Identification Of Persistent Defects In Insulin Receptor Structure And Function In Capillary Endothelial Cells From Diabetic Rats, Ching Fai Kwok, Barry J. Goldstein, Dirk Muller-Wieland, Tian-Shing Lee, C. Ronald Kahn, George L. King

Insulin actions and receptors were studied in capillary endothelial cells cultured from diabetic BB rats and their nondiabetic colony mates. The endothelial cells from diabetic rats of 2 mo duration had persistent biological and biochemical defects in culture. Compared with normal rats, endothelial cells from diabetic rats grew 44% more slowly. Binding studies of insulin and insulin-like growth factor I (IGF-I) showed that cells from diabetic rats had 50% decrease of insulin receptor binding (nondiabetic: 4.6 +/- 0.7; diabetic: 2.6 +/- 0.4% per milligram protein, P < 0.01), which was caused by a 50% decrease in the number of binding sites per milligram protein, whereas IGF-I binding was not changed. Insulin stimulation of 2-deoxy-glucose uptake and alpha-aminoisobutyric acid uptake were also severely impaired with a 80-90% decrease in maximal stimulation, in parallel with a 62% decrease in insulin-stimulated autophosphorylation (P < 0.05). 125I-insulin cross-linking revealed an 140-kD alpha subunit of the insulin receptor similar to …

Shiga toxin, Shiga-like toxin I (SLT-I) and Shiga-like toxin II (SLT-H) are cell-associated cytotoxins that kill both Vero cells and HeLa cells, whereas Shiga-like toxin II variant (SLT-llv) is an extracellular cytotoxin that is more cytotoxic for Vero cells than for HeLa cells. The basis for these differences in cytotoxin localization and host cell specificity were examined in this study. The A and B subunit genes of Shiga toxin and the SLTs were recombined by two methods so that hybrid toxins would be formed in vivo. Complementation of heterologous subunits was accomplished by cloning the individual A and B subunit …

This thesis explores how support groups can reduce perceived stress among caregivers of individuals with Alzheimer’s disease. Linda Blewer Swindle, a registered nurse, provides a comprehensive look at both the emotional and practical challenges faced by those providing care, particularly as the disease progresses through its stages. The work begins with a clinical overview of Alzheimer’s, differentiating it from normal aging and emphasizing the disease’s slow, irreversible, and deeply disruptive nature.

Activation of the insulin receptor tyrosine kinase in vitro and in vivo was studied in two rat models of insulin resistance: hypercortisolemia and streptozotocin-induced diabetes. In control rats, intravenous insulin administration resulted in dose-dependent in vivo activation of the skeletal muscle insulin receptor kinase. Chronic cortisone treatment did not affect the kinase activity of receptors solubilized from muscle after activation with insulin in vivo or in vitro. In contrast, diabetes depressed skeletal muscle insulin receptor kinase activation in vivo by ~50%. Diabetes impaired the in vitro insulin stimulation of receptor kinase activity from skeletal muscle and liver towards the substrate …

This edition of Quality/Calidad/Qualité gives information about a postnatal family planning program in the Sfax Center of the Tunisia Maternity and Newborns Hospital. This program, developed jointly by the national family planning program and the Maternity staff, has had extraordinary success in getting more mothers to return after six weeks for their postnatal visit. During the visit, the period between births is discussed; and family planning services are offered as an important means for achieving both the recovery and good health of the mother, as well as the physical and mental development of the infant. The purpose of this study was to determine if nurses who were given a tool to facilitate assessment of thought processes would collect more assessment data, diagnose "alteration in thought processes" more frequently, and include a care plan for this diagnosis more often. A quasi-experimental pre-treatment/post-treatment design was used. An inservice program about the Level of Cognitive Function Scale (LOCF) was the treatment.; Of the 40 randomly selected subjects from the 199 adult critical care nurses at a metropolitan hospital, 20 completed the study with pre-treatment and post-treatment data. The skeletal remains of an adult male associated with desiccated tissue and a coprolite were recovered from an apen-air midden deposit at the NAN Ranch Ruin (LA15049)) a large Mimbres site in Grant County) New Mexico. The find dates to about A.C. 1000-1100. Identifiable macroscopic elements in the caprolite consist offi nely fragmented corn and tiny seed fragments of an unknown plant. High amounts of willow (Salix) and mustard (Brassicaceae) pollen may indicate the ingestion of medicinal plants to combat a deteriorating health condition. The individual was approximately 35 -40 years old at the time of death and suffered from …

One of the main problems encountered in the review of the bioarcheology of Region 3 has been the limited number of sites where human skeletal material has been adequately recovered and analyzed. In the preceding chapter it was documented that less than 30% of the burials recovered from recorded sites have been reported in published literature. It was further estimated that of the 323 sites with burials, no more than 80 sites have published detailed bioarcheological reports on the burials recovered. Only 50 of these 80 reports provide individual descriptions of each burial which facilitate subsequent analyses and evaluation.

Health care professionals continue to be concerned about the relief of pain and anxiety associated with childbirth. There are recent anecdotal reports in the literature which describe showering as an effective nonpharmaceutical alternative for labor pain management. The effectiveness of showering can most likely be explained physiologically by the "gate control" theory of pain.; The purpose of this descriptive-correlational study was to examine the labor patient's perception of the effectiveness of showering as a comfort measure and the relationship between showering in labor and outcome variables such as analgesia use, anesthesia use, and childbirth satisfaction. Methylxanthines (MX), such as theophylline, are commonly used in the treatment of recurrent apnea due to their stimulant effects on the respiratory center. Structure-activity studies have demonstrated that substitution of alkyl groups on the 3-position of the xanthine nucleus results in increased bronchodilator potency, while substitution on the 1-position is important for adenosine antagonism and CNS stimulation. Three different alkylxanthines were studied to determine the structural requirements for respiratory stimulation. Respiratory rates and volumes were determined in 4- to 7-day-old rats using a volume displacement body plethysmograph. Ten patients with idiopathic thrombocytopenic purpura (ITP) were studied before and following a rise in circulating platelets subsequent to infusions of intravenous gammaglobulin (400 mg/kg/day x 5 days). We quantitated the amount of circulating IgG capable of binding to normal donor platelets in vitro using an 125I-monoclonal anti-human IgG assay, as well as the amount of IgG associated with the patients' platelets before and following therapy. We found no evidence for a decrease in platelet-specific IgG antibodies in these patients undergoing an acute response to therapy. These data suggest that the short-term efficacy of intravenous gammaglobulin is due to effects …
