Interpersonal Dependency Inventory by Hirschfeld, Klerman, Gough, Barrett, Korchin, & Chodoff was utilized. The individual therapist of each participant independently assessed the participant's response to hospitalization at forty-five and ninety days using a five point Leikert type scale. The study demonstrated no significant changes in dependency levels at forty-five and ninety days. The therapist's …

The purpose of this survey was to assess the aseptic techniques currently used in 197 accredited dental hygiene programs in the United States. A self-designed, 51-item questionnaire examined the aseptic techniques used in: (1) preparation of the dental hygiene clinic and unit, (2) instrument preparation, (3) preparation of the student practitioner, (4) preparation of the patient, (5) preparation of radiographic equipment, and (6) treatment of the hepatitis patient.
The questionnaire and a letter of transmittal were mailed to the clinical supervisor of each accredited dental hygiene program. Data were obtained from 173 returned questionnaires and analyzed via modal and percentage …

Follicular center cell lymphoma with sclerosis is a significant clinicopathologic entity which has received little attention to date. Its uniqueness has been obscured by quarrels over classification of the non-Hodgkin's lymphomas; and by the reluctance of some to accept it as a bona fide pathologic subtype in the retroperitoneum, its most common site of presentation. This thesis examines the clinical and pathological findings in a series of 22 cases of retroperitoneal follicular center cell lymphoma with sclerosis obtained from the hematopathology files of Yale-New Haven Hospital. Strange (unfamiliar) males affect survival of progeny of reproductive female rodents in a variety of ways. In mice, strange males block pregnancy before implantation of embryos (Bruce, 1959) but only in females that are not lactating (Bruce and Parkes, 1961). In voles these males can block pregnancy both before and after implantation (Stehn and Richmond, 1975; Marks and Schadler, 1979; Schadler, 1981). Strange males have also been shown to kill nursing pups in lemmings (Mallory and Brooks, 1978) and mice (Labov, 1980 and vom Saal and Howard, 1982).

Zoonoses are infections transmitted by animals to man. Of the more than 50 such infections that are clinically significant, several are discussed in the articles by McIntosh and Madhavan and by Meyer and Adar in this issue of the Journal (pp. 7-13). These include brucellosis, leptospirosis, relapsing fever, Colorado tick fever, toxoplasmosis, psittacosis, and echinococcosis. The McIntosh and Madhavan article reviews the experience at Henry Ford Hospital with 60 patients, while the article by Meyer and Adar reports on an unusual case of echinococcosis of the spine and retroperitoneum that occurred at Haim Sheba Medical Center in Israel.

Antibody raised against the purified human erythrocyte glucose transporter specifically precipitated four proteins from normal and Rous sarcoma virus-transformed chicken embryo cells: a major protein of Mr 41,000 and minor proteins of Mr 68,000, 73,000, and 82,000. The Mr 41,000 and 82,000 proteins were found only in a membrane fraction, not in the soluble fraction, and displayed a heterogeneous mobility on NaDodSO4/polyacrylamide gel electrophoresis, suggesting glycosylation. The Mr 41,000 and 82,000 proteins were increased in amount after malignant transformation in direct proportion to the increase in hexose transport rate, and the increase was dependent on the expression of the src …

Millions of dollars are spent annually on special educational programs for children whose severe behavior disorders prevent them from participating in the regular school setting despite average or above average intellectual capacity. A growing body of research indicates that some of these behavior disorders are related to nutritional problems. (Pfeiffer and Iliev 1972; Kittler 1973; Mayron 1979; and Buckley 1977), and many clinicians support the view that no matter what the etiology of behavioral disorders, nutritional programs can improve the baseline data on medical, social and intellectual achievement, and on personality measures (Palmer 1978).
