Selection Of Genetic Variants Of Simian Immunodeficiency Virus In Persistently Infected Rhesus Monkeys, Dawn P. Wooley, Ronald C. Desrosiers

Genetic and antigenic variation may be one means by which lentiviruses that cause AIDS avoid elimination by host immune responses. Genetic variation in the envelope gene (env) was studied by comparing the nucleotide sequences of 27 clones obtained from two rhesus monkeys infected with molecularly cloned simian immunodeficiency virus. All 27 clones differed from each other and differed from the input clone in the gp120 (SU) portion of the envelope gene. Nucleotide substitutions were shown to accumulate with time at an average rate of 8.5 per 1,000 per year in SU. Surprisingly, the majority of nucleotide substitutions (81%) resulted in …

The purposes of this study were: (a) to describe differences , if any, in sex-role traits, age and number of children, proportionate income, and responsibility for household tasks and childcare between female registered nurses who return to college and those who do not; and (b) to extend the research regarding the adult learner's return to college in the specific area of baccalaureate degree completion by registered nurses. Chi square and analysis of variance of survey data, and interviews were used in determining differences between the student and nonstudent groups.

The purpose of this study was to determine the effects of scaling and root planing with the modified P-10 CavitronR tip and curet instruments on root surface topography. Sample specimens included 20 periodontally involved extracted human teeth. Specimens were carved into a three split root design, each sample specimen acting as its own control. Control and experimental surfaces were randomly assigned to each specimen. Scaling techniques were applied on the assigned experimental surfaces for 2 minutes and 14 seconds. The purpose of this study was to investigate whether practicing dental hygienists promote their profession as a desirable career. A self-designed, 48 item questionnaiire, Dental Hygienists' Participation in Career Recruitment, was mailed to a random sample of 600 dental hygienists residing in the Commonwealth of Virginia. Data were obtained from 393 dental hygienists for a 65.5 percent response rate. Responses from each item were analyzed by frequency distributions, percentages and chi-square test for independence at a 0.05 level of confidence. The purpose of this study was to identify the range of problems and coping strategies as perceived by antepartal patients experiencing medical treatment to decrease risk of preterm labor. In this qualitative, interpretive study problems and coping strategies were identified through semi-structured interviews with a convenience sample of seven antepartal patients. Using content analysis of interview data, identified problems were categorized, and for each problem, coping strategies were identified and grouped using the categories of the Jalowiec Coping Scale. The health-promoting behaviors, perceived health status, and perceived health education needs of older women were identified in this non-experimental, descriptive, correlational study. Fifty women over 60 years of age, living in three housing complexes for the elderly, disabled and handicapped in two southeastern United States cities were asked to complete The Health-Promoting Lifestyle Profile, The Perceived Health Status Questionnaire, The Health Education Needs Questionnaire, and Demographic Data Sheet. This study uses the post-test only design to assess the preliminary effects of a community-based policy initiative, the Norfolk State University Resource Mothers Program (RMP), on the gestational ages, birth weights, and survival rates of infants born to participating adolescent mothers.

The RMP uses experienced mothers or paraprofessionals similar in race and socio-economic status to the participating teenagers. These persons are trained to assist pregnant adolescents and teen parents with nonmedical dimensions of pregnancy and child care. They recruit teens into the RMP, encouraging them to get early prenatal care. Resource Mothers provide teen mothers and their families with practical …

There is an expressed need to develop a greater variety of safe contraceptive methods which would find acceptance worldwide, particularly in developing countries. Immunization against spermatozoa might be such a method as judged by the accumulated evidence from studies on human females and in a number of female animal species.

Two extraction techniques were used for sperm membrane antigen isolation. The first technique involved NP-40 detergent for antigen extraction from human motile sperm and the second technique employed homogenized human testis for antigen extraction. Recent animal studies have shown significant mercury absorption from dental fillings and resulted in unfavorable media attention. Yet, an FDA advisory committee has found no evidence of Risk to dental patients, and many dentists believe that patients are being unnecessarily alarmed. The paper reviews the history of amalgam fillings through the recent animal studies and concludes that the Risk, whatever it may prove to be, is sufficiently high to warrant permitting patients to choose between amalgam and alternative dental filling materials.

Case management strategies for the nutritional support of patients infected with the human immunodeficiency virus (HIV) are evolving as the disease becomes less rapidly fatal and more chronic. Nutritional status changes in advanced HIV infection are similar in many respects to protein-calorie malnutrition. Current clinical effort and research focuses on the beneficial effects of preserving lean body mass and keeping asymptomatic patients in good nutritional status by preventing micronutrient deficiencies and by treating preexisting nutritional problems rather than attempting to intervene late in the disease's course, after secondary malnutrition has already developed. Treatment with human growth hormone (GH) at Henry Ford Hospital began in 1964 and has included 145 patients. A total of 68 patients were treated with extracted hormone and 77 with recombinant GH. The appearance of Creutzfeldt-Jakob disease, now five cases worldwide, in patients treated with extracted hormone abruptly stopped its use in the United States. The development of recombinant GH in 1985 has resulted in greater availability of treatment. Diagnostic criteria are now more liberal and certain patients without GH deficiency, i.e., those with the Turner syndrome, are now being successfully treated with GH. GH is expensive and its …

Determining the cause of Cushing's disease and correcting the abnormality presents a continuing challenge to the clinician despite remarkable advances in diagnostic and therapeutic techniques. Contemporary Approach To Thyroid Disease Emphasizing Use Of High-Sensitivity Thyrotropin Assays, Carolyn S. Feldkamp, Malachi J. Mckenna

Capabilities of new high-sensitivity immunoradiometric assays for thyroid-stimulating hormone (TSH-IRMA) to distinguish among hypothyroid, euthyroid, and hyperthyroid subjects and patient groups with low TSH for nonthyroidal causes suggested an algorithmic approach (directed TSH) to the evaluation of patients with suspected thyroid disease. Utilizing the algorithm, a TSH-IRMA result outside normal limits (0.5 to 5.0 mUlL) generates follow-up tests on the same sample. The interpretation of thyroid function tests (TSH-IRMA, thyroxine, resin uptake, free thyroxine index) and associated studies in the context of different clinical settings is reviewed. The approach is a cost-effective and efficient utilization of laboratory services.

Studies were undertaken to investigate acquired resistance to cisplatin in human ovarian cancer cells. The cell lines A2780 and A2780/CP70 were studied to assess their respective characteristics of drug accumulation and efflux, cytosolic inactivation of drug, and DNA repair. All experiments were performed using 1-h drug exposures. The A2780/CP70 cell line was 13-fold more resistant to cisplatin than A2780 cells. When studied at their respective IC50 doses, drug accumulation rates were similar for the two cell lines. However, the resistant cell line was twofold more efficient at effluxing drug, which was associated with reduced total drug accumulation for equivalent micromolar …

To facilitate this study an in vitro human model system was established that exhibited many aspects of normal osteoblasts. The human osteosarcoma cell line (TE85 cells) expressed a skeletal alkaline phosphatase activity (an accepted bone cell differentiation marker) which was stimulated by 1,25(OH)2D3 (a potent differentiating agent), under serum-free conditions in a dose-dependent, time-dependent, and cell density-dependent manner. Cytochemical analysis of the stimulation of ALP activity by 1,25(OH)2D3 showed that 1,25(OH)2D3 increased the number of TE85 cells that expressed detectable ALP activity, suggesting that 1,25(OH)2D3 promoted the process …

Identification of factors that influence obese adolescents' health behaviors is necessary if obese adolescents are to benefit from therapeutic interventions. The purpose of this descriptive-correlational study was to identify the relationships between selected basic conditioning factors (BCFs)--perceived health status (PHS), perceived self-efficacy (PSE), family satisfaction (FS), and life events (LE)--and the self-care agency (SCA) and self-care practices (SCP) among obese adolescents. The relative predictive strength of each BCF in relation to the outcome variables was also explored. Orem's self-care theory provided the conceptual framework for this study. An alternative approach to structurefunction analysis of vesicular stomatitis virus (VSV) gene products and their interactions with one another during each phase of the viral life cycle is described. We showed previously by using the vaccinia viruslT7 RNA polymerase expression system that when cells expressing the nucleocapsid protein (N), the phosphoprotein (NS), and the large polymerase protein (L) of VSV were superinfected with defective interfering (DI) particles, rapid and efflicient replication and amplification of DI particle RNA occurred. Interieukin-1, a mediator of inflammation, or tumor promoting phorbol esters induce transcription of stromelysin, a metalloproteinase that degrades extracellular matrix molecules and that is overexpressed in diseases such as rheumatoid arthritis. Sequences required for induction of transcription of the human stromelysin promoter are contained on a 46 base pair fragment. This fragment contains a sequence with a high degree of similarity to the binding site for the transcription factor activator protein-1 (AP-1) and indeed, the AP-1 sequence of this fragment is necessary but not sufficient for the maximal response to phorbol 12-myristate 13-acetate (phorbol) or interleukin-1. The impact of renal insufficiency on the dispositions of 300 mg of orally administered ceftibuten-cis, a new broad-spectrum oral cephalosporin, and its primary metabolite ceftibuten-trans was characterized in 30 adult subjects. Subjects were divided into five groups of six subjects each on the basis of their 24-h ambulatory creatinine clearances (CLCR). Extracellular tetraethylammonium (TEA) inhibits currents in Xenopus oocytes that have been injected with mRNAs encoding voltage-dependent potassium channels. Concentration-response curves were used to measure the affinity of TEA; this differed up to 700-fold among channels RBK1 (KD 0.3 mM), RGK5 (KD 11 mM), and RBK2 (KD greater than 200 mM). Studies in which chimeric channels were expressed localized TEA binding to the putative extracellular loop between trans-membrane domains S5 and S6. Site-directed mutagenesis of residues in this region identified the residue Tyr379 of RBK1 as a crucial determinant of TEA sensitivity; substitution of Tyr in the equivalent positions of RBK2 …
