Establishing Caenorhabditis Elegans As A Platform For Bioaccumulation And Selectivity Screening Of Anthelmintic Compounds, Jiarui Li

To address the cuticular barrier in anthelmintic discovery, this study establishes a bifurcated framework in Caenorhabditis elegans. First, HPLC-based quantification of 14 PIM inhibitors revealed that cuticular penetrance is primarily dictated by phylogenetic lineage (Clade V) rather than ecological niche. Specifically, C. elegans demonstrated a significant pharmacokinetic correlation with the hookworm Ancylostoma ceylanicum (Clade V) but not the whipworm Trichuris muris (Clade I), validating it as a superior surrogate model for hookworm bioaccumulation. Within this small molecule inhibitor library, SGI-12was identified as a lead candidate with superior potency against C. elegans and bioaccumulation. Second, target validation using a C. …

There is much interest in the role of sweeteners such as table sugar (sucrose) and high-fructose corn syrup in obesity and metabolic disease. Both sweeteners consist of glucose and fructose, two six-carbon isomeric sugars. Whereas glucose ingestion may promote obesity through its effects to stimulate insulin secretion, fructose has unique metabolic effects that promote triglyceride synthesis and fat accumulation. These effects arise from fructose's well-known role as a signal of metabolic plenty. Under modern conditions of overnutrition, chronic excess fructose drives features of metabolic syndrome. To identify metabolites as potential biomarkers of fructose vs glucose consumption and related metabolic changes, we conducted exploratory metabolomic profiling of fasting blood samples from participants in a double-blind, parallel-arm trial involving 31 male and female adults with overweight or obesity, both before and after supplementation with glucose- or fructose-sweetened beverages. Orthogonal Partial Least Square Discriminatory Analysis (OPLS-DA) was used to identify metabolites that could discriminate between the 2 intervention groups. Background: Epidemiologic data on the association between maternal fish intake and birth defects are sparse. Our objective was to assess associations between maternal fish intake and 52 different birth defects, most of which have not been assessed previously.

Methods: Using logistic regression, data was analyzed for 29,242 mothers of infants with birth defects and 10,973 mothers of control infants who participated in the National Birth Defects Prevention Study (NBDPS) and delivered between 1997 and 2011. Adhesion G protein-coupled receptors (aGPCRs) constitute a structurally and functionally distinct group within the superfamily of GPCRs. In 2015, the International Union of Pharmacology invited the Adhesion GPCR Consortium to publish a comprehensive review about aGPCRs and establish a unified nomenclature. Since then, substantial progress has been made in delineating the biological roles, molecular architecture, biochemical properties, expression profiles, ligand repertoire, and activation and signaling strategies of aGPCRs. Commensurate with these advances, their relevance to human pathophysiology has become increasingly apparent. Background: Relapsed medulloblastoma remains a significant therapeutic challenge as it is near universally fatal. The tumor microenvironment of medulloblastoma plays a critical role in tumor progression, influencing tumor growth, immune evasion, and therapeutic resistance. We hypothesized that defining tumor-immune interactions in diagnostic and relapsed medulloblastoma may uncover mechanisms of immune evasion and identify novel therapeutic targets.

Methods: We analyzed paired primary and recurrent RNA-sequencing data from 140 medulloblastoma patients to profile immune cell composition and validate spatial relationships within the TME. Histomonas meleagridis is a parabasalid protozoan and the causative agent of histomonosis, or blackhead disease, in turkeys. Withdrawal of the last remaining in-feed arsenical from the market in 2015 led to the re-emergence of the disease. As a result, there is a need to investigate alternatives, such as dietary inclusion of postbiotic and phytogenic compounds, to reduce H. meleagridis transmission and disease severity in commercial turkey flocks. Paralysis following spinal cord injury (SCI) has traditionally been considered permanent due to the disruption of supraspinal input to spinal motor circuits. However, growing evidence suggests that spinal networks below the level of injury retain intrinsic motor-generating capabilities through central pattern generators. Transcutaneous spinal cord stimulation (scTS) is a noninvasive neuromodulation technique that delivers electrical stimulation through the skin to reactivate these spinal networks. The goal of this study was to increase general knowledge of Blue Zones as well as the implementation of healthy habits to increase the longevity of the population of Calloway County, Kentucky through educational sessions of elementary students. Escherichia coli is a leading cause of neonatal sepsis, with infection occurring in approximately one in every 1,000 live births. However, with E. coli colonization beginning soon after birth and defects in neonatal host defence maturation, an alternative consideration is why infection does not occur even more frequently. Here we show that newborn babies with E. coli sepsis have selectively reduced vertically transferred natural antibodies that recognize E. coli, mechanistically explaining their susceptibility to infection. Complementary preclinical studies show that preconceptual intestinal colonization with probiotic E. coli Nissle 1917 (EcN) primes anti-E. coli immunoglobulin G (IgG) antibodies with broad cross-reactivity …

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants with widespread human exposure and well-established links to adverse health outcomes. However, despite increasing epidemiological and experimental evidence, the molecular mechanisms underlying PFAS-induced neurotoxicity remain incompletely defined. This dissertation investigates PFAS toxicity across biological scales, integrating protein biophysics with neuronal cell models to elucidate pathways relevant to neurodegeneration. At the molecular level, we demonstrate that PFAS disrupts the structure and function of globular proteins, including the impairment of α-lactalbumin's calcium-binding capacity and of β-lactoglobulin's retinol binding, through concentration-dependent conformational destabilization. Activation of T lymphocytes is the main event that occurs in the adaptive immune response after engagement of the T cell receptor (TCR) with the peptide of the major histocompatibility complex (pMHC) molecules that are demonstrated by antigen presenting cells [1]. A key step in this process is the recruitment and activation of the zeta chain associated protein of 70 kDa (ZAP-70). This dissertation explores that question by examining how farming practices shape the nutritional quality of meat—and what, if anything, those differences mean for the people who consume it. Alzheimer’s disease is a progressive disorder involving neuronal death and cognitive decline, such as memory loss and changes in personality. One feature of the disease is buildup of abnormal protein called amyloid-beta in the brain. Amyloid-beta may activate support cells called astrocytes, triggering them to release large amounts of chemicals, possibly harming nearby cells. This study set out to examine the roles of the channels on astrocytes involved in releasing these chemicals, called connexin 43 hemichannels. Smoking tobacco is the leading avoidable cause of death and drives many health problems. Nicotine, the main chemical in tobacco products, keeps people using it. This study used the Rat Electronic Nicotine Delivery System (RENDS), an inexpensive and freely shared, to examine e-cigarette use in adolescent and adult female rats by exploring four phases of nicotine delivered and not delivered to understand use patterns among ages. Our study found that both adolescent and adult female rats chose to inhale nicotine when given the opportunity, showing drug-seeking behavior. Overall, nicotine use was similar between ages. Western dietary patterns are characterized by nutrient-poor, calorie-rich diets and have been linked to poor health outcomes and gut inflammation phenotypes. Probiotics are often taken to support gut health, but many beneficial bacteria do not survive transport through the digestive system to produce any effect. Spore-forming bacteria, such as Bacillus coagulans, are more resilient due to their hardy outer coat which aids in survival through many obstacles encountered in the gastrointestinal tract, including acidic stomach conditions. Colorectal cancer is one of the most common and deadly cancers worldwide, with a lifetime risk of approximately one in twenty-three for men and one in twenty-five for women. Although early detection significantly improves survival, current diagnostic approaches such as tissue biopsy are invasive, reliant on subjective interpretation, have extended turnaround times, and risk missing flat or depressed lesions that are more difficult to detect than raised polyps. Background Over the past decade, neuropsychopharmacology has shifted from stagnation to momentum, with first-in-class mechanisms and biomarker-enabled trials spanning psychiatry and neurology.

Methods We narratively synthesized advances from 2013 to 2026 across central nervous system (CNS) discovery and development, including pivotal trials, regulatory actions, digital/real-world evidence, genetics, artificial intelligence (AI), and implementation/global-access themes that are endorsed by international societies.

In the chronic stage of stroke recovery, neuroplasticity is thought to require external influence, and is often considered to be experience-dependent, or “Hebbian,” plasticity. Current studies have been investigating multiple ways to induce plasticity to enhance recovery, including investigating the role of sensory stimulation in cortical priming to enhance the effects of rehabilitation interventions.

This case study of an 80-year-old female with aphasia after L-MCA ischemic stroke included 18 neurotherapeutic sessions over 10 weeks. BACKGROUND: Synovial fluid biomarkers reflect the inflammatory response after joint injury and surgery. Previous studies have identified synovial fluid biomarker concentrations in the knee after both anterior cruciate ligament (ACL) injury and ACL reconstruction (ACLR).

BACKGROUND: Preterm birth increases the long-term risk of diabetes and chronic kidney disease (CKD), yet its impact on diabetic kidney disease (DKD) is unclear. We previously showed that male preterm mice with diabetes develop early features of DKD, including reduced podocyte density, decreased renin expression, activation of angiogenesis pathways, and impaired endothelial-podocyte signaling. Here, we examine whether preterm birth similarly accelerates DKD progression in female mice and compare structural and transcriptomic outcomes in the females to the prior male cohort to assess sex-specific differences.

Introduction: Inflammatory bowel disease involves aberrant immune responses and is associated with both cardiovascular disease risk and altered intestinal blood flow. However, little is known about how inflammatory bowel disease affects regulation of perivascular nerves that mediate blood flow. Previous work found perivascular nerve function is impaired in mesenteric arteries with Inflammatory bowel disease. Prophylactic brivaracetam (BRV) was evaluated in a rat TBI model for effects on behavior and serum tau. BRV reduced anxiety-like behavior with optimal dose-dependent effects at 2.1mg/kg, but did not affect working memory. Serum tau was detectable early post-injury, supporting its use as a biomarker.

Background: High dietary salt intake is a well-established modifiable risk factor for hypertension and cardiovascular disease. Salt sensitivity – a blood pressure (BP) phenotype defined by exaggerated BP response to salt loading, remains underrecognised in normotensive populations despite its strong link to adverse cardiovascular outcomes. The burden and determinants of salt sensitivity amongst young Nigerians remain poorly understood.

Food insecurity remains a persistent public health concern throughout New Mexico, impacting both rural and urban populations and contributing to significant health disparities. Limited access to affordable, locally sourced fresh foods exacerbates chronic disease burden and undermines community health. The purpose of this Doctor of Nursing Practice project was to conduct a comprehensive policy analysis to identify feasible state-level strategies to improve food access through strengthening local food systems.
