An estimated 350 million people are living with chronic Hepatitis B virus (HBV) worldwide. Preventative HBV vaccination in infants has reduced the disease burden; however, insufficient immunization programs and access obstacles leave vulnerable populations at risk for infection in endemic regions. Gene electrotransfer (GET) using a noninvasive multielectrode array (MEA) provides an alternative platform for DNA vaccination in the skin. DNA vaccines are nonlive and nonreplicating and temperature stable unlike their counterparts. In addition, their simple engineering allows them to be manufactured quickly at a low cost. In the current work, we present the combination of GET and moderate heating …

Cold atmospheric plasma, including plasma jet and surface plasma, can promote the apoptosis of cancer cells without causing significant damage to surrounding normal cells, which was hopeful to be applied to the clinical cancer therapy. However, experimental plasma devices used directly to clinical experiments has challenges in technology and methods, especially the difference in killing tumor cells efficiency of these two common plasma sources. Therefore, it is great necessity to explore the differences in treating tumors between different plasma sources. This paper achieved good killing efficiency by using two kinds of cold atmospheric plasma generating devices, namely plasma jet and …

The Role Of Neutrophils Against Pythium Insidiosum, Apichaya Sriwarom
The Role Of Neutrophils Against Pythium Insidiosum, Apichaya Sriwarom
Chulalongkorn University Theses and Dissertations (Chula ETD)
Pythium insidiosum (P. insidiosum) is an oomycetes fungus-like microorganism causing pythiosis infection in humans. The zoospores (infective stage) of this pathogen contaminate aquatic environments such as moist soil and swampy area, which is a natural habitiat. Humans are infected with P. insidiosum via cutaneous route which is the main entry of the zoospores. Recent studies have shown that patients with pythiosis shows high morbidity and mortality rates particularly those with hematologic diseases. As neutrophils are the first line of immune defense during fungal infections, this study therefore investigated the activity of neutrophils in response to P. insidiosum. The neutrophil killing …

Alteration Of Extracellular Matrix Components In Anterior Pituitary Gland Of Neonatal Rats Induced By Maternal Bisphenol A (Bpa) Diet During Pregnancy, Bumpenporn Sanannam
Chulalongkorn University Theses and Dissertations (Chula ETD)
Bisphenol A (BPA) is a well-known endocrine-disrupting chemical that is widely used in the resin and plastic manufacturing sectors. Prenatal exposure to this chemical results in changes of pituitary hormone-producing cells. However, there have been no reports of the alterations of extracellular matrix (ECM) which play a role in controlling cell activities in the pituitary gland. In the present study, a daily oral dose of 5,000 µg/kg BW of BPA was administered to pregnant rats. ECM-producing cells, i.e., pericytes and folliculostelate cells, ECM products, and ECM regulators were investigated in the neonatal anterior pituitary at day 1 at the histology …

A Novel Review Of Heat Shock Protein 110 Kda: A Basis For Research And Continued Experimentation Through Biochemical Analysis, Crist W. Cuffee
Theses and Dissertations
Heat shock protein 110 kDa, Hsp110, is a distinct cellular protector, different in form and function from Hsp70, a close relative of Hsp110. Functioning primarily as a holdase or in tandem with other molecular chaperones, a review of current accomplishments elucidates the uniqueness of this protein and the continued mysteries that surrounds it. Found only in eukaryotes, Hsp110 has been linked to many diseases, ranging from parasitic infection to neurodegenerative disorders. While still lacking, studies of this protein have provided much in the realm of speculation on the mechanisms behind Hsp110s’ contribution to different pathologies. This review will serve as …

Is Sexuality A Choice? An Analysis Of The Facts And Factors That Influence One’S Sexual Orientation, Brandon Scott Hodges
Is Sexuality A Choice? An Analysis Of The Facts And Factors That Influence One’S Sexual Orientation, Brandon Scott Hodges
PCOM Capstone Projects
Sexual orientation is often viewed as the sex or gender to which one is attracted to; however, it is not that simple. Also, though many believe the default is for one to be attracted to the opposite sex and any other claim of sexual attraction is a choice; that is not true. Sexual orientation is the result of a complex combination of one’s genes, environment, and hormones. As a result, not only is sexuality not a choice, but just as there are so many different colors, there are so many different sexual orientations beyond heterosexual, such as homosexual, bisexual, and …

Karst features in the Silurian dolomites of Taylorsville MetroPark (Dayton Metropolitan Area, Ohio, United States) were explored from 2017 to 2018 to identify sites of paleontological interest. Initial landscape surveys recovered 124 skeletal elements (from 12 sites) that were attributed to 17 vertebrate species--including evidence of such extirpated animals as bobcats (Lynx rufus) and rattlesnakes (Crotalus sp.). Of the 12 sites, 9 sites contained remains from the historical era and 3 sites contained much older remains (n = 17) that were radiocarbon dated to approximately 1,400 years before present (YBP). Human remains at one site, butchered bones at another, and …

Because of growth in the craft brewing industry, farmers in the eastern United States are planting winter malting barley (Hordeum vulgare L.) to meet demands for locally sourced grain. However, given that barley is a relatively new crop in this region, basic agronomic information relating to stand assessment is needed. This is particularly relevant in this region, as climatic variability from extreme temperature fluctuations during the winter and spring can reduce a barley stand, creating the need for farmers to estimate grain yield potential. The objective of the research was to evaluate the relationship between spring stem counts, fractional …

Over the past three decades, monoclonal antibodies (mAbs) have revolutionized the landscape of cancer therapy. Still, this benefit remains restricted to a small proportion of patients due to moderate response rates and resistance emergence. The field has started to embrace better mAb-based formats with advancements in molecular and protein engineering technologies. The development of a therapeutic mAb with long-lasting clinical impact demands a prodigious understanding of target antigen, effective mechanism of action, gene engineering technologies, complex interplay between tumor and host immune system, and biomarkers for prediction of clinical response. This review discusses the various approaches used by mAbs for …

Drug resistance, a major challenge in cancer therapy, is typically attributed to mutations and genetic heterogeneity. Emerging evidence suggests that dynamic cellular interactions and group behavior also contribute to drug resistance. However, the underlying mechanisms remain poorly understood. Here, we present a new mathematical approach with game theoretical underpinnings that we developed to model real-time growth data of non-small cell lung cancer (NSCLC) cells and discern patterns in response to treatment with cisplatin. We show that the cisplatin-sensitive and cisplatin-tolerant NSCLC cells, when co-cultured in the absence or presence of the drug, display dynamic group behavior strategies. Tolerant cells exhibit …

With recent advances in sequencing technologies, large scale genome wide sequencing has advanced our understanding of disease risk and etiology. These tests, however, often provide secondary data beyond the scope of the original sequencing objectives, and our understanding of how and if participants want to receive these secondary results is important as clinical and research-based sequencing becomes commonplace. To address this, we completed a systematic review of quantitative research that measures attitudes and preferences on the return of genomic sequencing results. Based on that review, we studied the return of genetic ancestry since these results, popularized in direct-to-consumer sequencing companies …

ATP-binding cassette (ABC) transporters are a large class of integral membrane proteins that contribute to key physiological functions in all organisms by utilizing ATP binding and hydrolysis to transport diverse substrates across membrane barriers. P-glycoprotein (P-gp/ ABCB1) and Multidrug Resistance protein 1 (MRP1/ABCC1) are widely reported ABC transporters associated with multidrug resistance in cancer. Multidrug resistance (MDR) mediated by P-gp and MRP1 is responsible for treatment failures of many metastatic cancers as a result of reduced accumulation, bioavailability and diminished potency of anticancer drugs. Currently, known P-gp and MRP1 inhibitors are limited due to toxicity, lack of selectivity and low …

Background/aim: The aim of this study was to evaluate the efficiency of in vitro embryo splitting (IES) procedures. We also assessed the quality of the blastocysts developed from embryos obtained from different sources. Materials and methods: Good quality embryos at 6-8-cell stages were categorized according to their fertilization sources: 1) frozenwarmed donated embryos, 2) chromosomally abnormal embryos, 3) parthenogenetic embryos, and 4) embryos derived from fertilization of in vitro matured oocytes (rescue IVM). After IES, splitting and developmental efficiency was assessed. Furthermore, the quality of the developed blastocysts was evaluated by Hoechst and propidium iodide (PI) staining. Results: The data …

Background/aim: Currently, there is not any specific treatment for chronic pancreatitis (CP). It was aimed to investigate the effects of melatonin administration on endoplasmic reticulum (ER) stress, oxidative stress, fibrosis, biochemical and histopathological parameters, and Abcc2,Abcc5, and Abcg2 gene levels in an experimental rat CP model. Materials and methods: Forty rats were randomized into five groups: Sham, CP, CP+25 mg/kg melatonin, CP+50 mg/kg melatonin, and CP+placebo. In all rats, except the sham group, a model of chronic pancreatitis was accomplished with intraperitoneal caerulein administration. In treatment groups, melatonin was used as a therapeutic agent. Serum TGF-ß, TNF-α, MDA and GPx …

There has been a recent surge in popularity for cannabidiol (CBD), a major non-psychotropic constituent of cannabis, due to numerous claims of potential therapeutic properties, which include, but are not limited to, anxiolytic, antinociceptive, and anti-inflammatory effects. However, as previous scientific literature on CBD's effectiveness in providing such therapeutic effects is limited, this project was aimed to evaluate the potential beneficial properties of a hemp derived 99% pure CBD compound provided from Ellipse Analytics (Denver, CO) in a rodent model. We analyzed the pharmacokinetics of this CBD product as well as the behavioral outcomes after acute and chronic administration. Pharmacokinetics …

Integrative Genome Assembly Of Staphylococcus Epidermidis And Staphylococcus Hominis Strains, Cana L. Brown
Honors Theses
Due to rapid advances in sequencing technology, it is becoming increasingly easier to assemble unknown genomes from millions of short sequencing reads of nucleotides taken from the full genomic sequence (Hernandez, 2008). In this study, we used various computational programs to align reads from unsequenced strains of the bacteria Staphylococcus epidermidis and Staphylococcus hominis into a hitherto undefined, single contiguous genomic sequence. We used SPAdes to assist with template free assembly (Bankevich, 2012), BLAST to identify a suitable reference genome from closely related species (Madden, 2013), Bowtie2 to align our reads to the reference genome, SAMtools to sort and organize …

Theses and Dissertations--Toxicology and Cancer Biology
Neurotensin (NT) is a nutrient-regulated gut hormone that plays important roles in lipid absorption, obesity, metabolic disorders, and normal and neoplastic growth in the intestine. In this study, we 1) elucidate the mechanisms regulating NT release from endocrine cells, 2) examine the role of NT on proliferation and stem cell function in the small intestine and 3) define the effects of NT on colorectal cancer stem cells. We report that NT release from endocrine cells is enhanced by the MAPK scaffold protein Kinase Suppressor of Ras 1 (KSR1) and the exocyst complex component 70 (Exo70). Moreover, free fatty acid stimulated …
