Small Molecules Binding To Serpins, Junaid Afridi

Serpins are a unique breed of proteins due to their enzymatic mechanism. Two systems were closely monitored during fluorescent binding studies, the ACT-CHY along with the AT:TRY interaction. Four different conformational variants of each system were studied including the native, cleaved, latent and complex forms. Three different fluorescent dyes were used to identify the conformations including ANS, TNS, and bis-ANS. SI studies and protease assays utilizing both Suc-AAPF-pNA and L-BAPNA were instrumental in determining conformations along with gel electrophoresis studies. The hydrophobic dyes bound to the different serpins with varying KD and ΔFmax due to structural variations among the conformers …

Variability of motor unit discharge is a likely contributor to the greater force fluctuations observed in old adults at low muscle forces. We sought to determine whether the variability of motor unit discharge rate underlies the fluctuations in force during steady contractions across a range of forces in young (n = 11) and old (n = 14) adults. The coefficient of variation (CV) for discharge rate and force were measured during a force-matching task as the first dorsal interosseous muscle performed isometric contractions. Stretching has been a part of warm-up for a very long time. Some recent research has shown that stretching could possibly hinder performance and others have shown it enhances performance. The purpose of this study was to examine whether stretching has an effect on vertical jump, and if so, how long the effects last. Twenty Western Kentucky students (10 M and 10 F) performed three trials each. There was a non-stretching group, a pre-stretching group and a between-stretching group; each of the subjects performed all three. In addition, the paper is intended to inform, guide, and encourage policy makers, program developers, and school counselors as they develop school counseling interventions for the disease. The focus of the paper is limited to children/adolescents because they bear the greatest burden of HIV/ AIDS infections. The purpose of this paper was to explore and define the concept of home through an extensive review of the literature. The concept analysis framework of Walker and Avant (1995) was used to structure the paper and the theories of Johnson (1980) and Maslow (1954) were used for operationalization. The literature search examined characteristics of home applicable to all individuals. The concept’s defining attributes were discovered and applied to the home experiences of those living with serious and persistent mental illness. Infants need to develop effective, secure attachment to their primary caregivers in the first year of life. Researchers have not been able to identify all the factors that may influence the development of infant attachment. Most of the studies in this area have been done without regard to infant feeding as a potential factor. Objectives: Muscle stiffness increases during muscle contraction.The purpose of this study was to determine the strength of the correlation between myotonometric measurements of muscle stiffness and surface electromyography (sEMG) measurements during various levels of voluntary isometric contractions of the biceps brachii muscle. Subjects: Eight subjects (four female; four male), with mean age of 30:6 ± 8:23 years, volunteered to participate in this study. Methods: Myotonometer and sEMG measurements were taken simultaneously from the right biceps brachii muscle.Data were obtained: (1) at rest, (2) while the subject held a 15 lb (6.8 kg) weight isometrically and, (3) during a maximal voluntary …

Informing adolescents about appropriate and acceptable behaviors, and ways to protect themselves against unwanted and unprotected sex, has proved problematic in Kenya. Education programs for in- and out-of-school adolescents are lacking, there is controversy about providing services to sexually active adolescents, and a pervasive concern that sexuality education and contraceptive services leads to promiscuity. Unbiased and accurate information and services are needed if adolescents are to delay becoming sexually active, to resist pressures to engage in nonconsensual sex, and to protect themselves against unwanted pregnancies and infections if they do have sex. This study represented an initial effort to investigate how a case study methodology could be used in the context of critical care nursing education. The study hoped to improve students' content specific knowledge of critical care nursing and facilitate critical thinking processes vital to clinical decision making. The study describes the transformation of a critical care-nursing course from a traditional lecture-based approach to a case study approach.

Byatt's short fiction, from her first collection Sugar and Other Stories (1987) to Elementals (1998) is marked by a continuous preoccupation with the body: the body as the product of social signifying practices, the body as the site of desires, fears, and anxieties, the body as the source of experience, the body as creator and created. In brief, the body is treated as the centre of existence, consciousness and identity. As the athletic training profession continues to expand, it is necessary to evaluate the preparedness of the entry-level certified athletic trainer (ATC). The purpose of this research is to determine if entry-level certified athletic trainers perceive that they have the knowledge and skills necessary to perform specific tasks related to therapeutic exercise.

An online questionnaire was sent out to 450 entry-level certified athletic trainers in District Four. A total of 69 subjects returned the survey for a response rate of 15.3%. Disruption of the growth hormone/insulin-like growth factor-1 (GH/IGF-1) axis has been reported and studied in menopause, hypothalamic amenorrhea, and anorexia nervosa, but not in weight-stable amenorrheic athletes. We investigated the effects of short-term transdermal estradiol on basal and exercise-stimulated serum GH, IGF-1, and associated binding proteins (IGFBP-1 and IGFBP-3) in seven weight-stable female amenorrheic athletes with percentage body fats greater that 12%. Each subject received a 72h placebo patch followed by 144h of transdermal estradiol. Serum samples for GH, IGF-1, IGFBP-1, and IGFBP-3 were obtained at baseline (t1), 72hr (0), 144hr (6), and during three 90-minute trials of aerobic exercise. …

Athletic trainer is a well-established profession in the U.S., but its definition is unclear in Japan because of no certification until recently. Japan Amateur Sports Association (JASA) has started in curriculum to prepare, educate and certify athletic trainers since 1994. This study researched the current trends in athletic training education and certification in Japan. It defines the historical and present athletic training situation, and clarifies education and certification with questionnaires. Questionnaires were sent to 43 sports medicine professors at JASA athletic trainer accredited and non-accredited program schools, and 26 (61%) responses were returned. Eccentric contractions, where a muscle is repeatedly lengthened while generating torque, result in decreased muscle function and muscle soreness. This study was designed to determine whether there was a difference in muscle response of the elbow flexors from untrained subjects (n = 12) between a bout of high intensity eccentric exercise at 30°•s-1 (LVE) compared to the equivalent at 210°•s-1 (HVE). Subjects performed 120 seconds of eccentric exercise of the elbow flexors using a Cybex 6000 Isokinetic Dynamometer. At 30°-s-1, a total of 30 repetitions were required whilst at 210°•s-1, 210 contractions were performed (at a 1:7 work/rest ratio). Both …

The purpose of this study was to identify the frequency and nature of perceived restrictions by certified athletic trainers (ATCs) and to determine the frequency of perceived restrictions for each practice setting within states that have legislation. This was a qualitative-descriptive study performed with a web-based survey. 10,000 ATCs receiving electronic mail from the National Athletic Trainers' Association (NATA) were invited to participate in this study. 1158 subjects participated in the study, and 724 subjects (295 female and 429 male; age=33.2 ± 8.3yrs) remained after exclusions. Equally severe natural disasters that hit equally populous areas produce vastly different impacts and outcomes depending on preexisting political and economic conditions. International relief organizations and civil defense disaster response teams employ effective life saving protocols in the immediate aftermath of a disaster. However, there is little information that addresses the ongoing needs or assesses appropriate responses to the persistent hardships and ill health remaining months after a disastrous event hits an impoverished nation. Within South Africa there is much interest in expanding youth center programs, particularly in the nongovernmental sector, in part because of recognition that the HIV/AIDS crisis is disproportionately affecting young South Africans. This study was designed to give implementing agencies and donors a broad view of how the youth centers function, who they reach, and the quality of information and services. The assessment underscored the importance of monitoring the performance of programs and understanding who is being reached with what interventions.
