Cleavage and polyadenylation of substrate RNAs containing the herpes simplex virus type 1 (HSV-1) thymidine kinase (tk) gene polyadenylation signal region were examined in HeLa cell nuclear extract. 3'-End RNA processing was accurate and efficient and required ATP and Mg2+. Cleavage, but not polyadenylation, occurred in the presence of EDTA or when ATP was replaced with 3' dATP (cordycepin) or AMP(CH2)PP, a nonhydrolyzable analog of ATP. Processing in vitro and in vivo showed the same signal element requirements: a series of substrates containing linker scanning, internal deletion, and small insertion mutations was processed with the same relative efficiencies and at …

The study was designed to investigate the hypothesis that various environmental stimuli would elicit specific individual responses, as reflected in autonomic patterns. It then became the aim of the study to see whether the stimuli which controlled the physiological pattern would also control the verbal response. Three female and two male students volunteered for the single-subject experiment, where they were exposed to a 15-slide sequence of environment stimuli. Results indicated that when allowed to verbally respond without rating restrictions, responses varied greatly, limiting the possibilities of comparison. The sera from patients with the CREST (calcinosis, Raynaud's phenomenon, esophageal dysmotility, sclerodactyly, telangiectasia) variation of the autoimmune disease scleroderma contain autoantibodies that specifically recognize the kinetochore by immunofluorescence. Two major antigens of molecular masses 18 and 80 kD are consistently identified by Western blotting of proteins of isolated chromosomes using CREST sera. In this paper, the possible roles that these two proteins play in the interaction of metaphase chromosomes with tubulin and microtubules are examined using two different procedures. The purposes of this study. were: to explore job-related perceptions of stress in a group of military critical care nurses; and to determine if differences in anxiety and stress levels existed between nurses working in coronary care (CCCU) and medical/surgical intensive care (ICU) settings using the State/Trait Anxiety Inventory and the Questionnaire of Stressful Factors in Intensive Care. A free response question and a force-response checklist were also used to identify and rank work-related stressors. The sample of Navy nurses (n=105) consisted of 39 CCU nurses, 62 medical/surgical ICU nurses and 4 critical care supervisors. The results of the mailed …

The anatomical relationship of the mandibular canal and related structures were examined in 29 dried human mandibles (21 mature and eight immature). First and second premolars and molars were sectioned in a buccal-lingual plane. The molars were separated into mesial and' distal sections. A total of 322 sections were examined according to age (immature or mature), left or right, and type of tooth/root. Thirteen measurements were taken of each section and the results were statistically analysed.

Early cellular changes in pulps whose blood supplies were severed were studied using transmission electron microscopy (TEM). Twenty-eight extracted teeth were divided into seven equal groups. Except for the 0 hour group, all other teeth were incubated in Tyrode solution at 37°C, after extraction. After 1/4, 1/2, 1, 2, 4, and 8 hours, teeth were split open and the pulps were processed for light and transmission electron microscopy and evaluated for cellular changes. Except for nuclear pyknosis in the 4 and 8 hour groups, light microscopic examination revealed very little changes when compared with that of TEM examination. With the …

The present study evaluated the efficacy of a four-week seminar which emphasized the principles of Agentive Theory. This theory, which is compatible with theories of a phenomenological/ existential perspective, was first developed by C. T. Warner, an American philosopher. Agentive Theorists/Therapists emphasize that our negative emotions, ie., depression, anger, etc. , are assertions or judgments we make and not merely feelings we are responsible for controlling or expressing. Forty-eight outpatients who sought help with personal/emotional problems from a department of behavioral medicine were assigned to either a treatment or waiting-list control Group. Distribution Of Vertebrates Of The Bighorn Canyon National Recreation Area, Stanley H. Anderson, Wayne A. Hubert, Craig Patterson, Alan J. Redder, David Duvall
Great Basin Naturalist
During a survey of the vertebrates in the Bighorn Canyon National Recreation Area, 46 mammal, 210 bird, 9 reptile, 5 amphibian, and 28 fish species were found. Habitat structure and moisture were two environmental variables associated with species numbers across this 1,820-ha area. Eight distinct habitats were evaluated. In coniferous forests, 27% of the birds and 54% of the mammals were observed. Sagebrush/grasslands and upland shrublands were very arid and had relatively few vertebrates. Five introduced game birds occurred in sagebrush habitat. Riparian and wetland habitats had the largest number of unique vertebrates, 38 and 77 respectively. The establishment of …

Using chlorine as a drinking water disinfectant may have potential health effects due to its reactivity with organic amino nitrogen compounds found in the stomach. Organic N-chloramines have been shown to form in the stomachs of laboratory rats. The possible reactions of N-chloramines in the stomach fluid were examined in this study using a model radiolabeled N-chloramine. 36Cl-N-Chloropiperidine, was synthesized and purified to remove 36Cl-chloride. Stomach fluid was obtained from Sprague-Dawley rats which had been first fasted for 24 or 48 hours and then administered 3 mL of deionized water. The present study evaluated the implications of a family systems approach for preventing psychiatric hospitalization by comparing the behavior of field clinicians who professed a systems view versus clinicians who held more traditional nonsystems views. The focus of the investigation was the clinical assessments and decisions of 31 clinicians who worked at five Virginia public mental health centers as they prescreened (i.e., evaluated need for inpatient treatment) 171 candidates for hospitalization. Q-technique was employed to determine theoretical orientation.

Eighteen cases of medullary thyroid carcinoma (MTC) (11 hereditary) had parallel measurements of basal plasma levels of somatostatin (SRIF), levels of SRIF extracted from fresh tumor tissue (radioimmunoassay), and immunohistochemistry (peroxidase-antiperoxidase method) using an anti-SRIF antibody on paraffin sections. Immunohistochemistry disclosed some cell positivity in nine cases (50%). Plasma levels of SRIF were above normal (> 50 pg/mL expressed in terms of the 14 amino acid SRIF peptide) in only two of 12 patients. Extraction of fresh tumor tissue detected SRIF in 60% of our cases: two patients > l.200pg/mg (equivalents 14). with the other positive values ranging between 13 and …

In this retrospective study of 68 cases of medullary thyroid carcinoma (MTC), morphological features were studied, which could be helpful in prognosis and in the distinction between hereditary and sporadic forms of the disease. Necrosis within the tumors seemed to carry a poor prognosis. Bilateral MTC. unquestionable C-cell hyperplasia, glandular or follicular pattern, and thyroglobulin-positive cells seemed to be more prevalent in hereditary forms of the disease. A smaller proportion of calcitonin-immunoreactive cells were encountered in hereditary tumors than in sporadic tumors.

We have employed the TT cell line, a model for the human medullary thyroid carcinoma cell, lo study the regulation of peptide hormone production by glucocorticoids. Complementary DNA probes were used to measure the calcitonin (CT), CT gene-related peptide (CGRP), and somatostatin (SRIF) mRNA levels. Dose-response experiments in serum-free medium showed that dexamethasone (six-day treatment) lowered somatostatin (to 1% of basal) and CGRP mRNA (to 50% of basal) and stimulated CT mRNA (threefold to thirteenfold) with a half-maximal effective concentration of 10−8 M. Ten pheochromocytomas (five from patients with multiple endocrine neoplasia type 2 and five sporadic) were stained immunohistochemically with antibodies to calcitonin (CT) and calcitonin gene-related peptide (CGRP) by means of the peroxidase-antiperoxidase method. CGRP positive cells were found in variable numbers in all of the investigated tumors. No demonstrable difference was noted between the hereditary tumors and the sporadic tumors. Staining with CT antibody also showed cells with a positive reaction in eight of ten tumors. The distribution of the two peptides was similar, and in some cells their coexistence was visualized. Doubling times of basal and stimulated calcitonin (CT) levels and of random carcinoembryonic antigen (CEA) levels were compared over time in 11 patients with residual medullary thyroid carcinoma (MTC)—five with the sporadic MTC and six with the hereditary MTC. Four patients with an indolent form of the disease showed little change in CT levels over three to eight years and little change in CEA doubling times (42 to 70+ months). A tissue culture explant and a primary cell culture of malignant C-cells have been estabUshed to study the secretion pattern of calcitonin (CT), katacalcin (KC), and CT gene-related peptide (CGRP) in vitro under different conditions. Within the first five days of culture the spontaneous secretion of the peptides in both systems dropped to and remained at a constant level. With increasing calcium (Ca) concentration in the medium, a dose dependent release of CT, KC and, in cell culture, of CGRP was observed. A radiolabeled anticalcitonin monoclonal antibody (MoAb) was testedfor tumor-localizing capability in WAG/Rij rats allografted with cultured medullary thyroid carcinoma cells (line 44-2). 131I-labeled anticalcitonin IgG was taken up by the tumors in a time-dependent and dose-dependent manner. Tumor tissue uptake of the injected MoAb was 0.7 to 1.3%/g of tumor tissue when tested with a dose range of 40 lo 250 μg/kg of body weight. Distribution ratios of the radiolabeled MoAb were 1.2:1 for tumor-to-blood and 2-20:1 for tumor-to-organs. In rats injected with control IgG, disiribution ratios of radioactivity were 0.4:1 for tumor-to-blood, and 0.6-2:1 for tumor-to-organs. These results suggested …

The steroid hormone, 1 alpha,25-dihydroxyvitamin D3 (calcitriol), has been shown to inhibit T cell proliferation, primarily through inhibition of interleukin 2 (IL-2) production. In these experiments, we show that calcitriol also markedly inhibited production of the lymphokine, gamma interferon (IFN-gamma), by activated human T lymphocytes. Regulation of both IL-2 and IFN-gamma production as well as transferrin receptor (TfR) expression by calcitriol was apparent at the messenger RNA (mRNA) level as determined by Northern blotting. The decrease in IL-2 and IFN-gamma mRNA that occurred with calcitriol treatment was coordinate and not apparent up to 12 h after phytohemagglutinin stimulation, whereas decreased …

The correction of anterior open bite in dolichofacial individuals with magnetic forces has been studied. The attempt was made to intrude buccal segments by using interarch magnets set in repulsion in order to correct the anterior open bite of six dolichofacial patients. It was expected to intrude the posterior sections with these magnetic forces, to autorotate the mandible and to close the anterior open bite by decreasing the lower facial height and rotating the facial axis closed.
