The Application Of Macrophage-Targeted, Anti-Inflammatory Celecoxib Nanoemulgel For Nerve Regeneration In Peripheral Nerve Injury, Lu Liu Aug 2025

The Application Of Macrophage-Targeted, Anti-Inflammatory Celecoxib Nanoemulgel For Nerve Regeneration In Peripheral Nerve Injury, Lu Liu

Electronic Theses and Dissertations

Peripheral nerve injury (PNI) refers to damage to the nerve located outside the brain and spinal cord. PNI is commonly caused by a physical accident (trauma) or medical operation1, 2. The impact on the quality of life of PNI patients is directly correlated to neuropathic pain, resultant depression, and increased stress2. Even after recovering from the PNI, patients still experience phantom pain and lose the ability to work due to the loss of tactile sensory or motor function2-4. However, unlike damage to the central nervous system (CNS), peripheral nerves can regenerate because macrophages clean …

Polyphenols For The Mitigation Of Vascular Calcification; In Vitro And In Vivo Studies, Abdullah Altuhami Aug 2025

Polyphenols For The Mitigation Of Vascular Calcification; In Vitro And In Vivo Studies, Abdullah Altuhami

All Theses

Globally, cardiovascular diseases, in most cases associated with calcification, are the main cause of death. However, currently, the only effective cure is to undergo surgery to augment or receive a healthy donor heart, which could be very difficult due to the long waiting list and the associated risks. The possibility of body rejection of the foreign organs is attributed to immunoreaction or the risk of infection, since the donor must be on immunosuppressant drugs that lower the body's immunity and facilitate the body's acceptance of the new organ as one of its own. Taking that into consideration, using tissue-engineered vascular …

Effects Of Flavonoids On Reducing Colon Cancer Risk Via Macrophage Polarization, Gracien Jules Aug 2025

Effects Of Flavonoids On Reducing Colon Cancer Risk Via Macrophage Polarization, Gracien Jules

Graduate Theses and Dissertations

Colorectal cancer is one of the most fatal diseases in the United States. Chronic tissue inflammation can cause cell proliferation, which leads to tumor formation and cancer development. Macrophages are responsible for maintaining tissue homeostasis and for inflammatory responses. M1 macrophages exhibit proinflammatory responses while M2 macrophages exhibit anti-inflammatory responses. Imbalance between M1 and M2 macrophages is closely related to inflammation-associated disease development. The anti-inflammatory effects of apiaceous vegetable constituents, specifically flavonoids, have been studied for their preventative effects towards reducing disease risk. However, sufficient evidence is lacking on the mechanisms by which flavonoids impact M1/M2 macrophage polarization. RAW 264.7 …

2020-Current year OA Pubs

Reverse genetics approaches in mice are widely used to understand gene functions and their aberrations in diseases. However, limitations exist in translating findings from animal models to human physiology. Humanized mice provide a powerful bridge to understanding human physiology and mechanisms of disease pathogenesis while maintaining the feasibility of working with small animals. Methods for generating humanized mouse models that allow scientists to probe contributions of particular genes have been rudimentary. Here, we established an efficient method for generating genetically modified human cord blood-derived CD34

Lipid droplets (LDs) are evolutionarily conserved organelles that play important roles in metabolism. Each LD is enclosed by a monolayer of phospholipids, distinct from bilayer membranes. The composition of LD surface phospholipids and their impact on LD growth and function remain to be defined. Phosphoinositides mark cellular organelles and regulate organellar function. Here, we demonstrate that PI(4)P decorates a subset of LDs to recruit and activate CIDE proteins. Enhanced expression of ORP2 and ORP5, LD-associated lipid transfer proteins that remove PI(4)P from LDs, abolished the localization and function of CIDE proteins. Blocking the synthesis of PI(4)P on the LD surface …

Pediatric high-grade gliomas (pHGGs), particularly diffuse midline gliomas (DMGs), are among the most lethal brain tumors due to poor survival and resistance to therapies. DMGs possess a distinct genetic profile, primarily driven by hallmark mutations such as H3K27M, ACVR1, and PDGFRA mutations/amplifications and TP53 inactivation, all of which contribute to tumor biology and therapeutic resistance. Developing physiologically relevant preclinical models that replicate both tumor biology and the tumor microenvironment (TME) is critical for advancing effective treatments. This review highlights recent progress in in vitro, ex vivo, and in vivo models, including patient-derived brain organoids, genetically engineered mouse models (GEMMs), and …

Distinguishing tumor maintenance genes from initiation, progression, and passenger genes is critical for developing effective therapies. We employed a functional genomic approach using the Lazy Piggy transposon to identify tumor maintenance genes in vivo, and applied this to SHH medulloblastoma (MB). Combining Lazy Piggy screening in mice and transcriptomic profiling of human MB, we identified the voltage-gated potassium channel KCNB2 as a candidate maintenance driver. KCNB2 governs cell volume of MB-propagating cells, with KCNB2 depletion causing osmotic swelling, decreased plasma membrane tension, and elevated endocytic internalization of EGFR, thereby mitigating proliferation of MB-propagating cells to ultimately impair MB growth. …

Music As Medicine: The Impact Of Music On Pain Reported After Surgery, Anastasia Pike, Gregory T. Chesnut May 2025

Music As Medicine: The Impact Of Music On Pain Reported After Surgery, Anastasia Pike, Gregory T. Chesnut

Journal of Global Awareness

The impact of music on subjective reporting of pain and anxiety has been increasingly described in medical literature. Multimodal methods of alleviating postoperative pain and anxiety, including the use of live or recorded music in perioperative settings, have been shown to improve patient-reported pain levels and, in some studies, to result in decreased opioid usage. The overprescription of opioids after surgery can contribute to long-term abuse or trafficking of medications, and the use of non-opioid medications, alternative methods of pain relief, and environmental/auditory stimuli offer new mechanisms to promote safe opioid prescribing while improving both pain control and patient wellbeing. …

Identification And Preclinical Assessment Of Novel Therapeutic Modalities In Environmental Exposure-Induced Lung Disease, Aaron D. Schwab

Theses & Dissertations

Environmental lung diseases are preventable respiratory conditions either caused or made worse by inhaled environmental exposures. Contemporarily, environmental lung diseases are most associated with workplace exposures given specific occupational processes aerosolize inflammatory agents that can be inhaled at high concentrations. Although a considerable amount is known regarding immunoglobulin (Ig)E-mediated responses to environmental exposures, little is known about non-IgE mediated respiratory conditions resulting from lipopolysaccharide (LPS)-enriched organic dust exposure(s). Chronic respiratory diseases such as asthma, hypersensitivity pneumonitis, chronic obstructive pulmonary disease (COPD), and pulmonary fibrosis have all been identified as environmental lung diseases caused or exacerbated by such environmental dusts. Therapeutic …

International Journal of Health and Allied Sciences

Learning processes in medical practices in medicinal schools put more emphasis on the hard sciences than they do on developing soft skills like empathy and communication. For this reason, the medical humanities emerged to provide a "different" take on the creative process. Medical ethics, the doctor-patient relationship, clinical research, mental illness, and professionalism are just a few of the topics that have been utilized to discuss in medical school. A strong connection between classroom learning and clinical experience is crucial for medical school graduates. Students graduating from a medical program are expected to have experience with patient care, clinical procedures, …

Preliminary Study: Minimizing The Gap Between Pre-And Post-Spaceflight, Caleb E. Scheideger, Louis A. Diberardino

Aurora

Long-duration space missions pose significant challenges to astronaut health, primarily due to the detrimental effects of microgravity on musculoskeletal and cardiovascular systems. This research proposal aims to address the critical gap between pre- and post-spaceflight physiological conditions, focusing on the marked decline in muscle mass, bone density, and cardiovascular function. Existing countermeasures, such as resistive exercise, are insufficient to prevent muscle atrophy and bone demineralization, highlighting the need for novel solutions. Neuromuscular Electrical Stimulation (NMES) emerges as a promising intervention, capable of eliciting muscle contractions to maintain function and mitigate atrophy. By integrating NMES into existing exercise regimens, we hypothesize …

Objectives: Circulating Epstein-Barr virus antibodies (EBV-Ab) are used as a biomarker of chronic stress in high-income settings, but their relevance in environments with a high burden of infectious disease, nutritional constraints, and limited resources is less clear. We investigated EBV-Ab as a biomarker of adversity in a setting where local ecology and economy may affect immune development differently than in wealthy countries.

Methods: We measured EBV-Ab in finger-prick dried blood spots collected from Indigenous Shuar (n = 433) and non-Indigenous Colonos (n = 84) ranging from < 1 to 87 years old in Amazonian Ecuador. For a subset of adults (≥ 15 years, n = 210), we collected socioeconomic information (income, education, and occupation) and assessed household-level market integration. We determined the most important predictors of EBV-Ab for adults and children using multi-model averaging of linear regression models.

Results: Male children (< 15 years) had lower EBV-Ab than female children (model averaged β [SE]: -0.238 [0.066]). For adults, Shuar had higher EBV-Ab than Colonos (model averaged β [SE]: 0.235 [0.113]), and high systolic blood pressure was an important predictor of elevated EBV-Ab (model averaged β [SE]: 0.088 [0.047]). Individuals who reported unpaid domestic work as their primary occupation had higher EBV-Ab than agricultural workers (model averaged β [SE]: 0.302 [0.113]). Individuals living in houses with more market-sourced infrastructure had lower EBV-Ab (model averaged β [SE]: -0.088 [0.068]).

Conclusions: Circulating EBV-Ab may capture context-specific aspects of socioecological adversity …

Angiotensin-converting enzyme 2 (ACE2) and the neutral amino acid transporter B0AT1, encoded by SLC6A19, are membrane proteins with pivotal roles in human physiology. ACE2 is involved in regulating blood pressure and serves as the cellular entry receptor for SARS-CoV-2, while B0AT1 facilitates amino acid absorption in the intestine. The interplay between ACE2 and B0AT1, particularly their physical interaction and co-expression in the intestine, underscores their relevance in both normal physiology and disease. Dysregulation of these proteins has been implicated in conditions such as hypertension, and Hartnup disease and they have been usurped by SARS-CoV-2 to cause COVID-19. Despite their …
