Although the prostate specific antigen (PSA) test is widely used in clinical settings for prostate cancer (PCa) diagnosis and post-treatment follow-up monitoring, false positive PSA test results, which contribute to over-diagnosis of PCa, and false negative results, which miss some patients with aggressive PCa, remain problems of clinical importance.
Our study demonstrates that radiation therapy, which is widely used for treatment of localized PCa, generates TNF-α in tumor cells and stromal fibroblasts, redox dependently. Interestingly, TNF-α rapidly and transiently triggers the RelA-mediated NF-κB canonical pathway, but its effect on RelB expression is more robust and long lasting, which leads to …

Cofilin’s activity to nucleate actin filament assembly, is regulated by phosphorylation at a single site on the amino terminus, Serine 3. Phosphorylation at this site abolishes the ability of ADF/cofilin to bind to F-actin and inhibits its severing function. This work characterizes the ability of dephosphorylated cofilin (mutation at Serine 3 site) to navigate prostate cancer actin cytoskeleton and metastatic properties in response to TGF-β. TGF-β increased Lim Domain Kinase 2 (LIMK-2) activity leading to cofilin phosphorylation and decrease actin filament severing in wild type cofilin (WTCFL) PC-3 cells. The astrocyte-elevated gene-1 (AEG-1, also known as metadherin) is associated with various aspects of tumour malignancy; however, little knowledge is available related to the role of AEG-1 in ductal and lobular carcinomas. The aim of this study was to investigate the relationship of AEG-1 with the prognostic parameters in invasive ductal and lobular carcinomas. Materials and methods: This study was conducted on a total of 72 paraffin-embedded breast tumour samples. They consisted of 61 ductal and 11 lobular carcinomas. The aim of this study was to determine the probable effects of cellular phone usage on apoptosis and oxidative/antioxidative balance in testicular tissue, and the probable protective role of vitamin C and E on these processes. Materials and methods: Forty-two male rats were divided into 6 groups: control, antioxidant, calling, stand-by, calling plus antioxidant, and stand-by plus antioxidant. To compare the efficiency of extracorporeal shock wave (ESW) treatment with hyaluronic acid (HA) viscosupplementation in an experimental rabbit cartilage defect model. Materials and methods: A total of 24 New Zealand rabbits were randomly divided into 4 groups: HA, ESW, ESW + HA, and control. Chondral defects were created in the left knees of the rabbits. HA viscosupplementation was performed on the HA and ESW + HA groups, and after 24 h, 0.16 mJ/mm2 ESW was performed on the ESW and ESW + HA groups. In this study, we designed and used 2 different type-specific polymerase chain reaction (PCR) methods for the detection of Brucella abortus. Materials and methods: There were 2 different primer sets (B4/B5 and AF/AR) selected and used for the amplification of 2 different genes that are present in all biovars of Brucella species, including bcsp31, encoding 31-kDa immunogenic cell surface proteins, and omp25, which is known to be one of the virulence factors encoding outer membrane proteins of 26–23 kDa. To determine the urease inhibitory potential of extracts and isolated compounds from Zizyphus oxyphylla Edgew. Materials and methods: Methanolic extracts and subsequent fractions of Z. oxyphylla stem and 3 isolated compounds from the roots of Z. oxyphylla were obtained through column chromatography and subjected to in vitro urease inhibition. Results: Three fractions (n-hexane, ethyl acetate, and butanol fractions) showed good to excellent activity, while the chloroform fraction showed nonsignificant activity. Amongst the isolated compounds, oxyphylline D (1) was the most active of the 3 isolated cyclopeptide alkaloids, followed by nummularin R (3) and nummularin C (2). Conclusion: Z. oxyphylla has …

This study was designed to investigate the beneficial effects of green tea administration on angiotensin converting enzyme (ACE) gene expression in CCl_4-induced liver cirrhosis in rats. It is known that desflurane and propofol protect liver tissue via different mechanisms. The aim of this study was to compare the protective effects of these 2 agents against hepatic ischemia-reperfusion injury. Materials and methods: Rats were randomly divided into 2 groups: ischemia and ischemia-reperfusion. Each group was further divided into 3 subgroups: ketamine, desflurane, and propofol. To evaluate the utility of immunohistochemical staining of cytokeratin 19 (CK19), HBME-1, and galectin-3 in cell block preparations of fine-needle aspiration cytology specimens as ancillary techniques for the differential diagnosis of papillary thyroid carcinoma. Materials and methods: Immunohistochemical detection of CK19, HBME-1, and galectin-3 was completed in 45 thyroid aspiration cytology specimens fixed in paraffin-embedded blocks consisting of the following cytologic diagnoses: 1) 16 cases of papillary carcinoma, 2) 16 cases suspicious for papillary carcinoma, and 3) 13 cases involving nonneoplastic lesions. To investigate the oxidative and antioxidative status in diabetic patients with cataracts and nondiabetic patients with cataracts as compared to controls. Materials and methods: There were 149 subjects included in the present study. Blood was collected from type 2 diabetic patients suffering from cataracts (diabetic cataract; n = 42), senile nondiabetic patients suffering from cataracts (nondiabetic cataracts; n = 60), and healthy subjects of a control group (n = 47). To determine the frequency with which the CCR5-\Delta32 mutant allele caused resistance to HIV, which is main agent of AIDS in all geographic regions of Turkey. Materials and methods: Randomly selected human blood samples from 400 healthy individuals were collected. The polymerase chain reaction method was used to determine the CCR5-\Delta32 polymorphism and samples were screened using 2 primers. Results: The frequency of wild alleles was found to be 0.9738 and the frequency of mutant alleles was found to be 0.0262. Ingestion of fructose and mental arithmetic can increase blood pressure (BP); the present study assessed the effect of fructose ingestion on cardiovascular (CV) response to mental stress. Effects were compared with sucralose used as a control. Materials and methods: Ten healthy, white European males were studied twice. A Finometer recorded CV parameters, noninvasively, throughout the study period. After the 30-min baseline, a mental arithmetic task (MAT), a series of arithmetic questions displayed as PowerPoint slides, was presented. Background and Significance: Previous studies have shown that cells infected with Chlamydia pneumoniae (Cpn) exhibit altered gene expression consistent with that observed in Alzheimer’s disease (AD). Furthermore, AD neurodegeneration has been linked to dysregulation of intracellular calcium and calcium-related processes. Therefore, we hypothesize that one mechanism by which pathogenesis evolves in AD is through infection-induced changes in expression of calcium-related genes. An 8-month-old crossbred ewe, normal upon physical examination, was humanely euthanized for tissue collection. After approximately 3 weeks in tissue culture, fungi began budding out of cells obtained from the choroid plexus. After an additional 3 weeks, budding was observed in kidney cell cultures and eventually in monocyte cultures as well. Serum from the lamb was submitted to the Veterinary Diagnostic Laboratory at Colorado State University for fungal diagnosis and was found negative for Aspergillus, Blastomyces, Coccidioidomycosis and Histoplasmosis. In a genome-wide siRNA screen, we recently identified the interferon (IFN) inducible protein 35 (IFI35, also known as IFP35) as a factor required for VSV infection. Studies reported here were conducted to further understand the role and requirement of IFI35 in VSV infection. Consistent with the siRNA screening data, we found that depletion of IFI35 led to reduced VSV replication at the level of viral gene expression. Although no direct interaction of IFI35 with the viral replication machinery was observed, we found that IFI35 negatively regulated the host innate immune response and rescued poly(I:C)-induced inhibition of VSV replication. Infectious myonecrosis virus (IMNV) is a significant and emerging pathogen that has a tremendous impact on the culture of the Pacific white shrimp Litopenaeus vannamei. IMNV first emerged in Brazil in 2002 and subsequently spread to Indonesia, causing large economic losses in both countries. No existing therapeutic treatments or effective interventions currently exist for IMNV. RNA interference (RNAi) is an effective technique for preventing viral disease in shrimp. Here, we describe the efficacy of a double-stranded RNA (dsRNA) applied as an antiviral therapeutic following virus challenge. α-Herpesvirinae subfamily members, including herpes simplex virus type 1 (HSV-1) and bovine herpes virus 1 (BHV-1), initiate infection in mucosal surfaces. BHV-1 and HSV-1 enter sensory neurons by cell-cell spread where a burst of viral gene expression occurs. When compared to non-neuronal cells, viral gene expression is quickly extinguished in sensory neurons resulting in neuronal survival and latency. The HSV-1 latency associated transcript (LAT), which is abundantly expressed in latently infected neurons, inhibits apoptosis, viral transcription, and productive infection, and directly or indirectly enhances reactivation from latency in small animal models. The philosophies behind many martial arts often claim that by practicing martial arts individuals can gain better control over stress. We tested this idea by using controlled physical stressors to elicit an acute stress response from martial artists (n=15) and non-martial artists (n=18). To measure the extent of the stress response, we looked at changes in heart rate, respiratory sinus arrhythmia, and galvanic skin level. These three measures explore both parasympathetic and sympathetic responses, and changes in these variables continue to be explored in studies of stress and reactive aggression. Abstract
This paper reports a case of endometriosis of the lung in a 29-year-old woman with long-term periodic catamenial hemoptysis. A chest computed tomography image obtained during menstruation revealed a radiographic opaque lesion in the lingular segment of the left superior lobe. During bronchoscopy, bleeding in the mucosa of the distal bronchus of the lingular segment of the left superior lobe was observed. Histopathology subsequent to an exploratory thoracotomy confirmed the diagnosis of endometriosis of the left lung. The 2-year follow-up after lingular lobectomy of the left superior lobe showed no recurrence or complications.

Endothelial dysfunction precedes pathogenesis of vascular complications in diabetes. In recent years, the mechanisms of endothelial dysfunction were investigated to outline strategies for its treatment. However, the therapies for dysfunctional endothelium resulted in multiple clinical trial failures and remain elusive. Oxidative stress-induced endothelial dysfunction plays a key role in ischemia/reperfusion injury. Recent evidence indicates that endothelial progenitor cell-derived microvesicles (EPC-MVs) can promote angiogenesis of endothelial cells (ECs). Here, we investigated the potential effects of EPC-MVs on hypoxia/reoxygenation (H/R) injury in human brain microvascular ECs (hb-ECs). MVs were prepared from EPCs cultured in a serum deprivation (SD) medium (starving stress, sEPC-MVs) or SD medium containing tumor necrosis factor-α (TNFα) (apoptotic stress, aEPC-MVs). H/R injury model of hb-ECs was produced by 6 hr hypoxia (1% O2) and 24 hr reoxygenation. The H/R hb-ECs were co-cultured with EPC-MVs. Results showed that …

Microparticles are emerging as an integral part of the vascular system. The microparticles are derived from stimulated or apoptotic endothelial cells, platelets, and leukocytes. They are involved in coagulation and regular cell function. Excessive numbers of microparticles contribute to artherosclerosis, chronic renal failure, and metabolic syndrome. Microparticle levels may be reduced by medications, relieving symptoms for awhile. Studies are being done to predict disease by counting microparticles. Further research must be done to understand and make use of microparticles.

To evaluate the amount of intestinal parasite infections over a 10-year period in the Gaza Strip. Materials and methods: Data from the Epidemiology Department, Ministry of Health were collected and analyzed statistically. The prevalence of intestinal parasites was monitored and studied for the period 1998-2007. Results: The present study shows results including the records of 471,688 patients (all ages) who had every provided 1 stool specimen to the laboratories of primary health care centers in one of the 5 governorates of the Gaza Strip. To detect the prevalence and type distribution of human papillomavirus (HPV) in different cytological diagnostic categories. Materials and methods: Between 2007 and 2010, a total of 1014 liquid-based thin preparations of cervical smears were selected and classified according to cytology results. HPV DNA polymerase chain reaction (PCR) was also performed using these samples. HPV DNA-positive samples were genotyped by DNA sequencing. Results: Of those enrolled in the study, 45.3% were negative cytologically, 36.4% had atypical squamous cells of undetermined significance, 0.3% had atypical squamous cells preventing the exclusion of a high-grade squamous intraepithelial lesion, 16.8% showed a low-grade squamous intraepithelial …

To determine the association between women's menopausal symptoms and their marital adjustment. Materials and methods: A descriptive cross-sectional study was conducted on 263 Turkish women chosen using an improbable randomized sampling method. A questionnaire, the Menopause Symptom Rating Scale (MSRS), and the Dyadic Adjustment Scale (DAS) were used to collect data. Percentage distributions, the t-test in independent groups, the one-way ANOVA test, Pearson correlation analysis, and the Kruskal-Wallis test were used in the evaluation of the data. To investigate the left atrial dilatation in an undetermined etiological group according to Trial of Org 10172 in Acute Stroke Treatment (TOAST) and to evaluate the relationship between left atrial dilatation and stroke volume and localization. Materials and methods: Files of patients admitted to our hospital (2000-2010) with acute ischemic stroke diagnosis were investigated.
