BACKGROUND: Impulsivity is among the strongest correlates of substance involvement (i.e. a broad continuum of substance-related behaviors), and distinct domains (e.g. sensation seeking [SS] and urgency) are differentially correlated, phenotypically and genetically, with unique substance involvement stages. Examining whether polygenic influences for distinct impulsivity domains are differentially predictive of early substance use initiation - a major risk factor for later problematic use - may improve our understanding of the role of impulsivity in addiction etiology.

METHODS: Data collected from participants of genetically inferred European ancestry enrolled in the Adolescent Brain Cognitive Development Study

The field of single-cell biology is growing rapidly, generating large amounts of data from a variety of species, disease conditions, tissues, and organs. Coordinated efforts such as CZI CELLxGENE, HuBMAP, Broad Institute Single Cell Portal, and DISCO allow researchers to access large volumes of curated datasets, including more than just scRNA-seq data. These resources have created an opportunity to build and expand the computational biology ecosystem to develop tools necessary for data reuse and for extracting novel biological insights. We highlight achievements made so far, areas where further development is needed, and specific challenges that need to be overcome.

Regenerative medicine is a multidisciplinary domain focused on restoring, repairing, or replacing damaged tissues and organs by harnessing the body’s inherent healing abilities with sophisticated biological and technical techniques. Mesenchymal stem cells (MSCs) have become fundamental among many stem cell types because to their self-renewal, multipotency, paracrine signaling, and immunomodulatory capabilities. Their therapeutic adaptability has been shown in orthopedic, cardiovascular, neurological, dermatological, and immune-mediated illnesses, underscoring their clinical significance. Dental polymers play a central role in modern dentistry, offering versatility in applications ranging from removable dentures and prosthetic frameworks to implant abutments and bioactive scaffolds. Common materials like PMMA, PEEK, and nylon are valued for their formability, cost-effectiveness, and aesthetic properties, while emerging biopolymers including chitosan and alginate offer promising biocompatible alternatives. However, both conventional and bio-based polymers exhibit limitations in terms of chemical stability, mechanical wear, and microbial adhesion. Dysfunction at the centrosome-cilium interface underlies a broad range of ciliopathies. Here, we identify biallelic variants in CEP76, encoding a centrosomal protein, in eight unrelated individuals presenting with neurodevelopmental, ocular, and variable additional multisystem features. Proband-derived fibroblasts and CEP76-depleted RPE1 cells display ciliary deficits, including impaired cilium formation and length, disrupted transition zone architecture, and impaired IFT88-mediated anterograde intraflagellar transport. Zebrafish cep76 mutants recapitulate key clinical phenotypes, and in vitro complementation assays confirm pathogenicity for all tested human disease-associated variants. The population of Puerto Rico has been heavily affected by chronic diseases; however, data remains limited on the influence of diabetes mellitus (DM), hypertension, and obesity on the probability of COVID-19 diagnosis. The purpose of this quantitative study was to evaluate the association between non-communicable diseases that included DM, obesity, hypertension, and COVID-19 diagnosis, controlling for age and sex and person exposure in Puerto Rico. The sample consisted of 1,463 participants who underwent a polymerase chain reaction (PCR) test for COVID-19 from April 1, 2020, to March 31, 2021. Regorafenib-induced pneumonitis and acute respiratory distress syndrome (ARDS) are rare and poorly characterized toxicities. We describe a 43-year-old male with metastatic colorectal cancer who developed hypoxic respiratory failure after two cycles of regorafenib. Imaging revealed cavitary lung lesions and diffuse ground glass opacities. Despite antibiotics and high-dose corticosteroids, his condition deteriorated, requiring ICU admission and mechanical ventilation. Bronchoalveolar lavage revealed bacterial pathogens, and he was treated for pneumonia. After multidisciplinary discussion, he was assessed to have regorafenib-induced pneumonitis as a major driver of his respiratory failure, and after limited steroid response, infliximab was administered with marked respiratory improvement. Lysosomes are essential organelles for cellular homeostasis and signaling, with dysfunction linked to neurological disorders, lysosomal storage diseases, and cancer. While proteomics has advanced our understanding of lysosomal composition, the structural characterization of lysosomal membrane proteins in their native environment remains a significant challenge. Here, we developed a cryo electron tomography workflow to visualize lysosomal membrane proteins within intact, native lysosomal membranes. We isolated endolysosomes by independently targeting two lysosomal membrane proteins, transient receptor potential mucolipin 1 and transmembrane protein 192, enriching organelles that exhibited the expected morphology and proteomic composition of the endolysosomal system. Background and Aim: Opioid use disorder (OUD) during pregnancy has become a major public health issue, with its prevalence rising significantly in recent years. The incidence of neonatal abstinence syndrome (NAS) has also surged, from 1.5 cases per 1000 hospital births in 1999 to 6.0 cases per 1000 in 2013. Clonal hematopoiesis (CH) is defined by the expansion of a lineage of genetically identical cells in blood. Genetic lesions that confer a fitness advantage, such as leukemogenic point mutations or mosaic chromosomal alterations (mCAs), are frequent mediators of CH. However, recent analyses of both single cell-derived colonies of hematopoietic cells and population sequencing cohorts have revealed CH frequently occurs in the absence of known driver genetic lesions. INTRODUCTION: Children from refugee families resettled in the USA face higher risks of serious mental health challenges compared with their native-born peers. Research shows that refugee youth in high-income countries frequently suffer from trauma-associated disorders such as post-traumatic stress disorder (PTSD), depression and anxiety. The high prevalence of trauma-associated mental health problems among these youth may be attributed to their own trauma exposure, especially if born in conflict zones, and post-resettlement challenges like poverty, acculturation difficulties, racism and discrimination. SNP paternity testing panels offer high-resolution capabilities for determining livestock paternity, thereby enhancing the reliability of genetic verification and ensuring pedigree record accuracy. In this study, we aimed to develop a SNP-based paternity test panel utilizing a 50K SNP chip for paternity testing and pedigree verification in the Eşme sheep breed. Additionally, the study sought to validate the paternity test panel. Based on data from 26 rams and 524 lambs of the Eşme sheep breed, the results indicated that the 50K SNP chip used demonstrated a very high genotyping rate, with the majority of its 53,738 SNPs exhibiting significant polymorphism. …

In-hospital strokes are often underrecognized, leading to treatment delays and poorer outcomes compared to community-onset strokes. This scholarly project evaluated whether integrating the Epic Stroke Narrator into inpatient code stroke protocols could improve documentation of last known well time (LKWT) and reduce time to intervention. A 6-week quasi-experimental pilot study guided by the Iowa Model of Evidence-Based Practice and Rogers’ diffusion of innovations theory was conducted on a neuroscience telemetry step-down unit at a comprehensive stroke center in Richmond, Virginia. This review provides an overview of the health and fitness benefits attributed to sauna exposure. The specific focus explores muscle performance and flexibility, heat acclimation for performance, cardiovascular and respiratory health parameters, all-cause mortality, endocrine response, inflammatory modulators, and exercise recovery. The overarching mechanisms underpinning the health and fitness benefits attributed to sauna use stem from hyperthermia, which activates thermoregulatory cardiovascular, neuroendocrine, and cytoprotective responses that promote homeostasis and adaptation to stress. Muscle performance improvements are attributed to plasma volume expansion, enhanced thermoregulatory efficiency, and upregulation of heat shock proteins. Feeding is a significant co-occupation of young children and their caregivers, unfolding in a social context. While addressing feeding challenges comprehensively because multiple contributing factors, the social-emotional dimension is often overlooked. This paper proposes a relationship-based feeding guideline to help occupational therapists incorporate this critical component. Centered on the child-caregiver dyad, the guideline prioritizes supporting the developmental, social-emotional, and environmental needs of the child.
