Enterotoxigenic Escherichia coli (ETEC) infections are a leading cause of diarrheal illness, responsible for an estimated 100,000 deaths annually. ETEC pathogenesis is driven by various virulence factors, including toxins, adhesins, and noncanonical factors such as the protease EatA. The first line of host defense against intestinal pathogenic bacterial infections is the protective intestinal mucus layer. Here, we demonstrate the mechanism by which EatA degrades the core mucus component MUC2, thereby facilitating access to the epithelial cell surface and promoting infection. We identify the specific cleavage site region localized at the C-terminal of MUC2. EatA's protease activity depends on the interaction …

Carbon ion radiation therapy (CIRT) is a high-LET radiotherapy, which distinguishes itself from traditional low-LET radiation, such as photons and protons, through its unique physical aspects, biological attributes, and the dramatically increased damage it causes within cellular DNA. Given its distinctive characteristics, it is expected to improve the therapeutic ratio of radiation treatments and enhance treatment outcomes in traditionally radiation-resistant tumor histologies. Despite these unique properties, much remains to be understood regarding the clinical use of CIRT before its full potential can be realized. In this review, we summarize the distinct advantages of CIRT with regard to its physical and …

Background: The circadian clock is an evolutionarily conserved system that orchestrates 24-h physiological rhythms through transcriptional and translational feedback loops. Mounting evidence suggests a bidirectional relationship between circadian rhythm alteration and disease progression, positioning the circadian clock as a potential therapeutic target. Due to the scarcity of high-resolution temporal omics data, it remains very challenging to elucidate the underlying regulatory mechanisms of the circadian system. As a practical alternative, public untimed transcriptomic datasets offer the potential to infer gene expression oscillations retrospectively. However, existing computational approaches for circadian phase estimation often suffer from limited predictive accuracy, reducing their ability to …

Chemotherapy-induced bone loss is a debilitating and common side effect of cancer treatment, though its underlying mechanisms remain poorly understood. Here, we show that, despite the systemic administration of chemotherapy, cellular senescence is restricted to bone marrow adipo-lineage cells specifically Cxcl12-abundant reticular (CAR) cells and bone marrow adipocytes (BMAds). Induction of senescence within these populations promotes RANK ligand (RANKL)-mediated osteoclastogenesis, leading to significant bone loss. Notably, we find that inhibition of the p38MAPK-MK2 pathway suppresses the senescence-associated secretory phenotype (SASP), including RANKL production abrogating bone loss. Furthermore, treatment with the senolytic combination dasatinib and quercetin (D + Q) selectively eliminates …

Growing evidence suggests that psychiatric disorders are characterized by a prolonged inflammatory state, which may influence the efficacy of compounds targeting serotonin. Serotonin is a key signaling molecule in neuroplasticity of the adult hippocampus and involved in antidepressant action. Recent in vitro studies indicate the neurotransmitter may also facilitate the response to inflammation and potentially modulate microglial function towards neuroprotection. Using Tph2−/− rats depleted of brain serotonin, we examined microglial expression of various serotonin receptors (5-HTRs) in vivo in both the hippocampus and prefrontal cortex and assessed mRNA levels of cytokines and brain-derived neurotrophic factor (BDNF). We observed age-dependent and …

Hepatocellular carcinoma (HCC), the common liver cancer, exhibits higher incidence in males. Here, we report that mice lacking bile acid (BA) regulators, Farnesoid X Receptor (FXR also termed NR1H4) and Small Heterodimer Partner (SHP also termed NR0B2), recapitulate the sex difference in liver cancer risk. Since few therapeutic options are available, we focused on understanding the intrinsic protection afforded to female livers. Transcriptomic analysis in control and NR1H4 and NR0B2 double knockout livers identified female-specific changes in metabolism, including amino acids, lipids, and steroids. To assess translational relevance, we examined if transcriptomic signatures obtained from this murine HCC model correlate …

Recent evidence highlights the significance of a new type of tumour suppressors, which are not frequently mutated but inhibited by metabolic cues in cancers. Here, we identify BATF2 as a tumour suppressor whose expression is epigenetically silenced by glutamine in Head and Neck Squamous Cell Carcinomas (HNSCC). BATF2 correlates with type-I interferon and Th1 signatures in human HNSCC, with correlation coefficients even stronger than those of the positive control, STING. The phosphorylation of BATF2 at serine 227 promotes the oligomerization of STING. BATF2 deficiency or high glutamine levels result in higher oxygen consumption rates and metabolic profiles unfavorable for …

Effect Of Red Wine Immersion On The Flexural Strength Of Glass Ionomer Cement: An In Vitro Study, Niyola Crasta, Upasana Reddy, Jayaprakash Kukkila
International Arab Journal of Dentistry
Background: Alcoholic beverages such as red wine are widely consumed for social and cultural reasons. Ethanol and other constituents in red wine might alter the physical properties of dental restorative materials, particularly those exposed in the oral environment. Glass ionomer cements (GICs) are commonly used in restorative dentistry for their beneficial properties, but their mechanical strength under exposure to acidic or alcoholic solutions is a concern.
Objectives: To evaluate how exposure to red wine over time affects the flexural strength of glass ionomer cement (Shofu FX Ultra Mini).
Methods: Thirty rectangular specimens of glass ionomer cement (Shofu FX Ultra Mini), …

BACKGROUND: Microglia, the brain’s resident immune cells, play a pivotal role in Alzheimer’s disease (AD) pathogenesis. These cells exhibit diverse transcriptional states in response to neuroinflammatory stimuli, and characterizing these states is essential for understanding AD mechanisms. METHODS: We integrated single-cell and spatial transcriptomic datasets from multiple cohorts and brain regions, encompassing both experimental models and human tissues. Findings were validated through immunostaining of human brain samples. RESULTS: Our comprehensive atlas revealed pronounced heterogeneity among microglial states, with disease-associated microglia (DAM) significantly enriched in AD brains compared to controls. Spatial transcriptomics and immunohistochemistry demonstrated that DAM predominantly localize to external …

Objectives: Dental at-home bleaching is a widely recognized and effective treatment for improving the shade of anterior teeth. However, the impact of varying application times of carbamide peroxide (CP) concentrations on whitening efficacy and tooth sensitivity (TS) remains unclear.
Methods: In this split-mouth, randomized clinical trial, 80 participants were divided into two groups (n = 40 each) to receive either 10% or 16% CP gel for home bleaching. Different application times were tested. Tooth color was measured using the Optishade colorimeter, and TS was evaluated via a five-point analog numerical rating scale. The Friedman test with post hoc Tukey test …

Objectives: This in vitro study compared the solubility of three root canal sealers, AH Plus (epoxy resin-based), MTA Fillapex (MTA-based), and Cerafill RCS (bioceramic), in two solvents: Xylene and Endosolv.
Methods: Sixty standardized cylindrical specimens were prepared using the three sealers of interest. The specimens were divided into six groups (n = 10) and immersed in either Xylene or Endosolv for 10 minutes. Solubility was assessed by calculating the percentage weight loss before and after immersion. Data were analysed using one-way ANOVA and Tukey’s post hoc test (α= 0.05).
Results: All sealers exhibited statistically significant solubility in both solvents (p …
The Effect Of Loading On The Success Of Dental Implants After Immediate Extraction Compared To Delayed Dental Implants: A Systematic Review, Hassen Merzouk, Samir Habibi, Ben Ali Boutabout
The Effect Of Loading On The Success Of Dental Implants After Immediate Extraction Compared To Delayed Dental Implants: A Systematic Review, Hassen Merzouk, Samir Habibi, Ben Ali Boutabout
International Arab Journal of Dentistry
Introduction: The study focuses on dental implants, which are used to replace missing teeth. A key decision in implant dentistry is when to place the implant relative to tooth extraction. This review compares two main approaches: immediate implant placement (placing the implant during the same surgery as tooth removal) and delayed implant placement (placing the implant after the extraction site has healed, typically 6 to 12 months later).
Objectives: The study aims to compare the success rates and complications associated with these two-timing strategies, especially concerning the implant’s ability to withstand chewing forces. The primary objective was to compare post-extraction …
