Cell-specific regulation of endogenous G protein-coupled receptors (GPCRs) is crucial for understanding their roles in physiological processes. We present chemogenetic tools using shield-1-dependent irreversible protein switches to regulate peptide agonist activity. To demonstrate this platform, we engineered chemogenetically regulated pituitary adenylate cyclase activating polypeptide (cPACAP), which exhibited >15-fold chemical-dependent regulation of endogenous receptor activity. In vivo application of cPACAP allowed neuronal activation via the endogenous receptor for PACAP, engaging neural circuits that control respiratory and feeding behaviors. By integrating cPACAP with transgenic mice, we selectively activated endogenous PACAP receptor signaling in hypocretin-expressing neurons of the lateral hypothalamic area (LHA), revealing …

We present a large-scale study of structural variation (SV) in the Qatari population, based on short-read whole-genome sequencing (WGS) of 6,141 individuals, identifying 153,946 variants across 5 classes reflecting the region's diversity and evolutionary history. Leveraging consanguinity and biobank phenotypes, we identify >180 putative gene knockouts, and use proteomics to show functional consequences in homozygotes. Conversely, 52 genes show significant depletion of homozygous deletions, eight of which cause severe pediatric disease or murine embryonic lethality. Examining phenotypic extremes uncovers several non-exonic homozygous deletions with large effect, including in SPIRE2 (creatinine), MAGI2 (leanness) and a chr19 microRNA cluster (extreme obesity). Purpose: Physical Therapy (PT) and Behavioral Health (BH) often overlap in patient care; however, collaborative treatment is always not utilized by providers. This case study details the interdisciplinary treatment of a 69-year-old female with a fear of falling; emphasizing the importance of interprofessional collaborative care in treating Fear of Falling (FOF) in individuals at risk for fall-related injury with Cognitive Behavioral Therapy (CBT) and PT interventions.

Abstract
Purpose: This article proposes an opportunity for interprofessional collaboration between mental health practitioners (MHPs) and occupational therapy practitioners (OTPs) for the treatment of women experiencing mental health disorders (MHDs) associated with reproductive health (RH) changes throughout the lifespan. This collaborative approach have the potential to optimize women’s health care and mitigate the risk of fragmented care, as these conditions are highly complex.

International Journal of Transformative Teaching and Learning in Higher Education
This study presents a content analysis of scholarly works on collaborative online international learning (COIL) published between 2015 and 2023. Conducted by an interdisciplinary faculty group, the research explores how COIL literature frames student learning outcomes within internationalization-at-home efforts. Findings reveal that COIL most frequently supports assignments/projects, as well as intercultural communication and global competence, particularly at the undergraduate level. The analysis highlights COIL’s potential to democratize global learning by addressing access barriers for both students and faculty. The study calls for expanded research on the long-term impacts of COIL, inclusive pedagogies, and cross-institutional collaborations to advance global citizenship in …

The article probes into the interrelationships among cyborg technology, sexual violence, and toxic colonialism represented in the Chinese SF novel Waste Tide by Chen Qiufan. It applies methodologies in posthumanism and ecofeminism and situates the text in the Chinese as well as a global context. It intends to provide a non-Western perspective to understand cyborg feminism by associating the Chinese historical reality with trans-species thinking. Optimisation of machining parameters is crucial to achieve an excellent surface finish, with surface roughness serving as the primary indicator of product quality. This study investigates the application of a one-dimensional Convolutional Neural Network (1D CNN) to predict surface roughness in Computer Numerical Control (CNC) turning processes. The operation of photovoltaic/thermal (PV/T) systems is highly susceptible to the operating temperature of the photovoltaic (PV) panel, as elevated temperatures significantly reduce electrical efficiency and overall system performance. To address this critical issue, a PV/T system incorporating a serpentine channel with fins was carefully designed and experimentally tested using water and Al2O3/water nanofluids at 0.25 and 0.5%. The system’s thermal, electrical, and exergy efficiencies were systematically evaluated under different cooling conditions, along with detailed analyses of entropy generation and exergy losses. Capsaicin, a naturally occurring polyphenol, is known to affect energy expenditure and muscle fatigue and modulate contractions in skeletal muscle. Despite the nexus between cardiovascular health and frailty, the relevance of high-sensitivity cardiac troponin I (hs-cTnI), a biomarker of myocardial injury, to frailty is poorly understood. We examined whether hs-cTnI concentrations were associated with frailty in a well-characterized cohort of older women. A total of 1151 community-dwelling women from the Perth Longitudinal Study of Aging Women (mean age ± SD = 75.2 ± 2.7 years) were included. Frailty was operationalized using a validated frailty index (FI) of cumulative deficits and a modified Fried phenotype. Plasma hs-cTnI were categorized into quartiles. Introduction:
In this study, we introduce the design and implementation of PDBMine, a large-scale, queryable platform for mining sequence-structure statistics from the Protein Data Bank (PDB). PDBMine enables rapid analysis of local conformational trends across proteins by extracting dihedral angles and sequence patterns at scale. In addition to the design and implementation of PDBMine, we also present results validating its ability to return structurally meaningful information.

Traditionally, courts and practitioners assert that the antitrust laws are simply about protecting competition, not competitors. In essence, these parties perceive competition to be inherently desirable, rather than recognizing that it can be beneficial or harmful depending on its intensity and scope. As a result of this mistaken belief, courts hold that certain conduct is legal under the antitrust laws simply because they determine it to be procompetitive, while conduct deemed anticompetitive is condemned. Courts seldom provide definitive boundaries to this pro-competition trope and instead assert it as if it were a well-established truism and universal societal good.

The Caenorhabditis genus produced three independently-evolved hermaphroditic species, C. elegans, C. briggsae, and C. tropicalis. This convergence happened by the independent co-option of male programs for use in a female body. This required two distinct steps: (1) mutations in the sex-determination pathway that activated spermatogenesis in XX animals, and (2) mutations that expressed one of the redundant sperm activation signals in XX animals. My project focused on characterizing how sex-determination genes work in C. briggsae and C. tropicalis. Loss-of-function alleles were used to compare the core sex-determination pathway to that of C. elegans. I found that the somatic sex-determination pathway …

Effective and safe treatments for neglected tropical diseases caused by parasites, such as Chagas disease and sleeping sickness, remain lacking, posing a significant challenge for researchers worldwide. The rational design of dimeric compounds inspired solely by the pharmacophoric core of benznidazole (2-nitroimidazole) has proven to be a promising strategy for antiparasitic development. Thus, in the present work, it was increased the linker between the active units (2-nitroimidazole) to improve the interaction with TcNTR, facilitating the bioactivation of the longest dimers. The Densest Subgraph Discovery (DSD) problem is a prevalent problem in the field of graph mining, aiming to find the cohesive subgraph. Given a graph �� = (��,��) and an integer �� ≥ 2, the goal is to find a vertex sub- set �� ⊆ �� whose induced subgraph �� (��) maximizes the ��-clique density, defined as the number of ��-cliques per vertex. Larger val- ues of �� capture higher-order connectivity patterns beyond edges, enabling the discovery of more cohesive structures. There have been many solutions to this problem. This monograph challenges the Department of War to reframe the conversation about humans’ involvement in lethal autonomous weapons systems by codifying a human-centric framework built on the employment pillars of certification, authority, restriction, and accountability. Although an ample body of literature discusses lethal autonomous weapon systems, this monograph takes a novel approach by proposing a theoretical framework and applying it to historical and hypothetical practical scenarios involving weapons with autonomous characteristics. The endocrine system coordinates and integrates cellular activity throughout the body by regulating cellular and organ function and maintaining homeostasis. Homeostasis, the dynamic maintenance of internal balance despite changing external or internal conditions, is essential for proper cellular function. The endocrine system achieves this through a complex regulatory network of hormone-mediated signaling among multiple endocrine organs, relying on precisely regulated synthesis and release of hormones and specific hormone-receptor interactions. Endocrine-mediated actions coordinate critical physiological processes, including growth, metabolism, reproduction, stress adaptation, and circadian rhythms throughout the lifespan. Introduction: Hepatocellular carcinoma (HCC) can develop in individuals with metabolic dysfunction-associated steatotic liver disease (MASLD) without cirrhosis, especially in those with elevated liver enzymes. However, there is currently no low-cost, scalable screening tool for this population. To address this need, we investigated the association between serial changes in FIB-4 (fibrosis-4) scores over a 3-year period and the risk of developing HCC in a large, diverse cohort of patients with MASLD without cirrhosis.

The role of law enforcement officers is to restore peace and safety when chaos occurs, which can be life-threatening and stressful as officers respond to the critical incidents unfolding. For this study, a qualitative approach and phenomenological framework were utilized to gain insight into the first-hand psychological experiences of officers responding to critical incidents. Police officers work in demanding environments that can lead to mental health conditions such as depression and post-traumatic stress disorder. Peer support and communication among coworkers have emerged as a potentially important factor, as officers would share traumatic events with other officers. Therefore, my research focused …

The state and federally endangered light-footed Ridgway’s rail (LFRR; Rallus obsoletus levipes) is central to regional conservation, yet the drivers of its population fluctuations remain poorly understood. The fragmented nature of coastal wetlands in Southern California is threatening the existence of endemic species like the LFRR. Tidal inundation, severe storms, and overpredation threaten the survival of LFRR. This study addresses population fluctuations by analyzing the spatial and temporal drivers of LFRR populations to inform effective, site-specific management.
To do this, a 45-year (1980–2024) LFRR census data set across 16 wetlands throughout its range in California was synthesized. A multi-faceted …

Phenotypic responses to climate affect individual fitness, but the extent to which this translates into effects on population dynamics remains poorly understood. We assemble 213 time series on phenotypes and population sizes of wild vertebrates globally and match them with local climate data. Our meta-analysis shows that morphological traits are mostly climate insensitive. However, phenology is earlier in warmer-than-average years, which contributes positively to population growth in most species. At lower latitudes, temperature has weaker effects on phenology but stronger direct negative effects on population growth, likely because these populations are less capable of tracking climate via plasticity. Objectives: To determine whether changes in caregiver self-efficacy (beliefs about one's ability to manage dementia-related problems and access help) differed by caregiver race and ethnicity across all participants enrolled in a large pragmatic trial of comprehensive dementia care.
Methods: In the Dementia Care Study (D-CARE), community-dwelling older adults with dementia and their unpaid family caregivers were randomized to receive usual care, community-based, or health system-based comprehensive dementia care. Caregiver self-efficacy was assessed at baseline, 6 months, and 18 months using a 4-item scale (range 4-20, higher scores indicate greater self-efficacy). Biologic products are widely used in the treatment of various disease states, including rheumatoid arthritis, inflammatory bowel disease, cancers, and genetic diseases. These products are derived from proteins and genes produced by living organisms and are subsequently used to treat specific disease states. Due to the complexity of biological products, the costs of research and production are high. Currently, the United States approval pathway requires extensive testing for biologics to be designated as biosimilars, meaning there are no clinically meaningful differences in safety or purity of their active components. Individuals with history of chemo- or radiotherapy frequently exhibit somatic mosaicism in the blood, often involving mutations in genes responsible for DNA damage responses (DDR), such as CHEK2. However, the mechanisms by which CHEK2 mutations promote the expansion of mutant cells following chemo- or radiotherapy remain poorly understood. Here, we demonstrate that loss of CHEK2 confers resistance to chemotherapy in hematopoietic stem and progenitor cells (HSPCs). Through a CRISPR-based screen, we identified CHEK2 as a gene whose loss enhances resistance to cytotoxic chemotherapies. A complementary drug screen revealed that CHEK2-mutant cells are also resistant to DNA hypomethylating agents. Chek2-deficient HSPCs …
