Accuracy Of Race And Ethnicity Data In The Pediatric Electronic Health Record: A Concordance And System Adequacy Study, John Cowden, Rachel Drake, Jessi Johnson, Kataina Kelty, Mehwish Ahmed

Manuscripts, Articles, Book Chapters and Other Papers

Introduction: Conventional race and ethnicity categories and analysis are reductive and prone to inaccuracy. Because race and ethnicity data validity is essential to health equity efforts, we measured the accuracy of race and ethnicity data in a pediatric electronic health record (EHR) to identify areas for improvement in data collection and use. Methods: Patients and their caregivers reported patient race and ethnicity via in-person survey in four pediatric settings (inpatient, emergency room, urgent care, and primary care). Race and ethnicity data from the EHR were compared with survey data to calculate four measures of EHR data accuracy. The U.S. Census …

Kusurkar, Sherry Lee, Wei Jiang, Avinoam Nevler, Matthew Mccoy, Michael J. Pishvaian, Rosalie C. Sears, Jonathan R. Brody, Charles J. Yeo, Aditi Jain

Department of Surgery Faculty Papers

Pancreatic ductal adenocarcinoma (PDAC) remains one of the deadliest cancers demanding better and more effective therapies. BARD1 or BRCA1-Associated -Ring Domain-1 plays a pivotal role in homologous recombination repair (HRR). However, its function and the underlying molecular mechanisms in PDAC are still not fully elucidated. Here, we demonstrate that BARD1 is overexpressed in PDAC and its genetic inhibition suppresses c-Myc and disrupts c-Myc dependent transcriptional program. Mechanistically, BARD1 stabilizes c-Myc through ubiquitin-proteasome system by regulating FBXW7. Importantly, targeting BARD1 using either siRNAs or CRISPR/Cas9 deletion blocks PDAC growth in vitro and in vivo, without any signs of toxicity to mice. …

Novel Application Of Orthopedic Suture Tape For Sternal Closure After Upper Hemisternotomy, Vishal N. Shah, Joshua R. Chen, Konstadinos A. Plestis

Department of Surgery Faculty Papers

The FiberTape (FT) System (Arthrex, Naples, FL, USA), initially developed for orthopedic surgery, is a new, nonmetallic, suture-based alternative to standard stainless-steel wires for sternal reapproximation. Publications on FT use after cardiothoracic surgery are limited. The FT System has not been used in upper hemisternotomy (UHS) closure. Consequently, we report the use of the FT System for sternal closure in a 70-year-old man after UHS aortic valve replacement and discuss several advantages of its application.

Concurrent pulmonary artery (PA) blood flow/pressure was measured. RV pressure-volume (P-V) loops were generated with a conductance catheter. Pulmonary vascular resistance was used to stage PE severity into mild, moderate, and severe groups.

Results: All sheep developed graded RHF. For …

Histomorphology Of Sharp-Force Trauma On Partially Cremated Remains, Horaceena Findley May 2025

Histomorphology Of Sharp-Force Trauma On Partially Cremated Remains, Horaceena Findley

Student Theses

Identifying sharp-force trauma in thermally altered human remains presents a significant challenge in forensic investigations, particularly in arson-related deaths where traditional autopsy techniques may be limited. This study explores the efficacy of histological methods in detecting sharp-force trauma on partially cremated remains using pig ribs (Sus scrofa) as a human analog. Simulated tool marks from a knife, screwdriver, and flathead screwdriver were inflicted at varying angles (60° and 90°) and subsequently exposed to high temperatures in an open fire setting. Histological analyses, including Hematoxylin and Eosin Y (H&E) and Gomori’s Prussian Blue staining, were employed to assess tissue structure and …

Joint Modelling Of Longitudinal Egfr Trajectory And Time To Acute Kidney Injury In Lung Transplant Patients, Samiha Zakir May 2025

Joint Modelling Of Longitudinal Egfr Trajectory And Time To Acute Kidney Injury In Lung Transplant Patients, Samiha Zakir

Theses and Dissertations

Progressive declines in estimated glomerular filtration rate (eGFR) often precede acute kidney injury (AKI), yet the relationship between eGFR trends and AKI risk remains unclear. This study investigates longitudinal eGFR changes and their association with AKI in 459 lung transplant patients followed for up to 7 years (n = 6419). We applied a piecewise linear mixed-effects model to evaluate eGFR trajectories and a Cox proportional hazards model to assess time to AKI. A joint model was used to explore the interplay between longitudinal and survival processes. Key covariates included gender, age at transplantation, antibody-mediated rejection (AMR), and pre-transplant eGFR. Males …

Host Genetics Role In Viral Disease Susceptibility, Christian Willis Eaton May 2025

Host Genetics Role In Viral Disease Susceptibility, Christian Willis Eaton

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Viruses and their hosts exist in an evolutionary arms race, with evolutionary promotion of host genetic-based defense mechanisms and evolution of virus genetic-based mechanisms to overcome host defenses. Viruses are capable of much more rapid evolution and adaptation than their hosts, often leading to the use of host cellular mechanisms to promote viral infection and replication. In this study, we describe the generation of a detection assay for atypical porcine pestivirus (APPV), designed to target the viral 5’ UTR, a region of the viral genome which sees less mutations than the protein coding regions. This assay was generated to be …

Men’S Fertility Beliefs, Intentions, And Contraceptive Use, Meagan Kunitzer

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Despite advancements in reproductive technology, gender-inclusive approaches remain limited, and research on men’s reproductive health is scarce. Specifically, little is known about how men conceptualize reproductive health and how perspectives vary by race and social class. As such, the purpose of this study is to further the field’s understanding of men’s reproductive health, including fertility beliefs, intentions, and behaviors. Guided by the theory of conjunctural action, this study uses survey-driven narrative construction to explore how 35 men think about their reproductive health and how this varies by race and class. The main benefit of using this method is that it …

Interferon Regulatory Factor 3 In Interferon Gamma Receptor Signaling And Cancer Immunology, Zachary P. Guinn May 2025

Interferon Regulatory Factor 3 In Interferon Gamma Receptor Signaling And Cancer Immunology, Zachary P. Guinn

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Every year millions of people die from cancer throughout the world, and tens of millions of people are diagnosed with cancer worldwide. When surgery is not an option, treatment options commonly rely on the host immune system for optimal treatment efficacy. The expression of Interferon gamma (IFN-γ) by host immune cells is often associated with favorable antitumor immune responses. How IFN-γ is optimally induced in immune cells, and how IFN-γ promotes the transcription of genes downstream of the Interferon gamma receptor (IFNGR) is incompletely understood. Understanding how to induce immune cells to express optimal IFN-γ …

Electronic Theses and Dissertations

Kentucky Cancer Registry (KCR) prostate cancer (PCa) cases between the years of 1994 and 2021 were analyzed to examine differences between rural and urban populations’ primary stage at diagnosis and PCa-specific mortality. After adjusting for covariates, rural populations did not experience significantly higher odds of stage 3 diagnosis when compared to urban populations (OR = 1.02, 95% CI = 0.95 – 1.06). Rural populations had 16% increased odds of stage 4 diagnosis when compared to urban populations (OR = 1.16, 95% CI = 1.10 – 1.23). Rural populations had a 17% increased rate of PCa-specific death when compared to urban …

The Dynamic Role Of Hyaluronan In Acute Cardiac Wound Healing., Danielle T. Little May 2025

The Dynamic Role Of Hyaluronan In Acute Cardiac Wound Healing., Danielle T. Little

Electronic Theses and Dissertations

During acute myocardial infarction (MI), the composition of the extracellular matrix (ECM) undergoes significant changes. While collagen accumulation is one of the most well-documented ECM alterations, hyaluronan (HA) is equally abundant. However, the cellular sources, enzymatic regulation, and functional significance of HA accumulation remain largely unexplored. Here, we hypothesized that activated cardiac fibroblasts produce HA via hyaluronan synthase 2 (HAS2), facilitating scar formation and stability. Our findings demonstrate that HA accumulates post-MI and persists for at least four weeks. Through HA sizing techniques, we determined the molecular weight of this persistent HA. Additionally, RNA sequencing analysis of failing human hearts …

Improving Brain Health: Exercise As A Tool For Enhancing Executive Function And Reducing Stress In Older African Americans, Destiny Ellise Ramos

Theses, Dissertations and Culminating Projects

Older African American (AA) adults face disproportionately higher risk of stress and Alzheimer's Disease and Related Dementias (ADRD) compared to other racial and ethnic groups, yet they remain underrepresented in intervention-based research. Previous research has suggested aerobic exercise may reduce stress and benefit executive functioning, which is among the first cognitive domains to be affected in ADRD; however, no studies to date have directly tested the impact of aerobic vs. non-aerobic exercise in older AAs at high risk for developing ADRD. The current study evaluated the effects of a group-based cardio dance fitness (CDF) exercise intervention versus a strength, flexibility, …

The Role Of Endogenous Estrogen In Effects Of Non-Invasive Brain Stimulation On Cognitive Performance, Samantha Walsh May 2025

The Role Of Endogenous Estrogen In Effects Of Non-Invasive Brain Stimulation On Cognitive Performance, Samantha Walsh

Theses, Dissertations and Culminating Projects

High-definition transcranial direct current stimulation (HD-tDCS) directly modulates brain activity, allowing the study of brain-behavior relationships in cognitive processes. However, cognitive effects of tDCS vary widely, partly due to individual differences in baseline brain activity. Estrogen levels, which fluctuate during the menstrual cycle, are known to affect cortical excitability and impact neural response to tDCS, but no research to date has examined the potential moderating role of endogenous estrogen in tDCS effects on cognition. In this study, N = 16 healthy female adults completed four sessions of a working memory (WM) task while receiving tDCS (two active sessions, two sham) …

Advancing Drug-Drug Interaction Prediction Using Multi-Modal Feature Integration With Graph Neural Networks, Ernest C. Chianumba

Theses, Dissertations and Culminating Projects

Pharmaceutical treatments are essential for managing medical conditions, but drug-drug interactions (DDIs) pose significant risks to patient safety and healthcare outcomes. This research integrates Knowledge Graphs and Graph Neural Networks to predict DDIs by exploring complex drug relationships. We construct a comprehensive knowledge graph using DrugBank data (1,000 drugs, 155,774 interactions) enriched with molecular features from PubChem. Our methodology introduces a novel multi-modal approach by integrating transformer-based embeddings (ChemBERTa, SPECTER, and SBERT) to create 1152-dimensional feature vectors that capture structural, biomedical literature, and semantic properties of drugs. Formulating DDI prediction as a link prediction task, we compare three Graph Neural …

Haematopoietic stem cells maintain blood production throughout life1. Although extensively characterized using the laboratory mouse, little is known about clonal selection and population dynamics of the haematopoietic stem cell pool during murine ageing. We isolated stem cells and progenitors from young and old mice, identifying 221,890 somatic mutations genome-wide in 1,845 single-cell-derived colonies. Mouse stem cells and progenitors accrue approximately 45 somatic mutations per year, a rate only approximately threefold greater than human progenitors despite the vastly different organismal sizes and lifespans. Phylogenetic patterns show that stem and multipotent progenitor cell pools are established during embryogenesis, after which …

Effects Of Different Blue Light Conditions On Saos-2 Cell Viability, Differentiation, And Mineralization, Jamielyn Alcantara Jarvis

Theses and Dissertations

Photobiomodulation (PBM) is a low-intensity light therapy that uses non-ionizing forms of energy to induce a directed cellular response. Prior work has investigated the effects of blue light exposure at energy densities ranging from 1 J/cm2 to 10 J/cm2 on osteogenic differentiation of human gingival mesenchymal stem cells (hGMSCs), human dental pulp stem cells (hDPSCs), and stem cells from the apical papilla (SCAPs) and shown promising outcomes at 4 J/cm2 and then having a detrimental impact at higher energy densities. One key caveat in prior studies is that the blue light energy density was modulated by changing the light exposure …

Phase II clinical trials play an important role in drug development, providing key data that guide decision-making for promising therapeutic candidates. An important objective of phase II is to establish proof of concept by demonstrating that the drug produces its intended biological effects in the target population. Phase II trials also evaluate pharmacokinetics, pharmacodynamics, safety, and dose-response relationships. Ultimately, the goal is to generate the evidence needed to inform go/no-go decisions for further development. However, as we will discuss, phase II studies have inherent limitations and cannot fully predict phase III outcomes. In this JACC: Basic to Translational Science and …

Deep brain stimulation (DBS), a proven treatment for movement disorders, also holds promise for the treatment of psychiatric and cognitive conditions. However, for DBS to be clinically effective, it may require DBS technology that can alter or trigger stimulation in response to changes in biomarkers sensed from the patient's brain. A growing body of evidence suggests that such adaptive DBS is feasible, it might achieve clinical effects that are not possible with standard continuous DBS and that some of the best biomarkers are signals from the cerebral cortex. Yet capturing those markers requires the placement of cortex-optimized electrodes in addition …

Deep-learning models like Variational AutoEncoder have enabled low dimensional cellular embedding representation for large-scale single-cell transcriptomes and shown great flexibility in downstream tasks. However, biologically meaningful latent space is usually missing if no specific structure is designed. Here, we engineered a novel interpretable generative transcriptional program (iGTP) framework that could model the importance of transcriptional program (TP) space and protein-protein interactions (PPI) between different biological states. We demonstrated the performance of iGTP in a diverse biological context using gene ontology, canonical pathway, and different PPI curation. iGTP not only elucidated the ground truth of cellular responses but also surpassed other …
