To investigate serum HLA-G levels and its correlation with oxidative stress and antioxidant parameters in breast cancer patients. Materials and methods: The patient group consisted of 25 individuals with breast cancer who were operated on in the General Surgery Department of Gazi University’s Faculty of Medicine. Thirty-one individuals who had no history of any kind of cancer, allergic diseases, diabetes, or rheumatic and immunological disease constituted the control group. Results: Results showed that malondialdehyde (MDA), advanced oxidation protein products (AOPPs), and HLA-G levels in the patient group were statistically and significantly high compared to the control group (P < 0.001). While vitamin C levels were significantly lower in the patient group (P < 0.05), an insignificant difference in glutathione peroxidase (GPx) and total sulfhydryl group (R-SH) levels in both groups was observed (P > 0.05). To be able to differentiate benign vertebral bone marrow lesions from malign lesions according to apparent diffusion coefficient (ADC) values, enabling quantitative assessment, and to determine the sensitivity and the specificity in differentiating benign and malign lesions according to the optimal cutoff ADC value. Materials and methods: In 99 patients, 133 lesions in total were included in the study, 59 of which were benign (acute vertebral compression fracture depending on osteoporosis or trauma, spondylitis, atypical hemangioma), and 74 malign (malign compression fracture, metastasis). To evaluate the routine use of chromogenic media together with matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF) in gastrointestinal infections caused by Aeromonas spp. as rapid, practical, cost-effective and reliable methods. Materials and methods: Between August 2007 and March 2012, a total of 13,194 stool specimens of patients were included in the study. The stool samples and the reference strains were inoculated onto Cefsulodin-Irgasan-Novobiocin agar, sheep blood agar, eosin-methylene blue agar, and Hektoen enteric agar media together with CHROMagar Salmonella Plus. The In Vitro Determination Of Genotoxicity In Peripheral Lymphocytes Of Welders Exposed To Fumes From Metal Arc Welding, Ceylan Şener, Hali̇l Erhan Eroğlu

Gas metal arc welding is a widely used method in many industrial areas. In this study, the peripheral blood lymphocytes of 23 welders and 25 nonexposed subjects were monitored for genotoxicity. Materials and methods: The blood samples obtained from the subjects were incubated with a culture medium at 37 °C for 72 h. Cytochalasin-B and BrdU were added for micronucleus and replication index. After incubation, the cultures were harvested, stained, and examined. Results: The micronucleus rate (2.165 ± 1.645) of the welders was quite higher than that of those not exposed (1.352 ± 0.978) (P < 0.01). There was a significant difference between micronucleus rates of smoking and nonsmoking subjects. It was found that as the time of exposure to welding fumes increased, so did the micronucleus rates (2.938 ± 1.723) (P < 0.05). A positive correlation was observed between micronucleus frequency and age. The replication index rate (1.026 ± 0.020) of the welders was higher than that of those not exposed (1.019 ± 0.016) (P = 0.01). Conclusion: It is advised that welders who work with gas metal arc welding be instructed and that necessary protective measures be taken because of the risk caused by metal arc welding fumes.

Our aim was to investigate the usefulness of serum interleukin-8 (IL-8) and the neutrophil-to-lymphocyte ratio (NLR), either by themselves or as adjuncts to CA-125, in the diagnosis of various stages of endometriosis. Materials and methods: This study was a single-centre, prospective, group-comparative clinical trial. Serum IL-8 and the NLR, either alone or as adjuncts to CA-125, were analysed using receiver-operating characteristic (ROC) analysis to diagnose endometriosis. Fifty patients with endometriosis and 50 patients with benign ovarian cysts were included in this study. Asymmetric dimethylarginine (ADMA) is the major endogenous inhibitor of nitric oxide synthase. We aimed to investigate the ADMA levels in allergic rhinitis (AR) and nasal polyposis (NP). Materials and methods: A total of 29 AR patients, 21 NP patients, and 30 healthy subjects to be used as a control group were enrolled in the study. ADMA was measured in the AR, NP, and control groups with enzyme-linked immunosorbent assay. Mycoplasma pneumoniae is involved in the etiology of many respiratory tract infections. This prospective study describes the epidemiology and clinical properties of M. pneumoniae infections in hospitals of the Tabriz University of Medical Sciences. Materials and methods: From May 2009 to November 2010, 200 patients with respiratory tract infections were enrolled into this study. Due to antibiotic use 51 patients were excluded. The Epstein-Barr Virus (EBV) is a human herpes virus that infects B lymphocytes latently and immortalises the cells due to transformation. EBV infection is asymptomatic in childhood, while it may cause a self-limiting lymphoproliferative disorder in adolescence. To assess the effect of maternal demographic variables on development of hyperemesis gravidarum (HG) in pregnant Turkish women. Materials and methods: Two hundred consecutive women with HG were defined as the study group, and 200 consecutive pregnant women without any signs or symptoms of HG matched for age, parity, and gestational age were defined as the control group. Personal information, including lifestyle, educational level, occupation, and economic status were obtained via questionnaire. Due to the blood-brain barrier, delivery of many drugs to the brain has required intracranial surgery which is prone to complication. Here we show that Dopamine Neuron Stimulating Peptide 11 (DNSP-11), following non-invasive intranasal administration, protects dopaminergic neurons from a lesion model of Parkinson’s disease in the rat. A significant and dose-dependent increase in an index of dopamine turnover (the ratio of DOPAC to dopamine) was observed in the striatum of normal young adult Fischer 344 rats by whole-tissue neurochemistry compared to vehicle administered controls.

Changes in both sleep architecture and cognition are common with age. Typically these changes have a negative connotation: sleep fragmentation, insomnia, and deep sleep loss as well as forgetfulness, lack of focus, and even dementia and Alzheimer’s disease. Research has shown that psychosocial stressors, such as isolation from family and friends or loss of a loved one can also have significant negative effects on sleep architecture and cognitive capabilities. This leaves the elderly in a particularly vulnerable situation: suffering from cognitive decline and sleep dysregulation already, and more likely to respond negatively to psychosocial stressors. Estrogens, such as the biologically active 17-b estradiol (E2) have many actions in the male and female brain. Not only does E2 regulate reproductive behavior in adults, it organizes and activates the brains of younger animals in a sex-specific manner. In addition, many human studies have shown E2 to provide protection against a variety of neurological disorders, including stoke. These studies have been controversial and depend largely on the type and timing of hormone replacement. Traumatic brain injury (TBI) is a prevalent CNS neurodegenerative condition that results in lasting neurological dysfunction, including potentially debilitating cognitive impairments. Despite the advancements in understanding the complex damage that can culminate in cellular dysfunction and loss, no therapeutic treatment has been effective in clinical trials, highlighting that new approaches are desperately needed. A therapy that limits cell death while simultaneously promoting reparative mechanisms, including post-traumatic neurogenesis, in the injured brain may have maximum effectiveness in improving recovery of function after TBI. Epileptogenesis is the complex process of the brain developing epileptic acitivity. Due to the role of glutamate and the hippocampus in synaptic plasticity a dysregulation in glutamate neurotransmission and hippocampal dysfunction are implicated in the process of epileptogenesis. However, the exact causal factors that promote epileptogenesis are unknown.
We study presynaptic proteins that regulate glutamate neurotransmission and their role in epileptogenesis. The presynaptic protein, tomosyn, is believed to be a negative regulator of glutamate neurotransmission; however, no one has studied the effects of this protein on glutamate transmission in vivo. In many chronic inflammatory diseases, patients suffer from skeletal muscle weakness, exacerbating their symptoms. Serum levels of tumor necrosis factor-alpha (TNF) and sphingomyelinase are increased, suggesting their possible role in the progression of this weakness. This dissertation focuses on the role that reactive oxygen species (ROS) and nitric oxide (NO) play in mediating TNF-induced skeletal muscle weakness and to what extent sphingolipid signaling mediates cellular response to TNF.

The rapid increase in antibiotic resistance demands the identification of novel antibiotics with novel targets. One potential antibacterial target is the biosynthesis of peptidoglycan cell wall, which is both ubiquitous and necessary for bacterial survival. Both the caprazamycin-related compounds A-90289 and muraminomicin, as well as the capuramycin-related compounds A-503083 and A-102395 are potent inhibitors of the translocase I enzyme, one of the key enzymes required for cell wall biosynthesis. The caprazamycin-related compounds contain a core nonproteinogen b-hydroxy-a-amino acid referred to as 5’-C-glycyluridine (GlyU). Chronic ethanol exposure results in neuroadaptations that drive the progression of an alcohol use disorder (AUD). One such driving force is alcohol-induced neurodegeneration. Neuroinflammation has been proposed as a mechanism underlying this damage. Although neuroinflammation is a physiological response to damage, overactivation of its pathways can lead to neurodegeneration. A hallmark indicator of neuroinflammation is microglial activation, but microglial activation is a heterogeneous continuum of phenotypes that can promote or inhibit neuroinflammation. Because it is a multistep process, in carcinogenesis it is necessary to determine key genes such as cyclin D1, p27, p21, bcl-2, and p53 that are responsible for uncontrolled proliferation and other mechanisms. The aim of this study was to examine the correlation between the anomalous accumulation of these proteins and the prognosis of laryngeal carcinomas. To explore the roles of leptin and melatonin in early pubertal maturation. Materials and methods: Wistar female rats were used as experimental animals. Leptin was subcutaneously infused for 28 days through osmotic minipumps starting from postnatal day 15 (preweaning). Pinealectomy was performed on postnatal day 21 (weaning), and the animals were decapitated when estrus was detected by vaginal smearing. Results: Chronic preweaning, by peripheral infusion of leptin through subcutaneous routes, advanced the onset of puberty in leptin-treated sham and pinealectomized groups as determined by vaginal opening, while causing no significant change in serum estradiol levels and uterus weight. We investigated the effect of the number of cytosine-adenine (CA) repeats in the ADAMTS9 promoter region on breast cancer lymphatic metastasis. Materials and methods: Thirty-one postoperative breast cancer patients were selected and examined retrospectively. The patients were classified into 2 groups: metastatic or nonmetastatic. Thirty healthy women were selected as the control group, and their peripheral blood was obtained. The health and maintenance of rod photoreceptors depends on the high-fidelity vectorial transport of rhodopsin from its site of synthesis in the inner segment to its final resting place in the outer segment disks. Multiple molecular interactions play a role in this process, including the interaction of the small GTPase rab11a with rhodopsin that was first described in vertebrates in amphibians. We found that the interaction of the small GTPase rab11a and rhodopsin is conserved in mammalian retina. Studies suggest that inflammation impairs reverse cholesterol transport (RCT). We investigated whether serum amyloid A (SAA) contributes to this impairment using an established macrophage-to-feces RCT model. Wild-type (WT) mice and mice deficient in SAA1.1 and SAA2.1 (SAAKO) were injected intraperitoneally with 3H-cholesterol-labeled J774 macrophages 4 hr after administration of LPS or buffered saline. 3H-cholesterol in plasma 4 hr after macrophage injection was significantly reduced in both WT and SAAKO mice injected with LPS, but this was not associated with a reduced capacity of serum from LPS-injected mice to promote macrophage cholesterol efflux in vitro. The main psychoactive and therapeutic effects of Δ9-tetrahydrocannabinol (THC) are mediated through cannabinoid type 1 receptors (CB1Rs). The therapeutic uses of THC are mitigated by the development of tolerance to these therapeutic effects, whereas tolerance does not readily develop to some of the side-effects of THC, like motor impairment and reward. The development of tolerance occurs through adaptations at CB1Rs, which include desensitization (G-protein uncoupling) and downregulation (receptor degradation). Brain region-dependent differences in THC-mediated adaptations are proposed to explain the differences in tolerance to various THC-mediated effects. Pax3 has numerous integral functions in embryonic tissue morphogenesis and knowledge of its complex function in cells of adult tissue continues to unfold. Across a variety of adult tissue lineages, the role of Pax3 is principally linked to maintenance of the tissue’s resident stem/progenitor cell population. In adult peripheral nerves, Pax3 is reported to be expressed in nonmyelinating Schwann cells, however, little is known about the purpose of this expression. To evaluate the effects of insulin resistance (IR) on outcomes of doxazosin treatment for lower urinary tract symptoms (LUTS) due to benign prostatic hyperplasia (BPH). Materials and methods: This prospective study included 64 patients with LUTS due to BPH. Potentially pathogenic free-living amoebae have a cosmopolitan distribution in soil, dust, air, and water. Generally, environmental free-living amoebae do not threaten human health. This study aimed to investigate the presence of Acanthamoeba in well waters drawn from different locations in Kayseri, Turkey, which are mainly used for drinking or irrigation by the residents in the region.
