Right-sided congestive heart failure (brisket disease) commonly occurs in cattle raised at elevations >2,500– 3,500 m. We investigated clinical cases resembling brisket disease at a western Nebraska feedyard at a moderate altitude (1,369 m). Over a 15-mo period (2009–2010), we examined 17 cases (16 steers and 1 heifer), all purebred Angus. All animals had clinical right-sided heart failure: brisket and ventral abdominal edema, and severe chronic passive congestion of the liver. Gross examination confirmed right ventricular hypertrophy (left ventricle plus septum: right ventricle weight ratio mean: 1.33 vs. 2.8–4.0 reference interval). Innate CD8 + T cells express a memory-like phenotype and demonstrate a strong cytotoxic capacity that is critical during the early phase of the host response to certain bacterial and viral infections. These cells arise in the thymus and depend on IL-4 and IL-15 for their development. Even though innate CD8 + T cells exist in the thymus of WT mice in low numbers, they are highly enriched in KO mice that lack certain kinases, leading to an increase in IL-4 production by thymic NKT cells. น้ำวุ้นลูกตาถูกนำมาใช้เป็นตัวอย่างทางเลือก เนื่องจากทนต่อการเปลี่ยนแปลงหลังการเสียชีวิต รวมถึงมีส่วนประกอบที่เป็นเซลล์น้อยจึงสามารถนำมาเตรียมตัวอย่างได้ง่าย การเตรียมตัวอย่างทางนิติพิษวิทยาสามารถเตรียมได้หลายวิธี เช่น Solid Phase Extraction (SPE) และLiquid-Liquid Extraction (LLE) แต่มีข้อเสียคือใช้เวลาในการเตรียมนาน มีค่าใช้จ่ายสูงและใช้สารที่เป็นอันตรายในปริมาณมาก เทคนิค modified QuEChERS ถูกนำมาพัฒนาให้เข้ากับงานทางนิติพิษวิทยา ซึ่งเทคนิคนี้ใช้เวลาในการเตรียมน้อย ทำได้ง่าย ค่าใช้จ่ายถูก มีประสิทธิภาพ มีความทนทานและมีความปลอดภัย เครื่องมือตรวจวเคราะห์ Liquid Chromatography -tandem mass spectrometry (LC-MS/MS) สามารถวิเคราะห์สารประกอบได้หลากหลาย มีความจำเพาะสูงและการเตรียมตัวอย่างเพียงเล็กน้อยก็สามารถนำไปวิเคราะห์ได้ งานวิจัยนี้มีวัตถุประสงค์เพื่อตรวจสอบความใช้ได้ของวิธีการวิเคราะห์คัดกรองสารเสพติดในน้ำวุ้นลูกตาด้วยเทคนิค modified QuEChERS และตรวจด้วย LC-MS/MS ทำการทดสอบความใช้ได้ (validation) ทั้งหมด 6 พารามิเตอร์ โดยนำน้ำวุ้นลูกตามา spike สารเสพติดลงไปทั้งหมด 24 ชนิด จากนั้นนำไปสกัดด้วยเทคนิค modified QuEChERS และตรวจวิเคราะห์ด้วย LC-MS/MS หลังจากนั้นนำเทคนิคที่ผ่านการทดสอบแล้วไปทดสอบกับตัวอย่างน้ำวุ้นลูกตาจริงทั้งหมด 10 ตัวอย่าง และเปรียบเทียบผลที่ได้กับการทดสอบในตัวอย่างเลือด ผลการศึกษาความใช้ได้พบว่า การทดสอบความใช้ได้ทั้ง 6พารามิเตอร์มีค่าอยู่ในช่วงที่ยอมรับได้ทั้งหมด และผลการทดสอบ selectivity ไม่พบการรบกวนของสารอื่น ๆ ในการทดสอบและเมื่อเปรียบเทียบผลการทดสอบตัวอย่างน้ำวุ้นลูกตากับตัวอย่างเลือดก็ให้ผลไม่แตกต่างกัน

Pertussis is a human respiratory disease, primarily caused by the Gram-negative pathogen Bordetella pertussis. The infection is most severe and can be life-threatening in young children and infants where it manifests as a series of paroxysmal coughs. Lung cancer is a leading cause of cancer-related death worldwide. Every year it claims more lives than other common malignancies, breast, prostate, and colorectal cancers, combined. And despite recent improvements in disease diagnostics and treatment, drug resistance and tumor relapse remain major contributors to low patient survival. Both issues have been largely attributed to the existence of cancer stem-like cells (CSCs) or tumor initiating cells (TICs). Fungal peritonitis (FP) is a fatal complication in peritoneal dialysis (PD). Detection of fungal infection using standard culture is time-consuming and has a low sensitivity. While, Polymerase Chain Reaction (PCR) are often inapplicable in remote hospital and laboratory. In this study, Loop-mediated Isothermal Amplification (LAMP) assay was developed for rapid and specific detection of fungi in peritoneal dialysis. LAMP primers were designed based on the conserved sequence of fungal 18s rRNA gene. The reaction was optimized for 60 min at 64 ◦C. Fluoxetine เป็นยากลุ่ม SSRI (selective serotonin reuptake inhibitor) ที่ใช้ในการรักษาโรคซึมเศร้า ยาหลายตัวในกลุ่ม SSRI สามารถจับแบบจำเพาะ sigma-1 receptor แต่ในกรณี fluoxetine จะมีกลไกออกฤทธิ์จำเพาะต่อ serotonin transporter (Ki = 0.81 nM) มากกว่า sigma-1 receptor (Ki = 214–240 nM) หลายงานวิจัยรายงานว่า sigma-1 receptor แสดงออกบนเซลล์มะเร็งหลายชนิด และมีรายงานว่า antagonist สามารถลดการเพิ่มจำนวนและเหนี่ยวนำให้เกิด apoptosis แต่การทำงานร่วมกันระหว่าง fluoxetine กับ sigma-1 receptor ต่อเซลล์มะเร็ง ยังไม่ทราบกลไกการออกฤทธิ์ที่ชัดเจน งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาฤทธิ์ของ fluoxetine ในการยับยั้งการแบ่งตัวและเหนี่ยวนำให้เกิด apoptosis ผ่าน sigma-1 receptor ในเซลล์มะเร็งเม็ดเลือดขาว ผลการทดสอบพบว่า fluoxetine สามารถลดเปอร์เซ็นต์การรอดชีวิตของเซลล์ Jurkat และ U937 ได้ โดยมีค่า IC50 เท่ากับ 28 ± 1.4 และ 36 ± 1.6 uM ตามลำดับ เมื่อเซลล์ Jurkat ถูกทดสอบด้วย fluoxetine ตามระดับความเข้มข้นและระยะเวลาที่เพิ่มขึ้น พบว่าfluoxetine สามารถยับยั้งการแบ่งตัวของเซลล์ Jurkat ได้อย่างมีนัยสำคัญทางสถิติ โดยมีกลไกหยุดยั้งวัฎจักรเซลล์ที่ระยะ G2/M และลดการหลั่งไซโตไคน์ IL-2 รวมทั้งเหนี่ยวนำให้เกิด apoptosis โดยการลดการแสดงออกของโปรตีน pro caspase 9 ผ่านการยับยั้งการเกิด phosphorylation ของ AKT ในวิถี PI3K/Akt และ การถูกกระตุ้นของ sigma-1 receptor …

Ricin, Shiga toxin (Stx), exotoxin A, and diphtheria toxin are AB toxins that act within the host cytosol and kill the host cell through pathways involving the inhibition of protein synthesis. Background/aim: Changes in collagen metabolism and fibroblastic activity may play a role in the pathogenesis of brucellosis. The prolidase enzyme plays an important role in collagen synthesis. We aimed to investigate the association of prolidase levels with brucellosis.Materials and methods: Serum prolidase levels in 20 patients newly diagnosed with brucellosis were compared with levels in 30 healthy control subjects. Patients with brucellosis were reassessed 3 months later for prolidase, other laboratory measurements, and response to treatment. Parkinson’s disease (PD) is a disorder of the nervous system that causes problems with movement (motor symptoms) as well as other problems such as mood disorders, cognitive changes, sleep disorders, constipation, pain, and other non-motor symptoms. Ischemic stroke is the leading cause of disability world-wide and affects over 800,000 people per year in the United States. The majority of these strokes are ischemic due to a blockage of blood flow to the brain. Damage to the brain occurs at the onset of stroke, neuronal cell death is irreversible and therefore, quick treatment to remove blockage is critical factor in the recovery from stroke. Mechanical thrombectomy as a treatment for ischemic stroke provides an ideal opportunity to collect blood distal and proximal to the cerebral thrombus to examine neurochemical changes occurring during stroke.

Methyl group transfer from S-adenosyl-l-methionine (AdoMet) to various substrates including DNA, proteins, and natural products (NPs), is accomplished by methyltransferases (MTs). Analogs of AdoMet, bearing an alternative S-alkyl group can be exploited, in the context of an array of wild-type MT-catalyzed reactions, to differentially alkylate DNA, proteins, and NPs. Patients admitted to intensive care unit (ICU) are known to develop significant impairments in physical function. Patients with critical illness suffer up to 30% reductions in muscle size within the first ten days of admission to the ICU. Muscle strength testing, Medical Research Council-sum score, is current gold-standard to diagnosis ICU-acquired weakness and predicts risk of mortality and long-term physical function. Muscle power different from muscle strength in that it accounts for velocity of movement, is potentially a better independent predictor of function that has not been studied in this population. Membrane proteins belonging to the ATP-Binding Cassette (ABC) superfamily are responsible for transporting substrates into or out of cells. ABC transporters, such as P-glycoprotein (Pgp/ABCB1) defend cells by the active efflux of many chemically diverse cytotoxic compounds. The ABC protein, CFTR/ABCC7, is a passive chloride ion channel that provides fluid homeostasis. Post translational modification of proteins plays a crucial role in regulating their function, and the role of one such modification, termed O-GlcNAcylation, is understudied. O-GlcNAcylation involves the dynamic cycle of adding and removing an O-linked N-acetylglucosamine (O-GlcNAc) by the enzymes O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA), which are highly expressed in the hippocampus. Enzymes that catalyze O-GlcNAcylation are found at both presynaptic and postsynaptic sites, and O-GlcNAcylated proteins localize to synaptosomes. The kidneys are complex, multi-faceted organs that are responsible for regulatory processes, such as fluid homeostasis, hormone production, blood pressure regulation, and systemic toxin removal. Sudden disruption of these processes by acute kidney injury (AKI) causes rapid decline in renal function as well as significant morbidity and mortality. ST6Gal-I adds α2-6 sialic acids to select N-glycosylated cell surface receptors, thereby modulating receptor function and intracellular signaling. ST6Gal-I is upregulated in various carcinomas and confers cancer stem cell (CSC) properties evidenced by tumorspheroid growth, chemoresistance and tumor initiating potential. In pancreatic ductal adenocarcinoma (PDAC), ST6Gal-I conferred gemcitabine resistance by abrogating DNA damage and altering expression levels of gemcitabine metabolism genes. Further, ST6Gal-I promoted resistance to chronic gemcitabine treatment. Additionally, metastatic clones of a PDAC cell line had increased ST6Gal-I expression and ST6Gal-I knockdown enhanced gemcitabine sensitivity. Purpose:

In furthering the understanding of the process of spermatogenesis in the greater cane rat, this study describes the ultrastructural spermiogenic transformation and acrosomal formation in the testes of this hystricomorphic rodent that is currently undergoing domestication in parts of West Africa.

Materials and Methods:

Testicular samples were obtained from ten sexually mature cane rats that were perfused-fixed using Karnovsky's fixative (phosphate buffered 2% paraformaldehyde – 2.5% glutaraldehyde fixative at pH 7.4). The samples were processed for ultrastructural analysis and examined under the transmission electron microscope.

Allogeneic haematopoietic stem cell transplantation currently represents the primary potentially curative treatment for cancers of the blood and bone marrow. While relapse occurs in approximately 30% of patients, few risk-modifying genetic variants have been identified. The present study evaluates the predictive potential of patient genetics on relapse risk in a genome-wide manner. We studied 151 graft recipients with HLA-matched sibling donors by sequencing the whole-exome, active immunoregulatory regions, and the full MHC region. Background/aim: We aimed to investigate the associations between endothelial nitric oxide synthase(eNOS) gene polymorphisms [G894T (rs1799983)], intron 4 (27-bpTR) variable number tandem repeat (VNTR) and T786C (rs2070744), and ischemic stroke in the Anatolian population.Materials and methods: This case-control study included 112 patients with "stroke of undetermined etiology" and 160 controls. Real-time polymerase chain reaction (RT-PCR) analysis was used to analyze these polymorphisms. Between-group frequencies of alleles and genotypes were compared using binary logistic regression analysis.Results: No significant difference was observed between the two groups in terms of the genotype and allele distributions of the eNOS G894T (rs1799983) polymorphism (P > 0.05). …

Background/aim: The aim of this study is to present the results of modified combined decongestive therapy (CDT) in patients with lower extremity lymphedema (LEL). Materials and methods: We retrospectively reviewed 95 patients aged 55.84 ± 15.70 years who had been diagnosed with LEL between May 2015 and May 2017. The patients were treated for 4 weeks with modified CDT, including self-manual lymphatic drainage, self-bandaging, decongestive exercises, and skin care.Results: The mean reduction amounts of edema volume before and after treatment were 296.05, 784.92, and 1038.50 mL for stages 1, 2, and 3 respectively (P = 0.001). Background/aim: The aim of this study was to determine antibiotic resistance profiles and spa types of Staphylococcus aureus strains isolated from food business employees in Çanakkale, Turkey.Materials and methods:S.aureus isolates were collected from hand and nasal swabs of 300 individuals working in 17 food businesses and 9 hospital kitchens.
